Analysis of Composition and Activity of Essential Oil from Chrysanthemum zawadskii var. latilobum and C. indicum against Antibiotic-Resistant Pathogenic Bacteria

  • The composition of essential oils from Chrysanthemum zawadskii var. latilobum and C. indicum were analyzed and compared. The results of gas chromatography-mass spectrometry revealed there were distinctly different compositional patterns between C. zawadskii var. latilobum and C. indicum essential oils. The combinatorial effect of the oil of C. zawadskii var. latilobum and C. indicum, with various antibiotics was assessed against antibiotic-susceptible and -resistant strains of Staphylococcus aureus and Streptococcus pneumoniae. The essential oil fraction significantly inhibited most of the tested antibiotic-susceptible and -resistant strains of S. pneumoniae, with minimum inhibiting concentrations (MICs) ranging from 0.5 to 4.0 mg/ml. The fractional inhibiting concentration indices (FICIs) of the oils when combined with antibiotics against S. aureus and S. pneumoniae ranged from 0.26 to 0.75, and showed synergistic or additive effects.

Volatiles of Chrysanthemum zawadskii var. latilobum K.

  • The volatile aroma constituents of Chrysanthemum zawadskii var. latilobum K. were separated by hydro distillation extraction (HDE) method using a Clevenger-type apparatus, and analyzed by gas chromatography-mass spectrometry (GC/MS). The yield of C. zawadskii var. latilobum K. flower essential oil (FEO) was 0.12% (w/w) and the color was light green. Fifty-five volatile chemical components, which make up 88.38% of the total aroma composition, were tentatively characterized. C. zawadskii var. latilobum K. FEOs contained 27 hydrocarbons, 12 alcohols, 7 ketones, 4 esters, 1 aldehyde, 1 amine, and 3 miscellaneous components. The major functional groups were terpene alcohol and ketone. Borneol (12.96), (${\pm}$)-7-epi-amiteol (12.60), and camphor (10.54%) were the predominant volatiles. These compounds can be used in food and pharmaceutical industries due to their active bio-functional properties.

  • Chrysanthemum zawadskii var. latilobum (Compositae), which is called Gu-Jul-Cho, has been used as traditional medicine in pneumonia, bronchitis, common cold, pharyngitis, bladder-related disorders, women's diseases, and hypertension, etc. In this study we have investigated various biological activities of methanol extracts and their main component, linarin isolated from this herb. The antipyretic effect of them was tested by Brewers yeast method and the antiinflammatory activities, with arachidonic acid, o-tet-radecanoylphorbol 13-acetate and carrageenan. Their antipyretic and antiinflammatory activities were more potent than acetaminophene and aspirin. They also showed stronger hepatoprotective activities than ones of silymarin in mice.

  • This study was made on three taxa of Chrysanthemum that grew on the Ewha Womans University campus on the basis of gross morphology, pollen epidermal patterns and chromosomes. The three taxa were recognized as Chrysanthemum zawadskii subsp. zawadskii var. acutilobum(2n=54); Chrysanthemum zawadskii subsp. latilobum (2n=36), and possible hybrid(2n=45) between the two taxa in view of leaf morphorlogy and chromosome number counts.

구절초와 쑥부쟁이 정유의 휘발성 향기성분 분석 (Analyses of the Volatile Flavor Composition of Essential Oils from Chrysanthemum zawadskii var. latilobum Kitamura and Aster yomena Makino)

