감귤 embryogenic callus 원형질체 배양에 의한 식물체 재분화 (Plant regeneration from protoplasts-derived from embryogenic callus of Citrus)

  • 감귤에 있어서 원형질체 융합에 의한 체세포잡종체 생산을 위해서는 우선 캘러스 원형질체로부터의 식물체 재분화가 가능하여야 하기 때문에 온주밀감의 캘러스 원형질체로 부터 배형성과정을 통한 식물체 재분화에 관한 실험을 수행하였다. 흥진조생의 어린 미성숙 배주의 주심조직으로부터 유기된 배형성 캘러스를 사용하여 건강한 원형질체를 분리하고 0.6 M $BH_3$ 배지를 배양배지로 사용하여 배양방법에 따른 plating efficiency를 비교해 본 결과 liquid over solid 배양시 원형질체로부터의 미소괴 형성율이 더 높음을 알 수 있었으며, 적정 배형성 배지 선발에서는 1500 mg/L malt extract 첨가 배지에서 배형성율이 높았다. 자엽형 배로부터의 신초유기를 위한 배지비교에서는 1.0 mg/L GAB 첨가배지에서 발근과 함께 정상적으로 재분화된 신초를 얻을 수 있었다. 원형질체 배양으로부터 재분화된 식물체들은 순화과정을 거쳐 온실 육묘중에 있으며, 생장상이나 형태적인 특성에 있어서 모본 식물체와 차이가 없음을 확인할 수 있었다. 본 실험으로 얻어진 원형질체 배양에 의한 식물체 재분화 체계를 바탕으로 이종속간 감귤의 원형질체 융합 기술을 이용하여 교배육종이 불가능한 품종들로부터 우수한 형질을 지닌 감귤 품종 및 대목용 품종 생산에 활용하고자 한다.

떫은감 진피 복합추출물의 급성 역류성 식도염 개선 효과 (Improving Effects on Rats with Reflux Esophagitis Treated with Combined Extract of Young persimmon fruit and Citrus peel)

  • Objectives : The present study was conducted to evaluate protective effects of Combined Extract of young persimmon fruit and citrus peel (PCM) in Reflux Esophagitis(RE) rats.Methods : Twenty-four Sprague-Dawley (SD) rats were divided four groups and each group had six rats ; Normal group, RE control group, RE group treated PCM 50 ,100 mg/kg body weight group. Reflux esophagitis was induced that tied the pylorus and fundus in SD rats stomach. PCM was administered at 50, 100 mg/kg body weight 2 hrs prior to induction of RE. After 6 hrs, the effects of PCM treated rats were compared with those of normal and control rats. We have performed an analysis such as pH of stomach secretion, oxidative stress biomarkers in serum, and western blot.Results : The increased esophageal mucosa damage by RE was markedly improved by PCM treatment in a dose-dependent manner. Also, the administration of PCM decreased the elevated serum reactive oxygen species (ROS) and peroxynitrite (ONOO-) in serum. The protein expressions of anti oxidant such as SOD, catalase, GPx exhibited down-regulation by PCM treatment in tissues. And, PCM effectively reduce inflammatory cytokines such as inflammation-related proteins cyclooxygenase-2 (COX-2), inducible nitric oxide synthase (iNOS), Tumor necrosis factor-α (TNF-α), interleukin-6 (IL-6) in RE rats. In addition, NFκB and p-IκBɑ were decreased in PCM-adiministrated RE rats. But there was no difference on stomach secretion pH between reflux esophagitis rats and PCM administration rat group.Conclusions : In conclusion, administration of PCM (50, 100 mg/kg body weight) made esophagus have less inflammation and injury by decreased NFκB path way. These findings suggest that PCM could have Improving effects on reflux esophagitis.

