Clinical and pathological studies on the feedlot cattle caused by Clostridium perfringens type A infection (Clostridium perfringens A형균 감염증에 의한 비육우의 임상 및 병리학적 연구)

  • Enterotoxemia caused by Clostridium perfringens(Cl. perfringens)has occurred sporadically with devastating effect in Korean cattle. The disease is confirmed by detecting bacterial toxin the interstinal contents. however diagnosis has been challenging since the toxin is short lived and it must be demonstrated within one hour following death. The purpose of this paper is to provide additional clinicall and pathological information derived from observation of cattle administered with Cl. perfringens type A and/or its toxins isolated from natural case. Clinically cattle died suddenly in lateral recumbency with dyspnea tachycardia and muscle tremor without increased rectal temperature. hematology showed leukopenia with absolute decrease of all leukocytes. Blood glucose AST and ALT were increased. Grossly lungs were hyperemic haemorrhagic and edematous. Small intestine especially ileum was hyperemic and hemorrhagic with diffuse areas of necrosis. This study demonstrated that death without increased rectal temperature combined with leukopenia increased serum glucose pulmonary haemorrhage and edema plus necrotizing ileitis are important features associated with enterotoxemia due to Cl. perfringens type.   • A total of 100 swelled, springered or flippered canned meat and fish products were studied the degree of contamination with clostridias and serological relationships to Hobbs'13 "heat resistant" types, heat resistance of spores and susceptibility of Clostridium perfringens isolates to several antibiotics. Samples examined in this study were collected from Seoul area from June to October, 1975 and prepared in Korea. Clostridias were isolated from 46(46%) of these samples; 19 strains of Cl. perfringens, 9 strains of Cl. oedematiens A, B, 5 strains of Cl. sordelli, each 3 strains of Cl. chauvoei, Cl, oedematiens C.E, and Cl. difficile, 2 strains of Cl. sporogenes. The highest percentage of contamination by Cl. perfringens was found in beef products(26.5%), and the following(5.2%) in mackerel pike and none in baitop shell. whale, manna brand. and top shell. One of 19 isolates of Clostridium perfringens found in meat products was shown to produce heat resistant spores which resist $100^{\circ}C$ for 60 minutes and others were heat labile strains which is killed at $90^{\circ}C$ for 30 minutes. The distribution of Hobbs' serotype of 19 isolates were each 4 strains of type 6, 8, and 11, 1 strain of type 13 and others untypable. 19 Strains of Cl. perfringens were shown a marked susceptibility to cefamezin, lincomycin and minocin and relatively sensitive to vibraimycin, geopen, and chloramphenicol. A marked resistance to kanamycin, colimycin, and gentamycin were shown. Aerobic enteropathogens from samples were not recovered.

  • This study was conducted to investigate the biochemical characteristics, the population and antibiotic susceptibility test of Clostridium perfringens isolated from intestinal contents of slaughter cattle in Kyung-ju and Po-hang. 1. In slaughter cattle Cl perfringens were isolated from intestinal contents of 51 of 101 cases(50.4%) and the population were $\leq$$10^5$cfu/ml of 44 cases(86.3%). 2. In antibiotic susceptibility test, ampicillin, bacitracin, cephalothin, penicillin polymyxin B were highly susceptible, ohloramphenicol, erythromycin, tetracycline were lowly susceptible, gentamicin, kanamycin, amikacin, streptomycin, sulfamethoxine, sulfamethazine were resistant. 3. In leaving test intestinal contents leaved for 0, 4, 8, 16, 32 hours in room temperature and population of Cl perfringens were gradually increased.

Effects of Edible Herbs on the Growth of In Vitro Intestinal Microorganisms (산채류가 장내세균의 In Vitro 생육에 미치는 영향)

  • This study was aimed to screen edible herbs which control the composition of intestinal microflora. With in vitro experiments, we screened the water or ethanol extracts of about 60 edible herbs and wild plants in terms of the inhibition activity on the growth of the harmful Clostridium perfringens and growth promoting activity for the beneficial Bifidobacteria. The water extracts of mugwort and small water dropwort inhibited the growth of Cl.perfringens both in agar diffusion method and broth culture. On the other hand, the water extracts of petasites, mugwort, yellow day-lily and bitter cress have shown the promotion effect on the growth of Bifidobacterium longum. In the culture test using human feces as starter, the extracts of the above selected herbs increased the population of Bifidobacteria and Lactobacillus while they reduced the numbers of Cl.perfringens and E.coli.

  • Eighteen strains of Clostpidium perfringens were isolated from the piglets with hemorrhagic enteritis. The characteristics of the outbreaks, clinical signs and lesions were examined. The biochemical properties, type of toxins and susceptibility to antimicrobial agents against the isolates were investigated. 1. The incidence of diarrhea was appeared in 97(22.4%) of 432 piglets examined. 2. The isolation rate of Cl perfingens from the 97 diarrheal faeces were 18.5%(18 strains) 3. The population of Cl perfingens in feces were ranged $10^{8-9}$cfu/g in 5(32.5%) and $10^{3-7}$cfu/g in 13(67.4%) of 18 samples. 4. The toxin type of the 18 isolates investigated by mouse inoculation test was all type C strains of Cl perfringens. 5. As a results of antimicrobial susceptibility test, 18 isolates were higly susceptible to cephalothin, tetracycline and penicillin.

