발효 전복 추출물의 피부자극에 관한 연구 (The Study on Skin 1st Patch Test of Fermented Hydrolysate from Abalone(Haliotis discus hannai Ino))

  • 본 연구에서 남해안산 전복으로 부터 얻어진 일반 열수 전복 추출물과 발효추출물의 화장품 원료로서의 사용가능성을 평가하고자 기본적인 피부 안전성 시험으로 추물물과 이를 이용한 액정크림 제형에서의 피부 일차 첩포 테스트를 수행하였다. 비교군으로 정제수, 1,3-부틸렌글리콜, 액정크림으로 하였고 시료군은 전복추출물과 전복추출물을 함유한 액정크림 제형이었다. 피부 일차 첩포 테스트 결과, 비교군과 시료군 모두 피부의 자극이 없는 것으로 나타났다. 그러므로 전복 추출물이 화장품 원료로서 피부 안전성을 확인하였다.

Effect of Fermented Sea Tangle on the Alcohol Dehydrogenase and Acetaldehyde Dehydrogenase in Saccharomyces cerevisiae

  • Sea tangle, a kind of brown seaweed, was fermented with Lactobacillus brevis BJ-20. The gamma-aminobutyric acid (GABA) content in fermented sea tangle (FST) was 5.56% (w/w) and GABA in total free amino acid of FST was 49.5%. The effect of FST on the enzyme activities and mRNA protein expression of alcohol dehydrogenase (ADH) and acetaldehyde dehydrogenase (ALDH) involved in alcohol metabolism in Saccharomyces cerevisiae was investigated. Yeast was cultured in YPD medium supplemented with different concentrations of FST powder [0, 0.4, 0.8, and 1.0% (w/v)] for 18 h. FST had no cytotoxic effect on the yeast growth. The highest activities and protein expressions of ADH and ALDH from the cell-free extracts of S. cerevisiae were evident with the 0.4% and 0.8% (w/v) FST-supplemented concentrations, respectively. The highest concentrations of GABA as well as minerals (Zn, Ca, and Mg) were found in the cell-free extracts of S. cerevisiae cultured in medium supplemented with 0.4% (w/v) FST. The levels of GABA, Zn, Ca, and Mg in S. cerevisiae were strongly correlated with the enzyme activities of ADH and ALDH in yeast. These results indicate that FST can enhance the enzyme activities and protein expression of ADH and ALDH in S. cerevisiae.

Production performances and antioxidant activities of laying hens fed Aspergillus oryzae and phytase co-fermented wheat bran

  • Objective: Wheat bran (WB) was co-fermented with Aspergillus oryzae and phytase (Phy) to determine whether co-fermentation improve WB phosphorus and fiber utilization in Isa-brown layers. Methods: A total of 112 Isa brown layer were randomly divided into 7 treatments with 8 replicates per a treatment and 2 hens per a replicate. The treatments included basal diet (control), basal diet supplemented with 250 unit/kg Phy (control+Phy), diet with 10% WB (10% WB), diet with 5% WB and 250 unit/kg Phy (5% WB+Phy) diet with 10% WB and 250 unit/kg Phy (10% WB+Phy), diet with 5% fermented WB supplemented with molasses and phy (PCFWH) and 125 unit/kg Phy (5% PCFWH), and diet with 10% PCFWH (10% PCFWH). The intestinal microbial population, intestinal morphology, serum antioxidant enzyme activities, and excreta phosphorus content were assessed. Results: In PCFWH, spore counts, protease activity, xylanase activity, and ferulic acid were 8.50 log/g dry matter (DM), 190 unit/g DM, 120 unit/g DM, and 127 ㎍/g, respectively. Xylobiose and xylotriose were released in PCFWH, while they were not detectable in WB. Antioxidant capacity was also enhanced in PCFWH compared to WB. The 10% WB+Phy and 10% PCFWH groups produced higher egg mass, but hens fed 5% WB+Phy had the lowest amount of feed intake. Eggs from 10% PCFWH had better eggshell weight, eggshell strength, and eggshell thickness. Birds fed with 10% PCFWH also had higher serum superoxide dismutase and catalase activities. Compare to control, 10% PCFWH significantly reduced excreta phosphorus content. Conclusion: Diet inclusion of 10% PCFWH improved egg quality, antioxidant status, and excreta phosphorus content of laying hens.

Effect of Fermented Chub Mackerel Extract on Lipid Metabolism of Rats Fed Diets without Cholesterol

  • The present study was conducted to evaluate the effect of fermented chub mackerel extract (FCME) on lipid metabolism in rats fed diets without cholesterol. Four week-old male rats were divided into three groups of 10 rats with 0, 1% or 2% FCME supplementation to the diets. Purified diets were used in the present study. Feed and water were fed ad libitum. FCME supplementation had no effect on the activities of acetyl-CoA carboxylase, fatty acid synthetase, and the content of free cholesterol, triglyceride and phospholipid in the liver (p>0.05). 1% FCME supplementation significantly increased serum triglyceride (p<0.05) and hepatic 3-hydroxy-3-methylglutaryl-CoA reductase activity (p<0.05) with no effect on serum total cholesterol, free cholesterol and phospholipid concentration. FCME supplementation significantly reduced serum LDL+VLDL-cholesterol (p<0.01) and atherogenic index (p<0.01) with no effect on HDL-cholesterol. The current study showed that FCME inclusion might reduce the risk of atherosclerosis in rats fed diet without cholesterol.

