Characteristics of Intestinal Microflora and Their Extracellular Pretense of Sardine, Sardinops melanosticta (정어리 내장세균의 특성과 균체외 단백분해효소에 관한 연구)

  • Sardine, Sardinops melanosticta, has been caught more than fifty thousand metric tons every year in adjacent sea of Korea, but most of them used for uneatable fish meal because of their rapid spoilage. Usually it is known that the main reason of putrefaction of foods is caused by the maicro-organisms included in them. Therefore, this experiment was carried out to identify the micro-organisms isolated from the intestine of fresh sardine and characterize their proteolytic enzymes produced from them. Aerobic cell count ranged from $1.7{\times}10^4\;to\;3.6{\times}10^5/g$, while anaerobic cell count, from $2.9{\times}10^4\;to\;5.5{\times}10^5/g$. Most of the isolated strains were psychrotrophic mesophiles. Among the two hundred and eighty strains isolated from the fresh samples, fifty-six strains ($20.0\%$) were proteolytics, one hundred and seventy-five strains ($62.5\%$) were lipolytics and tenty-nine strains ($10.5\%$) had the ability to produce hydrogn sulfide. The most predominantly isolated microbial groups from the fresh sardine were Moraxella ($31.4\%$) and Pseudomonas sup. ($28.6\%$). Flavobacterium-Cytophaga, Vibrio, Acinetobacter, Micrococcus spp. and Enterobacteriaceae appeared from $7.9\%\;to\;5\%$ out of total tested strains. The average bacterial count in the spoiled samples (stored at about $18^{\circ}C$ for 48 hours) was increased to the level of $2.9{\times}10^8/g$ for aerobes, $1.5{\times}10^8/g$ for anaerobes, then one hundred and ten strains, corresponding to $52\%$, out of two hundred and thirteen strains submitted to the test were proteolytics. The strongest proteolytic bacterium among the two hundred and eighty strains was identified as Pseudomonas 101 which grew best at $25^{\circ}C$. The optimum condition for the activity of the proteolytic enzyme produced by Pseudomonas 101 appeared $35^{\circ}C$ and pH 9.0, but the activity was relatively unchanged between 5.0 and 11.0 of pH and between $30^{\circ}C\;and\;50^{\circ}C$ of temperature.

Reaction Mechanism and Curing Characteristics of Chicken Feather-Based Adhesives and Adhesive Properties of Medium-Density Fiberboard Bonded with the Adhesive Resins (닭털로 제조한 접착제의 반응기작 및 경화 특성과 이를 이용하여 제조한 중밀도섬유판의 접착 특성)

  • In this study, reaction mechanism and curing characteristics of adhesives formulated with NaOH- and $H_2SO_4$-hydrolyzed chicken feather (CF) and formaldehyde-based crosslinkers were investigated by FT-IR and DSC. In addition, adhesive properties and formaldehyde emission of medium-density fiberboards (MDF) applied with the adhesives were measured. CF-based adhesives having a solid content of 40% and over were very viscous at $25^{\circ}C$, but the viscosity reduced to $300{\sim}660m{\cdot}Pa{\cdot}s$ at $50^{\circ}C$. Consequently, the adhesives could be used as a sprayable resin. Through the FT-IR spectra of liquid and cured CF-based adhesives, addition reaction of methylol group and condensation reaction between the functional groups with the use of formaldehyde-based crosslinkers were identified. From the analysis of DSC, it was elucidated for CF-based adhesives to require a higher pressing temperature or longer pressing time comparing to commercial urea-formaldehyde (C-UF) resin. MDF bonded with CF-based adhesives, which was formulated with 5% NaOH-hydrolyzed CF (CF-AK-5%) and PF of formaldehyde to phenol mole ratio of 2.5 (PF-2.5), and pressed for 8 min had higher MOR and IB than those with other CF-based adhesives. MOR and IB of MDF bonded with the CF-based adhesives regardless of formulation type and pressing time were higher than those with C-UF resin. When the values compared with the minimum requirements of KS standard, IB exceeded the KS standard in all formulations and pressing time, but MOR of only MDF bonded with CF-AK-5% and PF-2.5 and pressed for 8 min satisfied the KS standard. What was worse, 24-TS of MDF bonded with all CF-based adhesives did not satisfied the KS standard. However, MOR and 24-TS can be improved by increasing the target density of MDF or the amount of wax emulsion, which is added to improve the water resistance of MDF. Importantly, the use of CF-based adhesives decreased greatly the formaldehyde emission. Based on the results, we reached the conclusion that CF-based adhesives formulated under proper conditions had a potential as a sprayable resin for the production of wood panels.

