Green shoot cuttings of 10 different grapevine species were inoculated with Agrobacterium vitis to find disease-related metabolites in the grapevine. Crown galls formed 60 days after inoculation varied in gall severity (GS) evaluated by gall incidence (GI) and gall diameter (GD), which were classified into three response types as RR (low GI and small GD), SR (high GI and small GD), and SS (high GI and large GD), corresponding to resistant, moderately resistant, and susceptible responses, respectively. In this, 4, 4, and 2 Vitis species were classified into RR, SR, and SS, respectively. Gas chromatography mass spectrometry (GC-MS) analysis of the grapevine stem metabolites with A. vitis infection showed 134 metabolites in various compound classes critically occurred, which were differentially clustered with the response types by the principal component analysis. Multivariate analysis of the metabolite profile revealed that 11 metabolites increased significantly in relation to the response types, mostly at post-inoculation stages, more prevalently (8 metabolites) at two days after inoculation than other stages, and more related to SS (7 metabolites) than RR (3 metabolites) or SR (one metabolite). This suggests most of the disease-related metabolites may be rarely pre-existing but mostly induced by pathogen infection largely for facilitating gall development except stilbene compound resveratrol, a phytoalexin that may be involved in the resistance response. All of these aspects may be used for the selection of resistant grapevine cultivars and their rootstocks for the control of the crown gall disease of the grapevine.

The CD4 and CDS coreceptors, in conjunction with the T cell receptor (TCR) , make important contributions to the differentiation of thymocytes. They have been shown to be involved in the clonal deletion and positive selection processes during T cell development in thymus. To further analyze the role of CD4 and CDS proteins during T cell differentiation, we have generated transgenic mice constitutively expressing high levels of a native CD4 and a CD4{CDSa hybrid protein. The hybrid protein is composed of CD4 extracellular domain linked to the CD8a transmembrane region and cytoplasmic tail. The transgenes were driven by human beta-actin promoter, and therefore, they were expressed in all tissues examined including thymus, spleen, and lymph nodes. The resulting CD4 and CD4{CD8${\alpha}$transgenic mice were found to express the CD4 and CD4{CD8${\alpha}$ respectively, in developing thymocytes and peripheral T cells. The expression levels of transgenic proteins were 5-10 times higher than that of endogenous CD4 in thymus. However, total surface CD4 expression (CD4 or CD4{CD8${\alpha}$ transgenic protein plus endogenous CD4) of the transgenic mice were main. tained at similar levels compared to control littermates. Surface CD4 expression on CDS T cells, however, was significantly lower than that on cells expressing endogenous CD4. These results suggest that a total avidity between developing thymocytes and thymic stromal cells is impor. tant for differentiation of thymocytes.
"Early Valley", is an early maturing potato cultivar with high yield potential. "Early Valley" is a clonal selection resulting from the cross between 'Suncrisp' and 'A87109-10'. It has medium plant height and light green foliage. "Early Valley" has medium flowering habit and white flowers. Tubers are smooth, yellow skin, light yellow flesh, round tuber shape, medium eye depth, and medium dormancy and good keeping quality. It has stable yield under wide range of climatic conditions. "Early Valley" is resistance to late blight, but moderately susceptible to common scab and hollow heart. This cultivar is also resistant to potato rotting at harvesting during the raining season. "Early Valley" has high level of antioxidant activity (about three times higher) and vitamin C (higher by 40%) than the 'Superior'. This cultivar has high level of tuber uniformity and capable of yielding 36.56 t/ha which is 17.07% higher than the control potato cultivar 'Superior' under optimum agronomical practices.
Background: Immunoglobulin (Ig) light chain repertoire has been implicated as a critical determinant in regulation of autoreactive B cells and production of pathogenic anti-DNA antibodies in systemic lupus erythematosus (SLE). We analyzed the impact of Ig ${\lambda}$ chain repertoire on development of autoimmunity in patients with SLE. Methods: We obtained genomic DNA from individual peripheral CD19+ B cells of 3 untreated active SLE patients, and amplified $V{\lambda}$ rearrangements from each single cell by polymerase chain reaction. Results: A total number of 208 $V{\lambda}J{\lambda}$ rearrangements were analyzed. Analyzed sequences included 158 productive rearrangements and 50 nonproductive rearrangements. The differences in $V{\lambda}$ gene usage in the productive and nonproductive repertoire of SLE patients were found compared to the non-autoimmune individuals. $V{\lambda}$ gene, 9A was significantly overrepresented in nonproducative repertoire of SLE patients (P=0.016). In the productive repertoire, $V{\lambda}$ genes, 3L and 1E were found more often in the SLE patients (P=0.001, P=0.043). When the productive and the nonproductive repertoires were compared, 9A was found significantly less in the productive repertoire in the SLE patients (P=0.000). There were no significant differences in the $J{\lambda}$ gene usage between SLE patients and non-autoimmune individuals, but $J{\lambda}2/3$ gene was the most frequently used in SLE, whereas $J{\lambda}7$ gene was the most frequently used in the normal subjects. In the productive SLE $V{\lambda}$ repertoire, 9.4% of the total sequences employed identical CDR3. It was particularly striking to find 7 identical versions of the 1G-$J{\lambda}2/3$$V{\lambda}J{\lambda}$ rearrangements from one patient and 3 of the same sequence from another patient. Notably, identical $V{\lambda}$ junctions in the SLE patients utilized significantly more homologous joining compared to $V{\lambda}$ junctions of the normal adults (P=0.044). Conclusion: These data demonstrate regulation of ${\lambda}$ light chain expression in the SLE patients by selection of unique $V{\lambda}$ genes. Also, biased selection and clonal expansion of particular $V{\lambda}$ rearrangements are apparent in the SLE ${\lambda}$ repertoire.
