Expression of colSR Genes Increased in the rpf Mutants of Xanthomonas oryzae pv. oryzae KACC10859

  • The rpf genes and $colS_{XOO1207}/colR_{XOO1208}$ were known to require for virulence of Xanthomonas oryzae pv. oryzae (Xoo). In Xoo KACC10331 genome, two more colS/colR genes, $colS_{XOO3534}$ (raxH)/$colR_{XOO3535}$ (raxR) and $colS_{XOO3762}/colR_{XOO3763}$ were annotated. The $colS_{XOO3534}/colR_{XOO3535}$ were known to control AvrXa21 activity and functions of $colS_{XOO3762}/colR_{XOO3763}$ were unknown in Xoo. To characterize the relationship between rpf and colS/colR genes, expression of colS/colR genes in Rpf mutants of Xoo were analyzed with quantitative reverse transcription PCR (qRT-PCR). Expressions of all three colS/colR genes increased in the rpfF mutant in which DSF synthesis is defective. Expression of $colS_{XOO1207}/col-R_{XOO1208}$, $colS_{XOO3534}/colR_{XOO3535}$ and $colS_{XOO3762}/colR_{XOO3763}$ increased 2, 2-7, 3-13 folds respectively. Expression of $colS_{XOO3534}$ and $colS_{XOO3762}$ also increased 2-4 folds in the rpfG mutant in which the signal from DSF is no longer transferred to down-stream. Expression of the other colS/colR genes was not significantly changed in the rpfG mutant compared to the wild type. Since RpfF and RpfG are responsible for DSF synthesis and signal transfer from DSF to down-stream to regulate virulence gene expression, these results suggest that the DSF and DSF-mediated signal regulate negatively three colS/colR genes in Xoo.

Effects of the combination of bone morphogenetic protein-2 and nano-hydroxyapatite on the osseointegration of dental implants

  • Objectives: This study aimed to investigate the in vitro osteoinductivity of the combination of bone morphogenetic protein-2 (BMP-2) and nanohydroxyapatite (nHAp) and the in vivo effects of implants coated with nHAp/BMP-2. Materials and Methods: To evaluate the in vitro efficacy of nHAp/BMP-2 on bone formation, bone marrow-derived mesenchymal stem cells (BM-MSCs) were seeded onto titanium disks coated with collagen (Col), Col/nHAp, or Col/nHAp/BMP-2. Protein levels were determined by a biochemical assay and reverse transcriptase-polymerase chain reaction. Stem cell differentiation was analyzed by flow cytometry. For in vivo studies with mice, Col, Col/nHAp, and Col/nHAp/BMP-2 were injected in subcutaneous pockets. Titanium implants or implants coated with Col/nHAp/BMP-2 were placed bilaterally on rabbit tibias and evaluated for 4 weeks. Results: In the in vitro study, BM-MSCs on Col/nHAp/BMP-2 showed reduced levels of CD73, CD90, and CD105 and increased levels of glycosaminoglycan, osteopontin, and alkaline phosphatase activity. After 4 weeks, the Col/nHAp/BMP-2 implant showed greater bone formation than the control (P=0.07), while no differences were observed in bone implant contact and removal torque. Conclusion: These results suggest that a combination of BMP-2 and an nHAp carrier would activate osseointegration on dental implant surfaces.

The Effect of Trigonella foenum-graceum L. (Fenugreek) Towards Collagen Type I Alpha 1 (COL1A1) and Collagen Type III Alpha 1 (COL3A1) on Postmenopausal Woman's Fibroblast

