Optimization of Anti-glycation Effect of ʟ-Carnitine, Pyridoxine Hydrochloride and ᴅʟ-α-Tocopheryl Acetate in an Infant Formula Model System Using Response Surface Methodology (ʟ-Carnitine, pyridoxine hydrochloride, ᴅʟ-α-tocopheryl acetate를 이용한 분유모델시스템의 마이얄반응생성물 저감화 조건 최적화)

  • The Maillard reaction is a non-enzymatic reaction between amino and carbonyl groups. During milk processing, lactose reacts with milk protein through this reaction. Infant formulas (IFs) are milk-based products processed with heat-treatments, including spray-drying and sterilization. Because IFs contain higher Maillard reaction products (MRPs) than breast milk, formula-fed infants are subject to higher MRP exposure than breast milk-fed ones. In this study, we investigated the optimization of conditions for minimal MRP formation with the addition of $\small{L}$-carnitine ($\small{L}$-car), pyridoxine hydrochloride (PH), and $\small{DL}$-${\alpha}$-tocopheryl acetate (${\alpha}$-T) in an IF model system. MRP formation was monitored by response surface methodology using fluorescence intensity (FI) and 5-hydroxymethylfurfural (HMF) content. The optimal condition for minimizing the formation of MRPs was with $2.3{\mu}M$ $\small{L}$-car, $15.8{\mu}M$ PH, and $20.6{\mu}M$ ${\alpha}$-T. Under this condition, the predicted values were 77.4% FI and 248.7 ppb HMF.

Inhibitory Effect of Ophioglossum vulgatum on Free Radical and MMP Expression in UV-irradiated Human Dermal Fibroblasts (병이소초 추출물의 항산화 및 MMP 발현 저해 효과)

  • Human skin is constantly exposed to environmental irritants such as smoke, chemicals and ultraviolet (UV). Free radicals and reactive oxygen species (ROS) caused by these environmental irritants play critical roles in cellular damage. In this study, to investigate the skin cell protective effect of Ophioglossum vulgatum extract, we investigated its effects on intercellular antioxidative activity and UVA-induced MMP expression in human dermal fibroblasts (HDFs). The dried O. vulgatum was extracted in a mixture of ethanol and water (1 : 1) for 24 h at room temperature. The extract was filtered and concentrated in vacuo and lyophilized. For testing intracellular ROS scavenging activity the cultured HDFs were analyzed by increase in DCF fluorescence upon exposure to UVB $20\;mJ/cm^2$. After treatment of O. vulgatum extracts, intracellular ROS levels were measured by luminescence spectrophotometer. Enzyme linked immuno sorbent assay (ELISA), and RT-PCR techniques were used for evaluating the effects of O. vulgatumon on MMP protein and mRNA expression in UVA irradiated HDFs. O. vulgatum extract was found to have ROS scavenging activity with the $IC_{50}$ values of $18.2\;{\mu}g/mL$ against superoxide radicals in the xanthine/xanthine oxidase system. After treatment of O. vulgatum extracts, the oxidation of CM-DCFDA was inhibited effectively and O. vulgatum extracts showed a potent free radical scavenging activity by 30.4 % at $100\;{\mu}g/mL$ in UVB-irradiated HDFs. UVA induced MMP protein expression was reduced 37.7 % by treatment with O. vulgatum extract, and MMP-1 mRNA expression was reduced in a dose-dependent manner. Taken together, these results suggest that O. vulgatum extract prevents the skin cell damage induced by UV irradiation, and implies that O. vulgatum extract may be useful as a new ingredient for anti-aging cosmetics.

A study on Airborne Concentration of Welding Fumes and Metals in Confined Spaces of a Shipyard (모조선소의 밀폐된 작업장에서의 공기중 용접흄 및 중금속 농도에 관한 조사 연구)

