The Finite Element Analysis of the Mandrel Shape's Influence on the Residual Stress Distribution by Cold Expansion Method (형상봉의 모양이 홀확장 잔류응력 분포에 미치는 영향에 대한 유한요소해석)

  • Cold expansion method is used to protect a fatigue fracture from fastener hole in the structure and aerospace industry. Cold expansion is that an oversized tapered mandrel goes through the hole and produces a compressive residual stress as well as plastic deformation around the hole. Here, mandrel shapes are one of the factors which are influenced on the residual stress distribution by cold expansion method. This paper, according to mandrel shapes (diameter of mandrel, length of mandrel and length of taper), we are performed a finite element analysis of residual stress distribution by cold expansion method. From this study, it has been found that diameter of mandrel and length of taper are an important factor which was generated a low compressive residual stress surround of fastener hole by cold expansion method.

The Effects of 120Hz High Frequency Electroacupuncture on the Cold-Restraint-Induced Gastric Mucosal Damages (Cold-Restraint 스트레스 유발 위점막손상에 미치는 고빈도 120Hz 전침의 효과)

  • Objective : The present studies investigated the effects of 120Hz high frequency electroacupunctue(EA) on the stress-induced stomach dysfunction in relation to its effect on the level of stress hormone and gastric mucosal damages. The gastric mucosal injury was induced by cold-restraint stress and two acupoints corresponding to Zusanli and Sanyinjiao in man were used. Methods: Cold-restraint stress produced typical gastric lesions in all rats of the stressed groups, but he number of ulcers as well as the mean ulcer diameter were reduced by 120 Hz EA pre-treatment. The cold-restraint stress also induced an increase in catecholamine response involving epinephrine, norepinephrine and dopamine, but an slight decline were observed in EA pre-treated rats compared with cold-restrained rats. Results: The degranulation value of gastric mast cell was significantly higher in cold-restrained rats than in control ones. However, with the significant reduction of degranulation values of gastric mast cells in EA pre-treated rats compared with cold-restrained ones, $PGE_2$ content in the gastric mucosa of EA pre-treated rats was also different from that observed in cold-restrained rats. Cold-restraint stress induced an elevated mRNA expression of pro-inflammatory gene such as cyclooxygenases-2 and tumor necrosis factor(TNF)-${\alpha}$, but these expression were down-regulated in EA pre-treated rats. Immunohistochemecal analysis showed that while the inhibitory-${\kappa}B{\alpha}$ an TNF-${\alpha}$ immunoreaction in the surface epithelium of the stomach tended to increase, both reactions in the EA pre-treated rats showed similar pattern as observed in controls. Conclusions : These results suggest that 120 Hz EA may act as a therapeutical means for gastric mucosal damages through a activation of pituitary adrenal system. it could be concluded that 120 hz high frequency electroacupuncture affords a good protective potential against stress-induce gastrointestinal dysfunction.

  • Cold expansion method is retarded of crack initiation due to the compressive residual stress developed on the hole surface. Previous research has just been study about residual stress distribution in the hole surrounding. But, The purpose of this study was to improve the understanding of the residual stress effect in hole surrounding as crack growth from another hole. In this paper, it is shown that residual stress is redistributed due to the application of cold expansion process for CT specimen using finite element method. It is further shown that tensile stress increases in proportion to cold expansion ratio in the vicinity of crack. It is thought that stress intensity factor increases with cold expansion ratio.