  • Background: Biomarkers for cancer have several potential clinical uses, including the following: early cancer detection, monitoring for recurrence prognostication, and risk stratification. However, no biomarker has been shown to have adequate sensitivity and specificity. Many investigators have tried to validate biomarkers for the early detection and recurrence of lung cancer. To evaluate plasma G-CSF as such a biomarker, protein levels were measured and were found to correlate with the clinicopathological features of primary lung tumors. Methods: Between December 2006 and May 2008, 100 patients with histologically-validated primary lung cancer were enrolled into this study. To serve as controls, 127 healthy volunteers were enrolled into this study. Plasma G-CSF levels were measured in lung cancer patients using the sandwich ELISA system (R & D inc.) prior to treatment. Results: The mean plasma G-CSF levels were 12.2$\pm$0.3 pg/mL and 46.0$\pm$3.8 pg/mL (mean$\pm$SE) in the normal and in the cancer groups, respectively. In addition, plasma G-CSF levels were higher in patients with early lung cancer than in healthy volunteers (p<.001). Plasma G-CSF levels were higher in patients who were under 65 years old or smokers. Within the cancer group, plasma G-CSF levels were higher in patients with non small cell lung cancer than in patients with small cell lung cancer (p<.05). Overall, plasma G-CSF levels were shown to increase dependent upon the type of lung cancer diagnsosed. In the order from highest to lowest, the levels of plasma G-CSF tended to decrease in the following order: large cell carcinoma, squamous cell carcinoma, adenocarcinoma, and bronchioloalveolar carcinoma.   • Myristicin, l-allyl-3,4-methylenedioxy-5-methoxybenzene, was one of the major essential oils of nutmeg. However, its anti-allergic effect in the Th1/Th2 immune response was poorly understood. Recently, it was shown that T-bet and GATA-3 was master Th1 and Th2 regulatory transcription factors. In this study, we have attempted to determine whether myristicin regulates Th1/Th2 cytokine production, T-bet and GATA-3 gene expression in ovalbumin (OVA)-induced asthma model mice. Myristicin reduced levels of IL-4, Th2 cytokine production in OVA-sensitized and challenged mice. In the other side, it increased $IFN-{\gamma}$, Th1 cytokine production in myristicin administrated mice. We also examined to ascertain whether myristicin could influence eosinophil peroxidase (EPO) activity. After being sensitized and challenged with ovalbumin (OVA) showed typical asthmatic reactions. These reactions included an increase in the number of eosinophils in bronchoalveolar lavage fluid, an increase in inflammatory cell infiltration into the lung tissue around blood vessels and airways, and the development of airway hyper-responsiveness (AHR). The administration of myristicin before the last airway OVA challenge resulted in a significant inhibition of all asthmatic reactions. Accordingly, these findings provide new insight into the immunopharmacological role of myristicin in terms of its effects in a murine model of asthma.

Breeding of ‘Skyblueball’, a New Gentiana spp. Cultivar with the Same Time Flower and Violet Blue Petals (연한 청색의 동시개화성 절화 용담 ‘스카이블루볼’ 육성)

  • A new Gentiana spp. cultivar “Skyblueball” was bred by crossing between “Jinbu-65 line” with violet blue (VB 95B) colored petals as female and “Jinbu-1 line” with violet blue (VB 93B) petals as male in 2000 year. The superior line of 00-168-3 was selected in Daegwallyeong from 2001 to 2003 year, and its growth and flower characterization were conducted from 2004 to 2007 year in field of Daegwallyeong and Jinbu, Pyeongchang, Gangwon-do, Namwon, Jeollabuk-do and Jeju Special Self-Governing-do. The flower showed violet blue (VB 96C) color at opening, the petal of flower had not spot and flower stem color was purple. The flowers were bloomed at the same time from bottom to upside (Table 1). Plant height was 62.9 ㎝, number of flower stem per plant is 8.3, number of flower node per stem was 5.3, and number of flower per stem was 21.7 (Table 2). The flowering of ‘Skyblueball’ was bloomed from July 26 in Daegwallyeong, August 3 in Jinbu and Namwon, and June 28 in Jeju Special Self-Governing-do. ‘Skyblueball’ cultivar can be used for cut flower.

