The volatile composition of Kiyomi peel oil cultivated in Korea was studied by using gas chromatography and gas chromatography-mass spectrometry. The peel oil from the Kiyomi fruit was prepared by using a cold-pressing extraction method. Among the 65 components quantified in Kiyomi oil, 25 terpene hydrocarbons and 40 oxygenated compounds were identified, with peak weight percentages measuring 94.5% and 4.9%, respectively. Limonene was the predominant compound (87.5%), followed by myrcene (2.4%), sabinene (0.9%), $\alpha$-pinene (0.8%), $\beta$-sinensal (0.8%), (Z)-$\beta$-farnesene (0.7%), neryl acetate (0.6%), valencene (0.5%), $\alpha$-farnesene (0.5%), and $\alpha$-sinensal (0.5%). A unique characteristic of the volatile profile of the Kiyomi oil was the proportion of aldehydes (2.7%), which resulted from the relative abundance of $\alpha$- and $\beta$-sinensal. Another unique characteristic of the Korean Kiyomi oil was its relative abundance of $\beta$-sinensal, (Z)-$\beta$-farnesene, neryl acetate, valencene, $\alpha$-sinensal and nootkatone. Valencene and $\alpha$- and $\beta$-sinensal were regarded as the influential components of Korean Kiyomi peel oil.

The application of 112 samples of Citrus fruit peel, which is commonly cultivated in Korea as crude drug, has been limited due to the absence of criteria concerning chemical constituents and application of itself. In the context, this study is on the analysis of 11 flavonoid compounds. As a result, the flavonoid pattern could be classified into 5 groups. Furthermore, by utilizing this HPLC data, this study could provide a basis for fruit peel applications of not only Citrus unshiu, but also its cultivars, hybrids and some of Citrus plants which has been cultivated in Jeju Island for a long time. Moondan could be used for the same purposes as Chinese Huajuhong, and Poncirus trifoliata of Korean Jisil origin are effective enough, in terms of ingredients and function, compared with C. aurantium. Based on the content and composition of the PMF region in this study, C. unshiu and C. reticulata, as Tanaka System shows, should be considered as different hortcultural speicies. Also, Jikak, which is known as 'sour orange' until now, is confirmed as Chosen-daidai, C. tenuissima, that was reported by Tanaka in 1970, and this C. tenuissima could be used for the same purposes as sour orange.
Essential oils were isolated from the peel of mandarin (Citrus unshiu Marc. Var. Okitsu) cultivated in Cheju Island. Among three different isolation methods investigated, simultaneous distillation gave higher yield 1.14%(w/w) than solvent extraction or cold pressing. The densities of essential oils were between 0.8409 and 0.8530, which showed no significant effect of the isolation methods. Some variations in the number of constituents detected and the content of d-limonene in the peel oils were observed depending upon the isolation methods. The citrus fruit was collected seasonally from the field and used as a sample for analysis. The peel thickness and the peel content of citrus fruit were around 2 mm and about 17% on wet weight basis, respectively, in harvest season. The maximum yield of essential oils was obtained in September from citrus peels just prior to ripeness and thereafter the yield had a tendency of decrease but remained over 1%(w/w). The relative content of d-limonene, the major constituent of citrus peel oils. increased gradually with ripening up to 68.69%(relative peak area, RPA). The other minor constituents were ${\gamma}-terpinene,\;{\beta}-elemene$, farnesene, hexadecanoic acid, ${\alpha}-pinene,\; {\beta}-myrcene$ and linalool in the decreasing order of their own contents of 7.75, 2.96, 2.29, 1.76, 1.63, 1.56 and 1.46%(RPA).

Citrus and its peels, which are by-products from juice and/or jam processing, have long been used in Asian folk medicine. Citrus peels show an abundant variety of flavanones, and these flavanones have glycone and aglycone forms. Aglycones are more potent than glycones with a variety of physiological functions since aglycone absorption is more efficient than glycones. Bioconversion with cytolase converted narirutin and naringin into naringenin and hesperidin into hesperetin. Therefore, this study aimed to investigate the anti-oxidant and anti-inflammatory effects of bioconversion of Citrus unshiu (CU) peel extracts with cytolase (CU-C) in RAW264.7 cells. HPLC chromatograms showed that CU and CU-C had 23.42% and 29.39% total flavonoids, respectively. There was substantial bioconversion of narirutin to naringenin and of hesperidin to hesperetin. All citrus peel extracts showed DPPH scavenging activities in a dose-dependent manner, and CU-C was more potent than intact CU. RAW264.7 cells were pre-treated with $0{\sim}500{\mu}g/mL$ of citrus peel extracts for 4 h and then stimulated by $1{\mu}g/mL$ of lipopolysaccharide (LPS) for 8 h. All citrus peel extracts showed decreased mRNA levels and protein expression of LPS-induced inducible nitric oxide synthase (iNOS) and cyclooxygenase-2 (COX-2) in a dose-dependent manner. Especially, CU-C markedly inhibited mRNA and protein expression of iNOS and COX-2 compared to intact citrus peel extracts. All citrus peel extracts showed decreased NO production by iNOS activity. This result suggests that bioconversion of citrus peel extracts with cytolase may provide potent functional food materials for prevention of chronic diseases attributable to oxidation and inflammation by boosting the anti-inflammatory effects of citrus peels.