In vitro Study and Clinical Trial of Natural Essential Oils and Extract Against Malassezia Species

  • Background: Malassezia, a lipophilic yeast, is a causative agent for dandruff and seborrheic dermatitis. Many biological agents have been studied for anti-Malassezia effect but further studies are needed for their clinical application. Objective: The study was conducted to evaluate the inhibitory effect of different natural essential oils and a fruit extract on Malassezia species in an in vitro study and a clinical trial. Methods: The antifungal effects of natural essential oils and a fruit extract on Malassezia species (M. furfur and M. sympodialis) were evaluated by measuring the minimum inhibitory concentration (MIC) and minimum fungicidal concentration (MFC) and using the disc diffusion method. Natural essential oils of citron seed, lavender, and rosemary and citrus junos fruit extract were used for the in vitro study. The clinical trial was conducted with a shampoo containing four ingredients. A total of 22 subjects used the shampoo every day for 4 weeks and were evaluated using clinical photography, trichoscopy, and sebumeter at baseline, 2 weeks, and 4 weeks after treatment. Results: Antifungal activity of agents was relatively lower in lavender and rosemary essential oils at MIC and MFC. Disc diffusion method revealed same results. In the clinical trial, the amount of sebum decreased statistically significantly and erythema, dandruff, and lesion extent also improved. Conclusion: The natural essential oils and fruit extract are effective for suppressing Malassezia activity, therefore these might be used as an alternative for treatment of dandruff and seborrheic dermatitis.

Anti-inflammatory Polymethoxyflavones Isolated from the Branches of Shiranuhi Tree

  • Shiranuhi is a fruit of Citrus species widely cultivated in Jeju Island, Korea. From an extract of Shiranuhi tree branches were identified five polymethoxyflavones possessing anti-inflammatory effects; nobiletin (1), sinensetin (2), tetramethylscutellarein (3), 6-hydroxy-5,7,3',4'-tetramethoxyflavone (4) and 5-desmethylsinensetin (5). Evaluation of the activities was conducted by monitoring the production of nitric oxide (NO), prostaglandin E2 and pro-inflammatory cytokines (TNF-α, IL-1β, IL-6) as well as the levels of iNOS and COX-2 protein expression in LPS-induced RAW264.7 cells. Among the isolates, the compound 4 exhibited the most significant NO inhibition, and suppressed the levels of iNOS and related cytokines. Therefore, it was suggested that the extract and constituents from Shiranuhi tree branches could be useful as anti-inflammatory ingredient.

Biological Control of Phytophthora palmivora Causing Root Rot of Pomelo Using Chaetomium spp.

  • Phytophthora diseases have become a major impediment in the citrus production in Thailand. In this study, an isolate of Phytophthora denominated as PHY02 was proven to be causal pathogen of root rot of Pomelo (Citrus maxima) in Thailand. The isolate PHY02 was morphologically characterized and identified as Phytophthora palmivora based on molecular analysis of an internal transcribed spacer rDNA sequence. This work also presents in vitro evaluations of the capacities of Chaetomium spp. to control the P. palmivora PHY02. As antagonists, Chaetomium globosum CG05, Chaetomium cupreum CC3003, Chaetomium lucknowense CL01 inhibited 50~61% mycelial growth, degraded mycelia and reduced 92~99% sporangial production of P. palmivora PHY02 in bi-culture test after 30 days. Fungal metabolites from Chaetomium spp. were tested against PHY02. Results showed that, methanol extract of C. globosum CG05 expressed strongest inhibitory effects on mycelial growth and sporangium formation of P. palmivora PHY02 with effective dose ED50 values of $26.5{\mu}g/mL$ and $2.3{\mu}g/mL$, respectively. It is interesting that C. lucknowense is reported for the first time as an effective antagonist against a species of Phytophthora.

Reactive oxygen species-dependent apoptosis induction by water extract of Citrus unshiu peel in MDA-MB-231 human breast carcinoma cells

