Korean Red Ginseng extract ameliorates melanogenesis in humans and induces antiphotoaging effects in ultraviolet B-irradiated hairless mice

  • Background: Panax ginseng is a marvelous herbal remedy for all ailments of body. That may be why it is called Panax, which means "cure for all". Melanin is a pigment that gives color to our skin; however, increased melanin production can lead to tumor formation. Human exposure to ultraviolet B radiation has increased extensively owing to the increased sunlight due to global warming. Consequently, a phenomenon called photoaging has been observed for all skin colors and types. As a result of this phenomenon, a set of enzymes called matrix metalloproteinases, which serve as degradation enzymes for extracellular matrix proteins, mainly collagen, is increased, causing depletion of collagen and resulting in early wrinkle formation. Methods: Therefore, in our study, we used the murine melanoma cell line B16/F10 to study the inhibition of melanogenesis by Korean Red Ginseng (KRG) extract in vitro and HRM-2 hairless mice exposed to artificial ultraviolet B to examine the efficacy of KRG in vivo. We prepared a 3% red ginseng extract cream and evaluated its effects on human skin. Results: Our results demonstrated that KRG induced potent suppression of tyrosinase activity and melanin production in B16/F10 cells; moreover, it reduced the transcription and translation of components involved in the melanin production pathway. In the in vivo experiments, KRG potently suppressed the expression of matrix metalloproteinases, reduced wrinkle formation, and inhibited collagen degradation. On human skin, ginseng cream increased skin resilience and skin moisture and enhanced skin tone. Conclusion: Therefore, we conclude that KRG is an excellent skin whitening and antiaging product.

Effects of Mechanical Stimuli on the Cell Proliferation and Collagen Production on the Micropatterned Substrate

  • In relation to the tissue engineering, the cellular responses to the morphology of the scaffold surface are interesting topics. Human ligament fibroblasts (HLFs) were cultured on the micrpatterned silicone substrates subjected to cyclic stretch to simulate ligament motion. Groove and ridge width of silicone substrates was 10/50, 20/50, 20/10, and 20/20 ${\mu}m$ (groove/ridge ${\mu}m$) with a depth of $3{\mu}m$. Strain was applied over two days for 4 hours per day with a frequency of 0.5 Hz with the magnitudes of 4 or 8%. The purpose of this study was to evaluate ligament fibroblast alignment and cellular responses in relation to the pattern of microgrooved surface and stretching magnitude. Ligament fibroblasts in the microgrooved surface were elongated and aligned parallel to the microgrooves under no stretch. Uniaxial cyclic stretch induced cellular activities and their orientation rise in cellular response and the cells showed alignment and elongation perpendicular to the direction of the stretch. Biochemical analyses showed that the best cellular response was found on the $20/50{\mu}m$ under 8% stretch. The surface morphology and mechanical stretching were found to contribute to increase of proliferation, collagen production.

  • The radish skin and radish greens are an edible part of the radish. But they are removed before eating the radish and used as a byproduct or an animal feed material because of their tough and rough texture. Melanin is a pigment that gives colour to our skin. But increased production of melanin can turn into benign or malignant tumours. These days due to global warming, the amount of Ultra violet (UVB) rays has been extensively increased with sunlight. Due to this, a phenomenon called exogenous photo aging is widely observed for all skin colour and types. As a result of this phenomenon, a set of enzymes called matrix metalloproteinases (MMP's) that serves as degradation enzymes for extracellular matrix proteins mainly collagen is increased, causing depletion in collagen and resulting in early wrinkles formation. Therefore in our study we used the murine melanoma cell line B16/F10 to study the melanogenesis inhibition by Heated radish extract (HRE) in vitro and we used HRM-2 hair less mice exposed to artificial UVB for checking the efficacy of Heated radish extract in vivo. Furthermore, we prepared a 3% Heated radish extract (HRE) cream and checked its effects on human skin. Our results have clearly demonstrated that Heated radish extract (HRE) have potently suppressed the tyrosinase activity and melanin production in B16/F10 cells. It had also reduced the expression of components involved in melanin production pathway both transcriptionally and transitionally. In in vivo studies, HRE had potently suppressed the expression of MMP's and reduced the wrinkle formation and inhibited collagen degradation. Moreover, on human skin, ginseng cream increased the resilience, skin moisture and enhanced the skin tone. Therefore in light of these findings, we conclude that HRE is an excellent skin whitening and antiaging product.

