  • BACKGROUND: This study was performed to develop a single residue analytical method for new herbicide metazosulfuron in crops. METHODS AND RESULTS: Brown rice, apple, mandarin, Kimchi cabbage and soybean were selected as representative crops, and clean-up system, partition solvent and extraction solvent were optimized. Instrumental limit of quantitation (ILOQ), linearity of calibration curve and method limit of quantitation (MLOQ) were determined based on the chromatography and whole procedures. For recovery tests, brown rice, apple, mandarin, Kimchi cabbage and soybean samples were macerated and fortified with metazosulfuron standard solution at three levels (MLOQ, 10 MLOQ and 100 MLOQ). And then those were extracted with acetonitrile, concentrated, and partitioned with ethyl acetate. Then the extracts were concentrated again and cleaned-up through $NH_2$ (aminopropyl) SPE cartridge with acetone : dichloromethane (1% acetic acid) (20 : 80, v/v) before concentration and analysis with HPLC. CONCLUSION(S): ILOQ of metazosulfuron was 2 ng (S/N${\geq}$10) and good linearity was achieved between 0.05 and 12.5 mg/Kg of metazosulfuron standard solutions, with coefficients of determination of 0.9999. MLOQ was 0.02 mg/Kg. Good recoveries from 74.1 to 116.9% with coefficients of variation (C.V.) of less than 10% were obtained, regardless of sample type, which satisfies the criteria of Korea Food and Drug Administration (KFDA). Those results were reconfirmed with LC-MS (SIM). The method established in this study is simple, economic and efficient to be applied to most of crops as an official and general method for residue analysis of metazosulfuron.

  • Sulfate reduction rates (SRR) using $^{35}SO_4^{-2}$, sulfide producing rates (SPR) using gas chromatography, the number of sulfate reducing bacteria (SRB) using the most probable number (MPN) method, and soil components (moisture, ammonium, total nitrogen, total organic carbon, total carbon, total inorganic phosphorus, total phosphorus, and sulfate) using standard methods in the organic/conventional rice paddy soils, cleaned/polluted reservoir soils, and cleaned/polluted foreshore soils were studied with the change of seasons. The average SRR was more related to the number of SRB and soil components (especially nitrogen and phosphorus) than sulfate concentration. SRR was also recorded to be highest in October soil samples. However, SPR was higher in foreshore soils containing a high concentration sulfate than in fresh water soils, and it was also recorded to be higher in the polluted areas than in clean areas. From these results, we can conclude that the SRR and SPR of anaerobic environments were affected by the number of SRB, soil components and temperature.

  • Extensive previous studies on taxonomy, behavior/bionomics and control of Anopheles sinensis are reviewed and summarized. Recent molecular identification revealed that the population of An. sinensis complex includes An. sinensis, An. pullus, An. lesteri and at least two new species, and An. yatsushiroensis is synonmy of An. pullus. An. sinensis is the main vector specie of vivax malaria in Korea. Larvae of An. sinensis breed in wide range of habitats which are naturally-made clean water, stagnant or flowing; main habitats include rice fields, ditches, streams, irrigation cannals, marshes, ponds, ground pools, etc. Their host preferences are highly zoophilic. Human blood rate is very low ($0.7-1.7\%$); nevertheless An. sinensis readily feeds on man when domestic animals are not found near by. They feed on hosts throughout the night from dusk to dawn with a peak period of 02:00-04:00 hours; they are slightly more exophagic (biting outdoors); much larger numbers come into the room when light is on. Main resting places are outdoors such as grasses, vegetable fields and rice fields. A mark-release-recapture study resulted that $37.1\%$ was recaptured within 1 km, $29.4\%$ at 1-3 km, $21.1\%$ at 3-6 km, $10.3\%$ at 6-9 km and $2.1\%$ at 9-12 km distance. An. sinensis hibernate outdoors (mostly under part of dense grasses) during October-March. At the end of the hibernation period (March-April) they feed on cows at daytime. Until today any single measure to effectively control An. sinensis population has not been found. Indoor residual spray with a long-lasting insecticide can not reduce vector population densities, but shorten their life spans in some degree, so contributes to malaria control.