GBCK25, fermented ginseng, attenuates cardiac dysfunction in high fat diet-induced obese mice

  • The fermentation of medicinal herbs facilitated by microbes is assumed to exert promising therapeutic efficacy on the absorption, bioavailability, and pharmacological effects by speeding up the making or conversion of active constituents into their metabolites. We examined the cardioprotective potential of fermented ginseng, GBCK25, against high-fat diet (HFD)-induced metabolic and functional illnesses as following the essential analysis such as electrocardiographic parameters, alterations of body and organ weights, and echocardiographic studies. The results exhibited that body weights were significantly reduced and the gain of different organ weights were partly eased by GBCK25 treatment. Echocardiography results proposed the amelioration of heart function through normalized levels of left ventricle systolic pressure, ejection fraction, and fractional shortening. These outcomes deliver straight confirmation that GBCK25 could be a potential nutraceutical source for the relief of HFD-induced obesity mediated cardiac dysfunctions.

Isolation and Structural Determination of Aldose Reductase Inhibitor from Korean Fermented Soybean Paste

  • Aldose reductase catalyzes the conversion of glucose into sorbitol. Inhibiting this enzyme in diabetes mellitus can delay or prevent pathogenic process. Aldose reductase inhibitor was screened from Korean fermented soybean pastes (Doen-jang) and purified via sequential processes of ethanol extraction, HP-20 column chromatography, ethyl acetate extraction, silica gel column chromatography, and crystallization. Aldose reductase inhibitor was identified as genistein with molecular weight of 270 Da and molecular formula of $C_{15}H_{10}O_5$ based on UV spectrometry, $^1H$ and $^{13}C\;NMRs$, and mass spectrometry. Genistein inhibited aldose reductase of pig lens with $IC_{50}$ level of $20\;{\mu}M$. Because genistein was effective against aldose reductase of animal source, it may be a potential therapeutic agent for diabetic complications.

발효시간에 따른 미강의 기능성 성분 및 항산화 효과 - 락토바실러스 브레비스를 이용 - (Functional Components and Antioxidant Effects of Rice Bran by Fermentation Time - Using Lactobacillus brevis -)

  • The purpose of this study was to investigate nutritional and functional ingredients and antioxidant activity after fermenting rice bran using Lactobacillus brevis for 24, 36, and 48 hours. The results of the analysis of the nutritional ingredients revealed that there was no significant difference in the carbohydrate, crude protein, crude fat and ash content regardless of the fermentation process and fermentation time. The amount of dietary fiber was significantly different between the unfermented and fermented rice bran and was observed to be the highest after a 48-hour fermentation. The γ-oryzanol, gamma-aminobutyric acid (GABA) and total phenolic contents were significantly higher in the fermented rice bran compared to the unfermented rice bran (p<0.05) and the GABA and total phenolic contents increased significantly as the fermentation time increased (p<0.05). The 2,2-diphenyl-1-picrylhydrazyl (DPPH), 2,2'-azino-bis-3-ethylbenzothiazoline-6-sulfonic acid (ABTS), superoxide and hydroxyl radical scavenging activities used to measure antioxidant effects significantly increased as the fermentation time increased (p<0.05). From these results, it was confirmed that the antioxidant effect and functional components, namely γ-oryzanol, GABA, and the total phenolic content of rice bran improved with fermentation. Based on these results, fermented rice bran could be presented as a functional material for use in high value-added industries.

Hair Growth Effect of TS-SCLF from Schisandra chinensis Extract Fermented with Lactobacillus plantarum

  • This study investigated the hair growth effect of Schisandra chinensis extract (TS-SC) and TS-SC fermented by Lactobacillus plantarum (TS-SCLF) on human dermal papilla cells (hDPCs). The production of vascular endothelial growth factor (VEGF), insulin-like growth factor 1 (IGF-1), keratinocyte growth factor/fibroblast growth factor 7 (KGF/FGF-7) and hepatocyte growth factor (HGF), transforming growth factor beta 1 (TGF-β1) were examined. The secretion rates of VEGF and KGF/FGF-7 were high in TS-SC, and the secretion rates of IGF-1 and HGF were high in TS-SCLF. TGF-β1 was inhibited in a concentration-dependent manner in all samples. Gene expression of VEGF, IGF-1, KGF, HGF and alkaline phosphatase, relevant to hair growth, were examined. The data revealed that TS-SC and TS-SCLF successfully promoted hair growth in hDPCs. The IGF-1 gene was expressed in a dose-dependent manner in TS-SCLF. These results indicate that TS-SC and TS-SCLF fermented extract effectively promoted hair growth and gene expression relevant to hair growth in hDPCs. Used in clinical trials the test substance 'CMK-LPF01' showed a statistically significant increase in the number of hairs at 8 weeks, 16 weeks, and 24 weeks compared to before product use, and a change in hair growth, a secondary efficacy evaluation variable. Through additional research in the future, it is expected that "CMK-LPF01" can be developed as a functional material that can help alleviate symptoms of hair loss.