  • BACKGROUND/OBJECTIVES: Although several recent studies have reported the anti-cancer effects of extracts or components of Citrus unshiu peel, which has been used for various purposes in traditional medicine, the molecular mechanisms for their effects remain unclear. In the present study, the anti-cancer activity of a water-soluble extract of C. unshiu peel (WECU) in MDA-MB-231 human breast carcinoma cells at the level of apoptosis induction was investigated. MATERIALS/METHODS: Cytotoxicity was evaluated using the MTT assay. Apoptosis was detected using DAPI staining and flow cytometry analyses. Mitochondrial membrane potential, reactive oxygen species (ROS) assay, caspase activity and Western blotting were used to confirm the basis of apoptosis. RESULTS: The results indicated that WECU-induced apoptosis was related to the activation of caspase-8, and -9, representative initiator caspases of extrinsic and intrinsic apoptosis pathways, respectively, and caspase-3 accompanied by proteolytic degradation of poly(ADP-ribose) polymerase and down-regulation of the inhibitors of apoptosis protein family members. WECU also increased the pro-apoptotic BAX to anti-apoptotic BCL-2 ratio, loss of mitochondrial membrane potential and cytochrome c release from mitochondria to cytoplasm. Furthermore, WECU provoked the generation of ROS, but the reduction of cell viability and induction of apoptosis by WECU were prevented when ROS production was blocked by antioxidant N-acetyl cysteine. CONCLUSIONS: These results suggest that WECU suppressed proliferation of MDA-MB-231 cells by activating extrinsic and intrinsic apoptosis pathways in a ROS-dependent manner.

수박 과실썩음병 병원균(Acidovorax avenae subsp. citrulli)에 대한 식물유래 항균 활성물질 탐색 (Search for Plant-originated Antibacterial Compounds Against Pathogen (Acidovorax avenae subsp. citrulli) of Watermelon Bacterial Fruit Blotch)

  • 본 연구는 수박 과실썩음병의 원인균인 Acidovorax avenae subsp. citrulli 균에 대해서 항균활성을 갖는 친환경 유기농자재를 개발할 목적으로 약용식물 133종을 대상으로 진행되었다. 이들 133종 약용식물의 MeOH 추출물에 대한 bioassay를 통해 항균 활성을 검정한 결과 청피(Citrus unshiu Markovich) 추출물에서 강한 항균활성을 보였다. 청피(Citrus unshiu Markovich)로부터 항균활성물질을 구명하고자 용매분획을 하였고, 용매분획 중에서 hexane fraction이 가장 강한 활성을 나타내었다. Hexane fraction을 GC-MS로 분석하여 chromatogram상의 각각의 peak에 해당하는 mass spectrum과 Wiley library를 비교하여 profiling 한 결과, essential oil인 d-limonene, ${\gamma}$-terpinene, ${\beta}$-linalool, terpineol과 지방산인 palmitic acid, 9,12-octadecadienoic acid, linolenic acid 그리고 steroid 화합물인 stigmasterol이 검출되었다. 이들 검출화합물 중에서 항균 활성물질로 추정되는 d-limonene, ${\gamma}$-terpinene, ${\beta}$-linalool, terpineol의 항균 활성을 검정하기 위해 표준품을 사용하여 bioassay한 결과 두 화합물 d-limonene, ${\gamma}$-terpinene에서 높은 항균활성을 보였다. 따라서 본 연구를 통해 청피로부터 분리한 d-limonene과 ${\gamma}$-terpinene 화합물이 항균 활성물질인 것을 구명하였다. 항균력이 강한 청피 추출물 또는 d-limonene과 ${\gamma}$-terpinene을 주성분으로 하는 추출물을 수박 과실썩음병에 대한 친환경 방제용 자제로 개발이 가능할 것으로 판단되었다.

국내산 유자씨박 (Citrus junos seed shell) 추출물 및 분획물의 항산화 활성 평가 (Antioxidant activities of Citrus junos seed shell extract and fractions cultivated in Korea)

