Potentiometric Characteristics of Acidic Drug Selective membrane Electrodes using Di-2-pyridyl ketone (디-2-피리딜케톤을 이용한 산성의약품 선택성 막전극의 특성)

  • Acidic drug selective electrodes based on metal(II)-di-2-pyridyl ketone-acidic drugs ternary complex as electroactive material were prepared. The metal ions, $Fe^{2+}$, $Co^{2+}$, $Ni^{2+}$, $Cu^{2+}$ were used. Nitrophenyl ether series were used as plasticizers. The electrodes exhibit a fast stable and linear response for $5{\times}10^{-5}{\sim}10^{-3}mol/L$ mefenamic acid (MA) in borate buffer solution (pH 8.9) and ibuprofen(Ib) in phosphate buffer solution (pH 7.0). The recovery test for mefenamic acid and ibuprofen using standard addition method were 99.0% and 98.4% with relative standard deviation of 2.4% and 2.6% respectively.

Inhibition of Proliferation of Human Fibroblast by δ-Aminolevulinic Acid (ALA) Derivatives through the Induction of Mitochondria Membrane Depolarization (δ-Aminolevulinic acid (ALA) 유도체들의 미토콘드리아 탈분극 유도에 의한 인간 섬유아세포의 세포분열 억제)

  • ${\delta}$-Aminolevulinic acid (ALA) is an endogenous metabolite formed in the mitochondria from succinyl-CoA and glycine, and plays a key role in the living body as an intermediate of the compound in the porphyrin biosynthesis pathway. ALA has been commonly used in photodynamic therapy for several years, because ALA is of interest as a biodegradable mediator, a growth regulator, and an effective agent used in dermatology. Here, we determined which ALA derivatives were the most effective for the inhibition of the cell proliferation and growth of human fibroblast. As a result, we found that the treatment of ALA derivatives including ALA, ALAP (ALA phosphate salt), MAL (Methyl 5-aminolevulinate hydrochloride salt), PBGL (phophobilinogen lactam) and PBGH (phophobilinogen-HCl) could attenuate cell proliferation of human fibroblast cells. Among them, PBGH was the most effective derivative. In addition, PBGH treatment could induce mitochondrial membrane depolarization, leading to cell death of human fibroblast. These results suggest that mitochondrial membrane depolarization induced by ALA and PBGH treatment might be responsible for inhibition of cell proliferation and death. Taken together, our results propose the possibility that PBGH can be used as one of the effective drugs in human skin disease, psoriasis.

Influence of Various Substrates on the Growth and Yield of Organically Grown Ginseng Seedlings in the Shaded Plastic House (상토의 물리.화학성이 시설하우스 묘삼의 생육에 미치는 영향)

  • This research was conducted to investigate the influence of variouis organic substrates on growth and yield of organically grown ginseng seedlings in a shaded plastic house. In the investigation of optimal substrate, the eight substrate were formulated by adjusting blending rate of peatmoss, perlite, coir dust(coco peat), and vermiculite. Then, the changes in physico chemical properties of root substrates as well as their influences on the growth characteristics and yield were determined at six months after sowing. The elevation of the blending rate of peatmoss from 50% to 70% with decrease in the rate of inorganic component (mixture of perlite and vermiculite) from 50 to 30% resulted in the increase in container capacities and decrease in total porosities and air-filled capacities. The concentrations of $NH_4-N$, $P_2O_5$ and K increased as the incorporation rate of castor seed meal, phosphate ore, and langbenite, respectively, were elevated during the root medium formulations. The PPV-1 and PPV-4 substrates produced high stem length, stem diameter, shoot fresh weight, leaf area and root length among eight substrate. Root fresh weight was heaviest in PPV-4 compound nursery media. The results of this experiment will be utilized in the new substrate application for ginseng organic culture in shaded vinyl house.