배양 혈청이 섬유아세포의 증식 및 교원질합성에 미치는 영향 (Effect of Serum Media on Fibroblast Proliferation and Collagen Synthesis)

  • Expanding cells ex-vivo is very important in tissue-engineering. Culture medium is usually supplemented with fetal bovine serum(FBS) in most of the experiments. However, cells grown in bovine serum media may posses the possibilities of disseminating bovine diseases and/or stimulating the patient's immune reactions. To overcome these problems, autologous or homologous serum should be used instead of the FBS. The purpose of this study is to compare cell proliferation and collagen synthesis depending on the kind of sera mixed on media and to provide a guideline on applying established experimental data to clinical cases. Human dermal fibroblasts were obtained from four patients. Five thousand cells per well in 96-well plates were incubated DMEM/F-12 Nutrient with varying serum mixture; 10% autologous serum, 10% homologous serum, and 10% FBS. Five days after incubation fibroblast proliferation and collagen production were determined by MTT assay and CICP enzyme immunoassay. The mean cell number were; $3.95{\times}10^4/well$, $2.97{\times}10^4/well$ and $2.30{\times}10^4/well$, respectively. The average amounts of collagen synthesized were; 238.13 ng/ml, 204.88 ng/ml, and 163.88 ng/ml in each. These results show that the use of human serum mixture may contribute to, not only preventing disseminated infection of bovine diseases. but also increase cell proliferation and collagen synthesis without simulating the patient's immune reactions.

4-phenylbutyric Acid Regulates Collagen Synthesis and Secretion Induced by High Concentrations of Glucose in Human Gingival Fibroblasts

  • High glucose leads to physio/pathological alterations in diabetes patients. We investigated collagen production in human gingival cells that were cultured in high concentrations of glucose. Collagen synthesis and secretion were increased when the cells were exposed to high concentrations of glucose. We examined endoplasmic reticulum (ER) stress response because glucose metabolism is related to ER functional status. An ER stress response including the expression of glucose regulated protein 78 (GRP78), C/EBP homologous protein (CHOP), inositol requiring enzyme alpha (IRE-$1{\alpha}$) and phosphoreukaryotic initiation factor alpha (p-eIF-$2{\alpha}$) was activated in the presence of high glucose. Activating transcription factor 4 (ATF-4), a downstream protein of p-eIF-$2{\alpha}$ as well as a transcription factor for collagen, was also phosphorylated and translocalized into the nucleus. The chemical chaperone 4-PBA inhibited the ER stress response and ATF-4 phosphorylation as well as nuclear translocation. Our results suggest that high concentrations of glucose-induced collagen are linked to ER stress and the associated phosphorylation and nuclear translocation of ATF-4.

연속가교를 통한 피부 진피세포 담지 콜라겐 겔의 강도 제어 (Tuning the Stiffness of Dermal Fibroblast-encapsulating Collagen Gel by Sequential Cross-linking)

  • 본 연구에서는 생체재료인 콜라겐과 합성 단량체인 아크릴아마이드를 연속가교 하여, 하이드로젤 기반의 콜라겐 겔을 제조하였다. 아크릴아마이드의 함량 및 가교 정도에 따라, 1.5 kPa에서 3.0 kPa까지 다양한 강도(E)를 갖는 콜라겐 겔을 제조할 수 있었다. 또한, 콜라겐 겔에 다공성 기공을 도입하고 진피세포를 내부에 담지하여, 겔 강도에 따른 세포 성장 및 거동을 확인하였다. 상대적으로 강도가 높은 겔에서 세포의 성장은 느렸지만 GAG 합성 및 분비는 활성화되는 것을 확인하였다. 콜라겐 겔의 기계적 물성에 따라 세포의 성장 및 활성이 영향을 받는 것을 알 수 있었으며, 이는 향후 인공피부 제조 및 응용, 나아가 다양한 조직공학 분야의 기반 기술로 활용 가능하리라 기대된다.

  • 류마티스성 관절염의 동물 모델인 CIA(collagen induced arthritisd) 생쥐에 CRN을 복강 투여하여 CIA의 발병률 및 관절염지수 등에 대한 효과를 조사한 결과는 다음과 같다. RF(rhumatoid factor) 인자인 IgG와 IgM 생산량, 그리고 염증 사이토카인인 IL-6와 $TNF-{\alpha}$의 생산량이 감소하였다. 사이토카인 분석결과, 비장세포에서 Th1과 Th2 세포에서 분비되는 $IFN-{\gamma}$와 IL-4를 측정하여 CRN 투여로 Th1 세포에서 Th2 세포로 면역반응이 전환됨을 시사하는 것은 CRN의 치료효과로 사료된다. 혈청 중에서 RA 인자인 IgG와 IgM, 염증 사이토카인인 $TNF-{\alpha}$와 IL-6 및 type II bovine collagen antibody 생산량 이 대조군에 비하여 CRN 투여군이 감소하여 CRN이 관절염의염증 사이토카인을 억제하는 것으로 확인된다.