Evaluation of Various Characteristics of High Quality Rice Varieties That Could Potentially be Grown on Reclaimed Land in Jellabuk Province, Korea (전북지역 간척지에서 최고품질 벼 품종의 작물학적·이화학적 특성 비교 및 선발)

  • The main objective of this study was to identify the best quality rice for cultivation on reclaimed land in Jeollabukdo-Province based on yield, head rice ratio, protein content, amylose content, and Toyo taste value evaluations. Seven varieties of rice were grown for two years on the reclaimed land. The Sukwang variety had the earliest heading date. The tiller number for the Younghojinmi variety was 19 on average, and its variation in tiller number was the lowest, which showed that the safety for cultivation of the variety was high on the reclaimed land with respect to that of the other varieties. The culm and panicle lengths of the Sukwang and Hyunpum varieties were the highest at 81.8 and 21.3 cm, respectively. Shindongjin produced the highest thousand grain weight at 27.7 g, whereas Haepum produced the lowest at 22.3 g. The number of grains per panicle for 7 all varieties was statistically equal for both years. The yield order for the varieties was Shindongjin (590) > Hopum (575) > Younghojinmi (552) > Sukwang (551) > Hopum (543) > Mipum (534) > Haepum (498 kg/10a), which showed that there was no significant difference in average yield between the varieties when both years are taken into consideration. However, the annual difference in Haepum yield between 2015 and 2016 was the highest at 108 kg/10a, indicating low cultivation safety. The head rice ratio of Younghojinmi was higher than for the other cultivars in both 2015 and 2016. However, there was no significant difference between the 2 years when the averages for all varieties were analyzed. There was was a considerable difference in protein and amylose content between the varieties in the two years, although this was not apparent in the results when the average over 2 years for each variety was calculated. There was no significant difference between the two years with respect to the average head rice ratio, and the protein and amylose contents of the seven varieties. The Toyo taste values for the Sukwang and Mipum varieties were higher than for the other varieties. Based on the principle characteristics, such as Toyo value, the Sukwang variety is a suitable substitute for Shindongjin, which has been previously used to produce high-quality rice on the reclaimed land.

Hypoglycemic Effect of Smallanthus sonchifolius (Yacon) Extracts on Animals with Streptozotocin-induced Diabetes (야콘 추출물이 Streptozotocin으로 유도된 당뇨 동물모델에서 혈당강하에 미치는 효과)

  • This study was performed to investigate improvements in diabetes mellitus by extracts of yacon in streptozotocin (STZ)-induced diabetic rats. Fifty rats were divided into a normal group and four experimental groups. STZ (45 mg/kg) was injected intraperitoneally to induce type I diabetes in the four experimental groups. Yacon extracts were administered for 5 weeks. Forty-five ICR mice were also divided into one positive control group and four experimental groups for the oral glucose tolerance test (OGTT) after fed yacon extract. The control group did not eat any yacon extracts, while Group 1 (GI) was fed 125 mg/kg of yacon extracts, Group 2 (GII) was fed 250 mg/kg of yacon extracts, and Group 3 (GIII) was fed 500 mg/kg of yacon extracts. After treatment for 5 weeks, blood glucose in GIII group showed decreased tendency at the 5 week. In OGTT by glucose, the glucose level of yacon treatment group in diabetic rats was significantly decreased compared to the glucose level of the control group, but there was no difference in OGTT by maltose. In ICR mice, the glucose level of the experimental group in OGTT by maltose was significantly decreased compared to the control group. The area of the islets of Langerhans was increased by yacon treatment in a dose-dependent manner on diabetic rats. Insulin concentration of the GIII group was also decreased compared to the control group, while the concentration of fructosamine, total cholesterol, and triglycerides in serum showed no difference. OGTT by glucose or maltose in ICR mice or diabetic rats, area of the Islets of Langerhans, and insulin concentration improved. Yacon treatment may be a useful therapeutic and preventive strategy for diabetes mellitus.

