Growth Characters and Sugar Content During Grain Filling in New Hybrid, Chalok 1/Cooktail 51 Corn

  • 'Chalok 1/Cooktail 51' corns, supersweet corn gene controlled by either brittle-l (bt l) or shrunken-2(sh2) gene introduced into waxy corn, showed agronomic characteristics between supersweet corn and waxy corn. The ears were harvested at different development stages from 15 to 35 days after silking (DAS). Ear diameter of Cocktail 51 and 'Cocktail 51'/Chalok 1 increased from 15 DAS to 30 DAS and little increased thereafter, but that of Chalok 1/Cocktail 51 and 'Chalok l' increased until 35 DAS. Diameter of ear extension increased more in Cocktail 51 and Chalok 1/Cocktail 51 corn than Chalok 1. Ear fresh weight of Cocktail 51 decreased later 30 DAS but those of the other hybrids were vice versa. Rate of super-sweet kernels per ear of Chalok 1/Cocktail 51 corns was about 38 %. Development, and elongation of kernel were much more prominent in supersweet kernel than in waxy kernel, but fresh weight increased higher in waxy kernel than supersweet kernel. Moisture content in kernel decreased from 15 DAS to 35 DAS. Total sugar content of the kernel increased until 25 DAS, and that of Cocktail 51 kernel showed the highest among of them. After cooked by steam, flavor and mastication feeling rate cooked by steam of Cocktail 51 and Chalok 1/Cocktail 51 were increased from 15 DAS to 25 DAS and markedly decreased thereafter. But those of Chalok l/Cocktail 51 and, Chalok 1 were decreased after 30 DAS. These results suggested that the optimum harvest date for fresh supersweet corn (Cocktail 51), Cocktail 51/Chalok 1 seems to be about 20 DAS and Chalok 1/Cocktail 51 and waxy com (Chalok 1) was about 25 DAS.

  • Fiber Cocktail을 혼입유무에 따른 40${\sim}$100 MPa 고강도 콘크리트 구조부재의 고온영역에서 재료적 물리적 역학적 특성을 규명한 후, 유한요소해석법(ABAQUS)을 적용한 구조요소 화재거동 시뮬레이션 해석을 수행한 결과, 다음과 같은 결론을 얻었다. 첫째, 콘크리트 Fiber Cocktail을 혼입한 실험체는 무혼입 실험체에 비해 최대수축이 제어되는 것으로 해석되었으며, 그 제어 범위는 혼입실험체 대비 약 25%${\sim}$55%로 분석되었다. 이는 고강도 콘크리트 기둥에서 Fiber Cocktail 혼입을 통해 기둥변위 수축제어가 가능한 것으로 분석되었다. 둘째, 본 연구는 고강도 콘크리트의 내부 공극압에 의한 폭렬 특성이 반영되지 않은 화재 거동 해석으로서 향 후 콘크리트 내부 공극압에 대한 특성 반영, 다양한 고강도 콘크리트 및 강재의 강도별, 내부온도별 재료 특성 DB가 구축된다면 성능기반 내화구조 설계를 위한 기술적 기반이 이루어질 것으로 판단된다.

Supplementation of enzyme cocktail in chickens diet is an effective approach to increase the utilization of nutrient in wheat-based diets

  • This experiment was conducted to evaluate the effect of supplementing enzyme cocktail on growth performance, digestibility of nutrients, and monosaccharide concentration in ileum and ceca of broiler chickens fed wheat-based diets. A total of 600 male broilers (42.26 ± 1.76 g, 0 day old) were used for 35 days of feeding trial consisting of 2 phases (starter phase from d 0 to 21 and finisher phase from d 21 to 35). Four dietary treatments were prepared based on wheat diets containing four levels of enzyme cocktail supplementation at 0, 0.2, 0.3, and 20 g/kg. Overall, dietary enzyme cocktail supplementation decreased feed conversion ratio (linear p = 0.007; quadratic p = 0.013) and improved (linear p < 0.05) the apparent ileal digestibility of dry matter (DM), crude protein, and soluble and insoluble non-starch polysaccharides. The apparent total tract digestibility of DM and gross energy were increased (linear p < 0.01) with increasing supplementation levels of the dietary enzyme cocktail. The concentrations of arabinose, xylose, mannose, and glucose in ileal digesta were linearly increased (p < 0.01) with increasing enzyme cocktail supplementation levels. In addition, the quadratic effect was observed (quadratic p = 0.046) in mannose concentration of ileal digesta. The concentration of arabinose, xylose, mannose, and galactose in cecal digesta was increased (linear p < 0.05) with increasing dietary enzyme cocktail supplementation levels. The supplementation of enzyme cocktail efficiently increased the utilization of nutrients in broiler and there was no adverse effects of high dosage supplementation level.

