The Clinical Effectiveness of Atipamezole as a Medetomidine-Tiletamine/Zolazepam Antagonist in Dogs (개에서 Medetomidine-Tiletamine/Zolazepam 마취에 대한 Atipamezole의 길항 효과)

  • The cardiopulmonary and antagonistic effects of atipamezole, to medetomidine (30 ug/kg, IM)-tiletamine/zolazepam (10 mg/kg, IV) were determined. Twelve healthy mongrel dogs ,(4.00$\pm$0.53 kg, mean$\pm$SD) were randomly assigned to the four experimental groups (control, A30; atipamezole 30 ug/kg, A60; atipamezole 60 ug/kg, A150; atipamezole 150 ug/kg) with 3 dogs in each group. Atropine (0.03 mg/kg, IM), medetomidine, and tiletamine/zolazepam (TZ) were injected 10 minute intervals. Atipamezole was injected intravenously 15 minutes after TZ injection. Mean arousal time (MAT) was 52.50$\pm$4.98, 43.06$\pm$2.60, 32.83$\pm$8.13, and 14.36$\pm$1.60 minutes in control, A30, A60, and Al50 groups respectively. In Al50 group, MAT was significantly reduced (P < 0.05). but mean walking time (MWT) was similar to that in control group. In recovery period, the higher doses of atimapezole, the rougher recovery including head rocking, hypersalivation, and muscle twitching. Five of twelve dogs vomited within 5 minutes after medetomidine injection. In Control group, heart rate significantly decreased in all recording stages except 15 minutes after TZ injection, 10 minutes after medetomidine injection in all groups, and 40 minutes after atipamezole injection in A30 group (P < 0.05). In Al50 group, atipamezole reversed the respiratory depression induced by medetomidine. Arterial blood pressure was significantly decreased 10minutes after medetomidine injection and 15 minutes after TZ injection in almost dogs in this study (P < 0.05). From 10 minutes after atipamezole injection to arousal time, arterial blood pressure was progressively increased in A60 and A150 group. Any value of blood gas analysis and CBC, and serum chemistry values were not significantly changed except pH of Al50 at 10 minutes after medetomidine injection. As shown in present study, atipamezole(150 ug/kg) is considered to exert a useful reversal effect in dogs anesthetized with medetomidine-tiletamine/zolazepam combination.

Development of replacement diets for improved growth and survival rate of scallop juvenile Patinopecten yessoensis (큰가리비 Patinopecten yessoensis 치패의 성장 및 생존율 향상을 위한 대체 먹이원 개발)

  • This study was done to examine the effect of several diets (Phytoplankton = PHY, Shellfish Diet 1800 = INS, Oil type = OTE, Powder type = PTE) on growth, survival rate and biochemical composition of scallop juvenile Patinopecten yessoensis. The highest survival rate were observed in PTE + PHY (90%). The highest shell length and shell height was observed in PHY and PTE + PHY diet (P > 0.05). The growth with PTE and OTE diet was the lowest in shell length and shell height (P < 0.05). On the other hand, the shell width and meat weight were highest in PHY and PTE + PHY, while the lowest in PTE and OTE (P < 0.05). The content of fatty acids such as DHA and n-3 PUFA levels was significantly higher in the juvenile fed on PTE + PHY than in those fed on PHY and INS alone. Also, the total protein ranged 55.5 to 65.2% in PHY + INS, while 44.8%, 47.9% in PTE and OTE respectively. The RNA and DNA contents were the highest in PHY and PTE + PHY, while the lowest in PTE and OTE (P < 0.05). RNA/DNA ratio significantly higher in juvenile with PHY + INS than those with PTE and OTE alone (P < 0.05). The combination of PTE + PHY could improve the growth and survival of scallop juvenile. Our results suggested that PTE could partially replace live algae in bivalve laval rearing.

