Genetic and Environmental Effects on the Lactation Yield and Milk Compositions in Holstein Cow (Holstein종 유우(乳牛)의 비유량(泌乳量) 및 유조성분(乳組成分)에 미치는 유전(遺傳) 및 환경(環境)의 효과(效果))

  • This study was conducted to estimate the genetic and environmental effects on lactation yield and milk compositions in Holstein cows. The data analysis were the records of 159 cows rearing at Nation Animal Breeding Institute from 1990 to 1991. The least square means were estimated on milk and fat yield in lactation yield, and the percent of fat, protein, solids-not-fat and total solid in milk composition. The results obtained are summarized as follows: 1. The average yield of milk and fat in 305 days were $7,26.56{\pm}3,57.24$ and $254.65{\pm}44.94Kg$ and the percent of fat, protein, solids-not-fat and total solid were $3.69{\pm}0.43$, $3.32{\pm}0.41$, $9.15{\pm}0.49$ and $12.75{\pm}0.96$ and the coefficients of variation were 18.68, 17.64, 11.88 and 12.34% for milk yield, fat yield, fat percent and protein percent, respectively. 2. The effect of sires was highly significant at 1% level in milk and fat yield and fat percent, and significant at 5% level in protein and total solid percent. Among the sires, B, L and O sire were superior in milk yield with 7,571.22, 7,499.11 and 7,420.58 Kg, and A, F and K sire were superior in protein percent with 3.75, 3.64 and 3.65, respectively. 3. The effect of parity was highly significant at 1% level in milk yield, and significant at 5% level in fat yield. Among the parities, the 3rd parity was superior in milk and fat yield with 7,634.54 and 274.98 Kg, and the 4th and over was superior in fat and protein percent with 3.90 and 3.50, respectively. 4. The effect of calving seasons was highly significant at 1% level in milk yield, and significant at 5% level in fat yield, and the percent of fat, protein and total solid. Among the calving seasons, spring and winter were superior in milk yield with 7,310.31 and 7,364.57 Kg, also spring and winter were superior in protein percent with 3.68 and 3.52, respectively.

A STUDY OF DENTAL CROWDING AND ITS RELATIONSHIP TO MANDIBULAR INCISOR SHAPE BY MODEL ANALYSIS IN ADOLESCENTS (청소년 석고 모형 분석에 의한 하악절치 형태와 치아밀집의 상관관계에 관한 연구)

  • Mandibular incisor crowding is one of the most common features of malocclusion and is interesting characteristic in view of relapse and stability after orthodontic treatment. There are many potential factors in the etiology of lower anterior crowding. The tooth size variation is one of them, but biologic significance for the faciolingual width of the teeth has been overlooked. Peck and Peck reported that persons with ideal mandibular incisor alignment were shown to have incisor with smaller mesiodistal and larger faciolingual dimensions than persons with incisor crowding. On the basis of these findings they suggested MD/FL index as a clinical guideline for the assessment for lower incisor crowding. The present study was undertaken to examine the relationship between mandibular incisor crowding and mandibular incisor dimension, and determine their correlation with arch length discrepancy. 154 dental casts of people from 11 to 17 years of age were made, and were divided into normal group with irregularity index less than of 1, and crowding group with irregularity index greater than 1.The casts were measured and analyzed statistically. The results were as follows. 1. The mean mesiodistal width for mandibular incisor was larger in crowding group, and has significant difference in central inciosr measurement. There are no significant differences in the faciolingul width and MD/FL index. 2. Irregularity index has significant correlation coefficients with mesiodistal width and MD/FL index for mandibular incisor in crowding group, but no correlation with faciolingual width. It also has correlation with maxillary and mandibular arch length discrepancy, total tooth material, mandibular intercanine width, and mandibular inter first premolar width. 3. Upper and lower arch length discrepancy have significant correlation with mesiodistal width of mandibular incisor and overbite, but have no correlation with faciolingual width. Lower arch lenth discrepancy has significant correlation with MD/FL index for mandibular incisor and upper arch length discrepancy has correlation with MD/FL index for mandibular lateral incisor. 4. Significant differences were observed between normal and crowding group for the mandibular arch length discrepancy and overbite.