  • 고강도 콘크리트는 폭렬 발생을 제어하기 위하여 PP섬유와 나일론 섬유 등을 혼입하며, fiber cocktail로 PP섬유와 강섬유를 혼입한 형태도 많이 연구되어지고 있다. 본 연구에서는 Fiber-cocktail 내의 폴리프로필렌은 $1.5kg/m^3$, 강섬유는 각각 20, 30, $40kg/m^3$로 하여 시험체내의 온도를 살펴보았다. 실험결과 강섬유의 혼입이 증가하면서 콘크리트 내부 온도 및 철근의 온도가 낮아지는 것으로 나타나 강섬유는 고강도 콘크리트 화재 시 폭렬의 방지 및 온도상승 지연의 효과가 있는 것으로 나타났다.

Comparison of Gastrointestinal Permeability of Caffeine, Propranolol, Atenolol, Ofloxacin, and Quinidine Measured Using Ussing Chamber System and Caco-2 Cell Monolayer

  • The purpose of this study was to develop a cocktail approach for the measurement of the permeability of marker compounds, caffeine and propranolol (high permeability), ofloxacin (intermediate), atenolol (low), and quinidine (P-glycoprotein substrate), simultaneously. Then we compared the permeability in Caco-2 cells with that in rat intestinal segments. The difference between individual measurement and cocktail approach was less than 20 %, and the permeabilities of these compounds were similar to those previously reported, suggesting that the cocktail transport study and simultaneous drug analysis were successfully developed and validated in this study. Additionally, in the application of this cocktail method, the permeability of five drugs in rat jejunum was similar to that in ileum but different from that in colon, which was measured using the Ussing chamber system. Moreover, permeability in jejunum and ileum was similar to that in Caco-2 cells. In conclusion, the permeability in Caco-2 cells was equivalent to the permeability in rat jejunum and ileum determined with the Ussing system. Therefore, this newly developed cocktail assay and its application to the Ussing system can be a useful tool for robust and rapid screening for site-specific permeability in rat intestine, thus accelerating the prediction of absorption of new chemical entities.

Saccharomyces cerevisiae Strain Improvement Using Selection, Mutation, and Adaptation for the Resistance to Lignocellulose-Derived Fermentation Inhibitor for Ethanol Production

  • Twenty-five Saccharomyces cerevisiae strains were screened for the highest sugar tolerance, ethanol-tolerance, ethanol production, and inhibitor resistance, and S. cerevisiae KL5 was selected as the best strain. Inhibitor cocktail (100%) was composed of 75 mM formic acid, 75 mM acetic acid, 30 mM furfural, 30 mM hydroxymethyl furfural (HMF), and 2.7 mM vanillin. The cells of strain KL5 were treated with ${\gamma}$-irradiation, and among the survivals, KL5-G2 with improved inhibitor resistance and the highest ethanol yield in the presence of inhibitor cocktail was selected. The KL5-G2 strain was adapted to inhibitor cocktail by sequential transfer of cultures to a minimal YNB medium containing increasing concentrations of inhibitor cocktail. After 10 times of adaptation, most of the isolated colonies could grow in YNB with 80% inhibitor cocktail, whereas the parental KL5 strain could not grow at all. Among the various adapted strains, the best strain (KL5-G2-A9) producing the highest ethanol yield in the presence of inhibitor cocktail was selected. In a complex YP medium containing 60% inhibitor cocktail and 5% glucose, the theoretical yield and productivity (at 48 h) of KL5-G2-A9 were 81.3% and 0.304 g/l/h, respectively, whereas those of KL5 were 20.8% and 0.072 g/l/h, respectively. KL5-G2-A9 reduced the concentrations of HMF, furfural, and vanillin in the medium in much faster rates than KL5.