뽕잎 함유 대두발효물이 신생 당뇨유도쥐에 미치는 혈당강하효과 (Hypoglycemic Effect of Fermented Soybean Culture Mixed with Mulberry Leaves on Neonatal Streptozotocin-Induced Diabetic Rats)

  • 당뇨 환자를 위한 기존의 뽕잎 건강식품의 기능을 보완할 목적으로 뽕잎분말과 DNJ 생산균주인 Bacillus subtilis MORI균의 대두발효 혼합물을 신생 당뇨유도쥐에 4주간 경구투여 하여 먹이섭취량, 체중변화, 혈당량, 내당능, 혈중 지질함량, 장기무게 등을 조사하였다. 전체적으로 당뇨유도 시험군은 정상시험군(NC군)과 비교하여 사료섭취량은 많았으나 증체량이 적어 낮은 사료효율을 보였다. 그러나 뽕잎(M군) 및 뽕잎함유대두발효물(MM군)의 투여에 의해 사료효율이 개선되었으며 특히 MM-60군에서 가장 높은 사료효율을 나타내었다. 1주일 간격으로 4주간에 걸쳐 시행한 혈당량 측정 결과 뽕잎이나 뽕잎함유대두발효물의 투여는 공복혈당을 크게 감소시켰으며 특히 MM-60군과 MM-120군은 당뇨대조군(DC군)에 비해 유의적으로 낮게 나타났다. 4주간의 시료투여 후 경구당부하시험에 의한 내당능 조사 결과 뽕잎투여나 뽕잎함유대두발효물의 투여는 혈당량을 꾸준히 감소시켰으며 특히 포도당 투여 후 120분에서의 MM-60군과 MM-120군은 DC군에 비해 유의적으로 낮은 혈당량을 보여 뽕잎 단독보다는 대두발효배양물 첨가에 의해 내당능이 크게 개선된 것으로 나타났다. 혈중 총콜레스테롤 및 중성지질 역시 DC군에서 높은 함량을 나타냈으나 뽕잎이나 뽕잎함유대두발효물 투여에 의해 지질함량이 감소하였으며 그 감량 정도는 총콜레스테롤에서는 MM-60군과 MM-120군에서 그리고 중성지질의 경우에는 M군, MM-60군, MM-120군에서 DC군에 비해 유의적인 감소를 나타내었다. 당뇨 유발에 의해 쥐의 간, 신장, 비장, 심장 등 체내 주요장기의 무게는 크게 증가하였으나 뽕잎 또는 뽕잎함유대두 발효물 투여에 의해 이들 장기의 무게는 대부분 감소하였으며 특히 MM-60군, MM-120군은 DC군에 비해 유의적인 감소를 나타내었다. 이상의 실험결과 뽕잎분말과 DNJ 생산 균주인 B. subtilis MORI균의 대두발효물의 혼합물은 신생 당뇨유도쥐의 제반 당뇨증세 완화에 긍정적으로 작용하는 것이 확인되었으며 따라서 뽕잎함유대두발효물은 앞으로 당뇨환자들에 대한 건강식품 소재로 그 활용이 크게 기대된다 하겠다.

유산균을 이용한 홍고추의 발효를 통한 항비만과 항당뇨 효과 (Effects Against Obesity and Diabetes of Red Pepper (Capsicum annuum L.) Fermented with Lactic Acid Bacteria)

  • 고추(Capsicum annuum L.)는 한국 전통 음식의 중요한 채소 중 하나이며, 비타민 A, 비타민 C, 비타민 E, 폴리페놀 및 플라보노이드를 많이 함유하고 있다. 또한 고추는 높은 항산화능을 가지고 있으며, 비만, 당뇨병, 고혈압, 소화 장애, 스트레스 및 노화를 예방하는데 효과적이라고 알려져 있다. 본 연구에서는 발효 홍고추 및 발효하지 않은 홍고추의 항비만 및 당뇨병 효과를 조사하였다. 8 주 동안 45% HFD를 공급하여 비만 유도 시킨 C57BL / 6N 마우스는 HFD 사료와2.5% NRM, 1.25% FRM, 2.5% FRM을 함유한 HFD 사료를 8 주간 투여하였다. 식이섭취 시킨 후 7주 경과 후 경구당 부하검사(OGTT)를 진행하였고 8주 경과 후 체중, 간, 부고환 지방량, 혈청 인슐린 수치 및 HOMA-IR을 측정하고 지질 함량 검사를 시행하였다. 이 연구의 결과는 2.5% FRM이 HFD와 2.5% NRM보다 체중, 조직 중량, 지질 함량, 혈청 인슐린 수치 및 HOMA-IR을 감소시켰다. 이 결과를 통해 발효 홍고추는 항비만 및 항당뇨에 효과가 있는 것을 확인하였다. 또한, 본 연구는 발효 홍고추를 이용한 건강기능식품 및 기능성 생리활성증대 소재의 기초자료로 활용할 수 있을 것이다.