Spatial and Temporal Variability of Phytoplankton in Relation to Environmental Factors in Youngil Bay (영일만 수질환경과 식물플랑크톤의 시·공간적 분포)

  • We investigated the spatial and temporal variations of phytoplankton in Youngil Bay as well as the effect of water physico-chemical parameters. Water samples at three stations were collected and measured monthly from May to November in 2010. The taxa of phytoplankton observed in this study were classified as 33 Bacillariophyceae, 23 Dinophyceae, 1 Euglenophyceae, 2 Crysophyceae and 1 Cryptophyceae. The highest biomass of phytoplankton was observed at inner station in September, which was characterized high concentration of dissolved inorganic phosphate(DIP) in surface water after rainfall. Nutrient concentrations, chlorophyll-a and phytoplankton biomass values showed the marked trend to decrease from the inner bay to the outer bay. Pearson's correlation co-efficient between salinity and other water parameters including chlorophyll-a, pH and DIP showed the strong negative relationship r=-0.82, r=-0.78 and r=-0.75 (p<0.01), respectively. These results indicate that the water quality of Youngil Bay could be stimulated by nutrient enriched input from Hyeogsan River discharge, and the spatial and temporal distribution of phytoplankton biomass principally limited to DIP concentration from Hyeogsan river.

Inhibitory Effects of Boesenbergia pandurata on Age-Related Periodontal Inflammation and Alveolar Bone Loss in Fischer 344 Rats

  • Periodontitis, an infective disease caused by oral pathogens and the intrinsic aging process, results in the destruction of periodontal tissues and the loss of alveolar bone. This study investigated whether Boesenbergia pandurata extract (BPE) standardized with panduratin A exerted anti-periodontitis effects, using an aging model representative of naturally occurring periodontitis. In aged rats, the oral administration of BPE ($200mg{\cdot}kg^{-1}{\cdot}day^{-1}$) for 8 weeks significantly reduced the mRNA and protein expression of $interleukin-1{\beta}$, nuclear factor-kappa B, matrix metalloproteinase (MMP)-2, and MMP-8 in gingival tissues (p < 0.01). In alveolar bone, histological analysis with staining and micro-computed tomography revealed the attenuation of alveolar bone resorption in the BPE-treated aged group, which led to a significant reduction in the mRNA and protein expression of nuclear factor of activated T-cells c1 (NFATc1), c-Fos, tartrate-resistant acid phosphatase, and cathepsin K (p < 0.01). BPE not only increased the expression of osteoblast differentiation markers, such as alkaline phosphate, and collagen type I (COL1A1), but also increased the ratio of osteoprotegerin to RANKL. Collectively, the results strongly suggested that BPE is a natural resource for the prevention or treatment of periodontal diseases.

A NOTE ON THE REMOVAL OF PHYTATE IN SOYBEAN MEAL USING Aspergillus usami

  • Soybean meal was fermented by Aspergillus usami in order to reduce phytate content. Aflatoxin B1 was not detected in the fermented soybean meal. The contents of crude protein, crude fiber, ether extract and crude ash were slightly increased following fermentation with a concomitant reduction in nitrogen free extract. Though the fermentation partly degraded proteins in the soybean meal, there was small difference in amino acid composition between the soybean meal and the fermented soybean meal. The results showed that the fermentation did not affect nutritional value of protein in soybean meal. Approximately 55% of phosphorus extracted by trichloroacetic acid was inositol hexaphosphate (phytate) in the soybean meal. The content of inositol tetra to hexaphosphates was not detected in the fermented soybean meal. These results indicated that the fermentation almost completely eliminated phytate in soybean meal. Phytase activity was not detected in the unfermented soybean meal. However, the enzyme activity in the fermented soybean meal was 167.7 U/g. When the fermented soybean meal in supplemented in formula feeds, phytase in the fermented soybean meal might partly degrade the phytate in other ingredients in the digestive tract. The fermented soybean meal is possibly used as a phytate-free protein source of feed, which contains high available phosphorus.

  • Glucose is universal and essential fuel of energy metabolism and in the synthesis pathways of all mammalian cells. Glucose is the one of the major precursors of lactose synthesis using glycolysis result in producing milk fat and protein. During the milk fat synthesis, lipoprotein lipase (LPL) and CD36 are required for glucose uptake. Various morecules such as acyl-CoA synthetase 1 (ACSL1) activity of acetyl-CoA synthetase 2 (ACSS2), ACACA, FASN AGPAT6, GPAM, LPIN1 are closely related with milk fat synthesis. Additionally, glucose plays a major role for synthesizing lactose. Activations of lactose synthesize enzymes such as membranebound enzyme, beta-1,4-galactosyl transferase (B4GALT), glucose-6-phosphate dehydrogenase (G6PD) are changed by concentration of glucose in blood resulting change of amount of lactose production. Glucose transporters are a wide group of membrane proteins that facilitate the transport of glucose over a plasma membrane. There are 2 types of glucose transporters which consisted facilitative glucose transporters (GLUT); and sodium-dependent transport, mediated by the Na+/glucose cotransporters (SGLT). Among them, GLUT1, GLUT8, GLUT12, SGLT1, SGLT2 are main glucose transporters which involved in mammary gland development and milk synthesis. However, more studies are required for revealing clear mechanism and function of other unknown genes and transporters. Therefore, understanding of the mechanisms of glucose usage and its regulation in mammary gland is very essential for enhancing the glucose utilization in the mammary gland and improving dairy productivity and efficiency.

