Laminin as a Key Extracellular Matrix for Proliferation, Differentiation, and Maturation of Porcine Muscle Stem Cell Cultivation

  • Extracellular matrix (ECM) proteins play a crucial role in culturing muscle stem cells (MuSCs). However, there is a lack of extensive research on how each of these proteins influences proliferation and differentiation of MuSCs from livestock animals. Therefore, we investigated the effects of various ECM coatings-collagen, fibronectin, gelatin, and laminin-on the proliferation, differentiation, and maturation of porcine MuSCs. Porcine MuSCs, isolated from 14-day-old Berkshire piglets, were cultured on ECM-coated plates, undergoing three days of proliferation followed by three days of differentiation. MuSCs on laminin showed higher proliferation rate than others (p<0.05). There was no significant difference in the mRNA expression levels of PAX7, MYF5, and MYOD among MuSCs on laminin, collagen, and fibronectin (p>0.05). During the differentiation period, MuSCs cultured on laminin exhibited a significantly higher differentiation rate, resulting in thicker myotubes compared to those on other ECMs (p<0.05). Also, MuSCs on laminin showed higher expression of mRNA related with maturated muscle fiber such as MYH1 and MYH4 corresponding to muscle fiber type IIx and muscle fiber type IIb, respectively, compared with MuSCs on other ECM coatings (p<0.05). In summary, our comparison of ECMs revealed that laminin significantly enhances MuSC proliferation and differentiation, outperforming other ECMs. Specifically, muscle fibers cultured on laminin exhibited a more mature phenotype. These findings underscore laminin's potential to advance in vitro muscle research and cultured meat production, highlighting its role in supporting rapid cell proliferation, higher differentiation rates, and the development of mature muscle fibers.

골과 임플란트 접촉면의 조직학적 연구 (Histological Study on the Interface of Bone and Implant)

  • This paper reports the morphological nature of the remodelled interface process between implants and surrounding bone after 1, 4, 6, 8 and 12 weeks of implantation of smooth machined implants into rat tibias. After 4 weeks of implantation, histochemical analysis showed that the new bone was growing in direct contact with the implant. In the forming process, the activatived osteoblast cells migrated toward the interface and colonized the surface at the contact areas. This immature woven bone, rich in osteocyte lacunae, was deposited directly onto the implant surface. Osteoblast activity was found to continue ill 12 weeks of implantation The osteoblasts in lacunar areas developed numerous processes and synthesized bone matrix, after all, surrounded by secreting matrix. At the 12th week, the amount of newly formed bone matrix between bone and implant increased in mineralization. The mineralized mature bone contained well organized collagen fibers with characteristic banding pattern bone tissue formation around the implant.

황백약침이 콜라겐 유도 관절염 생쥐에 미치는 영향 (A Study on the Effect of Herbal-acupuncture with Phellodendri Cortex Solution on Collagen-induced Arthritis in DBA/1J Mice)

  • Objectives & Methods : The purpose of this study is to observe the effects of Phellodendri Cortex Herbal-acupuncture solution (PC-HAS) at Joksamni (ST36) on collagen II induced arthritis in DBA-1J mice. The author performed several experimental items to analyze arthritis evaluation, change of weight, spleen size and adhesion rate, change of cytokine level, IgG, IgM and anti-collagen II, chang of immunocyte count, histological change of CIA mouse joint. Results : 1. In the PC-HA group, arthritis index, the incidence of arthritis and joint edema were significantly decreased. 2. In the PC-HA group, the change of spleen size, spleen adhesion rate and the knee joint were significantly decreased. 3. The levels of $IL-1{\beta}$, IL-6 and INF- in serum of the CIA mouse were significantly decreased by PC-HA. 4. The levels of IgG, IgM and anti-collagen II in serum of the CIA mouse were significantly decreased by PC-HA. 5. In the CIA mouse spleen cell culture, the levels of IFN- , IFN- / IL-4, IL-10 were significantly decreased by PC-HA, but the level of IL-4 was significantly increased by PC-HA. 6. In the PC-HA group, the ratios of $CD3e^+$ to $CD45R^+$ cell, $CD4^+$ to $CD8^+$ cell and $CD4^+/CD25^+$ cell were similarly maintained as normal group in the CIA mouse spleen cell. 7. In the PC-HA group, $CD4^+CD25^+$ and $CD45R^+/CD69^+$ cell were significantly decreased in the lymph nodes. 8. In the PC-HA group, $CD3^+/CD69^+$ and $CD11b^+/Gr-1^+$ cell were significantly decreased in knee joint. 9. In histology, the cartilage destruction and synovial cell proliferation in the PC-HA group were similar with that of the normal group and the collagen fiber expressions in the PC-HA group were similar with that of the normal group. Conclusions : Form the result above, the results suggest that the PC-HA at ST36 has significant effect on collagen-induced arthritis, and can be put to practical use in the future rheumatoid arthritis clinic.