  • In the present investigation, we analyzed the data of 1,318 patients (2,636 images) who underwent mammographic examinations and obtained the distribution of the patient age and compressed breast thickness. We measured also average glandular doses (AGD) as function of compressed breast thickness. In order to obtain the values of AGD, we measured half value layer (HVL) and tube output (mR/mAs) for each kVp and target/filter combination. Entrance surface air kerma (ESAK) was calculated from the tube output as measured for each voltage used under clinical conditions and from the tube loading (mAs). AGD per exposure were calculated by multiplying the ESAK values by the conversion factors tabulated by Dance. We obtained in this study the following conclusions. The mean value of compressed breast thickness for cranio-caudal (CC) view was 35.8mm and that for medio-lateral oblique (MLO) view was 43.3 mm. The mean value of AGD for CC view was 1.55 mGy and that for MLO view was 1.70 mGy. The AGD for MLO view was 0.15 mGy (10%) higher than that for CC view because the thickness for MLO view was on average 4.8 mm higher than that for CC view. The values of AGD increased with increasing compressed brest thickness. The increased AGD value was on average 0.34 mGy per 10 mm in the thickness ranges $10{\sim}80\;mm$, therefore differences between the AGD values of each thickness were relative large. Thus, it is considered to need limited doses for mammography with the upper end of exposure range at several different compressed brest thickness.

The Flora of Habitats Distributed with Allium victorialis var. platyphyllum Populations in the South Korea (우리나라 산마늘 개체군 자생지의 식물상)

  • This study was conducted to investigate the flora of habitats distributed with Allium victorialis var. platyphyllum populations in the South Korea. The vascular plants were surveyed 4 times from March 2009 to June 2010. The flora of studied area was listed as 214 taxa; 64 families, 142 genus, 181 species, 2 subspecies, 30 varieties, 1 forma. According to the specific plant species by classes, class I has 17 taxa; 15 families, 16 genus, 15 species, 2 varieties. Class II has 16 taxa; 13 families, 16 genus, 15 species, 2 varieties. Class III has 20 taxa; 16 families, 19 genus, 18 species, 2 varieties. Class III has 20 taxa; 16 families, 19 genus, 18 species, 2 varieties. Class IV has 23 taxa; 20 families, 23 genus, 19 species, 4 varieties. Class V has 2 taxa; 2 families, 2 genus, 2 species. Rare plant was 12 taxa; 9 families, 11 genus, 10 species, 2 varieties. Endemic plant was 21 taxa; 17 families, 19 genus, 18 species, 3 varieties. Endangered plant was not present in study area. It was considered that the ecosystem was probably stabilized for nothing of naturalized species and ecosystem disturbance species there. The characteristics of growth environments in the habitats with Allium victorialis var. platyphyllum were considered to be related to valley species and subalpine species such as Actinidia polygama, Cornus controversa, Rhododendron brachycarpum and Sorbus commixta.

Effect of Dietary Metabolizable Energy and Crude Protein Concentrations on Growth Performance and Carcass Characteristics of Korean Native Ducks (사료의 에너지 및 조단백질 함량이 토종오리의 성장과 도체 특성에 미치는 영향)

  • This study was undertaken to assess dietary CP and ME concentrations for optimum growth performance and carcass characteristics of Korean Native male ducks. In a $3{\times}3$ factorial arrangement, 1-d-old Korean Native male ducks were completely randomized design to experimental diets with 3,000, 2,900, and 2,800 kcal of ME/kg of diet; each contained 23, 22, and 21% CP, respectively, from 0 to 3 wk of age. From 4 to 8 wk of age, experimental diets had 3,100, 3,000, and 2,900 kcal of ME/kg of diet, and each contained 19, 18, and 17% CP, respectively. Each dietary treatment was replicated 4 times, and feed and water were provided ad libitum. Body weight, feed intake and uniformity were measured at 3 wk and 8 wk, and carcass characteristics were evaluated at 8 wk. As CP increased from 21 to 23 %, the body weight, BW gain and feed conversion ratios (FCR) linearly improved (P<0.01) during 0 to 3 wk of age. From 4 to 8 wk of age, the body weight, feed intake, BW gain, FCR and uniformity were not different (P>0.05). Carcass ratios of birds fed 3,000 kcal of ME/kg diets from 3 to 8 wk of age were quadratically increased significantly (P<0.01). As ME and CP increased, respectively, relative weight of liver weight per 100 g of BW was linearly decreased (P<0.01). Relative weights of left breast, thigh and drumstick weight per 100 of BW were not different (P>0.05). Also, breast meat color, pH, cooking loss and shear force were not different (P>0.05). Thus, diets with 2,800 kcal of ME/kg and 23% CP or with 2,900 kcal of ME/kg and 17% CP at 0 to 3 wk and 4 to 8 wk, respectively, were used more efficiently. However, diets with 3,000 kcal of ME/kg at 4 to 8 wk in carcass rate was quadratically increased.
