Glycine max Fermented by a Novel Probiotic, Bifidobacterium animalis subsp. lactis LDTM 8102, Increases Immuno-Modulatory Function

  • Many probiotic species have been used as a fermentation starter for manufacturing functional food materials. We have isolated Bifidobacterium animalis subsp. lactis LDTM 8102 from the feces of infants as a novel strain for fermentation. While Glycine max has been known to display various bioactivities including anti-oxidant, anti-skin aging, and anti-cancer effects, the immune-modulatory effect of Glycine max has not been reported. In the current study, we have discovered that the extract of Glycine max fermented with B. animalis subsp. lactis LDTM 8102 (GFB 8102), could exert immuno-modulatory properties. GFB 8102 treatment increased the production of immune-stimulatory cytokines in RAW264.7 macrophages without any noticeable cytotoxicity. Analysis of the molecular mechanism revealed that GFB 8102 could upregulate MAPK2K and MAPK signaling pathways including ERK, p38, and JNK. GFB 8102 also increased the proliferation rate of splenocytes isolated from mice. In an animal study, administration of GFB 8102 partially recovered cyclophosphamide-mediated reduction in thymus and spleen weight. Moreover, splenocytes from the GFB 8102-treated group exhibited increased TNF-α, IL-6, and IL-1β production. Based on these findings, GFB 8102 could be a promising functional food material for enhancing immune function.

A Study on Anti-inflammatory and Skin Soothing Cosmetics Material of COMPAREX (COMPAREX 의 항염 및 진정 화장품 소재로서의 연구)

  • Recently, there has been a growing interest in the development of safer and more effective soothing materials to calm skin that has become sensitive to various external factors. The aim of this study was to investigate the skin soothing and anti-inflammatory effects of COMPAREX compared to Portulaca Oleracea extract. The results showed that COMPAREX significantly inhibited LPS-induced NO production and the expression of inflammatory factors iNOS, COX-2, TNF-α, and IL-6 more than P. oleracea extract. In addition, COMPAREX has been confirmed to have a more effective sedative effect by further inhibiting the gene and protein expression of IL-1α against SDS stimulation than the Portulaca Oleracea extract. Furthermore, COMPAREX inhibited the expression of inflammatory factors COX-2 and IL-8 increased by PM2.5 and suppressed H2O2-mediated carbonylated protein in hair cell. These results suggest that COMPAREX has shown the potential to be used as an improved natural soothing material over P. oleracea extract, and it is expected to be used as a derma cosmetic material in the future.

Production of Second Generational Cloning Embryos with Activated Oocytes in Rabbits (토끼에서 수핵란의 세포질 활성화에 의한 제 2세대 복제수정란의 생산)

  • Large scale production of cloned embryos requires the technology of multiple generational nuclear transfer(NT) by using NT embryos itself as the subsequent donor nuclei. In this work we investigated comparatively the effects of enucleated oocytes treated with ionomycin and 6-DMAP on the electrofusion rate and in vitro developmental potential in the first and second NT embryos. The embryos of 16-cell stage were collected from the mated does by flushing oviducts with Dulbecco's phosphate buffered saline(D-PBS) containing 10% fetal calf serum(FCS) at 47 hours after hCG injection. The recipient cytoplasms were obtained by removing the nucleus and the first polar body from the oocytes collected at 15 hours after hCG injection. The enucleated oocytes were pre-activated by 5 min incubation in 5$\mu$M ionomycin and 2 hours incubation in 2 mM 6-DMAP at 19~20 hours post-hCG before microinjection. In the first and second generation NT, the unsynchronized 16-cell stage embryos were used as nuclear donor. The separated donor blastomeres were injected into the enucleated activated recipient oocytes by micromanipulation and were electrofused by electrical stimulation of single pulse for 60 $\mu$sec at 1.25kV/cm in $Ca^2$+, $Mg^2$+ - free 0.28 M mannitol solution. In the non-preactivation group, the electrofusion and electrical stimulation was given 3 pulses for 60 $\mu$sec at 1.25 kV/cm in 100$\mu$M $Ca^2$+, $Mg^2$+ 0.28 M mannitol solution. The fused oocytes were co-cultured with a monolayer of rabbit oviductal epithelial cells in TCM-199 solution containing 10% FCS for 120 hours at 39$^{\circ}C$ in a 5% $CO_2$ incubator. The results obtained were summarized as follows: 1. In the first generational NT embryos, the electrofusion rate of preactivated and non-activated oocytes(80.4 and 87.8%) was not significantly different, but in the second generational NT embryos, the electrofusion rate was significantly(P<0.05) higher in the non-activated oocytes(85.7%) than in the preactivated oocytes(70.1%). 2) In the first and second generational NT embryos, the developmental potential to biastocyst stage was significantly(P<0.05) higher in the preactivated oocytes(39.3 and35.7%) than in the non-preactivated oocytes(16.0 and 13.3%). No significant difference in the developmental potential was shown between the first and second generational NT embryos derived from the preactivated oocytes. In conclusion, it may be efficient to use the oocytes preactivated with ionomycin and 6-DMAP for the multiple production of cloned embryos by recycling nuclear transfer.