Synergistic Effect of Combined Treatment of Bacteriocin Produced by Enterococcus faecalis MJ-231 and Potassium Sorbate on Growth of Food-Borne Pathogenic Bacteria (식중독균 생육에 대한 Enterococcus faecalis MJ-231의 박테리오신과 소르빈산칼륨의 혼합처리 효과)

  • The alone and combined effects of bacteriocin produced from Enterococcus faecalis MJ-213 and potassium sorbate against the food-borne pathogenic bacteria were studied. Bacteriocin minimal inhibitory concentration (MIC) values for Staphylococcus aureus ATCC 6538 and Salmonella enteritidis ATCC 13076 were 50 and 100 ${\mu}g$/ml, respectively. Bacteriocin (100 ${\mu}g$/ml) alone was active against S. aureus and S. enteritidis, but it was lower in antimicrobial effectiveness than the combination of bacteriocin (100 ${\mu}g$/ml) with potassium sorbate (100 ${\mu}g$/ml), which reduced initial counts (6 log cycle) of S. aureus and S. enteritidis by 1 and 3 log cycle, respectively. The bactericidal activity of bacteriocin of E. faecalis MJ-213 heated at $100^{\circ}C$ for 30 min or $121^{\circ}C$ for 15 min was markedly decreased as compared with the control. Moreover, the activity of bacteriocin was completely abolished by pepsin or protease II, but not affected by ${\alpha}$-amylase or lipase. The activity of bacteriocin adjusted to pH 6.0-8.0 showed almost the same inhibition ratio compared with the bacteriocin unadjusted pH, and though the inhibition ratio against pathogenic bacteria was reduced than the control, the bacteriocin was stable at pH 4.0 or 10.0, relatively. Furthermore, the combined treatment of bacteriocin and potassium sorbate than the alone treatment of bacteriocin significantly decreased (p<0.05) the viable cell counts of S. aureus or S. enteritidis inoculated on grind beef during storage at $4^{\circ}C$.

Study on Analysis of the Proper Ratio and the Effects of Low Impact Development Application to Sewage Treatment District (하수처리구역 내 LID 적용에 대한 적정비율 및 효과분석 연구)

  • Increase of impervious area caused by overdevelopment has led to increase of runoff and then the problem of flooding and NPS were brought up. In addition, as decrease of base flow made groundwater level to decline, a stream that dries up is issued. low impact development (LID) method which is possible to mimic hydrological water cycle, minimize the effect of development, and improve water cycle structure is proposed as an alternative. As introduction of LID in domestic increases, the study on small watershed is in process mainly. Also, analysis of property of hydrological runoff and load on midsize watershed, like sewage treatment district, is required, the study on it is still insufficient. So, area applying LID practices from watershed of Dongrae stream is pinpointed and made the ratio and then expand it to watershed of Oncheon stream. Among low impact development practices, Green Roof, Porous Pavement, and Bio- retention are selected for the application considering domestic situations and simulated with SWMM-LID model of each watershed and improvement of water cycle and reduction of non-point pollution loads was analysed. Improvement of water cycle and reduction of non-point pollution loads were analyzed including the property of rainfall and soil over long term simulation. The model was executed according to scenario based on combination of LID as changing conductivity in accordance with soil type of the watershed. Also, this study evaluated area of LID application that meets the efficiency of conventional management as a criteria for area of LID practices applying to sewer treatment district by comparing the efficiency of LID application with that of conventional method.