Spectrophotometric Determination of Soil Chemical Properties Using Soiltek® KA-P Spectrophotometer (Soiltek KA-P 분광광도계률 사용한 토양 화학적 성질의 분광학적 분석)

  • To enable rapid and convenient soil test, new soil analytical methods, which require only one instrument, UV/Vis spectrophotometer, were developed and named "Soiltek KA-P spectrophotometric methods". The Soiltek$^{(R)}$ KA-P spectrophotometric method was compared with standard method of RDA in analytical capability for soil chemical properties. Using the 78 soils collected from upland, paddy, orchard, and vinyl house soils, soil organic matter, exchangeable K, Ca, and Mg. CEC, available $SiO_2$, and nitrate were analyzed by the two methods. The color stability(ratio of the absorbance at elapsed time t to the absorbance at time t=0) of organic matter. Ca, Mg, and available $SiO_2$ decreased to about 2% within one hour. However, that of exchangeable K, CEC, and nitrate remained constant. The results obtained with Soiltek$^{(R)}$ KA-P spectrophotometric method showed highly significant correlation with those measured by the standard method of RDA($R^2$ >0.9501), in which the slopes were near unity of $1.0{\pm}0.05$. The standard deviation values of organic matter, exchangeable K, Ca, and Mg, CEC, available $SiO_2$, and nitrate were apparently lower than ${\pm}1.8gkg^{-1}$, ${\pm}0.05cmol^+kg^{-1}$, ${\pm}0.18cmol^+kg^{-1}$, and ${\pm}0.13cmol^+kg^{-1}$, ${\pm}1.0cmol^+kg^{-1}$, ${\pm}5.0mgkg^{-1}$, and ${\pm}10.0mgkg^{-1}$, respectively. All the measurements showed coefficients of variation of less than 7~17% and were within the confidence level of 95%, which means both the methods are precise. Considering the relative simplicity, low cost, precision and accuracy, the proposed Soiltek$^{(R)}$ KA-P spectrophotometric methods could be recommended as an alternative to standard method.

  • Fenoxycarb, pyriproxyfen and methoprene are juvenile hormone mimic insecticide. These insecticides have been widely used for mosquito, fly, scale insects, and Lepidoptera. The purpose of this study was to develop a simultaneous determination procedure of fenoxycarb, pyriproxyfen and methoprene residues in crops using HPLC-UVD/MS. These insecticide residues were extracted with acetone from representative samples of four raw products which comprised brown rice, apple, green pepper, and Chinese cabbage. The extract was diluted with saline water, and then n-hexane/dichloromethane partition was followed to recover these insecticides from the aqueous phase. Florisil column chromatography was additionally employed for final clean up of the extract. The analytes were quantitated by HPLC-UVD/MS, using a $C_{18}$ column. The crops were fortified with each insecticide at 3 levels per crop. Mean recovery ratios were ranged from 80.0 to 104.3% in four representative agricultural commodities. The coefficients of variation were less than 4.8%. Quantitative limit of fenoxycarb, pyriproxyfen, and methoprene was 0.04 mg/kg in crop samples. A HPLC-UVD/MS with selected-ion monitoring was also provided to confirm the suspected residues. The proposed simultaneous analysis method was reproducible and sensitive enough to determine the residues of fenoxycarb, pyriproxyfen and methoprene in the agricultural commodities.

Comparison of Sampling Methods for On-Farm Use Quick Test Procedure of Soil Nitrate (토양의 질산태질소 현장검정을 위한 시료 채취방법 비교)

