Contamination by pesticides is an everincreasing problem associated with fields of environmental management and healthcare. Accordingly, appropriate treatments are in demand. Pesticide detection methods have been researched extensively, aimed at making the detection convenient, fast, cost-effective, and easy to use. Among the various detecting strategies, paper-based assay is potent for real-time pesticide sensing due to its unique advantages including disposability, light weight, and low cost. In this study, a paper-based sensor for chlorpyrifos, an organophosphate pesticide, has been developed by layering three sheets of patterned plates. In colorimetric quantification of pesticides, the blue color produced by the interaction between acetylcholinesterase and indoxyl acetate is inhibited by the pesticide molecules present in the sample solutions. With the optimized paper-based sensor, the pesticide is sensitively detected (limit of detection =8.60 ppm) within 5min. Furthermore, the shelf life of the device is enhanced to 14 days after from the fabrication, by treating trehalose solution onto the deposited reagents. We expect the paper-based device to be utilized as a first-screening analytic device for water quality monitoring and food analysis.
A non-isotopic neutravidin-based reverse transcriptase (RT) assay adapted for high throughput screening of HIV activity is described. Using a 96-well microtitre plate, HIV particles are lysed and the RT enzyme released into a reaction mixture containing poly(A) RNA, biotinylated oligo d(T) and fluorescein-labelled dUTP (FI-dUTP). With poly(A) as a template and oligo d(T) as primer, the viron RT incorporates FI-dUTP into an elongating DNA strand. The resulting product is captured on a neutravidin-coated 96-well plate and the unincorporated nucleotides removed by a series of washing steps. A simple ELISA is subsequently performed using a monoclonal antifluorescein antibody conjugated to alkaline phosphatase. Quantification of RT activity is facilitated by a colorimetric readout. The assay was validated in the context of a diagnostic HIV-1 phenotyping assay. Using supernatants from HIV-1 infected lymphocyte cultures the assay was shown to be as sensitive as a radioactive assay and the RT activity correlated well with levels of cell-asociated HIV-p24. Importantly, even minor reductions of RT activity by virus variants with reduced fitness could be distinguished.

The NR90, NR50, MTT90 and MTT50 values of cadmium were 0.2mM, 19.5mM 1.0mM and 60.0mM, respectively.
The present study was carried out to evaluate the cytotoxicity of cadmium on cultured rat fibroblasts. The colorimetric assays of neutral red and tetrazolium MTT, the lactatedehydrogenase activity, the amounts of total protein, the rate of DNA synthesis, the amounts of unscheduled DNA synthesis, the frequency of sister chromatid exchange, the releasing rate of intracellular calcium, and light and electron microscopic studies were performed on cultured rat fibroblasts maintained in the media containing various concentrations of cadmium. The results were as follows: The neutral red(NR) and MTT values were decreased dose-dependently by cadmium, and the NR90, NR50, MTT90 and MTT50 values of cadmium were 0.2mM, 21.5mM, 1.0Mm and 60.0Mm, respectively.
In the present study, we have evaluated cytotoxic effects of the chloroform soluble fraction of the methanolic extract of Perilla frutescens in human oral epitheloid carcinoma cells. The light microscopic study showed morphological changes of the treated cells. Cell membrane damaging activity was measured by the lactate dehydrogenase (LDH) assay and disruptions in cell organelles were determined by 3-(4,5-dime-thylthiazol-2-yl)-2,5-diphenyl-2H-tetrazolium bromide (MTT), neutral red (NR) and sulforhodamine protein B (SRB) of colorimetric assay. These results suggest that Perilla frutescens retains a potential antitumor activity.