This study was designed to know difference in degree of dehardening and rehardening respectively by artificial high and low temperature treatments among different clonal seedlings and seedlings from different seed sources of Cryptomeria japonica which have been grown under the cold areas in Japan and Korea. High temperature treatment was done with 15 to $20^{\circ}C$ under 100% relative humidity for one to nine days and low temperature treatment was carried with $-7^{\circ}C$ for one to three days. Occasionaly, high temperature treatment was combined and followed by low temperature treatment. The ability of stem section to delay dehardening by high temperature treatment and/or to hasten rehardening by low temperature treatment was used as an indicator of adaptability under extreme temperature fluctuation in nature. Clones and seedlings from different seed sources which showed greater freezing resistance than others after artificial high and/or low temperature treatments were selected over two to three time periods: early winter, mid winter and early spring in 1977 to 1980. These were Seoul #7, and #9, Namboo #3, and #4, Sung-Kang #11, Chung-Sam #8 and Huek-Suk #9. These selected seedlings might have survival advantage to withstand early and late frost damage, especially the critical frost damage of the basal stem, since it was known to be induced by lowering freezing resistance of the basal part when exposed to the high temperature near the ground during the day. Large variation in freezing resistance and degree of dehardening and rehardening was found among clonal or seed sources and among individuals within a seed source, but was not related to the difference in climatic conditions where the parent trees was selected. These indicated the possibility of future breeding work for more cold resistant family of Cryptomeria japonica.
Recombinant Chinese hamster ovary (rCHO) cell lines expressing a high level of human thrombopoietin (hTPO) or its analog, TPO33r, were obtained by transfecting expression vectors into dihydrofolate reductase-deficient (dhfr) CHO cells and subsequent gene amplification in media containing stepwise increments in methotrexate (MTX) level such as 20, 80, and 320 nM. The parental clones with a hTPO expression level $>0.40\;{\mu}g/ml$ (27 out of 1,200 clones) and the parental clones with a TPO33r expression level $>0.20\;{\mu}g/ml$ (36 out of 400 clones) were subjected to 20 nM MTX. The clones that displayed an increased expression level at 20 nM MTX were subjected to stepwise increasing levels of MTX such as 80 and 320 nM. When subjected to 320 nM MTX, most clones did not display an increased expression level, since the detrimental effect of gene amplification on growth reduction outweighed its beneficial effect of specific TPO productivity ($q_{TPO}$) enhancement at 320 nM MTX. Accordingly, the highest producer subclones ($1-434-80^{*}$ for hTPO and $2-3-80^{*}$ for TPO33r), whose $q_{TPO}$ was 2- to 3-fold higher than that of their parental clones selected at 80 nM MTX, were isolated by limiting dilution method and were established as rCHO cel1 lines. The $q_{TPO}$ of $1-434-80^{*}\;and\;2-3-80^{*}\;was\;5.89{\pm}074\;and\;1.02{\pm}0.23\;{\mu}g/10^6$ cells/day, respectively. Southern and Northern blot analyses showed that the enhanced $q_{TPO}$ of established rCHO cell lines resulted mainly from the increased TPO gene copy number and subsequent increased TPO mRNA level. The hTPO and TPO33r produced from the established rCHO cell lines were biologically active in vivo, as demonstrated by their ability to elevate platelet counts in treated mice.