  • This study was performed to evaluate the exposure levels of worker exposed to welding fume and metals in confined spaces of a shipyard. The airborne concentration of welding fumes and metal elements in confined spaces were compared with those in open working areas. Results of the study were as follows. 1. The geometric mean of welding fume concentration in a confined space was $16.6mg/m^3$, which contained $3.9mg/m^3$ Fe, $1.2mg/m^3$ Mg, $0.8mg/m^3$ Zn, $0.008mg/m^3$ Cu, $0.008mg/m^3$ Pb, $0.005mg/m^3$ Ni, $0.003mg/m^3$ Cr, $0.003mg/m^3$ Cd. The geometric mean of welding fume concentration in open working areas was $5.2mg/m^3$, which contained $1.1mg/m^3$ Fe, $0.3mg/m^3$ Mg, $0.3mg/m^3$ Zn, $0.004mg/m^3$ Cu, $0.008mg/m^3$ Pb, $0.005mg/m^3$ Ni, $0.003mg/m^3$ Cr, $0.0003mg/m^3$ Cd. The geometric mean of welding fume concentration in confined spaces was 3,2 times higher than that in open working areas. The geometric mean concentrations of such metals as Fe, Mg, Zn, or Cu within fume in confined spaces were 2-4 times higher than those in open working areas, while little difference made such metals as Pb, Ni, Cr, Cd. 2. In 32 samples out of a total of 39 samples (82.1%) collected in confined spaces, the concentrations of welding fume exceeded TLV. while so did 19 samples out of 33 samples (57.6%) in open working areas. As for the concentrations of metals in welding flume from confined spaces, Fe exceeded TLV in 14 out of a total of 38 samples (36.8%), Mn exceeded TLV in 23 out of a total of 38 samples (60.5%). As for the concentration of metals in welding fume from open working areas, Fe exceeded TLV in 3 out of a total of 34 samples (8.8%), Mn exceeded TLV in 6 out of a total of 34 samples (17.6%). Considering additive effect among metals, in 31 out of a total of 39 samples (79.5%) collected in confined spaces, the concentrations of welding fume exceeded TLV, while so did 14 out of 38 samples (55.6%) in open working areas. 3. In respect of base metal and welding type the concentration of total welding fume by $CO_2$ gas W./mild steel was the highest, followed by semiauto MMA/mild steel, then followed by TIG or $CO_2$ gas W./stainless steel. ; as for concentration of metal within fume, a decreasing order was Fe, Zn, Mn, and Pb in $CO_2$ gas W./mild steel and semiauto MMA/mild steel, but Fe, Mn, Cr, and Ni in TIG or $CO_2$ gas W./stainless steel. 4. In case of welding base metal covered by paint, contents of Zn within red paint chip and within gray paint chip were 14.0% and 0.08% respectively, which showed a little difference, while the airborne concentrations of Zn within fume during welding base metal covered red paint and gray paint were $1.351mg/m^3$ and $1.018mg/m^3$ respectively, which showed little difference. As for Pb, contents of red paint chip and gray paint chip were 0.14% and 0.08% respectively, and the airborne concentrations within fume during welding base metal covered red paint and gray paint were $0.009mg/m^3$ and $0.007mg/m^3$ respectively, both of which showed little difference.

Protective Effects of Korean Panax Ginseng Extracts against TCDD-induced Toxicities in Rat (랫드에서 TCDD 투여에 의해 유도된 생체독성의 고려홍삼 추출물에 의한 억제 효과)

  • To achieve a better understanding of protective effects of water extracts of Panax ginseng against TCDD-induced toxicities, we monitored physiological and clinical changes in rat for 4 weeks after administrations of each Panax Ginseng extract or TCDD, and co-administration of the two materials. For this study, 120 male Sprague-Dawley (SD) rats weighing 190-210 g each (8 weeks old) were divided into four groups: TCDD-administered, co-administered group with TCDD and ginseng extract, ginseng extract-administered, and control group. The TCDD-administered group received single dose of TCDD in a corn oil vehicle ($25\;{\mu}g/kg$ body weight) by intraperitoneal administration on Day 1. The Panax ginseng extracts-administered group received intraperitoneally 100 mg/kg body weight every other day for one month. For the co-administered group with TCDD and ginseng extracts, Panax ginseng extracts were intraperitoneally administered to rats at 100 mg/kg body weight every other day for one month after a single intraperitoneal dose of $25\;{\mu}g$ of TCDD/kg body weight on Day 1. Panax ginseng extracts attenuated the mortality induced by TCDD administration. The extracts also slightly attenuated the TCDD-induced body weight loss. Administration of TCDD alone increased liver weight at 2, 5, and 16 days after administration of TCDD. Administration of Panax ginseng extracts rather decreased liver weight through whole the experimental period, but which was statistically insignificant. Administration of TCDD alone at $25\;{\mu}g/kg$ body weight increased both serum enzyme activities of alanine aminotransferase (ALT) and aspartate aminotransferase (AST) at 32 days, indicating that liver damage occurred maximally at that time. Ginseng extract administration caused insignificant changes in serum ALT, but gradually decreased in AST as the exposure time increased. Coadministration of TCDD and ginseng extracts caused serum AST activity to significant recovery to normal value at 16 days and 32 days after exposure to TCDD. The extracts also significantly decreased the TCDD-induced ALT activity after 16 days of TCDD administration. These results suggest that Panax ginseng extracts may possess a protective effect against TCDD-induced toxicities including hepatotoxicity in rats.