Bacteriophage Cocktail Comprising Fifi044 and Fifi318 for Biocontrol of Erwinia amylovora

  • Erwinia amylovora is a plant pathogen that causes fire blight on apples and pears. Bacteriophages, which are viruses that selectively infect specific species of bacteria and are harmless to animal cells, have been considered as biological control agents for the prevention of bacterial pathogens. In this study, we aimed to use bacteriophages that infect E. amylovora as biocontrol agents against fire blight. We isolated bacteriophages Fifi044 and Fifi318 infecting E. amylovora, and characterized their morphology, plaque form, and genetic diversity to use as cocktails for disease control. The stabilities of the two phages were investigated at various temperatures and pH values and under sunlight, and long-term storage experiment was conducted for a year. To evaluate whether the two phages were suitable for use in cocktail form, growth curves of E. amylovora were prepared after treating the bacterial cells with single phages and a phage cocktail. In addition, a disease control test was conducted using immature apples and in vitro cultured apple plantlets to determine the biocontrol effects of the phage cocktail. The two phages were morphologically and genetically different, and highly stable up to 50℃ and pH value from 4 to 10. The phages showed synergistic effect when used as a cocktail in the inhibition of host bacterial growth and the disease control. This study demonstrated that the potential of the phage cocktail as a biocontrol agent for commercial use.

Effects of dietary enzyme cocktail on diarrhea and immune responses of weaned pigs

  • Weaning is the most stressful event for nursery pigs because they are moved from familiar to unfamiliar environments. In addition, weaned pigs have immature digestive and immune systems. This situation makes weaned pigs susceptible to diseases and makes the absorption of nutrients from diets difficult. A feed approach, such as dietary enzyme supplementation, can be considered a solution. This study investigated the effects of dietary enzyme cocktail on diarrhea and immune responses of weaned pigs. A total 36 weaned pigs ($5.92{\pm}0.48kg\;BW$; 28 d old) were randomly allotted to 2 dietary treatments (3 pigs/pen, 6 replicates/treatment) in a randomized complete block design. The dietary treatments were a typical diet based on corn and soybean meal (CON) and CON with 0.05% enzyme cocktail (Cocktail; combination of xylanase, ${\alpha}-amylase$, protease, ${\beta}-glucanase$, and pectinase). Pigs were fed their respective diets for 6 wk. Incidence of diarrhea, packed cell volume (PCV), white blood cells (WBC) count, and immunoglobulin content were measured. A significantly lower incidence of diarrhea (p < 0.05) was observed in the Cocktail group as compared with the CON group. The Cocktail group also showed a decreased PCV (p < 0.1) on d 3 after weaning than the CON group. However, no differences were observed for number of WBC and contents of immunoglobulin G, M, and A between the Cocktail and CON groups. Consequently, inclusion of an enzyme cocktail in diets for weaned pigs had a positive influence on gut health by reducing the incidence of diarrhea in the present study.

  • Besides recent progresses in the physics-based modelling of fission gas and helium behaviour, the scarcity of experimental data concerning their combined behaviour (i.e., cocktail) hinders further model developments. For this reason, in this work, we propose a modelling methodology aimed at providing recommendations for accelerated experimental investigations. By exploring a wide range of annealing temperatures and cocktail compositions with a physics-based modelling approach we identify the most interesting conditions to be targeted by future experiments. To corroborate the recommendations arising from the proposed methodology, we include a sensitivity analysis quantifying the impact of the model parameters on fission gas and helium release, in conditions representative of high and low burnup.

Effects of dietary enzyme cocktail on growth performance, intestinal morphology, and nutrient digestibility of weaned pigs

  • Soybean, one of most widely used swine feed component in the world, contains non-starch polysaccharides (NSP). The digestive system of weaned pigs is not yet fully developed, and thus weaned pigs cannot easily digest diets based on corn and soybean meal. Dietary exogenous enzymes supplementation has been intensively investigated to assist digestion of anti-nutritional factors, such as NSP. This experiment was conducted to investigate the effects of dietary enzyme cocktail on growth performance, intestinal morphology, and nutrient digestibility of weaned pigs. A total 36 weaned pigs ($5.92{\pm}0.48kg\;BW$; 28 d old) were randomly allotted to 2 dietary treatments (3 pigs/pen, 6 replicates/treatment) in a randomized complete block design. The dietary treatments were a typical diet based on corn and soybean meal (CON) and CON with 0.05% enzyme cocktail (Cocktail; mixture of xylanase, ${\alpha}-amylase$, protease, ${\beta}-glucanase$, and pectinase). Pigs were fed their respective diets for 6 wk. Growth performance, morphology of ileum, apparent ileal digestibility (AID) and apparent total tract digestibility (ATTD) of dry matter, crude protein, and energy of weaned pigs were measured. No significant differences (p > 0.05) were observed for growth performance for the duration of the experimental period, and morphology of ileum, and nutrient digestibility between CON and Cocktail treatment groups. Therefore, the results from the current study indicated that enzyme cocktail supplementation in diets had no influence on growth performance, intestinal morphology, and nutrient digestibility of weaned pigs.