A colorimetric assay for $\gamma$-glutamyl transpeptidase ($\gamma$-CTP; E.C 2.3.2.2) employing 2, 4, 6-trinitrobenzene sulfonate (TNBS) to detect the amount of disappeared acceptor via transpeptidation, has been developed. Under the experimental conditions using L-$\gamma$-glutamyl ethyl ester and L-phenylalanine as $\gamma$-glutamyl donor and acceptor, respectively, it was found that the decreased absorbance of yellow color at 420 nm was strictly related to the amount of L-$\gamma$-glutamyl-L-phenylalanine (L-$\gamma$-Glu-L-Phe) formed, which was determined by DEAE-cellu-lose column chromatography. Concentrations of the enzyme and $\gamma$-glutamyl products were able to be determinedin the nanogram and nanomoles per milliliter range, respectively, with high precision and reliability. This novel assay system may therefore be a useful means for understanding of catalytic function of the $\gamma$-CTP spectrophotometrically without any usage of sophisticated instruments.

A method for the determination of nitrogen in ruminant feedstuffs, products and excreta (e.g. milk and urine) using a spectrophotometer is developed, where samples processed for P determination are also used to determine N. Samples are digested with sulphuric acid and subsequently with hydrogen peroxide in Kjeldahl tubes. Digested solutions along with phenol and buffered alkaline hypochlorite reagents are incubated in a water bath at $37^{\circ}C$ for 30 min and presented in the spectrophotometer. The spectrophotometer set at 625 nm measures the concentration of N of each sample. Nitrogen in 261 of the samples was also determined by the classical Kjeldahl method in order to develop a relationship between N determined by the Kjeldahl method (Y) and the colorimetric method (X). The mean value of Y was as high as that of X (0.92 vs. 0.96; p>0.05). The colorimetric method predicted Kjeldahl N highly significantly (Y=0.985X-0.024, $R^2=0.993$, p<0.001; or more simply Y=0.974X, $R^2=0.993$, p<0.001). An analysis of regression found no difference (p>0.05; both t-test and F-test) between colorimetric (0.96% N) and adjusted (0.96% N) N. In comparison with the Kjeldahl method, the analytical capacity of N by colorimetric method increases greatly, where 200-300 determinations of N are possible in a working day. In addition, the system provides an opportunity to use not only the same digested solution for both N and P determination of a particular sample, but also uses the same spectrophotometer to assay both N and P. Therefore, the system may be attractive in situations where both elements of a sample are to be determined. In conclusion, the speed of N determination, low cost, efficient use of labour, time and reagents, fewer items of equipment, and the reduction of environmental pollution by reducing effluent and toxic elements are the advantages of this method of N determination.
This study was designed to investigate the effect of ferulic acid on cell viability and cell adhesion activity in normal human gingival fibroblasts. The cell viability and cell adhesion activity of ferulic acid was measured by MTT assay or XTT assay, respectively, after normal human gingival fibroblasts were treated with or without ferulic acid for 48 hours. The cell viability of ferolic acid on normal human gingival fibroblasts did not show any decreasement by MTT assay and also, cell adhesion activity did not decreased by XTT assay, respectively, compared with control after cells were treated with various concentrations of ferolic acid for 48 hours. MTT/sub 50/ and XTT/sub 50/ were 2,130.0 μM and 1,773.7 μM ferolic acid, respectively. These results suggest that ferolic acid is non-toxic to normal human gingival fibroblasts by showing no significant differences in the cell viability and the adhesion activity compared with control by colorimetric assay.
In this study, newly improved Ferron assay test haved on timed spectrometry was used for the determination of hyolrolytic Al species presented in PACl coagulant. The color development reagent ferron was prepared by using conventional method and two newly developed methods. Then the ferron assay test was used to compare and analyze the distribution of Al(III) hydrolyzed species presented in the prepared PACl and alum. The preparing method of reagent A required an aging period of 7 days by adding a hydroxylamine hydroxide and a 1,10-phenanthroline monohydrate reagent, whereas the preparing method of reagent B was used as a coloring agent immediately without aging time. The regression analysis between UV absorbance and Al concentrations of conventional method and newly developed method of ferron reagents in low-concentration aluminum solutions and high-concentration aluminum solutions, showed the correlation coefficients of 0.999 or higher, as showing high correlations of conventional method and newly developed method. Applying Ferron assay test, Al species in the PACls and alum were classified as Ala(monomeric Al), Alb (polymeric Al), and Alc (colloidal and precipitated Al). Distribution of Al(III) hydrolyzed species according to the preparation of ferron colorimetric reagents was similar.