  • Trigonella foenum-graceum L. (fenugreek) is a phytoestrogen, a nonsteroidal organic chemical compound from plants which has similar mechanism of action to sex hormone estradiol-$17{\beta}$. This study aims to assess the effectivity of fenugreek seeds extract on collagen type I alpha 1 (COL1A1) and collagen type III alpha 1 (COL3A1) which are both decreased in aging skin and become worsen after menopause. This in vitro experimental study used old human dermal fibroblast from leftover tissue of blepharoplasty on a postmenopausal woman (old HDF). As a control of the fenugreek's ability to trigger collagen production, we used fibroblast from preputium (young HDF). Subsequent to fibroblast isolation and culture, toxicity test was conducted on both old and young HDF by measuring cell viability on fenugreek extract with the concentration of 5 mg/mL to $1.2{\mu}g/mL$ which will be tested on both HDF to examine COL1A1 and COL3A1 using ELISA, compared to no treatment and 5 nM estradiol. Old HDF showed a 4 times slower proliferation compared to young HDF (p<0.05). Toxicity test revealed fenugreek concentration of $0.5-2{\mu}g/mL$ was non-toxic to both old and young HDF. The most significant fenugreek concentration to increase COL1A1 and COL3A1 secretion was $2{\mu}g/mL$ (p<0.05).

  • PTC Thermistors specimens were fabricated by added $MnO_2$ as donors, and $Nb_2O_5$ as acceptors and sintered $1250^{\circ}C$/2hrs. Average grain size decreased with increased in added $MnO_2$, and increased with added in $Nb_2O_5$. But, appeared liquid phase as $Bi_2O_3$ and $TiO_2$, affect to grain growth. XRD result, peak strength waslowed then crystallization not well, but, secondary phase were not showed all specimens. All specimens resistance were so high, about $40M\Omega$ over, couldn't measured to those resistance and doesn't appear PTCR effect.

Streptococcus faecium의 급여가 육계의 성장과 장내 세균총 변화에 미치는 영향 (Effect of Dietary Streptococcus faecium on the Performances and the Changes of Intestinal Microflora of Broiler Chicks)

  • 본 실험은 육계에서 생균제 Streptococcus faecium C-68(SF)과 항생제 colistin (Col)의 단독 또는 혼합 급여가 증체율과 장내 세균총 변화에 미치는 영향을 조사하기 위하여 실시하였다. 암수 동수의 초생추 252수를 공시하여 Col과 SF를 첨가하지 않은 기초사료구(대조구), 기초사료+Col 10ppm첨가구, SF 0.04%구, SF 0.04%+col 10ppm구, SF 0.08%구, SF 0.08%+col 10ppm구 등 6개 처리구를 두었다. SF 0.04%구와 SF 0.08%구의 Streptococcus faecium 생균수는 사료 g당 각각 7$\times$10난, 1.4$\times$10개가 되도록 하였고, 사료는 옥수수와 대두학을 주원료로 하여 7주간 급여하였다. 사양실험이 진행되는 동안 처리구별로 분내 세균총의 변화를 알아보기 위하여 매주 말 신선한 분을 사료로 무균적으로 채취하여 조사하였고, 장내 세균총의 변화를 조사하기 위해 4주와 7주말에 처리당 9수씩 희생시켜 대장부분의 내용물을 채취하여 세균총의 분포를 조사하였다. 각 처리구 실험사료의 영양소 이용률을 조사하기 위하여 3주와 6주말 전분 채취법에 의하여 대사실험을 실시하였고, 7주말 각 처리구 별로 9수씩 희생시켜 소장의 길이와 무게를 측정하였다. 평균 증체량은 SF 0.08%(2.37kg)와 SF 0.08%+col (2.34kg)을 첨가한 처리구가 대조구(2.18kg)보다 유의적으로 높았으나 (P<0.05), 다른 처리구의 증체량은 대조구에 비하여 통계적인 차이가 없었다. 사료요구율은 SF 0.04%구를 제외한 모든 첨가구들이 대조구보다 더 개선되었고(P<0.05), 건물, 조단백질, 조지방, 총 탄수화물 등과 같은 영양소의 소화율은 전 기간 내내 SF나 Col의 첨가에 의한 변화를 볼 수 없었다. 예상했던 대로 SF첨가구의 분 중 Streptococci는 유의성 있게 증가되었는데 SF 0.04%구와 SF 0.08%구, SF 0.08%+col구에서는 통계적인 유의성이 인정되었다(P<0.05). 그러나 colistin의 단독첨가에 의한 Streptococci의 변화는 볼 수 없었고 Col과 SF와의 혼합 첨가구에서도 Col첨가가 Streptococci수의 변화에 미치는 영향은 볼 수 없었다. 분 중의 coliforms수는 SF와 Col의 첨가된 사료의 급여에 의해 현저히 감소되었다(P<0.05) 그러나 SF와 Col의 혼용효과는 관찰할 수 없었다. 장내 세균총의 분포 변화도 분중의 세인총 변화와 같은 양식을 나타냈다. 소장의 길이는 SF 0.08%구와 SF 0.08%+col 10ppm구에서 SF를 급여하지 않은 처리구보다 10%정도 유의하게 긴 것으로 나타났다(P<0.05). 그러나 소장의 무게(empty weight)는 대조구에 비하여 SF 0.08%구와 SF 0.08%+col구에서 가벼워졌고, Col과 SF를 단독 또는 혼용 급여한 모든 처리구의 단위길이 당 소장무게는 유의성 있게 감소된 것을 볼 수 있었다(P<0.05). 전체적인 결과를 볼 때 SF가 0.08%첨가된 구에서 육계의 증체량 개선효과가 가장 좋은 것으로 나타났다. 10ppm의 colistin첨가는 SF와 혼용했을 때나 10ppm 그 자체만으로는 육계의 증체효과 또는 양내 세란총 변화의 관점에서 볼 때 유익하지 못했고, SF와 Col은 소장의 벽 두께를 않게 변화시키는 효능이 있다고 추측된다.