Development of Peptides from the Germinated Black Rice and Applications as Cosmetics Ingredients (발아 흑미 유래 펩타이드의 개발과 화장품 응용에 대한 연구)

  • To develop novel anti-aging peptides from the germinated black rice, we treated with bromelain, papain and Pronase E. And we investigated the effects of the germinated black rice peptide (GBRP) as anti-aging cosmetic ingredients, and compared with the non-germinated black rice protein (NBRP). We investigated the effects on in vitro inhibition of matrix-metalloprotease (MMP), proliferation of human skin fibroblasts, stimulation of collagen synthesis and expression of UVA-induced MMPs in human skin fibroblasts, UVA induced MMP-1 expression and collagen contents in human skin fibroblasts were analyzed by enzyme-linked immunosorbent assay (ELISA). As a result, the molecular weight distributions of GBRP and NBRP were determined by gel permeation chromatography to be approximately 900 and 10,000 daltons. GBRP increased skin cell proliferation about 40% and reduced UVA-induced MMP-1 expression about 50%. Also the collagen protein level of cells, which were cultured with GBRP, was increased about 25%. These results suggest that the geminated plant seed peptides can be novel anti-aging ingredients for cosmetics.

Experimental Study on Fracture Pressure, Permeability Enhancement and Fracture Propagation using Different Fracture Fluids (다양한 파쇄 유체별 파쇄압력, 투과도 증진 및 균열전파에 관한 실험적 연구)

  • The hydraulic fracturing developed to improve permeability of tight reservoir is one of key stimulation technologies for developing unconventional resources such as shale gas and deep geothermal energy. The experimental study was conducted to improve disadvantage of hydraulic fracturing which has simple fracture pattern and poor fracturing efficiency. The fracturing experiments was conducted for tight rock using various fracturing fluids, water, N2, and CO2 and the created fracture pattern and fracturing efficiency was analyzed depending on fracturing fluids. The borehole pressure increased rapidly and then made fractures for hydraulic fracturing with constant injection rate, however, gas fracturing shows slowly increased pressure and less fracture pressure. The 3D tomography technic was used to generate images of induced fracture using hydraulic and gas fracturing. The stimulated reservoir volume (SRV) was estimated increment of 5.71% (water), 12.72% (N2), and 43.82% (CO2) respectively compared to initial pore volume. In addition, permeability measurement was carried out before and after fracturing experiments and the enhanced permeability by gas fracturing showed higher than hydraulic fracturing. The fracture conductivity was measured by increasing confining stress to consider newly creating fracture and closing induced fracture right after fracturing. When the confining stress was increased from 2MPa to 10MPa, the initial permeability was decreased by 89% (N2) and 50% (CO2) respectively. This study shows that the gas fracturing makes more permeability enhancement and less reduction of induced fracture conductivity than hydraulic fracturing.

Effects of Natural Herbal Extracts on Electrical Brain Activity and Autonomic Nervous System (천연 약초 추출 향이 뇌파 활성과 자율신경계에 미치는 영향)

  • In the literature, inhalation of cosmetic fragrances, including natural extracts or natural essential oils, has been reported to induce brainwave changes. Angelica gigas (AG) and Cnidium officinale (CO) are widely used in East Asian countries as traditional medicines to improve health, but there have been no previous reports of inhaling their aromas and assessing changes in brain activity through electroencephalogram (EEG) recordings. In this study, the scent of natural herbal extracts such as Artemisia princeps var. orientalis (Compositae), AG, and CO was inhaled and evaluated whether emotional state changes were caused by olfactory stimuli, and brain activity was confirmed through EEG records before and after inhaling the scent of natural herbal extracts in subjects aged 20 to 30. We also used an electrocardiogram (ECG) to record the electrical activity of the heart during fragrance administration. The test results indicated that inhaling the scents of Compositae, AG, and CO extracts, as well as the positive control lavender, significantly increased the relative alpha power and the alpha/beta ratio of brain waves across the brain regions. The ECG recordings indicated that the inhalation of Compositae fragrance significantly decreased the low-frequency/high-frequency ratio, and that the inhalation of traditional herbal extract fragrances resulted in comfort and relaxation.

