Bacteriophage Cocktail Comprising Fifi044 and Fifi318 for Biocontrol of Erwinia amylovora

  • Erwinia amylovora is a plant pathogen that causes fire blight on apples and pears. Bacteriophages, which are viruses that selectively infect specific species of bacteria and are harmless to animal cells, have been considered as biological control agents for the prevention of bacterial pathogens. In this study, we aimed to use bacteriophages that infect E. amylovora as biocontrol agents against fire blight. We isolated bacteriophages Fifi044 and Fifi318 infecting E. amylovora, and characterized their morphology, plaque form, and genetic diversity to use as cocktails for disease control. The stabilities of the two phages were investigated at various temperatures and pH values and under sunlight, and long-term storage experiment was conducted for a year. To evaluate whether the two phages were suitable for use in cocktail form, growth curves of E. amylovora were prepared after treating the bacterial cells with single phages and a phage cocktail. In addition, a disease control test was conducted using immature apples and in vitro cultured apple plantlets to determine the biocontrol effects of the phage cocktail. The two phages were morphologically and genetically different, and highly stable up to 50℃ and pH value from 4 to 10. The phages showed synergistic effect when used as a cocktail in the inhibition of host bacterial growth and the disease control. This study demonstrated that the potential of the phage cocktail as a biocontrol agent for commercial use.

A Study on Improving the Efficiency of the Survival Rate for the Offshore Accommodation Barge Resident Using Fire Dynamic Simulation (화재시뮬레이션을 이용한 해양플랜트 전용생활부선 거주자의 생존율 향상에 대한 연구)

  • The offshore plant crews that were commissioned in the commercial startup phase boarded the offshore plant in two shifts until the end of the project. The crews who were hired by the owner side stayed in the original offshore plant during the project. However, most of the offshore plant commissioned members who were dispatched from the shipyard were accommodated in the offshore accommodation barge. For this reason, they were exposed to many accidents since there are a lot of people staying in a small space. This study suggested a method for improving survival rate at offshore accommodation barge in terms of life safety. It is assumed that the fire accident among unfortunate events which take place in the offshore accommodation barge mainly occurred. So, this study analyzed the safety evacuation for offshore plant employees using fire simulation model based on both domestic and international law criteria. In particular, When fire occurs in the offshore accommodation barge, the periodically well trained crews are followed safety evacuation procedure. whereas many employees who have different background such as various occupations, cultural differences, races and nationality can be commissioned with improper evacuation behaviors. As a result, the risk will be greater than normal situation due to these inappropriate behaviors. Therefore, This study analyzed the Required Safe Escape Time (RSET) and Available Safe Escape Time (ASET). Also it was suggested the improvement of structure design and additional arrangement of safety equipment to improve the survival rate of the residents in offshore accommodation barge.

  • The development of high-rise firefighting vehicles warrants thorough structural analysis for ensuring vehicle stability. A few structural analyses were carried out using CAD data, material properties, load conditions, and boundary conditions for evaluating the structural stability of an overloaded multi-aerial platform for firefighting and rescue. Structural analysis was performed with an analytical model consisting of a turntable, six booms, two jib booms, and a basket structure. This model was operated in eight modes. All simulation was performed using NASTRAN, a commercial code. As a result, we confirm that the position of local stress exceeds that of the yield strength. Therefore, stress concentration relaxation is possible by introducing reinforcing boom structures, changing the shape, or imparting a larger moment of inertia to the booms' cross sections.

  • A direct-injection stratified-charge(DISC) engine has been considered as a promising alternative in spite of high unburned hydrocarbon emission levels during light load operation. In this paper investigation is made to characterize formation and combustion processes of stratified mixture charge in a simple constant volume combustion chamber. Both experimental and numerical analyses are performed for fluid and combustion characteristics with 3 different induction types for rich, homogeneous and lean mixture conditions. The commercial code FIRE is applied to the turbulent combustion process in terms of measured and calculated pressure traces and calculated distributions of mean temperature, OH radical and reaction rate. It turns out that the highest combustion rate occurs for the rich state condition at the spark ignition location due to existence of stoichiometric mixture and timing.

  • To co-fire with commercial LTCC (Low Temperature Co-fired Ceramic) materials at $850^{\circ}C{\sim}880^{\circ}C$, different contents of $B_2O_3$ were added to the $Bi_2O_3-ZnO-Nb_2O_5$ (BZN) ceramics. According to the test results, the cubic phase of BZN was transformed into orthorhombic in all the test materials. $BiNbO_4$ phase was formed in test materials with $2{\sim}5wt%$ of $B_2O_3$ addition. The phase transformation of cubic BZN was controlled during the synthesis process with excess ZnO content. The Cubic and orthorhombic phases of BZN could coexist and be sintered densely at $850^{\circ}C/2hr$.

  • For the design of Aluminium pool furnace, position of burner and pool depth effects on flow and temperature field in Aluminium pool furnace are examined by the commercial computational code, CFD-ACE+. From the results, position of burner which is on the same face in side wall is better to distribute the flow field in Al furnace. That yields temperature to distribute more uniformly. And the burner position is on upper wall, fire frame reach pool surface. Customer must consider that, because it make Aluminium to oxidize.

  • This study is aimed to develop fundamental technology on the smoke control method by simulation model and scale model simulation technique in underground space. Thereby, this research aimed to establish design elements and technologies required for smoke control system that is suitable to underground spaces of the high-rise residential-commercial and office buildings in order to minimize the loss of lives and property damage in case of fire.

Optimization of Heat Exchanger Network in the Steam Assisted Gravity Drainage Process Integration

  • The Steam Assisted Gravity Drainage (SAGD) process is an enhanced method to extract oil from bitumen which involves surface and central process facilities. This paper describes the Central Process Facilities (CPF) of SAGD and proposes several retrofit plans to the Heat Exchanger Network (HEN). In this approach, the process integration scheme is applied to estimate the energy saving in HENs, and various cases are modeled in favor of a commercial simulator. Throughout this work, a minimum approach temperature of 10℃ is assumed. The results reveal that, due to the HEN optimization using process integration, the heating and cooling duties can be reduced to 29.68MW and 1.886MW, respectively. Compared with the Husky case, all cases considered in this study indicate a potential reduction of at least 6% in total cost, including investment and operation costs.

  • This study deals with output characteristics of a hybrid power generation system for a vehicle such as a crane, a fire engine, and a wingbody. The proposed method obtains the commercial AC voltage of single phase 220V/60Hz by connecting a variable speed three-phase PMSG(Permanent magnetic synchronous generator) and an AC/DC/AC power converter to PTO (Power take off) or hydraulic motor. The proposed system is fabricated and tested to demonstrate the usefulness of the proposed system.

  • This paper present design and implementation of U-city infrared image surveillance system based on the digital media processor. The hardware is designed and implemented by using commercial chips such as DM642 processor and video encoder, video decoder and the functions of software are to analyze temperature distribution of a monitoring image and to monitor disaster situation such as fire. The required functions and performances are confirmed by testing of the prototype and we verified practicality of the system.