Hypoxia Inducible Factor-1α Directly Regulates Nuclear Clusterin Transcription by Interacting with Hypoxia Response Elements in the Clusterin Promoter

  • Differential transcription of the clusterin (CLU) gene yields two CLU isoforms, a nuclear form (nCLU) and a secretory form (sCLU), which play crucial roles in prostate tumorigenesis. Pro-apoptotic nCLU and anti-apoptotic sCLU have opposite effects and are differentially expressed in normal and cancer cells; however, their regulatory mechanisms at the transcriptional level are not yet known. Here, we examined the transcriptional regulation of nCLU in response to hypoxia. We identified three putative hypoxia response elements (HREs) in the human CLU promoter between positions -806 and +51 bp. Using a luciferase reporter, electrophoretic gel mobility shift, and chromatin immunoprecipitation assays, we further showed that hypoxia-inducible factor-$1{\alpha}$ (HIF-$1{\alpha}$) bound directly to these sites and activated transcription. Exposure to the hypoxia-mimetic compound $CoCl_2$, incubation under 1% $O_2$ conditions, or overexpression of HIF-$1{\alpha}$ enhanced nCLU expression and induced apoptosis in human prostate cancer PC3M cells. However, LNCaP prostate cancer cells were resistant to hypoxia-induced cell death. Methylation-specific PCR analysis revealed that the CLU promoter in PC3M cells was not methylated; in contrast, the CLU promoter in LNCap cells was methylated. Co-treatment of LNCaP cells with $CoCl_2$ and a demethylating agent promoted apoptotic cell death through the induction of nCLU. We conclude that nCLU expression is regulated by direct binding of HIF-$1{\alpha}$ to HRE sites and is epigenetically controlled by methylation of its promoter region.

Development of Smart Packaging for Cream Type Cosmetic (크림 제형 화장품용 스마트 패키징 기술 개발)

  • The degree of cosmetic's oxidation depends on the storage conditions and external conditions when using the product. The microbial contamination and oxygen exposure often results in the quality deterioration of cosmetics. In addition, the problem is that consumers often use cream-type cosmetics, which have short expiration period (6-12 months), even after the product is expired. When using the deteriorated cosmetics, it can be fatal to consumers' safety including some symptoms such as folliculitis, rashes, edema, and dermatitis. Therefore, it is necessary to develop sealed smart packaging for cosmetics to prevent the deterioration of cosmetics and improve consumer safety. In this study, we have developed smart packaging design for cosmetics that can measure the surrounding environment and expiration date for the cosmetics in the real time. In addition, the smart packaging includes sensor, which are linked to the mobile application. Users can find out the measurement results through the application. Also, the packaging design and functions were set up based on the survey results by the user and feasible model can be produced based on user choice. The measurement in the three environment has been done after manufactured the sensor, PCB, and mobile application. As a result, it works normally within a certain range under all three environmental conditions. It is believed that the information on expiration dates and storage environment can be efficiently delivered to the consumers through developed cosmetics smart packaging and applications. The development of UI/UX design for consumer is further studied. The UX/UI design of the application plays an essential role in achieving this goal through the commercialization the cosmetic products in the wide range.

Evolution of Mechanical Properties through Various Heat Treatments of a Cast Co-based Superalloy (주조용 코발트기 초내열합금의 열처리에 따른 기계적 특성 변화)

  • The effects of a heat treatment on the carbide formation behavior and mechanical properties of the cobalt-based superalloy X-45 were investigated here. Coarse primary carbides formed in the interdendritic region in the as-cast specimen, along with the precipitation of fine secondary carbides in the vicinity of the primary carbides. Most of the carbides formed in the interdendritic region were dissolved into the matrix by a solution treatment at $1274^{\circ}C$. Solutionizing at $1150^{\circ}C$ led to the dissolution of some carbides at the grain boundaries, though this also caused the precipitation of fine carbides in the vicinity of coarse primary carbides. A solution treatment followed by an aging treatment at $927^{\circ}C$ led to the precipitation of fine secondary carbides in the interdendritic region. Very fine carbides were precipitated in the dendritic region by an aging heat treatment at $927^{\circ}C$ and $982^{\circ}C$ without a solution treatment. The hardness value of the alloy solutionized at $1150^{\circ}C$ was somewhat higher than that in the as-cast condition; however, various aging treatments did not strongly influence the hardness value. The specimens as-cast and aged at $927^{\circ}C$ showed the highest hardness values, though they were not significantly affected by the aging time. The specimens aged only at $982^{\circ}C$ showed outstanding tensile and creep properties. Thermal exposure at high temperatures for 8000 hours led to the precipitation of carbide at the center of the dendrite region and an improvement of the creep rupture lifetimes.