Response of Ascorbate Peroxidase and Dehydroascorbate Reductase in Lettuce (Lactuca sativa L.) Leaves Exposed to Cold Stress (저온 처리한 상추의 잎 내에서 ascorbate peroxidase와 dehydroascorbate reductase의 반응)

  • To investigate the relationship between cold stress and the activity of ascorbate peroxidase(APX), dehydroascorbate reductase (DHAR), mRNA expression level of two enzymes, hydrogen peroxide content was studied in lettuce leaves under stress condition imposed by cold stress at $4^{\circ}C$ for 24 hr in the dark and following recovery at $20^{\circ}C$ from cold stress. Hydrogen peroxide content increased gradually in lettuce leaves during cold stress, but decreased slightly following recovery from cold stress. Soluble protein content, however, decreased gradually during cold stress, and then rapidly returned to normal levels following recovery. Total chlorophyll content decreased gradually during cold stress, and then keep constant following recovery. The patterns of chlorophyll a and b content similar to that of total chlorophyll content, and carotenoid content didn't change. The ratio of chlorophyll a and total chlorophyll was increased during cold stress, but decreased with rapid during cold stress, and then the ratio returned to normal levels following recovery. During cold stress, the activity of APX and DHAR in the lettuce leaves increased dramatically, and also transcript levels of mRNA of APX and DHAR, as determined by probing 32P-labeled single stranded RNA of APX and DHAR, highly increased and returned to normal levels following recovery, respectively. Relationship between APX and DHAR activity and hydrogen peroxide highly related ($R^2$=0.8715 and 0.8643), whereas between hydrogen peroxide and total chlorophyll content and soluble content related reversely ($R^2$=0.5021 and 0.8915).

  • Glutathione and ascorbic acid have been shown to fulfill many essential functions in animal and plant growth, development, defence and protection against oxidative damage. Effects of glutathione and ascorbic acid were examined in transgenic N. tabacum cells producing hGM-CSF to determine the effects of the vitamins on growth and cell viability. In lag phase, cell viability was preserved by glutathione and ascorbic acid. Therefore, recombinant protein productivity was increased. The purpose of present study is to investigate the role of antioxidants in cold stress-induced apoptosis in plant suspension cells. Cold stress lowered cell viability and increased total genomic DNA fragmentation. Supplementing the cell cultures with glutathione and ascorbic acid inhibited cold stress-induced decrease in cell viability and increase in total genomic DNA fragmentation.

Clinical Observation on Heart Rate Variability and Peripheral Blood Flow by Doppler Ultrasound in Healthy Subjects Stimulated by Cold Stress (정상인의 한냉자극에 대한 심박변이도 및 도플러 초음파 혈류계를 이용한 말초 혈류관찰)

  • Objectives: This study was performed to investigate the effect of cold stress on heart rate variability and peripheral blood flow by Doppler ultrasound in healthy subjects. Methods: We classified the subjects by their Yin or Yang tendency using a Yin-Yang questionnaire. Cold stress was taken on the left hand of subjects for 10 seconds. Heart rate variability and peripheral blood flow were measured on the right radial artery before and after cold stress. Results: The Yang tendency group showed a significant decrease of heart rate after cold stress which was not seen in the Yin tendency group. The Yang tendency group showed a decrease of LF, total power, and increase of HF after cold stress, while the Yin tendency group showed the opposite. However, there were no statistically significant differences of heart rate variability analysis between the results of the two groups before and after cold stress. Recovery time of blood flow velocity by ultrasound Doppler was significantly different between the Yin and Yang tendency groups after cold stress. Conclusion: These results suggest that people with Yin or Yang tendency may show different responses in the autonomic nervous system. Further modified studies may include the responses to various manipulation techniques in acupuncture treatment and the individual responses according to pattern identification in traditional Korean medicine.

Physiological and transcriptome analysis of acclimatory response to cold stress in marine red alga Pyropia yezoensis