Third, it is the levels of IL-6, $TNF-{\alpha}$, $IFN-{\gamma}$, $IL-{\beta}$, IgG, IgM and anti-collagen II in serum and the level of IFN-y,$IFN-{\gamma}$/IL -4 ratio in CIA mouse spleen cell culture. Fourth, it is histological analysis of the mice joint. Fifth, it is expression ratio of $CD3e^+$ to $CD19^+$+ cell, $CD4^+$ to $CD8^+$ cell, $CD69^+/CD3e^+$/cells, $CD11a^+/CD19^+$/cells, $CD11b^+/Gr-1^+$ cells and $CD4^+/CD25^+$ cells. Results & Conclusion : 1. In the EC-HA, the incidence of arthritis and arthritis index were significantly decreased. 2. In EC-HA, the levels of IL-6, $IFN-{\gamma}$, $TNF-{\alpha}$, $IL-1{\beta}$, IgG, IgM and anti-collagen II in serum of CIA mice and the level of $IFN-{\gamma}$, IL-4, $IFN-{\gamma}$/lL-4 ratio in CIA mouse spleen cell culture were significantly decreased. 3. In the histological study, the cartilage destruction and synovial cell proliferation were decreased in the EC-HA, and the collagen fiber expressions in the EC-HA were similar with that of the Normal group. 4. In the EC-HA, the expression ratio of $CD3e^+$ to $CD19^+$ cell and $CD4^+$ to $CD8^+$ cell were similarly maintained as Normal group in lymph nodes, and $CD69^+/CD3e^+$ cells and $CD11a^+/CD19^+$ cells were decreased in lymph nodes, and $CD11b^+/Gr-1^+$ cells and $CD4^+/CD25^+$ cells were decreased in synovium. These results suggest that EC-HA at ST36 has an effect to control synovial cell proliferation and cartilage destruction in rheumatoid arthritis, and to be put to practical use in the future rheumatoid arthritis clinic.

  • Objectives : The purpose of this study is to observe the effects of Carthami Flos herbal-acupuncture (CF-HA) at Joksamni($ST_{36}$) on arthritis in mice induced by Collagen II. Methods : The author performed several experimental items, including arthritis evaluation, change in weight, spleen size and stenosis rate, change in cytokine level, IgG, IgM and anti-collagen II, change of immunocyte count and histological change of the CIA mouse joint. Conclusions are as follows: Results : 1. In the CF-HA, the arthritis index and rate and the incidence of arthritis were decreased as the experiment proceeded. 2. In the CF-HA, spleen swell and stenosis, joint edema and change were decreased. 3. In the CF-HA, the level of $IL-1{\beta}$, IL-6, $TNF-{\alpha}$ and $IFN-{\gamma}$ in blood serum were significantly decreased. 4. In the CF-HA, the level of IgG, IgM and anti-collagen II were decreased. 5. In the CF- HA, $IFN-{\gamma}$, $IFN-{\gamma}/IL-4$, IL-10 of the culture fluid was decreased. 6. In the CF-HA, the cell rate of $CD3e^+$ and $CD45R^+$, $CD4^+$ and $CD8^+$, $CD4^+/CD25^+$ in spleen was similar to the cell rate of the normal group. 7. In the CF-HA, the cell rate of $CD4^+/CD25^+$, $CD45R^+/CD69^+$ in a lymph node was decreased as in the normal group. 8. In the CF-HA, the cell rate of $CD3^+/CD69^+$, $CD11b^+/Gr-1^+$ in joints was decreased as in the normal group. 9. In the CF-HA, the cartilage destruction and the inflammation cell growth in the H&E stain were decreased. The collagen fiber in the M&T stain were less destructed, therefore the result was similar to the normal group. Conclusions : These results suggest that CH-HA at $ST_{36}$ has an effect in controlling immune reaction and suppressing inflammation in rheumatoid arthritis therefore, the continuous flow of the following study is expected.