Post-thaw Development of Rabbits Pronuclear Embryos by Cryopreservation (토끼 전핵배의 동결보존 후 배발달률)

  • This study assessed development in vitro of pronuclear(PN) stage embryos cryopreserved by the method of either vitrification or slow freezing, by using of different cryoprotectants, and equilibration and cooling rate, in rabbit. Ethyleneglycol- ficoll- sucrose(EFS) or ethyleneglycol- polyvinylpyrrolidone - galactose- (EPG-I) for vitrification, and EPG- II for slow freezing as cryoprotectant were used. The pronuclear embryos were exposed to EFS for 0 to 5 min and diluted with D-PBS and/or pre-dilution with 0.5 M sucrose. To examine the viability of frozen-thawed embryos, PN embryos were co-cultured with bovine oviductal epitherial cell(BOEC) for 5 days to hatching blastocyst stage in 39 $^{\circ}C$ 5% $CO_2$incubator. The results obtained were as follows: The dilution with 0.5 M sucrose and D-PBS after the exposure to EFS for 1.0 min resulted in no significant(P<0.05) decrease in the development of PN embryos to hatching blastocyst(72.0%), compared with controls. The development of PN embryos cryopreserved to hatching blastocyst was not significantly (P<0.05) different between EFS for 1.0 min(72.0%), EPG-I for 1.0 min(72.0%) and EPG-II for 30 min(66. 7%). The post-thaw development of PN embryos to hatching blastocyst was similarly very low as 6.1% and 11.5% in vitrification with EFS and slow freezing with EPG-II, respectively. The incidence of post-thaw zona-crack in PN embryos cryopreserved by slow freezing with plunging to liquid nitrogen at -35$^{\circ}C$ was signicantly(P<0.05) higher(25.0%), compared with -85$^{\circ}C$ (1.9%). These results indicated that the rabbit PN embryos could be cryopreserved with either vitrification or slow freezing procedure, and frozen PN embryos could be successfully developed in vitro to haching blastocyst. but the post-thaw development of cryopreserved PN embryos was still very low under the present conditions.

Effect of Submergence and Air Exposure of the Shoot on Growth, Nutrient Uptake and Photosynthesis in Monochoria vaginalis Presl. (물달개비 경엽(莖葉)의 침수여부(沈水與否)에 따른 생장(生長), 양분흡수(養分吸收) 및 광합성(光合成) 비교(比較))

  • Growth, nutrient uptake and photosynthesis as affected by submersion of shoot in pickerel weed (Monochoria vaginalis Presl.) were determined. The shoots of pickerel weeds in hydroponic culture were subjected to the submerged or emerged condition at 3- or 5-leaf stage for 8 or 10 days. Under submerged condition, growth in plant height was enhanced, but leaf number, leaf area, fresh and dry weight were reduced compared to those under the emerged condition. Similar responses in growth to submergence were obtained with the pickerel weeds rooted in the soil. Under submergence, chlorophyll content increased during the first 2 days, but thereafter remarkably decreased at 3-leaf stage and after the first 4 days at 5-leaf stage. Compared to the emerged condition, uptakes of $NH_4\;^+$-N, $NO_3\;^-$-N, $P_2O_5$ and $K^+$ were reduced, but uptakes of $Ca^{++}$ and $Mg^{++}$ increased under the submerged condition. Photosynthetic rate of shoot under water, measured by $CO_2$electrode, showed the maximum by 210 ${\mu}$moles $HCO_3\;^-$/g F.W. at the 8th day after submergence(DAS) at 3-leaf stage and 320 ${\mu}$moles $HCO_3\;^-$/g F. W. at 6 DAS at 5-leaf stage. These results indicate that pickerel weeds grow much better when the shoot is air-exposed and are less tolerable to submergence at 3 leaf-stage than at 5-leaf stage.

  • Since the usage of tributyltin(TBT) has been banned, many chemicals including Sea-Nine 211, Cu-pyrithione, and Zn-pyrithione were developed to use as antifouling agents for ships and coastal structures. However, the toxicity of these antifouling chemicals have not been systematically evaluated in ecotoxicological and biological studies. In this study, we investigated the effect of four antifouling substances on survival of estuarine rockfish, Sebastes schlegeli and amphipod, Monocorophium acherusicum. Survival of S. schlegeli and M. acherusicum during the 96-h exposure period were used to estimate the median lethal concentrations(LC50s) of test chemicals for each test species. Among antifouling agents, Cu-pyrithione($56{\mu}g{\cdot}1^{-1}$;96-h LC50) was most toxic to S. schlegeli, followed by $TBT(73{\mu}g{\cdot}1^{-1}),\;Sea-Nine(184{\mu}g{\cdot}1^{-1})\;and\;Zn-pyrithione(l707{\mu}g{\cdot}1^{-1})$, while TBT($26{\mu}g{\cdot}1^{-1}$) was most toxic to M. acherusicum followed by Sea-Nine($49{\mu}g{\cdot}1^{-1}$), Cu-pyrithione($119{\mu}g{\cdot}1^{-1}$) and Zn-pyrithione($334{\mu}g{\cdot}1^{-1}$). Effect concentrations of the antifouling chemicals estimated in this study can be used when assessing the potential risks of these substances, of which usage is increasing in the coastal environment.
