본 연구는 probiotics mixture(Lactobacillus acidophilus PBS066(DSM 24936), Lactiplantibacillus plantarum PBS067(DSM 24937), Limosilactobacillus reuteri PBS072(DSM 25175))의 안전성 및 탄수화물 소화 효소 저해 효과를 in vitro로 평가하는 것을 목적으로 한다. 모든 균주는 유럽식품안전처(EFSA) 지침에서 권장하는 항생제 내성 프로필을 충족시켰으며, 모든 균주는 용혈 활성 및 세포 독성을 나타내지 않았다. Probiotics mixture, L. plantarum PBS067 그리고 L. reuteri PBS072는 α-amylase 활성을 억제하는 것으로 나타났으며, probiotics mixture와 이의 균주 3종 모두 α-glucosidase 활성을 억제하는 것으로 나타났다. 이로써 본 연구에 사용한 probiotics mixture 및 이의 단일 균주 3종의 안전성과 탄수화물 소화 억제 효과를 확인하였으며, 따라서 probiotics mixture 이의 단일 균주 3종은 섭취하여도 안전하며 섭취 시 체내 탄수화물 대사를 잠재적으로 조절하는 것을 확인하였다.

The anaerobically digestion and agricultural application of organic wastes was conducted using food wastes and cow dung. Twenty kilograms each of the feed stocks was added into two 30 liters-capacity batch digesters. The anaerobic digestion was carried out within a temperature range of $25-31^{\circ}C$ for a retention time of 51 days. The results showed a cumulative gas yield of 5.0 bars for food waste and no gas production for cow dung within the retention time. Bacteria such as Escherichia coli, Staphylococcus aureus, Pseudomonas aeruginosa, Proteus vulgaris and Clostridium sp were isolated. Fungi isolated included Aspergillus niger, Aspergillus nidulan, Trichophyton rubrum and Epidermophyton flocossum. The non-dispersive infrared (NDIR) analysis of the biogas produced confirmed that the gas consisted of $CH_4$, $CO_2$ and $H_2$. Statistical analysis revealed there was no significant correlation between temperature and biogas produced from the organic wastes (r= 0.177, p = 0.483).The organic wastes from the biogas production process stimulated maize growth when compared to control (soil without organic waste) and indicated maximum height. The study therefore reveals that food waste as potential substrates for biogas production has a moderate bio-fertilizer potential for improving plant growth and yield when added to soil.
동물 사체 처리가 축산업에 큰 부담이 되고 있다. 본 연구의 목적은 고온 혐기성 소화에 의한 동물 사체의 처리 타당성을 검토하는데 있다. 고기와 내장을 사용하여 수행된 회분식 실험 결과 동물 사체가 혐기성 고온 조건에서 분해 가능함이 확인되었다. 초기 기질 volatile solids (VS) 1.5~7.7% 조건에서 유기물 제거 효율과 메탄 생성 모두 52.7~58.5%, 220~243 ml/g VS의 양호한 효율을 보였다. 그러나 연속 실험에서는 VS 2.5% 이상의 조건에서 혐기성 미생물의 활동이 암모니아 독성에 의해 저해됨이 발견되었다. 암모니아가 동물 사체 내의 단백질이 분해되는 과정에서 생성되며 혐기성 조건에서 분해되지 않으므로, 암모니아 농도를 낮추는 방법은 희석이 유일하다고 판단된다. 따라서 고온 혐기성 소화를 적용할 시, 동물 사체의 단독 처리 보다는 질소 농도가 상대적으로 낮은 다른 폐수 또는 폐기물과의 혼합 소화를 고려하는 것이 타당할 것으로 사료된다.