Conclusion : These results suggest that MR-HA at ST36 has an effect to control synovial cell proliferation and cartilage destruction in rheumatoid arthritis, as well as prophylaxis is important to treat rheumatoid arthritis in clinic.

  • Objectives & Methods : The purpose of this study is to observe the effects of Angelicae Radix Pharmacopuncture(AR-P) at Joksamni(ST36) on collagen II induced arthritis in DBA/1J mice. The authors evaluated arthritis index, arthritis incidence and joint edema, and measured body weight, spleen size and stenosis rate, serum cytokine level, serum antibody level, immune cell populations In spleen, lymph node, and knee joint, and performed histological analysis of CIA mouse joint. Results : In the AR-P group, arthritis index, arthritis incidence and joint edema were decreased, and the enlargement, malformation and stenosis of spleen and the malformation of joint appeared milder than the control group. In AR-P group, the levels of $IL-1{\beta}$, IL-6, $TNF-{\alpha}$, $IFN-{\gamma}$ in serum were significantly decreased. And the level of anti-collagen II in serum was maintained lower in AR-P group than in the control group. In AR-P group, the level of $IFN-{\gamma}$ and IL-10, and $IFN-{\gamma}/IL-4$ ratio were significantly decreased, and the ratios of $CD3e^+$ cells to $CD45^+$ cells, $CD4^+$ cells to $CD8^+$ cells in spleen were similarly maintained as those of the normal group. In the AR-P group, the populations of $CD4^+/CD25^+$ cells in spleen and lymph nodes, $CD45R^+/CD69^+$ cells in lymph nodes, $CD3^+/CD69^+$ cells and $CD11b^+/Gr-1^+$ cells in knee joint were decreased. In the histological analysis, the cartilage destruction, synovial cell proliferation and the collagen fiber destruction were decreased in the AR-P group Conclusions : The results suggest that AR-P at right ST36 has a therapeutic effect on collagen-induced arthritis in mice.

여러 가지 가교제가 인체 무세포진피의 안정성에 미치는 영향 (Effects of Cross-Linking Agents on the Stability of Human Acellular Dermal Matrix)

  • Purpose: Human acellular dermal matrix(ADM) is widely used in the treatment of congenital anomalies and soft tissue deficiencies. But it is rapidly degraded in the body and does not provide satisfactory results. There is a need to improve collagen fiber stability through various methods and ultimately regulate the speed of degradation. Methods: The ADMs were added with various cross-linking agents called glutaraldehyde, dimethyl 3,3'-dithiobispropionimidate to produce cross-linked acellular dermal matrices. 1,4-butanediol diglycidyl ether solution was applied with a pH of 4.5 and 9.0, respectively. The stability of cross-linked dermal matrix was observed by measuring the shrinkage temperature and the degradation rates. The cross- and non-cross linked dermis were placed in the rat abdomen and obtained after 8, 12 and 16 weeks. Results: The shrinkage temperature significantly increased and the degradation rate significantly decreased, compared to the control(p<0.05). All of cross-linked dermises were observed grossly in 16 weeks, but most of non-cross linked dermis were absorbed in 8 weeks. Histologically, the control group ADM was found to have been infiltrated with fibroblasts and most of dermal stroma were transformed into the host collagen fibers. However, infiltration of fibroblasts in the experiment was insignificant and the original collagen structure was intact. Conclusion: Collagen cross-linking increases the structural stability and decreases degradation of acellular dermis. Therefore, decrease in body absorption and increase in duration can be expected.