Selection of Human-Originated Lactobacillus acidophilus For Production of Probiotics

  • Lactobacillus acidophilus KFRI 233, a strain isolated from human, was selected as a candidate for probiotics due to its excellent growth in MRS broth where no special anaerobic condition is required. Both simultaneous and deferred agar diffusion assays exhibited Lb. acidophilus KFRI 233 to possess an antagonistic effect against Clostridium perfringens. Its antagonistic effect was pH dependent Associative culture of KFRI 233 and Cl. perfringens in broth resulted in maximum 94.04% inhibition of Cl. perfringens. $\beta$-Galactosidase activity of KFRI 233 was higher than other tested strains that are sold as commercial probiotics. Survival of KFRI 233 in pasteurized skim milk (4$^{\circ}C$) and Sherbet mix (-15$^{\circ}C$) after 7 days of storage were 71.9 and 105.5%, respectively.

  • About Clostridium perfringens causing clinically necrotic enteritis or isolated from the intestinal contens of healthy chicken, We examined the usefulness of a rapid identification method by gas-liquid chromatography as well as the types of toxins. For this study, there were used 169 chickens including 116 broilers, 27 layers and 26 breeders which collected from 9 healty flock and 21 diseased flock showing necrotic enteritis. Among them, Cl perfringens was isolated from 30 chickens(17.8%) including 7 breeders(26.9%), 5 layers(18.5%) and 18 broilers(15.5%). Isolation of Cl perfringens was mainly from ceca (100%) and followed by small intestines(70.0%) and livers(16.7%), respectively. Average concentration of the pathogen in intestinal contents was $10^{3.8}CFU/g$ in cases occuring necrotic enteritis and on the contrary $10^{3.8}CFU/g$ in healthy cases. All isolates tested showed the same characterstics in biochemical tests compared to those in standard strain. Analysis of gas-liquid chromatography to volatile fatty acids produced by Cl perfringens in PYG broth showed the typical peaks of acetic and butyric acids compatible with the standard chromogram and was confirmed as a effective and reliable tool for rapid identification of the bacteria. Toxin types of 30 strains were mostly classified in A type(26 isolates) and the rest in C type(2 isolates) and unidentifed type(2 isolates). All the isolates were highly susceptible to amphicillin, amoxicillin and cephalothin.

  • The case reports for clostridium type A enterotoxemia in Formosan deer have rarely been reported. This paper describes a natural case of type A enterotoxemia in farmed Formosan deer in Cheongwon-gun. A dead, male 10-month-old Formosan deer was submitted to Chungbuk Livestock and Veterinary Research Institute, March 24, 2001 and examined. That deer was fed with assorted grain feed, oak leaves, acorn and bean curd. Grossly there was no visible external change. Despite of the carcass being examined within 12 hours of death, there was a quite degree of posonortem decomposition. There was severe hemorrhage in the serosa of abomasum and small intestine. Much blood tinged and watery contents were contained in those organs. Also there were severe swelling of spleen, some red foci in hepatic parenchyma. Microscopically there were severe congestion and hemorrhage in mucosa submucosa, muscular layer, and serosa of abomasum and small intestine. Also spleen and pancreas showed severe Congestion and hemorrhage. There were multifocal hemorrhage with hepatic necrosis in periportal area and focal mononuclear cell deposition in sinusoid. In bacterial culture for small intestine, Cl perfringens was isolated. By toxin typing for the strain, that had $\alpha$ -toxin belonged to type A. In electronmicroscopy for feces, no vims particle was detected. Considering clinical signs, gross lesions, microscopic lesions, bacterial culture, and toxin typing of the isolate, this case was diagnosed as enterotoxemia by Cl perfringens type A.

Quantitative Microbial Risk Assessment of Clostridium perfringens on Ham and Sausage Products in Korea (햄 및 소시지류에서의 Clostridium perfringens에 대한 정량적 미생물 위해평가)

  • This study was conducted for quantitative microbial risk assessment (QMRA) of Clostridium perfringens with consumption on ham and sausage products in Korea, according to Codex guidelines. Frame-work model as product-retail-consumption pathway composed with initial contamination level, the time and temperature in distributions, and consumption data sets for ham and sausage products and also used the published predictive growth and dose-response models for Cl. perfringens. The simulation model and formulas with Microsoft@ Excel spreadsheet program using these data sets was developed and simulated with @RISK. The probability of foodborne disease by Cl. perfringens with consumption of the ham and sausage products per person per day was estimated as $3.97{\times}10^{-11}{\pm}1.80{\times}10^{-9}$. There were also noted that limitations in this study and suggestion for development of QMRA in the future in Korea.

Effects of the Water-extract of Sinomeniiacuti Radix(Sinomenium acuturn) on the Growth of Some Intestinal Microorganisms (방기(Sinomenium acutum)의 물추출물이 주요 장내 미생물의 생육에 미치는 영향)

  • The water extract of Sinomeniiacuti Radix (Sinomenium acutum) was found out to have a strong inhibition activity on the growth of Clostridium perfringens. The anti-bacterial activity was stable at the range of pH 1 to pH 13 and kept in a thermal treatment at $121^{\circ}C$ for 15 minutes. The minmal inhibition concentration of the Sinomeniiacuti Radix extract on the growth of Cl. peifringens was 1000 ppm. The Sinomeniiacuti Radix extract also suppressed the growth of Cl. ramosum, Cl. paraputnficum, Cl. butyn'cum, Bifidobacterium blficum, Bacteriodes fragilis, Eubacterium limosum. The extract, however, did not inhibit the growth of Bif adolescentis, Blf. infantis, Bif longum, E. coli. Enterococcus faecalis and Staphylococcus aureus. On the other hand, the extract showed a promoting effect for the growth of Bif animalis and Lactobacillus acidophilus.