Comparative Studies of the Combined Dietary Animal and Vegetable Proteins on Growing and Blood Compositions the Weanling Rats and Mice (식이중의 동물성 및 식물성단백질의 종류와 배합비율에 따른 흰쥐 (Rat)와 흰생쥐 (Mouse)의 성장과 혈액성분에 관한 비교연구)

  • In order to investigate the effect of the combined dietary animal and vegetable proteins on growing female and male rats and mice. 25 female and 25 male rats of 4 weeks old weighing approximately 43-65g and 47-60g, respectively, were subjected to feeding trials for 4 weeks and then subsequently to metabolic trials for 10 days. In case of mice, 25 female and 25 male mice of 4 weeks old weighing approximately 12-17g and 12-16g, respectively, were equally treated. The dietary energy level was set as 3600 kcal ME/kg, and protein and fat provided respectively 12 and 5% of the dietary energy. The rest 83% of the energy level was supplied with appropriate amounts of starch and glucose. The following results were obtained. The body weight gain of female and male rats were increased as the combined dietary animal protein level increased. Whereas that of male mice was the highest for the diet E(Ap 10+Vp 90). Food efficiencies both of female and male rats were improved as combined dietary of animal protein increased. Male rats were superior to that of female. In the mice, the food efficiency value of male mice was superior to that of female. And the values of the male showed the same fashion as that in the growth rate, although it was not for the female. Protein efficiencies, both of female and male rats, were improved as combined dietary of animal protein level increased. That of male mice was in accord with the body weight gain and food efficiencies. But that of female mice was not accordant. Nitrogen retention of the experimental diets in the same protein level was in proportion to nitrogen intake. That of male mice was improved as animal protein level increased, but, that of female mice was not. Body protein utilizability was found to be superior in mice to rats. Blood serum protein level was not found to be affected by the experimental diets in the same protein level. The best combination ratio of animal and vegetable protein seems to be variable depending on the sex and species of animals, and the best diets for female and male rats and female and male mice were found to be experimental diet A(Ap 10+Vp 90) or diet B(Ap 75+Vp 25), diet C(Ap 50+Vp 50), and diet E(Ap 10+Vp 90), respectively. From the above-mentioned results that there was no difference in growth rate of body weight gain from the weanling period to puberty, and that the protein requirements of rats and mice were different from each other.

Toxicity Evaluation of 'Bt-Plus' on Parasitoid and Predatory Natural Enemies (기생성 및 포식성 천적에 대한 작물보호제 '비티플러스'의 독성 평가)

  • Effect of a new crop protectant 'Bt-Plus' on natural enemies was analyzed in this study. Tested natural enemies included two parasitic species of $Aphidius$ $colemani$ and $Eretmocerus$ $eremicus$, and four predatory species of $Harmonia$ $axyridis$, $Orius$ $laevigatus$, $Amblyseius$ $swirskii$, and $Phytoseiulus$ $persimilis$. 'Bt-Plus' was formulated by combination of three entomopathogenic bacteria ($Xenorhabdus$ $nematophila$ (Xn), $Photorhabdus$ $temperata$ subsp. $temperata$ (Ptt), $Bacillus$ $thuringiensis$ (Bt)) and bacterial metabolite (BM). All three types of 'Bt-Plus' showed significantly higher toxicities against fourth instar $Plutella$ $xylostella$ larvae than Bt single treatment. Two types of bacterial mixtures ('Xn+Bt' and 'Ptt+Bt') showed little toxicity to all natural enemies in both contact and oral feeding assays. However, 'BM+Bt' showed significant toxicities especially to two predatory mites of $A.$ $swirskii$ and $P.$ $persimilis$. The acaricidal effects of different bacterial metabolites were evaluated against two spotted spider mite, $Tetranychus$ $urticae$. All six BM chemicals showed significant acaricidal effects. The BM mixture used to prepare 'Bt-Plus' showed a high acaricidal activity with a median lethal concentration at 218.7 ppm (95% confidence interval: 163.2 - 262.3). These toxic effects of bacterial metabolites were also proved by cytotoxicity test against Sf9 cells. Especially, benzylideneacetone, which was used as a main ingredient of 'BM+Bt', showed high cytotoxicity at its low micromolar concentration.