  • A field experiment was conducted to investigate the performance of three legume species in a zero-tillage, non-fertilized rice field in a temperate zone. Before the experiment for 5 years, from 1995 to 1999, plant growth patterns of three legume species grown as over-wintering (October-May) cover crops on a paddy field were maintained to study N balance and $\textrm{N}_2$ fixation. Decrease in plant density accelerated from after winter to flowering from 1,090, 320, and 5 to 732, 232, and 6 plants $\textrm{m}^{-2}$ in Chinese milk vetch (CMV), white clover (WC), and hairy vetch (HV), respectively. Total dry weights of plants above-ground level were 0.05, 0.11, and 2.43 g $\textrm{plant}^{-1}$. in WC, CMV and HV respectively but steeply increased at ripening up to 0.77, 2.33, and 26 g $\textrm{plant}^{-1}$. The root dry weight of HV and CMV rapidly increased while in WC, root dry weight increased slightly towards flowering. The highest nodule numbers were recorded in CMV to April thereafter WC produced the highest. Nodule size was distributed within 7mm in CMV but it was larger in HV varying from 1 to 10mm. Shoot N (g $\textrm{m}^{-2}$) greatly increased from over-wintering to flowering in CMV, HV and WC and it ranged from 1.66, 0.5 and 1.92 to 12.6, 3.1 and 13.02 g $\textrm{m}^{-2}$, respectively. After wintering, the initial shoot N content (%) was more in CMV. Root N content (%) was constant or slightly decreased in HV and WC. Soil total N in the control plot (clean fallow) was the highest on Mar. 2 then decreased rapidly to flowering. Soil N content was constant in HV plots whereas it was low in WC plots for the entire growth period except just after winter. Maximum nitrogenase activities were 9, 37.8, and 131 mol $\textrm{C}_2\textrm{H}_4$ $\textrm{plant}^{-1}$ $\textrm{hour}^{-1}$. in CMV, HV, and WC, respectively. Nitrogenase activity showed a direct correlation with nodule number, size and fresh weight. As a cover crop preceding a rice crop, CMV is more suited to colder regions due to its earlier ripening characteristics. Hairy vetch and WC are recommended for regions with a mild winter and a long summer owing to their late ripening and great N fixation activity.

  • Ametryn is used in USA, China, and Japan, but not introduced in Korea yet. So, MRL (Maximum Residue Level), and analytical method of ametryn were not establishment in Korea. Therefore, this experiment was conducted to establish a determination method for ametryn residue in crops using HPLC-UVD/MS. Ametryn residue was extracted with acetone from representative samples of five raw products which comprised hulled rice, soybean, apple, green pepper, and Chinese cabbage. The extract was diluted with saline water, and dichloromethane partition was followed to recover ametryn from the aqueous phase. Florisil column chromatography was additionally employed for final clean up of the extract. The ametryn was quantitated by HPLC with UVD, using a Tosoh ODS 120T ($4.6{\times}250$ mm) column. The crops were fortified with ametryn at 2 levels per crop. Mean recovery ratio were ranged from 83.7% for a 0.2 mg/kg in soybean to 91.1% for a 1.0 mg/kg in hulled rice. The coefficients of variation were ranged from 1.2% for a 1.0 mg/kg in hulled rice to 3.6% for a 1.0 mg/kg in soybean. Quantitative limit of amatryn was 0.02 mg/kg in representative 5 crop samples. A LC/MS with selected-ion monitoring was also provided to confirm the suspected residue. Therefore, this analytical method was reproducible and sensitive enough to determine the residue of ametryne in agricultural commodities.

