Analysis of Degradation Products in Madder Dyed Fabrics in Selective Degradation Conditions (퇴화조건에 따른 꼭두서니 염색물의 퇴화물 연구)

  • The purpose of this investigation was to investigate the degradation products of the dye component extracted from madder dyed fabrics using the GC-MS analysis and to evaluate the change of color due to degradation treatment. Four different degradation protocols were used in this study,; refrigeration at $7^{\circ}C$ (LT), room temperature (RT), oven treatment at $100^{\circ}C$ (OV), and $H_2O_2/UV(PER)$ method. Degradation times for each thermal system were 6 hour, 24 hour, 48 hour, 1 week, 2 week, 4 week. Alizarin was detected from the control and degraded samples of both alizarin dyed and madder dyed fabrics. Benzoic acid, 2, 4-di-tert-butylphenol, phthalic anhydride were detected as the degradation products for both alizarin dyed and madder dyed fabrics. The result suggest that these products can be used as the fingerprints of GC-MS analysis for the identification of madder dye in archaeological textiles. Both alizarin dyed and madder dyed samples became less red and less yellow after degradation. In the PER degradation system madder dyed sample showed the greatest color difference even after 1 week of degradation treatment. Further research is necessary for investigating the color change in the exhumed textiles, which is caused by the dual action of dye fading and the staining of organic matters in the soil.

Identification of SNP(Single Nucleotide Polymorphism) from MC1R, MITF and TYRP1 associated with Feather Color in Chicken (닭의 모색 연관 유전자인 MC1R, MITF, TYRP1의 SNP(Single Nucleotide Polymorphism) 규명)

  • The Feather Color of chicken is considered as most obvious, and the purpose of this study is to identify the genotype following the SNP of MC1R, MITF and TYRP1, which are genes related to Feather Color, and develop a SNP marker that can be classified per breed. When a haplotype is observed through the combination of markers, a Korean Native Chicken can especially be distinguished when it is a CGG type in the SNP combination of the MC1R gene. In case of the TAG, TGG and TAA types, only Araucana was identified, and for the CAA type, Leghorn could specifically be distinguished. In the SNP combination of TYRP1 gene, only Leghorn was differentiated in case of the TTTCA and CCTCA types, and only Silky Fowl was identified in case of the CTTTA type. The SNP combination of MC1R gene enabled for Korean Native Chicken, Leghorn, and Araucana to be distinguished and each of the SNP and combination of TYRP1 gene allowed for all 4 breeds to be classified. If many researches are conducted about genetic polymorphism between breeds, then it is considered that the differences between breeds will be understood from a molecular biological aspect instead of simply distinguishing the breeds through Feather Color.

Ontogenetic Color Variation of Abudefduf notatus (Pomacentridae: Perciformes) Revealed by 16S rRNA Sequences Analysis (미토콘드리아 16S rRNA 염기서열 분석에 의해 밝혀진 동갈자돔 치어의 성장에 따른 체색변이)

  • Seven individuals (16.1~29.1 mm in length) which were estimated with fishes of genus Abudefduf were collected in Seogwipo, Jeju Island in the summer of 2011 and 2012. Among them, five individuals (20.8~29.1 mm SL) are similar to Abudefduf notatus, based on morphological characters such as yellow transverse band on the body, one small black spot on the base of the pectoral fin, and yellow caudal fin. On the other hand, two individuals (16.1 mm, 17.0 mm SL) differ from them in several light transverse bands and transparent rays of all fins except for pelvic fin, and anterior transparent head with only one (16.1 mm). According to the results of molecular analyses of 578 base pairs of mtDNA 16S rRNA sequences, these individuals correspond to A. notatus adult, with a high bootstrap value of 95% (genetic distance, d=0.002). Therefore, this study shows that the individuals more than 20.8 mm similar to adult body color but the 16.1 mm individual differs to that of adult. We confirm that this species changes to body color during their early life stages. This result regards as a survival strategy to protect themselves against their predator during their early life stages.