Establishment of Optimal Rearing Conditions for the Production of Tenebrio molitor Larvae (갈색거저리 유충 생산을 위한 효율적인 사육조건 확립)

  • Tenebrio molitor larvae contain large amounts of proteins, lipids and other functional materials, enabling this insect to be used as an edible food source in animal feeds and for industrialization. Although many efforts have been made to set up mass rearing systems, few studies have been conducted to establish optimal rearing conditions for the production of high quality T. molitor larvae. Herein we investigated 1) the effects of additional diets on the survival and fecundity of the insect, 2) the relationship between oviposition period and the uniformity of larval size, 3) the effects of rearing density and temperature on insect development, and 4) the storage stability of eggs and pupae at low temperatures given possible temporary production discontinuation. The addition of carrot and zucchini to the traditional wheat bran diet significantly increased the survival and fecundity rate of adult T. molitor. Of the three different oviposition sampling periods (3, 7, and 14 days) used to investigate the uniformity of the hatched larvae in each treatment, the period of 3 and 7 days provided higher uniformity than the 14 days oviposition period. Larval development was faster at $30^{\circ}C$ than at 20, 25, and $35^{\circ}C$. Interestingly, oviposition rates were highest at $20^{\circ}C$ but showed much slower larval development and lower uniformity at $30^{\circ}C$. Regarding the effect of larval rearing densities (1, 10, 20, 30, 40, and 50 larvae per 90 mm diam. dish), larval weight was significantly reduced at higher rearing densities, but larval longevity and length were not influenced by rearing density. The 30 larvae/dish is suggested to be a reasonable density to be applied to mass production systems. When kept at $4^{\circ}C$, T. molitor eggs showed a significant reduction in hatching rate; however, when stored under the same conditions, pupae emergence rates remained high until 10 weeks, suggesting that storage at low temperatures is more suitable for the pupal stage than the egg stage. Our findings suggest that an increase in T. molitor adult survival and fecundity rates and a uniformity of hatched larval development can be achieved with the following recommendations: a combination diet (including wheat bran), a 7-day oviposition period; a larvae-rearing temperature of $30^{\circ}C$, a rearing density of 30 larvae/dish, and the storage of pupal stages at low temperatures in the case of rearing discontinuation. This study serves as a strong foundation for the successful mass production of high quality T. molitor larvae.

A Comparative Study of Effect of Secondary Anti-tuberculosis Drugs in the Retreatment of Pulmonary Tuberculosis (폐결핵 재치료에서 이차항결핵제 복합처방의 효과에 관한 비교 연구)

  • Background : In the management of patients whose primary chemotherapy has failed, careful assessment is essential. It is important to find out as accurate a chemotherapy history as possible. Preferably it should contain the drugs which has never used before. The purpose of present study is establishment of retreatment regimen for pulmonary tuberculosis. The present report concerns the results of retreatment of pulmonary tuberculosis patients treated at National Masan Tuberculosis Hospital. Methods : Retrospective cohort study was made of 104 drug-resistant pulmonary tuberculosis patients who were treated by five regimens between Jan. 1994 and Nov. 1996. All the patients taken medicine for second anti-tuberculosis regimens for the first time. We separated the patients by three groups(Group I ; OFX+PTA+CS+PAS+Aminoglycoside, Group II : PZA+PTA+CS+PAS+Aminoglycoside, Group III : PZA+OFX+PTA+PAS+Aminoglycoside). Results : The age distribution was most frequent in fourth decade(36patients, 34.6%) and the mean age was 42.6 year. The sex distribution was more frequent in the males(81 patients, 85.7%). There was 31 patients(29.8%) with combined diseaes, 18 patients with complication and 24 patients(27.9%) with family history. Primary chemotherapy regimens were HERZ(S or K) in 48 patients (46.2%), HER(S or K) in 41 patients(39.4%) and others in 15 patients(14.4%). Result of drug sensitivity test showed that the resistance to INH and RFP is in 68 patients(65.4%), RFP is 12 patients(11.5%), INH is in 3 patients(2.9%) and all sensitive to INH and RFP is 3 patients(2.9%). The clinical symptoms on admission were coughing(89.4%), sputum(69.2%), dyspnea on exertion(37.5%), weight loss(33.7%) blood tinged sputum(15.4%) and others. The extent of disease on the radiograph was far-advanced in 73 patients(70.2%), moderate in 28 patients(26.9%) and minimal in 3 patients(2.9%). The side effects for drugs were gastrointestinal troubles in 31 patients(29.8%), arthralgia in 22 patients(21.2%), skin rash in 12 patients(11.5%) and others. The negative conversion rate on sputum AFB smear was 85.6%(87.5% in Group I, 80.0% in Group II and 90.5% in Group III). The average negative conversion time on sputum was 4 month(4.0 month in Group I, 4.6 month in Group II and 3.0 month in Group III). Conclusion : In the retreatment of pulmonary tuberculosis, ofloxacin is useful drug for the patients who are not available to use PZA and combination of PZA and OFX can be use effectively substituting for CS.