홍어껍질을 이용한 고기능성 콜라겐 펩타이드 소재 개발 (Development of High Functional Collagen Peptide Materials using Skate Skins)

  • 본 연구에서는 홍어껍질에서 추출한 콜라겐 펩타이드 이화학적 특성과 비만 유발 쥐에 대한 투여 실험을 통해서 지방조직의 지질 감소효과를 조사하였으며, 홍어 껍질 콜라겐 펩타이드를 원료로 한 응용 제품 개발을 통하여 산업적 적용을 검토하였다. 일반성분 분석 결과 홍어 껍질 콜라겐 펩타이드는 대부분이 단백질로 이루어져 있는 고단백 저지방 물질이며, 분자량 측정 결과 약 1000 Da.의 저분자량이고, 아미노산 분석에서는 전형적인 콜라겐 단백질의 특성을 나타내었다. 비만 유발 쥐에 홍어 껍질 콜라겐 펩타이드를 500 mg/kg씩 18일간 경구 투여한 실험 결과에서 사료 섭취량과 음수량이 상당량 줄어 들었으며, 지방조직의 중성지방과 총 콜레스테롤 수치가 유의적으로 감소하였다. 홍어 껍질 콜라겐 펩타이드를 대량생산을 한 결과 제품의 최종 수율은 원료대비 약 17.23%로 상당히 높은 수율을 나타내었다. 이를 원료로 한 기능성 화장품 및 생활용품을 생산 및 출시하였다. 따라서 홍어 가공 부산물인 껍질을 재이용한 기능성 단백질 소재 개발을 통하여 건강기능식품과 화장품 분야에서의 기능성과 가격경쟁력을 갖춘 소재 개발에 기여 할 것으로 기대된다.

Effects of Mechanically Deboned Chicken Meat (MDCM) and Collagen on the Quality Characteristics of Semi-dried Chicken Jerky

  • This study was conducted to determine the effects of using mechanically deboned chicken meat (MDCM) and collagen on quality characteristics of semi-dried chicken jerky. In experiment I, semi-dried chicken jerky was prepared with the replacement of chicken breast with MDCM (0, 10, 20, and 30%). The pH value of the jerky formulated with only chicken breast was 5.94, while the replacement of chicken breast with MDCM significantly increased the pH (p<0.05). The protein content and shear force of the jerkies decreased with increasing amounts of MDCM, whereas the fat, ash content and processing yield showed the opposite tendency (p<0.05). Replacement with up to 10% MDCM had no adverse effects on the sensory characteristics of the semi-dried chicken jerky. In experiment II, four levels of pork collagen (0, 1, 2, and 3%) were added to the semi-dried chicken jerky formulated with 90% chicken breast and 10% MDCM. The addition of collagen increased the moisture content, but decreased the ash content of the jerkies produced (p<0.05). The processing yield of the jerkies increased with increasing added amounts of collagen (p<0.05). It was found that the jerkies formulated with 0-2% collagen had significantly higher overall acceptance score than those prepared with 3% collagen (p<0.05). In conclusion, MDCM and collagen could be useful ingredients to reduce the production cost and improve the processing yield of semidried chicken jerky. The optimal levels of MDCM and collagen which could be added without adverse effects on the sensory characteristics were up to 10% and 2%, respectively.

Suppressive Effect of Solanum Iyratum Aqueous Extract Via Down-regulation of $TNF-{\alpha}$ and $IFN-{\gamma}$ Production on Collagen-induced Arthritis in Mice

  • Solanum Iyratum Thunb (Solanaceae) has multiple applications in korean traditional medicine because of its cytotoxic, immunological and anti-inflammatory activities. However, no study on the anti-arthritic activity of Solanum Iyratum Thunb has been reported in vivo. Rheumatoid arthritis (RA) is a systemic autoimmune disease with chronic inflammation characterized by hyperplasia of synovial cells in affected joints, which ultimately leads to the destruction of cartilage and bone. Cytokine production were assessed during CIA(collagen-induced arthritis) model mice in lymph node (LN), in knee joint and spleen, using ELISA. DBA/1j mice were immunized with bovine type II collagen. After a second collagen immunization, mice were treated with Solanum Iyratum Thunb (SLT) orally at 400, 200 mg/kg once a day for 4 weeks. The severity of arthritis within the knee joints was evaluated by histological assessment of cartilage destruction and pannus formation. SLT significantly suppressed the progression of CIA and inhibited the production of TNF-alpha and IFN-gamma in serum and spleen cell culture supernatant. The erosion of cartilage was dramatically reduced in mouse knees after treatment with SLT. In conclusion, our results demonstrates that SLT significantly suppressed the progression of CIA. This action was characterized by suppression of arthritis index, cartilage erosion and synovial cell infiltration and the decreased production of $TNF-{\alpha}$, $IFN-{\gamma}$, CD4+, CD19+, CD3+/CD69+ cells (in lymph node), CD11b+/Gr-1 + (in knee joint).