  • The hydrolysis of defatted rice bran using near-critical water was performed, and the feasibility of consequent hydrolyzate as a growth medium was investigated by the cultivation of Saccharomyces cerevisiae. The near-critical water hydrolysis was carried out through a series of batch experiments, and the contents of total carbohydrates, disaccharides, and monosaccharides, total organic carbon (TOC), total nitrogen (TN), pH of products were measured. The growth rate of Saccharomyces cerevisiae was measured with optical density. The yield of total carbohydrates, TOC, and TN increased with temperature below $240^{\circ}C$, however, decreased above $240^{\circ}C$. The decrease of yields above $240^{\circ}C$ was caused by the formation of organic acids, and it agreed with the change of pH of products. The yield of glucose was a maximum at $200^{\circ}C$ and it decreased dramatically at higher temperature. The growth rate of Saccharomyces cerevisiae cultivated in the hydrolyzate was similar with that in the commercial medium under certain conditions. The growth rate was correlated with the content of glucose in hydrolyzate.

  • This study explored an efficient modified Quick, Easy, Cheap, Effective, Rugged and safe (QuEChERS) method combined with liquid chromatography-electrospray ionization with tandem mass spectrometric detection for the analysis of residues of 76 pesticides in brown rice, barley and corn including acidic sulfonylurea herbicides. Formic acid (1%) acid in acetonitrile and dispersive solid phase extractions used for extraction of pesticides and clean-up of the extract respectively. Two fortified spikes at 50 and 200 ng $g^{-1}$ levels were performed for recovery test. Mean recoveries of majority of pesticides at two spike levels ranged from 73.2 to 132.2, 80.9 to 136.8, 66.6 to 143.5 for brown rice, barley and corn respectively with standard error (CV) less than 10%. Good linearity of calibration curves were achieved with $R^2$ > 0.9907 within the observed concentration ranged. The modified method also provided satisfactory results for sulfonylurea herbicides. The method was applied to the determination of residues of target pesticides in real samples. A total of 26 pesticides in 36 out of 98 tasted samples were observed. The highest concentration was observed for tricyclazole at 1.17 mg $kg^{-1}$ in brown rice. This pesticide in two brown rice samples exceeded their MRLs regulated for rice in republic of Korea. Except tricyclazole none of the observed pesticides' concentration was higher than their MRLs. The results reveal that the method is effectively applicable to routine analysis of residues of target pesticides in brown rice, barley and corn.

  • Nitrogen balance in the regional scale which was calculated the difference between nitrogen input and output was estimated to assess the impact of rice cultivation on water environment. Nitrogen balances in Gyeonggi province, where nitrogen concentration in irrigation water was high and in Chungnam province, where nitrogen absorbtion by rice was high, were -5.4 and -8.3 kg $-8.3kg\;ha^{-1}\;yr^{-1}$, respectively. Nitrogen balances of paddy field in Gangwon province, where nitrogen output was small and irrigation water was clean, and in Gyeongnam province, where organic matter content of soil was high and rice yield was low, were 4.9 and $14.0kg\;ha^{-1}\;yr^{-1}$, respectively. Average nitrogen balance and total nitrogen absorption of paddy field in Korea were estimated to $-0.3kg\;ha^{-1}\;yr^{-1}$ and $-3,315Mg\;yr^{-1}$, respectively. When the nitrogen concentration in irrigation water was increased by $1mg \;L^{-1}$, nitrogen balance of rice paddy changed by $-2.91kg\;ha^{-1}\;yr^{-1}$. Also, when nitrogen fertilizer applied was decreased from 110 to $90kg\;ha^{-1}$ and the same harvest was maintained, the nitrogen absorption by rice paddy from irrigation water was estimated to increase by 10,600 Mg per year in Korea. However, in cases, the harvest was reduced to either 90% or 85%, nitrogen balances were changed from -11.7 to -2.3 and $2.4kg\;ha^{-1}$, respectively. These results suggest that the reduction of nitrogen fertilizer use may not always lead to a negative nitrogen balance and sustainable agriculture can achieve by not cutting down the use of fertilizer only but by reduction of fertilizer application concurrently by maintenance of harvest and by utilization of environmental characteristics such as nutrient contents in irrigation water and soils.