  • The procedure of soil sampling for on-farm quick test of soil nitrate is very important to improve practical application without weighing or drying soil. To improve application of test strip reflectometer as a quick on-farm analytical procedure for the estimation of soil nitrate concentration, three sampling methods such as gravimetric sampling (GS), particle density sampling (PDS) and bulk density sampling (BDS) for on-farm analytical procedure were investigated with twelve soils of 45 to $281mg\;kg^{-1}$ nitrate nitrogen concentration. The nitrate nitrogen concentrations measured from different soils were compared with two analytical methods, ion electrode method as a standard laboratory analysis (SLA) and test strip reflectometer at three moisture conditions, viz. air dried soil, 20 and 40% of maximum water holding capacity (MWHC). Nitrate nitrogen concentration measured by test strip reflectometer was significantly correlated with that of SLA, and the coefficients of variation (CV) were in the range of 3.5 to 10.9%. These CV values less than 10.9% were thought to be acceptable for the measurement of soil nitrate as an on-farm real time analytical procedure. The nitrate nitrogen concentration by BDS for test strip reflectometer as well as ion electrode method was more similar to that of SLA compared with those by GS and PDS especially in case of moist soils. This result suggests that the BDS is more useful than GS and PDS in case of on-farm analytical procedure of soil nitrate for moist soils. Further the practical measurement by BDS could be improved by substituting the bottle cap with a larger container.

  • This investigation was carried out to define the cultivation state of Ginseng seedling and to study the growth of important characters by location and correlations between characters, the results obtained are summarized as follows; 1. Most of farmers culitvating ginseng seedling had not any special consideration for the improvement of soil, and they applied green manure only as basic fertilizer but they did not apply chemical fertilizer. 2. Any efforts on the improvement of ginseng seed were not found. Seed was harvested from 3 years old ginseng plant. Seedling density was 1.8 cm by 2 cm. 3. Growth of all characters showed significant differences among farms. Coefficients of variation among individual seedling plants was largest at root diameter and smallest at root length. 4. Gromth of all characters without root diameter by planted location under the shadow was poor at front row, however root diameter was shortest at back row. 5. Significant high correlatons were rocognized between root diameter and stem diameter, between leaf blade and stem diameter and between leaf blade and leaf length, but stem length were not correlated with any characters. It is noticable that negative correlations were found between stem length and root diameter, between stem length and root length and between stem length and stem diameter.

Estimation of forest Site Productivity by Regional Environment and Forest Soil Factors (권역별 입지$\cdot$토양 환경 요인에 의한 임지생산력 추정)

  • This study was conducted to develop regional site index equations for main tree species in Gangwon, Gyunggi-Chungcheong, Gyungsang, and Jeolla area of Korea, using environmental and soil factors obtained from a digital forest site map. Using the large data set obtained from the digital forest map, a total of 28 environmental and soil factors were regressed on site index by tree species for developing the best site index equations for each of the regions. The selected main tree species were Larix 1eptolepis, Pinus koraiensis, Pinus densiflora, Pinus thunbergii, and Quercus acutissima. Finally, four to five environmental and soil factors by species were chosen as independent variables in defining the best regional site index equations with the highest coefficients of determination $(R^2)$. For those site index equations, three evaluation statistics such as mean difference, standard deviation of difference and standard error of difference were applied to the data sets independently collected from fields within the region. According to the evaluation statistics, it was found that the regional site index equations by species developed in this study conformed well to the independent data set, having relatively low bias and variation. It was concluded that the regional site index equations by species had sufficient capability for the estimation of site productivity.

Determination of Fomesafen Residue in Agricultural Commodities Using HPLC-UVD/MS (HPLC-UVD/MS를 이용한 농산물 중 fomesafen의 분석)

  • Fomesafen is a selective herbicide, and used to control annual and perennial broad-leaf grass on soybean and fruit fields in USA and China, but not introduced in Korea yet. So, MRL (Maximum Residue Level), and analytical method of fomesafen were not establishment in Korea. Therefore, this experiment was conducted to establish a determination method for fomesafen residue in crops using HPLC-UVD/MS. Fomesafen residue was extracted with acetone from representative samples of five raw products which comprised hulled rice, soybean, apple, green pepper, and Chinese cabbage. The extract was diluted with saline water, and dichloromethane partition was followed to recover fomesafen from the aqueous phase. Florisil column chromatography was additionally employed for final clean up of the extract. The fomesafen was quantitated by HPLC with UVD, using a Shiseido CAPCELL-PAK UG C18 column. The crops were fortified with fomesafen at 3 levels per crop. Mean recovery ratio were ranged from 87.5% for a 0.4 ppm in hulled rice to 102.5% for a 0.4 ppm in apple. The coefficients of variation were ranged from 0.6% for a 2.0 ppm in hulled rice to 7.7% for a 0.04 ppm in green pepper. Quantitative limit of fomesafen was 0.04 mg/kg in representative 5 crop samples. A LC/MS with selected-ion monitoring was also provided to confirm the suspected residue. Therefore, this analytical method was reproducible and sensitive enough to determine the residue of fomesafen in agricultural commodities.