인체 섬유아세포 및 케라티노사이트에 대한 지방줄기세포 분비물의 세포생물학적 기능 (Cell Biological Function of Secretome of Adipose-Derived Stem Cells on Human Dermal Fibroblasts and Keratinocytes)

  • 피부재생에 대한 지방줄기세포 배양상등액(ADSC-CM)의 효능에 대한 연구를 진행하였다. ADSC-CM이 피부재생에 기여하는 기작은 명확하지 못하지만, ADSC-CM은 다양한 분비물을 포함하고 있고 따라서 피부트러블 처리를 위한 훌륭한 재료이다. 저 산소 상태에서 생산된 ADSC-CM, 즉 advanced adipose-derived stem cell protein extract (AAPE)는 피부재생에 보다 좋은 재료이다. 본 연구는 피부 재생에 결정적 역할을 하는 인체 primary 세포인 섬유아세포(HDF)와 케라티노사이트(HK)를 이용하여 AAPE의 효능을 검증하였다. 0.32 ${\mu}g/ml$ AAPE에서 콜라겐 합성이 관찰 되었으며 AAPE는 stress fiber 형성을 강화하였다. DNA microarray 결과에서는 세포증식, 세포이동, 세포부착, 상처반응에 관여하는 133개의 유전자 발현이 조절되는 것을 알았다. Antibody array를 통해 CD54, FGF-2, GM-CSF, IL-4, IL-6, VEGF, TGF-${\beta}2$, TGF-${\beta}3$, MMP-1, MMP-10, 그리고 MMP-19와 같은 MMP, 성장인자, 사이토카인등 25개의 알려진 단백질이 포함되어 있다는 것을 알았다. 따라서, AAPE는 HK의 세포생물학적 기능을 활성화 할 수 있다고 사료되며 HDF에서는 콜라겐 합성을 유도하였다. 이러한 결과는 AAPE가 피부재생에 임상적 적용이 가능하리라는 것을 의미한다.

밀어 (Rhinogobius brunneus)와 민물검정망둑 (Tridentiger brevispinis)의 흡반 구조에 관한 연구 (A Study of Structure of the Sucker of Common Freshwater Goby (Rhinogobius brunneus) and Triden Goby (Tridentiger brevispinis))

  • 본 연구는 좌 우의 배지느러미가 양쪽으로 합쳐져서 흡반을 형성하는 망둥어과(Cobiidae) 어류인 밀어 (Rhinogobius brunneus)와 민물검정망둑 (Tridentiger brevispinis)의 흡반의 형태 및 구조를 광학현미경, 주사전자현미경, 투과형 전자현미경을 이용하여 관찰하였다. 밀어 흡반의 외부형태는 첨부는 규칙적인 실타래 모양의 조밀한 굴곡을 형성하고 있으며, 중부와 기저부는 첨부에 비하여 조밀하지는 않으나 벌집모양의 규칙적인 구조를 갖는다. 세포의 경계와 세포 표면 내에 이랑을 형성하고 있었으며 구조물 사이에 점액분비세포(mucous cell)의 개구부(openings)가 다수 관찰되었다. 내부구조는 초생달모양의 지느러미살을 중심으로 조밀한 섬유층과 상피표면을 향하는 방사상의 성긴 탄력성 섬유층과 입방형 또는 원추형 상피 세포들로 이루어져 있다. 민물검정망둑의 흡반의 외부형태는 첨부는 실타래 모양의 굴곡을 가지나 규칙적이지 않다. 중부와 기저부는 밀어와 같이 벌집모양의 구조로 되어 있으며 구조물 사이에 점액분비세포의 개구부가 관찰되었으나 밀어에 비해 적다. 내부구조는 밀어 에서와 같이 지느러미살을 중심으로 조밀한 섬유층이 관찰되지만 방사상의 성긴 탄력성 섬유층은 관찰되지 않으며 편평상피 또는 다각형의 상피세포들로 구성되어 있어서 밀어에 비하여 흡반의 두께가 얇다.