  • Red macroalga Pyropia yezoensis is a high valuable cultivated marine crop. Its acclimation to cold stress is especially important for long cultivation period across winter in coasts of warm temperate zone in East Asia. In this study, the response of P. yezoensis thalli to low temperature was analyzed on physiology and transcriptome level, to explore its acclimation mechanism to cold stress. The results showed that the practical photosynthesis activity (indicated by ΦPSII and qP) was depressed and pigment allophycocyanin content was decreased during the cold stress of 48 h. However, the Fv/Fm and non-photochemical quenching increased significantly after 24 h, and the average growth rate of thalli also rebounded from 24 to 48 h, indicating a certain extent of acclimation to cold stress. On transcriptionally, the low temperature promoted the expression of differentially expressed genes (DEGs) related to carbohydrate metabolism and energy metabolism, while genes related to photosynthetic system were depressed. The increased expression of DEGs involved in ribosomal biogenesis and lipid metabolism which could accelerate protein synthesis and enhance the degree of fatty acid unsaturation, might help P. yezoensis thallus cells to cope with cold stress. Further co-expression network analysis revealed differential expression trends along with stress time, and corresponding hub genes play important roles in the systemic acquired acclimation to cold stress. This study provides basic mechanisms of P. yezoensis acclimation to cold temperature and may aid in exploration of functional genes for genetic breeding of economic macroalgae.

Acclimation responses of Tamarix chinensis seedlings related to cold stress

  • The purpose of this study was to investigate the acclimation responses of Tamarix chinensis to cold stress. We evaluated the acclimation responses by measuring biomass, daily elongation rate, chlorophyll content, and total soluble carbohydrate content. The plant samples comprised leaves from seedlings of 2 different ages (8 and 12 weeks); the leaves were collected 0, 2, and 4 weeks after cold treatment. We found that the cold-treated samples showed reduced daily elongation rates and chlorophyll content. Further, these samples showed more than 8-fold increase in the total soluble carbohydrate content. However, the seedling ages did not have a significant influence on the growth of cold-treated seedlings. On the basis of these findings, we can conclude that T. chinensis seedlings aged less than 1 year old show acclimation to cold stress by accumulating soluble carbohydrates. This study may help us understand how T. chinensis seedlings acclimatize to their first cold season.

Changes in Cell Ca2+ Distribution in Loquat Leaves and Its Effects on Cold Tolerance

  • Calcium has been associated with improved cold tolerance in many crops. The aim of this study was to investigate the changes in leaf cell $Ca^{2+}$ distribution and cell organelle ultrastructure of loquat (Eriobotrya japonica Lindl.) plants in response to cold stress at $-3^{\circ}C$, using transmission electron microscopy (TEM). Two loquat accessions, Zaozhong 6 (a commercial cultivar) and oakleaf loquat (a wild relative) were used. Cold tolerance, as measured by leaf browning rate, was higher in oakleaf plants, and calcium treatment improved cold tolerance in both species. Cold stress first induced inward transport of $Ca^{2+}$ from the intracellular space. Then, the imported $Ca^{2+}$ was aggregated around the chloroplast membrane, finally entering the chloroplast. This pattern of $Ca^{2+}$ distribution in leaf cells occurred earlier in Zaozhong 6 than in the wild loquat. With increasing time of cold exposure, the chloroplast membranes of Zaozhong 6 leaves were damaged, blurred and even disappeared, while those of wild oakleaf loquat leaves maintained their structure longer. In Zaozhong 6, cold stress induced a clear cavity between poorly structured granal thylakoids and vesicles appearing inside the chloroplast, while in oakleaf leaves cold stress had little effect on the ultrastructure of chloroplasts (although chloroplast membranes looked blurred). Loquat leaves accumulated free calcium ions around chloroplasts in response to cold stress, with earlier calcium accumulation occurring in the cold-sensitive cultivar Zaozhong 6 than in wild oakleaf loquat. These results demonstrate that these two loquat species have differences in both cold tolerance and calcium accumulation dynamics.

The regeneration and recombination of rice leaf discoloration in 146 pollen-derived lines, 70 pollen-derived lines from cold stress at $0^{\circ}C$ for 10 days, and 830 F$_2$ plants presented normal distribution curves with skewness in tolerance and no significant difference among 3 populations. Direct in vitro selection for rice cold tolerance through cold stress on primary pollen callus derived from anther culture, therefore, was revealed ineffective as a in vitro technology.