Secondary Metabolites and Morphological Diversity in the Leaves of Perilla Landrace from Korea

  • Screening and identification of genetic resources based on their phytoconstituents and morphological characters potentially provide baseline data for researchers, breeders, and nutraceutical companies who wish to formulate a nutrient-dense diet and health beneficial supplement. Thus, we evaluated the amount of total phenolic content and major phenolic compounds; examined if phenolic compounds could be used as distinguishing factors for perilla genetic resources; and investigated the relation between some quantitative and qualitative morphological characters with the contents of phenolic compounds in 360 accessions obtained from National Agrobiodiversity Center gene bank, Jeonju, Korea. Total phenolic content (TPC) was estimated using Folin-Ciocalteu colorimetric assay. Individual phenolic compounds were determined using an Ultra-High Performance Liquid Chromatography system equipped with Photodiode Array detector. Considerable variations were observed in TPC (7.99 to 117.47 mg GAE/g DE), rosmarinic acid (RA) (ND to 19.19 mg/g DE), caffeic acid (CA) (ND to 0.72 mg/g DE), apigenin-7-O-diglucuronide (ADG) (ND to 1.24 mg luteolin equivalent (LUE)/g DE), scutellarein-7-O-glucuronide (SG) (ND to 4.32 mg LUE/g DE), and apigenin-7-O-glucuronide (AG) (ND to 1.60 mg LUE/g DE). RA was the most dominant phenolic compound in most accessions (95.3%) followed by SG. The adaxial leaf color was light green, green and dark green in 13.8%, 65.0%, and 21.1 % of the accessions, respectively. 78.8% of the accessions had light green color at the abaxial side with the remaining being described as green. Most of the accessions (96.9%) were cordate shape, the remaining being eclipse. Intensities of green pigment at abaxial and adaxial leaf surfaces were correlated with contents of individual phenolic compounds and TPC whereas leaf length and width had no correlation with TPC, CA and RA, and negatively correlated with ADG, AG, and SG. Leaf shape was not related with content of phenolic compounds, color of leaves, or the length or width of leaves. Accessions IT57426, IT157434, IT267710, and IT267712 which contained relatively high contents of TPC and major phenolic compounds (RA and SG) could be used for further research in breeding and bioassay test. Our study result showed the contents of total phenolics and individual phenolic compounds along with the morphological characters could be useful distinguishing factors for perilla genetic resources.

2D Spatial-Map Construction for Workers Identification and Avoidance of AGV (AGV의 작업자 식별 및 회피를 위한 2D 공간 지도 구성)

  • In this paper, an 2D spatial-map construction for workers identification and avoidance of AGV using the detection scheme of the spatial coordinates based on stereo camera is proposed. In the proposed system, face area of a moving person is detected from a left image among the stereo image pairs by using the YCbCr color model and its center coordinates are computed by using the centroid method and then using these data, the stereo camera embedded on the mobile robot can be controlled for tracking the moving target in real-time. Moreover, using the disparity map obtained from the left and right images captured by the tracking-controlled stereo camera system and the perspective transformation between a 3-D scene and an image plane, depth map can be detected. From some experiments on AGV driving with 240 frames of the stereo images, it is analyzed that error ratio between the calculated and measured values of the worker's width is found to be very low value of 2.19% and 1.52% on average.

Identification of Five Strains of Bacteria Isolated from the Fermented Tobacco Leaves and Microbial Effects of them for the Quality Enhancement of the Greenish Tobacco Leaves. (발효담배에서 세균의 동정 및 그 세균처리가 청취엽품질에 미치는 영향)

  • In order to improve the quality of greenish tobacco leaves, fermentation experiments by microbial treatment and periodical variations of fermentation temperature were performed. More than twenty strains were isolated from Hyangcho ( Sun cured Korean native leaves ) and Perique tobacco leaves. Among them, five strains which showed good growth in Nicotine Broth medium were selected. Identification experiments of these strains as well as checking the effects of fermentation by these treatment on the quality and aging rate of greenish tobacco leaves were carried out. The results were like following ; 1. Selected strains were identified as pseudomona.f for H-82, for P-4 and Bacillus for H-81. p- 4 and P-7. 2. Among the five strains, strain H- 82 showed the best effect on the forced aging and the quality of greenish tobacco leaves, The rate of oxygen uptaking, pH and nicotine contents were decreased. However, total volatile acids and petroleum ether extracts were increased. 3. After fermentation, taste from greenish tobacco leaf were removed and smoking characteristics were improved. Color was also changed from greenish yellow to dark brown.