Cytotoxic Activities of Indigenous Plant Extracts in Cultured Human Cancer Cells

  • In continuous efforts for discovery of novel potent antitumor agents from natural products, fifty-seven methanolic extracts derived from indigenous Korean plants were primarily evaluated for in vitro cytotoxic activity in cultured human lung (A549) and colon (Col2) cancer cells. As a result, 16 plant extracts were found to be active against A549 cells and 15 extracts were active against Col2 cells in the criteria of $IC_{50}$<$50\;{\mu}g/ml$. In particular, the extracts of Calystegia soldanella $(IC_{50}$<$8.0\;{\mu}g/ml\;in\;A549;IC_{50}=27.4\;{\mu}g/ml\;in\;Col2)$, Heloniopsis orientalis $(IC_{50}=4.6\;{\mu}g/ml\;in\;A549; IC_{50}=4.5\;{\mu}g/ml\;in\;Col2)$, and Thuja koraiensis $(IC_{50}=1.2\;{\mu}g/ml\;in\;A549;IC_{50}=0.6\;{\mu}g/ml\;in\;Col2)$ showed a potent cytotoxic activity. Further study for the identification of active compounds from these lead extracts might be warranted.

Hydroxyapatite-collagen 나노복합재료에서의 불균질 핵생성 (Heterogeneous Nucleation of Hydroxyapatite-collagen Nanocomposite)

  • 공침법을 이용하여 수산화아파타이트[HAP]-콜라겐[COL] 나노복합재료를 제조하였다. HAP 결정과 COL 분자 사이의 화학결합형성을 확산반사법 FT-IR 및 투과전자현미경(TEM) 관찰로부터 확인하였다. 제조 시에 첨가되는 콜라겐 단백질의 농도가 높으면 미세한 아파타이트 나노결정의 콜라겐 복합체가 만들어지고 첨가되는 단백질의 농도가 낮으면 아파타이트 결정이 비교적 크게 발달하였다. FT-IR과 전자선 회절(electron diffraction)로부터 콜라겐 매체에 발달된 아파타이트 결정입자 들은 콜라겐 분자의 c 축을 따라 정렬하는 것임을 알 수 있었다. 칼슘이온 농도와 인산이온 농도를 일정하게 유지하는 수용액 계에 용해되어 있는 콜라겐 단백질의 농도는 아파타이트 결정의 발달을 위한 불균질 핵생성 위치를 제공하는 중요한 역할을 하고 있다. 콜라겐의 농도가 높으면 칼슘이온(Ca$^{2+}$)에 대한 핵생성을 위한 활성화 위치를 많이 제공하게 되며, 이는 핵생성 위치에 대한 칼슘이온 농도가 상대적으로 낮아지는 것에 대응하게 된다.