Objectives : Fermentation of herbs has been known to be helpful in improving the immune systems and protecting body against disease. The present study was conducted to evaluate anti-inflammatory effects of the fermentation extracts (FE) consisting of soybean, red ginseng andCitrus UnshiuPeel in lipopolysaccharide (LPS)-activated Raw264.7 cells.Methods : FE were prepared by the fermentation withBacillus Subtilisand then by extraction with ethanol (95%; prepared by the fermentation process). Cell viability was measured by MTT assay. Nitric oxide (NO) production was measured in culture media by Griess assay. The expression of nuclear factor (NF)-κB and inhibitory kappa B alpha (IκBα) was determined by Western blot.Results : LPS-induced production of NO and PGE2was dose-dependently decreased by the treatment of FE in Raw264.7 cells. These suppressive effects of FE on NO and PGE2production were related to the inhibition of inducible nitric oxide synthase (iNOS) and cyclooxygenase-2 (COX-2) expression. FE inhibited LPS-induced production of pro-inflammatory cytokines, TNF-α, IL-6, and IL-1βin a dose-dependent manner. Furthermore, FE inhibited the NF-κB signaling pathway through the prevention of LPS-induced degradation of IκBαin cytosol and the nuclear translocation of NF-κB.Conclusions : These findings suggest that FE could have anti-inflammatory effects on LPS-induced inflammatory responses in macrophages.
Objectives: Excessive melanin production leads to skin pigmentation, which causes various cosmetic and health problems. Citrus unshiu Markovich peel (CS) and Cimicifuga dahurica (CD) have long been widely used as a oriental medicinal plant because of their pharmacological properties including anti-ulcer, anti-oxidant, and anti-inflammatory properties. Methods: In this study, we investigated the inhibitory effects of CS, CD extracts or CS:CD=1:2 mixture on melanogenesis according to the various extraction methods. CS and CD extracted were prepared by ethanol extraction (EE), ultrasonification extraction (USE), Supercritical extraction (SCE), reflux extraction (RE), respectively. Results: DPPH radical scavenging and tyrosinase inhibitory activity of CD extracts or CS:CD=1:2 mixture were increased in dose-dependent manners. In addition, we evaluated the effect of CS, CD extracts or CS:CD=1:2 mixture on tyrosinase activity and melanogenesis in α-melanocyte stimulating hormone-induced B16-F10 melanoma cells. CS, CD extracts or CS:CD=1:2 mixture significantly inhibited tyrosinase activity and melanogenesis at 10-200 ㎍/mL. Conclusions: Therefore, our study suggests that CS and CD extracts have potential as a safe treatment for excessive pigmentation or as a natural ingredient in cosmetics.
BACKGROUND/OBJECTIVES: Citrus flavonoids have a variety of physiological properties such as anti-oxidant, anti-inflammation, anti-cancer, and anti-obesity. We investigated whether bioconversion of Citrus unshiu with cytolase (CU-C) ameliorates the anti-adipogenic effects by modulation of adipocyte differentiation and lipid metabolism in 3T3-L1 cells. MATERIALS/METHODS: Glycoside forms of Citrus unshiu (CU) were converted into aglycoside forms with cytolase treatment. Cell viability of CU and CU-C was measured at various concentrations in 3T3L-1 cells. The anti-adipogenic and lipolytic effects were examined using Oil red O staining and free glycerol assay, respectively. We performed real time-polymerase chain reaction and western immunoblotting assay to detect mRNA and protein expression of adipogenic transcription factors, respectively. RESULTS: Treatment with cytolase decreased flavanone rutinoside forms (narirutin and hesperidin) and instead, increased flavanone aglycoside forms (naringenin and hesperetin). During adipocyte differentiation, 3T3-L1 cells were treated with CU or CU-C at a dose of 0.5 mg/ml. Adipocyte differentiation was inhibited in CU-C group, but not in CU group. CU-C markedly suppressed the insulin-induced protein expression of CCAAT/enhancer-binding protein ${\alpha}$ ($C/EBP{\alpha}$) and peroxisome proliferator-activated receptor gamma ($PPAR{\gamma}$) as well as the mRNA levels of $CEBP{\alpha}$, $PPAR{\gamma}$, and sterol regulatory element binding protein 1c (SREBP1c). Both CU and CU-C groups significantly increased the adipolytic activity with the higher release of free glycerol than those of control group in differentiated 3T3-L1 adipocytes. CU-C is particularly superior in suppression of adipogenesis, whereas CU-C has similar effect to CU on stimulation of lipolysis. CONCLUSIONS: These results suggest that bioconversion of Citrus unshiu peel extracts with cytolase enhances aglycoside flavonoids and improves the anti-adipogenic metabolism via both inhibition of key adipogenic transcription factors and induction of adipolytic activity.