Identification of Verticillium tenerum Isolated from Potato and Pumpkin in Korea (감자와 단호박에서 분리한 Verticillium tenerum의 동정)

  • Verticillium tenerum was detected from the samples of potato (Solanum tuberosum L.) and pumpkin (Cucurbita maxima Duch.) in Korea. The samples on which this species was detected were potato tubers collected from Pyungchang and Gimjae, and fruits and seeds of pumpkin from Yeonchon in 2003. The conidiophores of the fungus produced mainly three to five phialides in verticil. The phialides were narrow, flask-shaped, only very slightly swollen at the base, and taper subulately to a narrow neck. They were hyaline, $10{\sim}26{\times}2{\sim}5\;{\mu}m$. The conidia were formed in slimy heads, and were oval to ellipsoidal, pale reddish-brown, $2.5{\sim}7.5{\times}2{\sim}4\;{\mu}m$. The colony on the substrata was unique, showing brick-red color. This is the first report on the distribution of Verticillium tenerum in Korea.

Morphological Characteristics of Medium-Leaf Type Zoysiagrasses (Zoysia spp.) and Their Classification Using RAPDs (중엽형 한국잔디(Zoysia spp.)류의 형태적 특성과 RAPDs를 이용한 분류)

  • Zoysiagrass, especially Jungji implicating medium-leaf type zoysiagrass has been widely used in South Korea recently. This study was carried out to classify 36 selected medium-leaf type zoysiagrass accessions compared to 5 basic zoysiagrass species using RAPDs. Morphological characteristics such as leaf width, leaf angle, leaf sheath length, existance of trichomes and stolon color were measured as useful characteristics for identification of species in Zoysia genus. Nineteen RAPD markers were identified using 8 selected random primers. The dissimilarity coefficient of variants ranged from 0 to 0.736. Three zoysiagrass groups were clustered by dissimilarity coefficient analysis. Group 1 consisted of Z. japonica and some US varieties including 'Zenith' and 'Meyer'. Group 2 consisted of Z. sinica, Z. macrostachya and Korean commercial varieties such as 'Anyang', 'Samdeock', and 'Pyeongdong' medium-leaf type grasses. Group 3 was genetically distinct from Group 1 and Group 2, and included Z. matrella and Z. tenuifolia. 'Anyang', 'Samdeock', and 'Pyeongdong' medium-leaf type zoysiagrasses showed very close genetic relationship with Z. sinica and Z. macrostachya.

Identification of an Entomopathogenic Bacterium, Serratia sp. ANU101, and Its Hemolytic Activity

  • Four different bacterial colonies were isolated from an old stock of an entomopathogenic nematode, Steinernema monticolum. They all showed entomopathogenicity to final instar larvae of beet armyworm, Spodoptera exigua, by hemocoelic injection. However, they varied in colony form, susceptibility to antibiotics, and postmortem change of the infected host insects. Biolog microbial identification and 16S rDNA sequence analyses indicate that these are four different species classified into different bacterial genera. Owing to high entomopathogenicity and a cadaver color of infected insect host, Serratia sp. was selected as a main symbiotic bacterial species and analyzed for its pathogenicity. Although no virulence of Serratia sp. was detected at oral administration, the bacteria gave significant synergistic pathogenicity to fifth instar S. exigua when it was treated along with a spore-forming entomopathogenic bacterium, Bacillus thuringiensis. The synergistic effect was explained by an immunosuppressive effect of Serratia sp. by its high cytotoxic effect on hemocytes of S. exigua, because Serratia sp. caused septicemia of S. exigua when the bacterial cells were injected into S. exigua hemocoel. The cytotoxic factor(s) was present in the culture medium because the sterilized culture broth possessed high potency in the cytotoxicity, which was specific to granular cells and plasmatocytes, two main immune-associated hemocytes in insects.

  • In a smart society, politicians analyze the big data of voters to build a favorable political positions. In other words, a variety of digital footprints uploaded in SNS or Internet are used to set the election strategies and political directions. In comparison, it is difficult for voters to extract intention information about how politicians are performing a political acts. Therefore, it is important that voters need to analyze what intention of politicians are like for two way interaction between voters and politicians. For this, in this paper, we want to do the identification by analyzing IT technologies to narrative styles of politicians who pursue relative advantages or gains compared to other competitors. The experiments will be carried out to identify about what relative advantages compared to other competitors by narrative styles of next presidential candidates who are expected to run into the next presidential election by analyzing the usual audio interviews.