Isolation and Characterization of Bifidobacterium longum subsp. longum BCBR-583 for Probiotic Applications in Fermented Foods

  • Recent human gut microbiome studies have supported that the genus Bifidobacterium is one of the most beneficial bacteria for human intestinal health. To develop a new probiotic strain for functional food applications, fourteen fecal samples were collected from healthy Koreans and the strain BCBR-583 was newly selected and isolated from a 25-year-old Korean woman's fecal sample using the selective medium for Bifidobacterium. Subsequent fructose-6-phosphate phosphoketolase (F6PPK) test and 16S rRNA gene sequencing analysis of the strain BCBR-583 confirmed that it belongs to B. longum subsp. longum. The stress resistance tests showed that it has oxygen and heat tolerance activities (5- and 3.9-fold increase for 24 h at 60 and 120 rpm, respectively; $78.61{\pm}6.67%$ survival rate at $45^{\circ}C$ for 24 h). In addition, gut environment adaptation tests revealed that this strain may be well-adapted in the gut habitat, with gastric acid/bile salt resistance ($85.79{\pm}1.53%$, survival rate under 6 h treatments of gastric acid and bile salt) and mucin adhesion ($73.72{\pm}7.36%$). Furthermore, additional tests including cholesterol lowering assay showed that it can reduce $86.31{\pm}1.85%$ of cholesterol. Based on these results, B. longum BCBR-583 has various stress resistance for survival during food processing and environmental adaptation activities for dominant survival in the gut, suggesting that it could be a good candidate for fermented food applications as a new probiotic strain.

Characterizations of High Early-Strength Type Shrinkage Reducing Cement and Calcium Sulfo-aluminate by Using Industrial Wastes

  • In this study, the utilization of the by-products of various industries was examined using raw materials of CSA high-functional cement such as coal bottom ash, red mud, phosphate gypsum, etc. Technology to improve energy efficiency and reduce $CO_2$ was developed as part of the manufacturing process; this technology included lower temperature sintering ($150{\sim}200^{\circ}C$) than is used in the OPC cement manufacturing process, replacement of CSA cement with the main raw material bauxite, and a determination of the optimum mix condition. In order to develop CSA cement, a manufacturing system was established in the Danyang plant of the HANIL Cement Co. Ltd., in Korea. About 4,200 tons of low purity expansion agent CSA cement (about 16%) and about 850 tons of the lime-based expansion agent dead burned lime (about 8%) were produced at a rate of 60 tons per hour at the HANIL Cement rotary kiln. To improve the OPC cement properties, samples of 10%, 13%, and 16% of CSA cement were mixed with the OPC cement and the compressive strength and length variation rate of the green cement were examined. When green cement was mixed with each ratio of CSA cement and OPC cement, the compressive strength was improved by about 30% and the expansibility of the green cement was also improved. When green cement was mixed with 16% of CSA cement, the compressive strength was excellent compared with that of OPC cement. Therefore, this study indicates the possibility of a practical use of low-cost CSA cement employing industrial wastes only.

  • We investigated the diffusive transport of bupivacaine HCl through the microchannels of microfluidic drug delivery devices. In the biodegradable microfluidic drug delivery devices developed in this research, the drug release rate can be controlled by simply modulating the geometrical parameters of the microchannels, such as the length, number, and cross-sectional area of the microchannels, when the microchannels are used as paths for drug release. However, the hydrophobic nature of a biodegradable polymer, 85/15 poly(lactic-co-glycolic acid), hinders the infiltration of a release medium (phosphate-buffered saline) through the microchannels into the reservoir of a device that contains bupivacaine HCl, at the early stage of drug release. This can have an adverse effect on the early stage release of local analgesic compounds from the device. In this study, microfluidic channels were surface-treated with surfactants such as PEG600 and Tween80, and the effects of the surfactants on the release performance are presented and analyzed.