Effects of $CO_2$ partial pressure($pCO_2$) on the characteristics of methane production rate and organic matter degradation in anaerobic digestion were investigated by using anaerobic chemostat type reactors at $35{\pm}1^{\circ}C$, at the HRT of 7days. The $pCO_2$ of the reactors was controlled in the range from 0.1 to 0.8 atm. Since the $pCO_2$ in an uncontrolled condition was about 0.4atm, $N_2$ was added for the reactors controlled of $pCO_2$ of between 0.1 and 0.4atm. At $pCO_2$ of 0.5 atm, the methane production rate was approximately 20% more that in an uncontrolled condition of $pCO_2$. Based on the carbon mass balance, it was concluded that methane production was related to the increment of removal organic carbon and consumption of $CO_2$. At $pCO_2$ of 0.5atm, the methane production by the increment of removal substrates increased 13.6%, on the orther hand, hand, the methane production by the conversion of $CO_2$ to methane increased 6.4%.
Anaerobic co-digestion of swine manure and food waste for biogas production was performed in serum bottles at 2% volatile solids(VS) concentration and various mixing ratios of two substrates(swine manure: food waste = 100 : 0 $\sim$ 0 : 100). Through kinetic mode of surface methodology, the methane production was fitted to a Gompertz equation. The specific methane production potential of swine manure alone was lower than that of food waste. However, maximum methane production potential increased up to 1.09-1.22% as food waste composition increased up to the 80%. The maximum methane production value of food waste was 544.52 mL/g VS. It was observed that the maximum methane production potential of 601.86 mL/g VS was found at the mixing ratio of 40:60.
Anaerobic digestion system is getting more attractive in that it produces biogas in the process of organic waste stabilization. Net energy production is important when biogas production is concerned. In this study, net energy production was evaluated with respect to biogas production and heat losses in a hypothetical digester. Under the condition of digester operation with slurry inflow of 5% of TS, additional fuel is required to maintain digester temperature during the winder season. Substrate therefore, needs to have higher VS contents through co-digestion of silage or food waste that has greater values of methane production rate. Heating input slurry is important in cold season, which covers over 80% of heating requirement. Heat recovery from digestate is valuable to reduce the use of biogas for heating. It seems desirable to minimize slurry inflow when temperature is very low. Psychrophilic digestion may be a feasible option for reducing heating requirement.
Si3N4 powder has been analyzed by inductively coupled plasma atomic emission spectrometry (ICP-AES). The sample was dissolved by high-pressure acid digestion with HF, H2SO4 (1+1), and HNO3 mix ture. This technique is well suited for the impurity analysis of Si3N4 because the matrix interference is eliminated. A round-robin samples trace elements, such as Ca, W, Co, Al, Fe, Mg, and Na, were determined. For the direct analysis, slurry nebulization of 0.96 mm Si3N4 powder also has been studied by ICP-AES. Emission intensities of Fe were measured as ICP operational conditions were changed. Significant signal difference between slurry particles and aqueous solution was observed in the present experiment. Analytical results of slurry injection and high-pressure acid digestion were compared. For the use of aqueous standard solution for calibration, k-factor was determined to be 1.71 for further application.
By introducing chemically enhanced primary treatment (CEPT) in the first stage of sewage treatment, organic matter in sewage can be effectively recovered. Because CEPT sludge contains a high biodegradable organic matter in volatile solids (VS), it is feasible to convert the collected CEPT sludge into energy through anaerobic digestion. This study examined the properties and biochemical methane potential (BMP) of the CEPT sludge obtained from a sewage treatment plant located in an ocean area. The CEPT sludge contains a VS content of 37,597 mg/L, which is higher than that of excessive sludge (ES), i.e., 33,352 mg-VS/L. In the methane generation reaction, the lag period was as short as 1 to 2 days. The BMP for the CEPT sludge was 0.57 ㎥-CH4/kg-VSremoved which is better than that of ES, i.e., 0.36 ㎥-CH4/kg-VSremoved. Unfortunately, the CEPT sludge showed a high salinity as 0.56~0.75% probably due to the saline sewage. Due to the salinity, repeated BMP testing in a sequencing batch reactor showed significantly low methane production rates and BMPs. Also, the ES showed a strongly reduced BMP when the salinity was adjusted from 0.20 to 0.70% by NaCl. The ES mixture with higher CEPT content showed a better BMP, which is suitable for co-digestion. Besides, anaerobic digestion for 100% CEPT sludge can be a considerable option instead of co-digestion.