Physicochemical properties and sensory evaluation of Satsuma mandarin produced in Cheju were investigated . Fruit index(width/Length), soluble solids and acid content of Citrus unshiu marc. var. miyagawa(miyagawa) and C. unshiu marc. var.(okitsu) harvested at 1st December were 1.16 and 1.23, 12.12 and 12.01, 0.97 and 1.09, respectively. and the fruit of miyagawa was showed somewhat round in shape, compared to ikitsu. the taste of miyagawa seemed to be better than okitsu. However, compared to miyagawa, peel thickness was thinner, flesh ratio was higher, and firmness was higher as 0.502kg-force in kitsu. Nevertheless there were some differences among panelist groups in sensory evaluation , middle size of citrus fruits were favorable to most panelists, but except very small or very large size of fruits, there were not so much differences in okitsu. The panelists referred to somewhat flat-shaped fruits from that sensory evaluation score was increased to 1.3 in fruit index. nevertheless there were not so much differences in full-colored fruits, the citrus peel color of reddish yellow was more favorable than greenish or pale yellow color. the score was increased more linearly according to citrusfruit produced in optimum ciltivation area, and was prefered to thinner in peel thickness , Correlation between soluble solids, acid content , Brix/acid ratio and flesh ratio with sensory evaluation score were not showed significantly, it was different to with citrus juice. This data obtained in this experiments are supposed to be applied to the quality evaluation of Satsuma mandarin produced in Cheju .
This study was performed to investigate the effect of whole mandarin, peel, and pulp intake of Citrus unshiu Marc on the antioxidative capacity of 15-month-old rats. Forty-eight male Sprague-Dawley rats weighing 621.9 $\pm$ 10.1 g were separated into four groups according to body weight. The rats were raised with diets containing 5% (w/w) dried mandarin powder for four weeks. Three powders were used, consisting of mandarin peel, pulp, and the entire fruit. Total flavonoids, antioxidant vitamins and dietary fiber was highest in the mandarin peel powder, followed by the whole mandarin powder and the mandarin pulp. The body weight gains of the whole mandarin and mandarin pulp groups were higher, while that of the mandarin peel group was lower than that of the control group. Food intake and ratios of liver, kidney and epididymal fat pad (EFP) weights to body weight were not significantly different among the groups, but ratios of EFP weights per body weight of the experimental groups tended to be lower than that of the control animals. Plasma and liver TBARS concentrations decreased in all the mandarin groups compared to the control group. Plasma and liver xanthine oxidase (XO) activity decreased in all of the mandarin diet groups. Erythrocyte and liver SOD activity in all of experimental groups was not significantly different from the control group. Plasma vitamin A concentration increased significantly in all of the mandarin diet groups. That of the mandarin peel group was 4 times higher than that of the control group. Plasma total carotenoids and vitamin C level also increased in the mandarin peel group. Plasma vitamin I level was not significantly different among the groups.

This study was performed to investigate the effect of whole mandarin, peel or pulp intake of Citrus unshiu Marc. on antioxidative capacity and oxidative DNA damage in fifteen-month aged rats. Forty-eight male Sprague-Dawley rats $(621.9\;{\pm}\;10.1\;g)$ were blocked into four groups according to their body weights as control group, whole mandarin powder group, mandarin peel powder group and mandarin pulp powder group. Rats were raised with diets containing 5% (w/w) freeze dried mandarin formulations for four weeks. Total polyphenol content and total antioxidant status (TAS) of mandarin formulations were highest in peel powder, followed by whole powder and then pulp powder. The 8-hydroxy2'-deoxyguanosine concentrations of kidney in all mandarin groups were significantly lower than that of control group, and that of mandarin peel group was much lower than whole powder and pulp groups. Plasma TAS levels of all the experimental groups were higher than that of control group, and among mandarin groups, peel group showed higher level than remaining two groups. In conclusion, all the mandarin formulations were effective on antioxidative capacity in fifteen-month aged rats, and the peel was most effective one among three formulations.