Protective Effects of Standardized Siegesbeckia glabrescens Extract and Its Active Compound Kirenol against UVB-Induced Photoaging through Inhibition of MAPK/NF-κB Pathways

  • Anti-photoaging effects of standardized Siegesbeckia glabrescens extract (SGE) and its major active compound kirenol were investigated using Hs68 human dermal fibroblasts and hairless mice, respectively. UVB-irradiated hairless mice that received oral SGE (600 mg/kg/day) showed reduced wrinkle formation and skinfold thickness compared with the UVB-irradiated control. Furthermore, SGE treatment increased the mRNA levels of collagen synthesis genes (COL1A1, COL3A1, COL4A1, and COL7A1) and activated antioxidant enzyme (catalase), while suppressing matrix metalloproteinase (MMP-2, -3, -9, and -13) expression. In Hs68 fibroblasts, kirenol also significantly suppressed MMP expression while increasing the expression of COL1A1, COL3A1, and COL7A1. Collectively, our data demonstrate that both SGE and kirenol attenuated UVB-induced photoaging in hairless mice and fibroblasts through inhibition of the mitogen-activated protein kinases and nuclear factor kappa B pathways, suggesting that SGE has potential to serve as a natural anti-photoaging nutraceutical.

RhoBTB3 Regulates Proliferation and Invasion of Breast Cancer Cells via Col1a1

  • Breast cancer is the leading cause of cancer-related death in women worldwide, despite medical and technological advancements. The RhoBTB family consists of three isoforms: RhoBTB1, RhoBTB2, and RhoBTB3. RhoBTB1 and RhoBTB2 have been proposed as tumor suppressors in breast cancer. However, the roles of RhoBTB3 proteins are unknown in breast cancer. Bioinformatics analysis, including Oncomine, cBioportal, was used to evaluate the potential functions and prognostic values of RhoBTB3 and Col1a1 in breast cancer. qRT-PCR analysis and immunoblotting assay were performed to investigate relevant expression. Functional experiments including proliferation assay, invasion assay, and flow cytometry assay were conducted to determine the role of RhoBTB3 and Col1a1 in breast cancer cells. RhoBTB3 mRNA levels were significantly up-regulated in breast cancer tissues as compared to in adjacent normal tissues. Moreover, RhoBTB3 expression was found to be associated with Col1a1 expression. Decreasing RhoBTB3 expression may lead to decreases in the proliferative and invasive properties of breast cancer cells. Further, Col1a1 knockdown in breast cancer cells limited the proliferative and invasive ability of cancer cells. Knockdown of RhoBTB3 may exert inhibit the proliferation, migration, and metastasis of breast cancer cells by repressing the expression of Col1a1, providing a novel therapeutic strategy for treating breast cancer.

INVOLVEMENT OF CYP2C9 ON CHLORPROPAMIDE 2-HYDROXYLATION IN HUMAN: IN VITRO AND IN VIVO EVIDENCE.

  • No report has been addressed to the CYP isoforms catalyzing chlorpropamide, a structural analogue of tolbutamide. To evaluate enzyme(s) mediating formation of 2-hydroxycWorpropamide, a major metabolite and identified by LC/Mass and NMR, incubation studies using human liver microsomes and cDNA expressed CYP were performed on the presence or absence of selective inhibitors of each CYP isoform.