Purpose: The objective of this study was to investigate the feasibility of anaerobic digestion of Chinese cabbage waste juice (CCWJ) and swine manure(SM). Methods: The anaerobic digestion test was conducted under batch and continuous conditions at mesophilic temperature ($36-38^{\circ}C$). The batch test was divided into Experiment I and II. In the Experiment I, biogas potential and production rate of CCWJ was evaluated. In Experiment II the effect of F/M ratio (2.0, 3.2, 4.9) at mixture ratio of 25:75(CCWJ: SM, % vol. basis) on biogas yield was studied. Results: CCWJ produced biogas and methane yield of 929 and 700 mL/g VS added respectively. The biogas yield from the mixture of CCWJ and SM was almost same at F/M ratio of 2.0 and 3.2 but dropped by 14% when F/M ratio increased from 3.2 to 4.9. In continuous test the mixture of CCWJ and SM (25:75, % vol. basis) produced biogas yield of 352 mL/g VS added which is around 11% higher compared to biogas yield from SM alone. Addition to biogas yield digester performance was also improved with co-digestion of CCWJ with SM. Conclusions: The results showed that the anaerobic digestion of CCWJ with SM could be promising for improving both the biogas yield and digester performance at mesophilic temperature.
Anaerobic digestion(AD) is the most promising method for treating and recycling of different organic wastes, such as organic fraction of municipal solid waste, household wastes, animal manure, agro-industrial wastes, industrial organic wastes and sewage sludge. During AD, i.e. organic materials are decomposed by anaerobic forming bacteria and fina1ly converted to excellent fertilizer and biogas which is a mixture of carbon dioxide and methane. AD has been one of the leading technologies that can make a large contribution to produce renewable energy and to reduce $CO_2$ and other green-house gas(GHG) emission, it is becoming a key method for both waste treatment and recovery of a renewable fuel and other valuable co-products. Currently some 80% of the world's overall energy supply of about 400 EJ per year in derived from fossil fuels. Nevertheless roughly 10~15% of this demand is covered by biomass resources, making biomass by far the most important renewable energy source used to date. The representative biofuels produced from the biomass are bioethanol, biodiesel and biogas, and currently biogas plays a smaller than other biofuels but steadily growing role. Traditionally anaerobic digestion applied for different biowaste e.g. sewage sludge, manure, other organic wastes treatment and stabilization, biogas has become a well established energy resource. However, the biowaste are fairly limited in respect to the production and utilization as renewable source, but the plant biomass, the so called "energy crops" are used for more biogas production in EU countries and the investigation on the biomethane potential of different crops and plant materials have been carried out. In Korea, with steadily increasing oil prices and improved environmental regulations, since 2005 anaerobic digestion was again stimulated, especially on the biogasification of different biowastes and agro-industrial biomass including "energy crops". This study have been carried out to investigate anaerobic biodegradability by the biochemical methane potential(BMP) test of animal manures, different forage crops i.e. "energy crops", plant and industrial organic wastes in the condition of thermophilic temperature, The biodegradability of animal manure were 63.2% and 58.2% with $315m^3CH_4/tonVS$ of cattle slurry and $370m^3CH_4/tonVS$ of pig slurry in ultimate methane yields. Those of winter forage crops were the range 75% to 87% with ultimate methane yield of $378m^3CH_4/tonVS$ to $450m^3CH_4/tonVS$ and those of summer forage crops were the range 81% to 85% with ultimate methane yield of $392m^3CH_4/tonVS$ to $415m^3CH_4/tonVS$. The forge crops as "energy crops" could be used as good renewable energy source to increase methane production and to improve biodegradability in co-digestion with animal manure or only energy crop digestion.