  • This study investigated the volatile flavor composition of essential oils from Chrysanthemum zawadskii var. latilobum Kitamura and Aster yomena Makino. The essential oils obtained by the hydrodistillation extraction method from the aerial parts of the plants were analyzed by gas chromatography (GC) and GC-mass spectrometry (GC-MS). One hundred (95.04%) volatile flavor compounds were identified in the essential oil from the C. zawadskii var. latilobum Kitamura. The major compounds were valencene (10.82%), ${\delta}$-cadinol (9.77%), hexadecanoic acid (8.70%), 2-methyl-4-(2,6,6-trimethylcyclohex-1-enyl) but-2-en-1-ol (3.67%), and 2-(2,4-hexadiynylidene)-1,6-dioxaspiro[4,4]non-3-ene (3.57%). Ninety-eight (93.83%) volatile flavor compounds were identified in the essential oil from the Aster yomena Makino. The major compounds were and 3-eicosyne (13.61%), 9,10,12-octadecatrienoic acid (7.8%), ${\alpha}$-caryophyllene alcohol (6.83%), 9-octadecynoic acid (6.03%), and ${\alpha}$-caryophyllene (5.74%). Although the two plants are apparently very similar, the chemical composition of the essential oils was significantly different in quality and quantity. In the case of C. zawadskii var. latilobum Kitamura, the sesquiterpene, valencene was found to be 10.82%, but it was not identified in A. yomena Makino. ${\delta}$-Cadinol appeared higher in C. zawadskii var. latilobum Kitamura than in A. yomena Makino. A clear characteristic of A. yomena Makino essential oil is that it has a high content of caryophyllene derivatives. The ${\alpha}$-caryophyllene alcohol contained in A. yomena Makino was relatively high at 6.83%, although the compound was not identified in C. zawadskii var. latilobum Kitamura. Also ${\alpha}$-caryophyllene was shown to be higher in A. yomena Makino than in C. zawadskii var. latilobum Kitamura.

  • 본 연구는 전통약재로 주로 사용되는 구절초에서 분리한 다당류(CZPS)가 선천 및 적응면역에서 중추적인 역할을 수행하는 대식세포의 활성화에 미치는 영향에 관하여 관찰하였다. 구절초에서 분리한 다당류를 마우스 유래 대식세포주인 RAW 264.7 cell에 처리하였을 때, iNOS의 발현을 조절하여 NO의 생성이 증가되었으며, 또한 면역활성 물질인 TNF-${\alpha}$, IL-$1{\beta}$ 및 IL-6 등의 cytokine의 분비능이 증가되었으며, 이러한 면역활성 매개자인 NO 및 cytokine의 증가의 원인에 관한 정확한 면역세포내 신호전달에 관하여 알아본 결과, CZPS 처리는 MAPKs(ERK, p38)의 인산화를 촉진시키며, 이로 인한 전사인자인 NF-${\kappa}B$의 인산화를 증가시켜, 대식세포의 활성화를 유도시키는 것으로 관찰되었다.

등대시호(Bupleurum euphorbioides Nakai) 개체군의 생태 (Ecology of Bupleurum euphorbioides Population)

  • This study was to analyze vegetation and soil characteristic, and ordination of Bupleurum euphorbioides community located in Mt. Seorak area, Mt. Sobaek area, and Mt. South Deogyu area. Bupleurum euphorbioides community was classified into Carex lanceolata subcommunity and Chrysanthemum zawadskii var. alpinum subcommunity, and Chrysanthemum zawadskii var. alpinum subcommunity was classified into Calamagrostis arundinacea and Carex breviculmis group. In the study sites, soil organic matter, nitrogen, available phosphorous, changeable potassium, changeable calcium, changeable magnesium concentration, cation exchange capacity, and soil pH were ranged from 9.0~15.2%, 0.32~0.47%, 3.0~8.6ppm, 0.3~0.9me/100g, 0.5~1.5me/100g, 0.2~0.6me/100g, 11.4~16.1me/100g, and 4.3~4.6, respectively. And, soil texture was sandy loam or loam. The Carex breviculmis group of Chrysanthemum zawadskii var. alpinum subcommunity was found in area of high elevation and had high exchangeable K, Ca, and Mg concentration. The Calamagrostis arundinacea group of Chrysanthemum zawadskii var.alpinum subcommunity was found in area of low elevation and had lower exchangeable K, Ca, and Mg concentration than in the other subcommunity. The Carex lanceolata subcommunity was found in area of medium elevation and had medium exchangeable K, Ca, and Mg concentration. The Carex lanceolata subcommunity was located in a path of top the mountain peak, and has been destroyed. We need any action to protect the subcommunity.