Effect of a Combined Extract of Orostachys japonicus with Medicinal Plants on the Lipid Composition of the Liver and Kidney from Streptozotocin-induced Diabetic Rats (와송과 생약재 복합물이 Streptozotocin 유발 당뇨쥐의 간장 및 신장 조직의 지질성분에 미치는 영향)

  • This study examined the biological activity and synergistic effects of an extract of Wa-song ($Orostachys$ $japonicus$, OE), a medicinal plant mixture (MPE) and a combination of both at different ratios (1:1, OMPE-1 and 3:1, OMPE-3). Extracts of the medicinal plants mixture were comprised of Baekbokyung, Changchul and Sa-in at the same ratio. The antioxidant activity of the extracts and their complex were tested $in$ $vitro$. The $in$ $vivo$ antioxidant activity was also analyzed by examining the lipid composition in the liver and kidney of streptozotocin (STZ)-induced diabetes rats. The nitric oxide radical scavenging activity was more than 50% in OMPE-3 at a 1,000 ${\mu}g$/mL concentration. Regarding metal ions, such as $Fe^{2+}$ and $Cu^{2+}$, the antioxidant activity of OMPE-1 and OMPE-3 was higher than that of OE and MPE. OMPE-1 and 3 had higher activity on $Cu^{2+}$ ions than $Fe^{2+}$ ions. The ${\alpha}$-glucosidase inhibition activity of the OE extract was higher than that of MPE and OMPE-1 but the relative activity of OMPE-3 was significantly higher than the others. Freeze-dried MPE, OMPE-1 and OMPE-3 were added to the diet at a level of 1% given to STZ induced diabetes rats for 4 weeks. The OMPE-1 and OMPE-3 administered groups showed significant decreases in the total lipid, total cholesterol and triglyceride levels in the liver and kidney. In these groups, the glycogen accumulation level of the liver was increased significantly. The content of thiobarbituric acid reactive substances (TBARS) in the liver and kidney was decreased but the DPPH radical scavenging activity was increased. These results suggest that Wa-song extract exhibits antioxidant and antidiabetic activity, which are enhanced by a complex with a medical plants extract.

Mechanical and Rheological Properties of Rice Plant (수도(水稻)의 역학적(力學的) 및 리올러지 특성(特性)에 관(關)한 연구(硏究))