Vaccinium oldhamii Miq. is a Korean native tree, which is deciduous and shrub tree with broad leaf. Fruits were used primarily for edible or medicinal purposes for bladder infection in Korea and China. However, there is a lack of information on the degree of variation of phytochemical contents and antioxidant activity of these native clonal. We investigated morphological characteristics, phytochemical contents (polyphenol, flavonoid, anthocyanin) and antioxidant activity for the fruits of 24 individual variation from 4 different regions in Korea. The results in terms of horticultural traits and biochemical characteristics showed significant differences among populations and among individuals within population. The total phenolic contents ranged from 4.40 to 10.58 mg GAE/g and the total flavonoid contents ranged from 2.02 to 8.09 mg NE/g. The total anthocyanin contents ranged from 232.5 to 684.3 mg CGE/100g. Reducing power fruits ranged from 0.15 to 1.43 and DPPH free radical scavenging activity ranged from 49.1 to 97.5% at 400 ppm. Results revealed that the region of origin greatly influenced the phytochemical contents and antioxidant activity of V. oldhamii, which might help for the selection and validation of the most productive cultivars in the food and pharmaceutical industries.

Growth characters such as height, DBH, crown width, branch length and branch diameter were measured from 23 clones of plus trees of Pinus koraiensis. The results were as follows; 1) Differences between the largest and smallest clones were amounted to 41% in height, 81% in DBH, 50% in crown width, 55% in branch length and 83% in branch diameter. 2) There were significant differences between clones in every growth characters and correlation coefficients between growth characters were positive and significant ($r=0.418^*{\sim}0.917^{**}$). 3) The broad sense heritabilities were 0.46 for height, 0.45 for DBH, 0.28 for crown width, 0.26 for branch length and 0.18 for branch diameter. 4) The genetic gains predicted for each characters were ranged from 4.1% to 11.1%, when selecting the best 3 out of 23 clones.
In order to investigate biomass yield of one-year-old hyblrid poplars, Pripulusalba x P. glandulosa $F_1$ and Populus nigra var. italica x P. maximowiczii $F_1$clones, as energy and fiber resources, dry matter yield, leaf area, leaf area index, dry matter production ability, specific gravity and fiber length and width were measured. Dry matter yield was 1.89 ton/ha for Poplus alba x P. glandulosa $F_1$ and 3.63 ton/ha for Populus nigra via.italica x P. merximowiczii $F_1$ clones in the planting density of 20,000 trees/ha and in the planting density of 40,000 trees/ha was 3.87 ton/ha for Populus alba x P. glandulosa $F_1$and 5.64 ton/ha for Populus nigra var. italica x P. maximowiczii $F_1$ clones. Leaf area index was 1.24mtim2 in the planting density of 20,000 trees/ha and 2.45 m31m3 in the density of 40,000 trees/ha for Populus alba x P. glandulosa $F_1$ clones and it was 1.96 m21m2 in the planting density of 20,000 trees/ha and 3.36 m21m2 in the density of 40,000 trees/ha for the hybrid $F_1$ Populus nigra var. italica x P. maximowiczii clones. The average specific gravity of the hybrid poplars was 0..36 when bark and pith were included and 0.31 when bark and pith were removed in the plot of 20,000 trees/ha and in the 40,000 trees/ha plot showed 0.35 and 0.31 respectively, for Populus alba x P. glandulosa $F_1$clones. It was 0.36 when bark and pith were included and 0.32 when bark and pith were removed in the 20,000 trees/ha plot and in the 40,000 trees/ha plot was 0.34 and 0.31 respectively for Populus nigra var. italica x P. maximowiczii $F_1$clones. The average fiber length was 0.57 mm in the 20,000 trees/ha plot and 0.58 mm in the 40,000 trees/ha plot for Poplus alba x P. glandulosa $F_1$clones and was 0.60 in both plots of 20,000 trees/ha and 40,000 trees/ha for Populus nigra var. italica x P. maximowiczii $F_1$ clones. There is a big clonal variation among those clones studied, showing high selection potential in both species.
The main objective of this breeding program is to develop high yielding, disease resistance and good processing potato cultivar. 'Gui Valley' is a clonal selection resulting from a cross between 'ND2471-8' and 'Cona'. It has medium plant height and light green foliage. 'Gui Valley' has medium flowering habit and light pink flowers. 'Gui Valley' is a medium maturing potato cultivar and tubers are smooth, yellow skin, light yellow flesh, long-oval tuber shape, distinct red eyes with medium depth and medium dormancy. It has high level of tuber uniformity and good keeping quality. 'Gui Valley' demonstrates resistance to potato virus Y (PVY), soft rot, but moderately susceptible to late blight and common scab. It is also resistant to most of the internal and external physiological disorders particularly dehiscence, hollow heart and internal brown spot. The specific gravity of 'Gui Valley' is significantly higher (1.097) than that of 'Shepody' (1.078). 'Gui Valley' has suitable for processing mainly French fries and chips. This cultivar has high level of tuber uniformity and capable of yielding 37.6 $t{\cdot}ha^{-1}$, which is 18.2% higher than the control potato cultivar 'Shepody' under optimum agronomical practices.