유방암세포에서 구절초 추출물의 암전이 억제 효과 (Chrysanthemum zawadskii var. latilobum Extracts Inhibits of TPA-induced Invasion by Reducing MMP-9 Expression Via the Suppression of NF-${\kappa}B$ Activation in MCF-7 Human Breast Carcinoma Cells)

  • Chrysanthemum zawadskii Herbich var. latilobum Kitamura (Compositae), colloquially known "Gujulcho" in Korea, has been used in traditional medicine for the treatment of various diseases, including cough, common cold, bladder-related disorders, gastroenteric disorders, hypertension, and inflammatory diseases, such as pneumonia, bronchitis, pharyngitis, and rheumatoid arthritis (RA) However, the effect of Chrysanthemum zawadskii var. latilobum on breast cancer invasion is unknown. In this study, we investigated the inhibitory effect of Chrysanthemum zawadskii var. latilobum extract (CZE) on 12-O-tetradecanoylphorbol-13-acetate (TPA)-induced matrix metalloproteinase-9 (MMP-9) expression and cell invasion, as well as the molecular mechanisms involved in MCF-7 cells. CZE were not cytotoxic up to 100 ${\mu}g/ml$ concentration in the MCF-7 cell line. CZE decreased MMP-9 expression. TPA substantially increased NF-${\kappa}B$ DNA binding activity. Pre-treatment with CZE inhibited TPA-stimulated NF-${\kappa}B$ binding activity and NF-${\kappa}B$ related protein expression. To identify invasion ability of MCF-7 cells decreased by CZE, we used martrigel invasion assay. As a result, it is significantly decreased cell invasion. These results indicate that CZE-mediated inhibition of TPA-induced MMP-9 expression and cell invasion involves the suppression of the NF-${\kappa}B$ pathway in MCF-7 cells. Chrysanthemum zawadskii var. latilobum may have potential value in restricting breast cancer metastasis.

Sclerotium rolfsii에 의한 구절초 흰비단병의 발생 및 특징 (Occurrence and Characterization of Stem Rot of Chrysanthemum zawadskii var. latilobum Caused by Sclerotium rolfsii)

  • 2009년 9월에 전북 완주군 상관면 조경포장에 재배중인 구절초의 줄기와 지제부위가 수침상으로 무르고 썩으면서 암갈색으로 시들어 말라 죽었다. 병반부와 지제부위의 토양 표면에 흰색의 균사가 자라고 갈색의 구형 또는 부정형의 균핵이 형성되었다. 균사의 생육적온은 $25{\sim}30^{\circ}C$이었으며, 균사의 폭은 4-9 ${\mu}m$이었고, 균핵의 크기는 0.8-3.5(평균 1.8) mm이었다. 균사특유의 clamp connection이 관찰되었다. 분리한 균은 구절초에 대한 실험결과 병원성이 인정되었다. 구절초에 발생한 병징과 분리균의 균학적 특징 및 병원성을 검정한 결과, 이 병을 Sclerotium rolfsii에 의한 구절초 흰비단병으로 명명하고자 한다.

6-Methoxyluteolin from Chrysanthemum zawadskii var. latilobum Suppresses Histamine Release and Calcium Influx via Down-Regulation of $Fc{\varepsilon}RI$ ${\alpha}$ Chain Expression

  • Mast cells and basophils are important effector cells in immunoglobulin-E (IgE)-mediated allergic reactions. Using the human basophilic KU812F cells, we assessed the inhibitory effects of 6-methoxyluteolin, isolated from Chrysanthemum zawadskii, in the $Fc{\varepsilon}RI$-mediated allergic reaction. We determined that 6-methoxyluteolin inhibited anti-$Fc{\varepsilon}RI$ ${\alpha}$ chain antibody (CRA-1)-induced histamine release, as well as elevation of intracellular calcium concentration $[Ca^{2+}]_i$ in a dose-dependent manner. Moreover, the inhibitory effects of 6-methoxyluteolin on the cell surface expression and the mRNA level of the $Fc{\varepsilon}RI$ ${\alpha}$ chain were determined by flow cytometric analysis and reverse transcription-polymerase chain reaction (RT-PCR), respectively. Therefore, these results show that 6-methoxyluteolin is a potent inhibitor of histamine release and calcium influx via down-regulation of the $Fc{\varepsilon}RI$ ${\alpha}$ chain.