Anti-obesity Effects of Ethanolic Extract of Polygonatum sibiricum Rhizome in High-fat Diet-fed Mice (고지방식이로 비만이 유도된 마우스에서 황정 주정 추출물의 항비만 효과)

  • We investigated the anti-obesity effects of ethanolic extract (ID1216) of Polygonatum sibiricum rhizome and its potential underlying mechanism in an animal model. ID1216 treatment decreased body weight gain and white adipose tissue weight in the prevention study. The mRNA levels of sirtuin-1 (SIRT1), peroxisome proliferator-activated receptor ${\gamma}$ coactivator-$1{\alpha}$ ($PGC1{\alpha}$), and peroxisome proliferator-activated receptor ${\alpha}$ ($PPAR{\alpha}$) significantly increased in the epididymal white adipose tissue of ID1216-administered mice. The stimulation effects of ID1216 on these gene expressions were also observed in a cell-based assay using differentiated 3T3-L1 adipocytes. In addition, similar to orlistat, ID1216 treatment improved weight gain and reduced epididymal fat in the treatment model. These results suggest that ID1216 has potential as an anti-obesity agent by modulating the expression of genes related to thermogenesis, lipid metabolism and fatty acid oxidation.

Cell migration and Anti-inflammatory Effect of Red Ginseng Extracts Fermented with Laetiporus Sulphureus (붉은덕다리버섯 균사체로 발효한 홍삼 배양액의 cell migration 및 항염 효능에 관한 연구)

  • Red ginseng (RG) contains specific ginsenosides (Rg2, Rg3) which show various pharmacological effects and absorption rate in the body better than panax ginseng. Therefore many people have been used it for health for a long time. Furthermore, many researchers have been studying its biological activities for a long times because fermentation generates lots of beneficial small molecules good for health. In this study, we fermented red ginseng with mycelium of Leatiporus sulphures var. miniatus for 7 days. As a result, we found that three ginsenosides Rg1, Re and Rb2 were decreased from 0.24, 0.25, 0.16 mg/g to 0.12, 0.1, 0.03 mg/g respectively HPLC analysis. In addition, we studied biological activities of fermented red ginseng (FRG) about skin ageing such as anti-inflammation, cell migration, anti-oxidation, collagen type 1 synthesis, and MMP-1 inhibition activities. As a result, FRG were shown higher anti-inflammatory and cell migration promoting activities than RG. FRG inhibited production of nitric oxide (NO) and mRNA expression of inducible nitric oxide synthase (iNOS) and decreased interleukin (IL)-6 induced by LPS stimulation in RAW 264.7 cells. In conclusion, this study suggest that FRG could be a potential source as a new natural anti-inflammatory agent.

Effects of Sex Steroid Hormones on Proliferation and Differentiation of Preadipocytes from Female and Male Pigs (스테로이드 성호르몬이 암, 수 돼지 지방전구세포의 증식과 분화에 미치는 영향)

  • The current study was undertaken to determine the effects of sex steroid hormones (estrogen, testosterone and 19-nortestosterone) on proliferation and differentiation of preadipocytes of female and male pigs. The preadipocytes were isolated from the backfat of new-born female and male pigs by collagenase digestion and cultured in the $CO_2$ incubator. The concentration of $10^{-7}M$ and 10-6M sex steroid hormones were treated to the cultured preadipocytes. Regarding the effects on preadipocytes proliferation, high concentration ($10^{-6}M$) of all the three hormones increased proliferation of female preadipocytes,and only estrogen and testosterone increased proliferation of male preadipocytes. Regarding the effects on preadipocyte differentiation, all the three hormones increased differentiation of pig preadipocytes, regardless of hormone concentrations and sex of preadipocytes. The degree of stimulation of cell differentiation by sex steroid hormones was greater than that of cell proliferation.

Effects of the Heat Therapy on Changes of Immune Activities in Human Body (온열요법이 면역활성의 변화에 미치는 효과)

  • The purpose of this study was to demonstrate immune-activities by heat therapy. To exam, furthermore, the immune effect is investigated from the healthy volunteer(male:15, female:15) by monitoring changes of immune substances such as complements(C1q, C3, and C4), a comparative study with warm water immersion($40.8{\pm}0.3^{\circ}C$) and infrared(250W) was carried out. The plasma analysis showed that the percentage of C1q, C3, and C4 was decreased in warm water immersion-stimulated group, but not in infrared-applied group, compared with control group from healthy volunteer. Therefore, these results suggest that the thermo-stimulation improved immune activity and inhibited complement-related inflammation through the warm water immersion-dependent classical pathway.