  • The solubility variations of Abies koreana Wilson wood and their correlations ammong the factors which effect the extraction were investigated. In hot water solubility for 10 hours, extracted contents were 2.33% in heart wood, 12.30% in sapwood. In base solubility (here, used NaOH), the solubility content was 8.23% only for 1 hour. In organo-soluble fractions, there was not any variation between the content 4.00% for 1 hour and 4.44% for 10 hours, about the temperature effect, in the neutral solvent, at temperature $25{\pm}3^{\circ}C$, $50{\pm}3^{\circ}C$ and $97{\pm}3^{\circ}C$ and the solubility contents were 2.73%, 3.29%, 7.32% respectably. In the pH variations, initinal pH of solution, 6.5 became as low 5.4 after 10 hours extraction. Generally, the correlation coefficients between solubility and hour, solubility and temperature, solubilities' pH and hours, solubility and part, were r=0.890, r=0.986, r=-0.955, r=0.840 respectably. It is suggested that the most serious factor of the extraction in this material is the temperature.

Studies on Measuring Volatile Terpenoids in Carrots Using the Direct Headspace Sampling Method (Direct Headspace Sampling 방법을 이용한 당근의 휘발성 Terpenoids 분석에 관한 연구)

  • Volatile terpenoids of carrots were measured by the direct headspace sampling method(DHS) with gas chromatography as a study for the breeding of high quality carrots. Using this method, 7 terpenoids such as ${\alpha}-pinene$, ${\beta}-pinene$, ${\beta}-myrcene$, ${\alpha}-terpinene$, limonene, ${\gamma}-terpinene$, and terpinolene were clearly separated. However ${\alpha}-phellandrene$ was not clearly separated from ${\beta}-myrcene$. In addition to this, higher boiling point compounds such as terpinen-4-ol, bornyl acetate, and ${\alpha}-bisbolol$ were not found. The coefficients of $determination(r^2)$ for the 7 terpenoids were higher than 0.99 and the standard curves were highly significant. Four replicated samples using this method demonstrated great reproducibility; the coefficidnt of variation (C.V.) for ${\alpha}-pinene$, ${\beta}-pinene$, ${\beta}-myrcene$, limonene, ${\gamma}-terpinene$, terpinolene, and total terpenoids were 6.8, 6.8, 8.4, 7.1, 3.8, 10.1, 7.1%. Sixty five carrot cultivars breeded worldwide were evaluated for the 7 terpenoids and total terpenoids; the range for ${\alpha}-pinene$, ${\beta}-pinene$, ${\beta}-myrcene$, ${\alpha}-terpinene$, limonene, ${\gamma}-terpinene$, terpinolene, and total terpenoids were $0.28{\sim}2.48\;ppm$, $0.35{\sim}1.87\;ppm$, $0.56{\sim}1.51\;ppm$, 0 ppm, $0.59{\sim}1.84\;ppm$, $0.87{\sim}3.33\;ppm$, $5.15{\sim}35.81\;ppm$, and $9.07{\sim}42.30\;ppm$, respectively. Big differences in each terpenoid and total terpenoids were found among cultivars. On the total terpenoids of the 65 cultivars, 5 cultivars(7.7%) contained less than 10 ppm, 15 cultivars (23.1%) $10{\sim}11.99\;ppm$, 14 cultivars(21.5%) $12{\sim}13.99\;ppm$, 9 cultivars(13.8%) $14{\sim}15.99\;ppm$, 10 cultivars(15.4%) $16{\sim}17.99\;ppm$, 4 cultivars(6.2%) $18{\sim}19.99\;ppm$, 5 cultivars(7.7%) $20{\sim}29.99\;ppm$, and 3 cultivars(4.6%) were higher than 30 ppm. Generally, cultivars developed in Japan contained less total terpenoids than cultivars developed in Europe and America.
