Effect of Glucose and Insulin on Human Gingival Fibroblasts and Periodontal Ligament Cells (포도당 및 인슐린이 인체 치은섬유모세포와 치주인대세포에 미치는 영향)

  • Diabetes mellitus is a systemic disease with profound effects on oral health and periodontal wound healing. Uncontrolled diabetes adversely affects surgical wound healing and is often associated with abnormal proliferation of fibroblasts. Human gingival fibroblasts and PDL cells were chosen because they are intimately involved in periodontal therapy and are important for the success of surgical procedure such as guided tissue regeneration. The aim of the present study was to elucidate whether cellular activity and collagen synthesis by glucose pre-treated human gingival fibroblasts and PDL cells are influenced by insulin, and whether healthy cells differ from glucose treated cells. Cells were cultured with DMEM at $37^{\circ}C$, 5% $CO_2$, 100% humidified incubator. To evaluate the effect of glucose on gingival fibroblasts and periodontal ligament cells, the cells were seeded at a cell density of $1{\times}10^4\;cells/well$ culture plates and treated with 20 and 50mM of glucose for 5 days. Then MTT assay was carried out. To evaluate the effect of insulin on glucose-pretreated cells, the cells were seeded at a cell density of $1{\times}10^4\;cells/well$ culture plates and treated with 20 and 50mM of glucose for 5 days. After incubation, $10^3$, $10^4$ and $10^5mU/l$ of insulin were also added to the each well and incubated for 2 days, respectively. Then, MTT assay and collagen synthesis assay were carried out. The results indicate that cellular activity of gingival fibroblasts significantly increased by glucose while periodontal ligament cells were unaffected and cellular activity of gingival fibroblasts and periodontal ligament cells were unaffected by insulin. Collagen synthesis of gingival fibroblast with 20mM glucose and insulin unaffected, but 50mM glucose and insulin increased than control. Collagen synthesis of periodontal ligament cell with 20mM glucose and $10^5mU/l$ insulin significantly increased than other groups and 50mM glucose pretreated PDL cells significantly increased at $10^3mU/l$ insulin but decreased at $10^4mU/l$ insulin. Our findings indicated that these cell types differed in their growth response to glucose, and the increase in collagen synthesis was significantly raised at insulin level of $10^3mU/l$ in gingival fibroblasts and periodontal ligament cells except 20mM glucose pretreated periodontal ligament cells.

  • Measuring in vivo rate of bone collagen synthesis has so far been technically difficult and often subject to quite large errors. In the present study, bone collagen synthesis rate was measured using a precursor-product method, based on the exchange of $^2$$H_2O$ into amino acids. Mass isotopomer abundance in hydroxyproline from bone collagen was analyzed by gas chromatography/mass spectrometry. The $^2$$H_2O$ labeling protocol consisted of an initial intraperitoneal injection of 99.9% $^2$$H_2O$, to achieve approximately 2.5% body water enrichment followed by administration of 4% $^2$$H_2O$ in drinking water for 9 weeks. Body $^2$$H_2O$ enrichments were stable at 2.7 ∼ 3.0% over labeling Period. In growing rats, the fractional synthesis rate ( $k_{s}$) of bone collagen was 0.066 $\pm$ 0.049 w $k^{-1}$ . The unique features of stable $^2$$H_2O$ pools and label incorporation allowed the precursor-product approach to be used for measuring bone collagen synthesis rate..

The Effects of Genistein on the Proliferation and Type I pN Collagen Synthesis in Aged Normal Human Fibroblasts (제니스테인에 의한 노화된 피부세포 활성화와 콜라겐 생성 효과)

  • We studied the effects of genistein obtained from glycolysis of genistin, a kind of phytoestrogen present in soybeans, on cell proliferation and type I pN collagen synthesis in normal human dermal fibroblasts(NHDF). Cell proliferation was increased significantly with genistein treatment at 54-year aged NHDF. Genistein increased cell proliferation more strongly in cells form old doner than young doner. The senescence-associated ${\beta}$-galatosidase activity was decreased in NHDF from 77-year old doner with genistein treatment. Type I pN collagen synthesis was increased with genistein treatement in UVA treated and non-treated NHDF. The increasement of collagen synthesis was more effective in aged cells than young cells. Type I pN collagen synthesis was also increased with genistein treatment in collagen matrix culture with NHDF from sun-exposed and non-exposed skin from 54-year old doner. Genistein treatment inhibited MMP-1 synthesis in old NHDF but not in young NHDF. In conclusion, genistein may be a useful agent for preventing intrinsic aging as well as photoaging.

  • Objectives: In this study, the author aimed to evaluate the effect of BuOH fraction(YK) from Cinnamomum loureirii on osteoblast proliferation in murine rat calvarial cells. Methods: The osteoblast separated from murine calvariae was cultivated for 10 days and evaluated the cell function. After the addition of YK on the culture medium, we determined the effect of YK on the cell proliferation, alkaline phosphatase activity, apoptosis of the cultivated osteoblast, protein synthesis and collagen synthesis. Results: YK increased the proliferation of rat calvarial osteoblast. YK increased ALP activity of rat calvarial osteoblast. YK did not change the survival rate of rat calvarial osteoblast. YK increased protein synthesis of rat calvarial osteoblast. YK increased collagen synthesis of rat calvarial osteoblast. Conclusions: This study suggests that YK might improve the osteoporosis resulted from augumentation of osteoblast proliferation.

  • A novel Nicotinoyl fused peptide, Nicotinoyl-LVH, was synthesized by solid phase peptide synthesis method, purified, and tested in cultured skin cells. Nicotinoyl-LVH enhanced the expression level of collagen mRNA and its fragments in fibroblasts. These data show that this novel Nicotinoyl peptide is a promising biomaterial in the anti-aging functional cosmetic market.

  • Activation of hepatic stellate cell has been identified as a critical step in hepatic fibrogenesis and is regulated by several factors including cytokines and oxidative stress. In this study, we assayed effects of saponin (CKS), inulin (CKI) and oligo-sugars (CKO) isolated from Platycodon grandiflorum A. DC, changkil (CK) on experimental cell cycle arrest and apoptosis in hepatic stellate cell line (HSC-T6). CKS induced cell arrest at G$_1$. CKS also reduced intercellular reactive oxygen species and collagen synthesis in hydrogen peroxide-induced oxidative stress and acetaldehyde-stimulated collagen synthesis, respectively, in HSC-T6 cells.   • Introduction: Liver fibrosis is defined unbalance of collagen metabolism, especially a stimulation of collagen synthesis and inhibition of collagen degradation, and antifibrotic effect is delaying or inhibition of new collagen synthesis and deposition in liver tissue. In this study, we investigated the antifibrotic and antioxidative effect of Solanum lycopersicum (SL) in liver fibrosis induced rats.