  • 본 연구에서는 건조된 유자씨박 70% 에탄올 추출물 및 에틸아세테이트 분획물을 제조하고, 이들의 항산화능을 평가하였다. 유자씨박 추출물 및 분획물의 수율은 각각 5.1 및 0.9%로 나타났다. 1,1-Phenyl-2-picrylhydrazyl (DPPH) 라디칼 시험법에서 자유 라디칼 소거 활성($FSC_{50}$)은 70% 에탄올 추출물과 에틸아세테이트 분획물이 각각 512.1 및 $514.0{\mu}g/mL$를 나타냈다. 라디칼 소거활성은 (+)-${\alpha}$-tocopherol($9.0{\mu}g/mL$)보다는 비교적 낮았다. $Fe^{3+}$-EDTA계를 이용한 총 항산화능 평가에서 유자씨박 70% 에탄올 추출물 및 에틸아세테이트 분획의 $OSC_{50}$은 242.9 및 $86.5{\mu}g/mL$이었다. 유자씨박 에틸아세테이트 분획은 70% 에탄올 추출물 보다 높은 총 항산화능을 보였지만, 대조군인 L-ascorbic acid ($1.7{\mu}g/mL$)보다 낮게 나타났다. $^1O_2$로 유도된 세포 손상에 대한 보호 효과(${\tau}_{50}$)에서, 70% 에탄올 추출물은 5, 10, 25, $50{\mu}g/mL$에서 농도 의존적인 세포 보호 효과를 보였다. 유자씨박 에틸아세테이트 분획은 $5{\mu}g/mL$에서 39.8 min으로 (+)-${\alpha}$-tocopherol (36.1 min)과 유사한 세포 보호 활성을 나타났다. 그러나 $10{\mu}g/mL$ 이상에서 농도가 증가함에 따라 낮은 세포 보호 효과를 나타냈다. 결과적으로, 유자씨박 에틸아세테이트 분획물이 라디칼 소거활성이 아닌 총 항산화 활성을 통해 낮은 농도에서는 세포 보호 효과를 나타냈으나 높은 농도에서는 세포 보호 효과가 농도 의존적으로 나타나지 않았다.

식물 추출물의 Pityrosporum ovale 및 Candida albicans에 대한 항진균 활성 (Antifungal Activity of Plant Extracts against Pityrosporum ovale and Candida albicans)

  • For the purpose of searching for the new antifungal agent from the plant origin, we have examined twenty seven species of medicinal plants for the antifungal activity against Candida albicans and Pityrosporum ovale. The whole MeOH extract of each plant material and the n-hexane and EtOAc fractions were evaluated individually for the antifungal activity. Among the test materials, the fractions from the root bark of Zanthoxylum schinifolium, Zanthoxylum piperitum and Citrus unshiu showed significant antifungal activities in a dose dependent manner against Pityrosporum ovale and Candida albicans, respectively.

진피 추출물에 의한 인간유방암 MDA-MB-231 세포의 apoptosis 유도에서 ROS 및 AMPK의 역할 (Relationship between Reactive Oxygen Species and Adenosine Monophosphate-activated Protein Kinase Signaling in Apoptosis Induction of Human Breast Adenocarcinoma MDA-MB-231 Cells by Ethanol Extract of Citrus unshiu Peel)

  • 한의학에서는 진피(陳皮)라고 칭하는 귤의 껍질(Citrus unshiu peel) 추출물은 항산화, 항염증 및 항균 특성을 포함한 다양한 약리학적 효능을 갖는 것으로 알려져 있다. 최근 그들의 항암 활성에 대한 가능성이 보고되었지만 정확한 기전 연구는 여전히 미비한 실정이다. 본 연구에서는 인간 유방암 MDA-MB-231 세포를 대상으로 진피 에탄올(EECU, ethanol extract of C. unshiu peel) 추출물의 항암 효능을 평가하고 그에 따른 기전 연구를 수행하였다. 본 연구의 결과에 의하면 EECU에 의한 MDA-MB-231 세포의 증식억제는 세포사멸(apoptosis) 유도와 관련이 있었다. EECU에 의한 apoptosis는 caspase-8, -9 및 -3의 활성화와 IAPs 계열의 발현 감소에 따른 PARP의 분해와 Bax : Bcl-2 비율의 증가와 연관이 있었다. 또한 EECU는 Bid의 truncation과 함께 미토콘드리아 막 잠재력의 감소와 세포질로 cytochrome c의 이동을 촉진시켰다. 아울러 EECU는 AMPK 및 ACC의 인산화를 촉진시켰으나, AMPK 효소 활성의 저해제는 EECU에 의한 apoptosis 유도와 생존력 저하를 현저하게 억제하였다. 부가적으로 EECU는 AMPK 활성화의 상류 신호로 작용하는 활성산소종(ROS)의 생성을 촉진시켰으며, 강력한 항산화제인 NAC는 EECU에 의한 AMPK의 활성화와 apoptosis를 역전시켰다. 결론적으로 EECU는 ROS/AMPK 의존적인 내인성 및 외인성 apoptosis 경로를 활성화시킴으로써 MDA-MB-231 세포 증식을 억제하였음을 알 수 있었다.