  • The mechanical and rheological properties of agricultural materials are important for engineering design and analysis of their mechanical harvesting, handling, transporting and processing systems. Agricultural materials, which composed of structural members and fluids do not react in a purely elastic manner, and their response when subjected to stress and strain is a combination of elastic and viscous behavior so called viscoelastic behavior. Many researchers have conducted studies on the mechanical and rheological properties of the various agricultural products, but a few researcher has studied those properties of rice plant, and also those data are available only for foreign varieties of rice plant. This study are conducted to experimentally determine the mechanical and the rheological properties such as axial compressive strength, tensile strength, bending and shear strength, stress relaxation and creep behavior of rice stems, and grain detachment strength. The rheological models for the rice stem were developed from the test data. The shearing characteristics were examined at some different levels of portion, cross-sectional area, moisture content of rice stem and shearing angle. The results obtained from this study were summarized as follows 1. The mechanical properties of the stems of the J aponica types were greater than those of the Indica ${\times}$ Japonica hybrid in compression, tension, bendingand shearing. 2. The mean value of the compressive force was 80.5 N in the Japonica types and 55.5 N in the Indica ${\times}$ Japonica hybrid which was about 70 percent to that of the Japonica types, and then the value increased progressively at the lower portion of the stems generally. 3. The average tensile force was about 226.6 N in the Japonica types and 123.6 N in the Indica ${\times}$ Japonica hybrid which was about 55 percent to that of the Japonica types. 4. The bending moment was $0.19N{\cdot}m$ in the Japonica types and $0.13N{\cdot}m$ in the Indica ${\times}$ Japonica hybrid which was 68 percent to that of the Japonica types and the bending strength was 7.7 MPa in the Japonica types and 6.5 MPa in the Indica ${\times}$ Japonica hybrid respectively. 5. The shearing force was 141.1 N in Jinju, the Japonica type and 101.4 N in Taebaeg, the Indica ${\times}$ Japonica hybrid which was 72 percent to that of Jinju, and the shearing strength of Taebaeg was 63 percent to that of Jinju. 6. The shearing force and the shearing energy along the stem portion in Jinju increased progressively together at the lower portions, meanwhile in Taebaeg the shearing force showed the maximum value at the intermediate portion and the shearing energy was the greatest at the portion of 21 cm from the ground level, and also the shearing strength and the shearing energy per unit cross-sectional area of the stem were the greater values at the intermediate portion than at any other portions. 7. The shearing force and the shearing energy increased with increase of the cross-sectional area of the rice stem and with decrease of the shearing angie from $90^{\circ}$ to $50^{\circ}$. 8. The shearing forces showed the minimum values of 110 N at Jinju and of 60 N at Taebaeg, the shearing energy at the moisture content decreased about 15 percent point from initial moisture content showed value of 50 mJ in Jinju and of 30 mJ in Taebaeg, respectively. 9. The stress relaxation behavior could be described by the generalized Maxwell model and also the compression creep behavior by Burger's model, respectively in the rice stem. 10. With increase of loading rate, the stress relaxation intensity increased, meanwhile the relaxation time and residual stress decreased. 11. In the compression creep test, the logarithmic creep occured at the stress less than 2.0 MPa and the steady-state creep at the stress larger than 2.0 MPa. 12. The stress level had not a significant effect on the relaxation time, while the relaxation intensity and residual stress increased with increase of the stress level. 13. In the compression creep test of the rice stem, the instantaneous elastic modulus of Burger's model showed the range of 60 to 80 MPa and the viscosities of the free dashpot were very large numerical value which was well explained that the rice stem was viscoelastic material. 14. The tensile detachment forces were about 1.7 to 2.3 N in the Japonica types while about 1.0 to 1.3 N in Indica ${\times}$ Japonica hybrid corresponding to 58 percent of Japonica types, and the bending detachment forces were about 0.6 to 1.1 N corresponding to 30 to 50 percent of the tensile detachment forces, and the bending detachment of the Indica ${\times}$ Japonica hybrid was 0.1 to 0.3 N which was 7 to 21 percent of Japonica types. 15. The detachment force of the lower portion was little bigger than that of the upper portion in a penicle and was not significantly affected by the harvesting period from September 28 to October 20. 16. The tensile and bending detachment forces decreased with decrease of the moisture content from 23 to 13 percent (w.b.) by the natural drying, and the decreasing rate of detachment forces along the moisture content was the greater in the bending detachment force than the tensile detachment force.
