Inhibitory Effect of Simazine on Photosynthetic Electron Transport Activity in Anabaena inequalis (Anabaena의 광합성 전자전달 활성에 미치는 Simazine의 억제효과)

  • Effects of simazine [2-chloro-4,6-bis(methylamino)-s-triazine] on the photochemical reactions of isolaed spinach chloroplasts and crude thylakoids of Anabaena inequalis UTEX B-381 were compared. Simazine inhibited photosynthetic O2 evolution and increased the chlorophyll fluorescence in whole cells of Anabaena. The electron transfer from diphenylcarbazide to 2,6-dichlorophenolindophenol was inhibited by simazine treatment in spinach chloroplasts, but not in crude thylakoids of Anabaena. In spinach chloroplasts, the chlorophyll fluorescence was increased by simazine treatment in the presence of diphenylcarbazide and ferricyanide, but not in the presence of diphenylcarbazide and silicomolybdate. In crude thylakoids of Anabaena, simazine treatment did not increase the chlorophyll fluorescence in the presence of either diphenylcarbazide and silicomolybdate, or diphenylcarbazide and ferricyanide. There results suggest that the inhibitory site of simazine on photosynthetic electron transport chain of anabaena is different from that of spinach chloroplasts. And there may be a possiblity that the inhibition site of simazine in Anabaena lies on the donor side of photosystem II, before the site of electron donation by diphenylcarbazide.

  • Chlorophyll (Chl) fluorescence imaging was used to investigate the effectiveness of in vivo incorporation methods for two chemicals, 3-(3',4'-dichlorophenyl)-1,1-dimethylurea (DCMU) and methyl viologen (MV) in rice, a monocot, and cucumber, a dicot, leaves. four different methods (vacuum infiltration, floating, transpiration-aided incorporation through petiole and spraying) were compared, and $F_i$ and $F_v$/$F_m$ were chosen for the imaging of the DCMU- and MV-treated leaves, respectively. The effects of the chemicals in plants were generally heterogeneous over the whole leaf area. Moreover, the effectiveness of the treatment of a chemical in plant leaves was dependent on chemical species, plant species, concentration of the chemical, the treatment method, the duration of the treatment, the existence of light and detergent, etc. In conclusion, we suggest that to achieve the proposed effects of chemicals in plants for an actual experiment, these factors must be considered before the chemical treatment, and the best method for the treatment of the chemicals tested was floating and vacuum infiltration in the dicot and the monocot leaves, respectively, as drawn from Chl fluorescence imaging analysis.

The Effects of Sulfite on the Greening of Etiolated Barley (Mordeum vuigare L.) Seedling (Sulfite가 보리(Hordeum vulgare L.) 유식물의 녹화에 미치는 영향)

  • To investigate the effects of sulfite on the chloroplast development, etiolated barley seedlings were treated with 100 mM sulfite solution every 3 hour by spraying during 96 hours greening Period. The effects were determined by chlorophyll a, b and carotenoids contents, photosynthetic electron transport activity, chlorophyll fluorescence yield and fluorescence quenching parameters. The contents of chlorophyll a and carotenoids were decreased than that of control by treatment of salfite over 48 hours greening. PS II Is more sensitive to sulfite than PS I Is. And by the addition of DPC to the chloroplasts of the barley seedling treated with sulfite, the photoreduction of DCPIP was not recovered. In greening with suite treated barley leaves, Fo, Fv and Nlh ratio were decreased with little difference from that of control. But qP, qNP and qR were lowed in comparison with those of controls whereas qE was markedly higher than that of control. Especially, It is Interesting that qR was decreased markedly compared to that of control. The results in the change of PS I activity, Nf and qP suggest that the strate of Inhibition by suite Is carbon dioxide reduction cycle.

The Effect of Light Intensity on the Growth and Chlorophyll Fluorescence Parameters of Three Ardisia Genus Native to Korea

  • This study investigated the growth and chlorophyll fluorescence reactions of three Ardisia genus grown under various indoor light intensity conditions with the aim of evaluating their suitability as indoor plants. Young seedlings of A. crispa (Thunb.) A.DC., A. pusilla DC., and A. japonica (Thunb.) Blume were used in the experiment. The plants were cultivated indoors for 10 weeks under different light intensities: 10, 50, 100, and 200 PPFD (μmol·m-2·s-1), and their growth was compared with that of plants cultivated in a greenhouse during the same period (mean value 236.8±20.4 PPFD at noon). Also, chlorophyll fluorescence analysis was investigated with a portable PAM fluorometer. The indoor plants were maintained at 12/12 h photoperiod, temperature at 25±1℃, and humidity at 55±3%. Irrigation frequency (once every three days) was the same for the indoors and the greenhouse. The results of growth in three Ardisia plants showed that almost all parameters except leaf number and chlorophyll content had similar levels regardless of light intensity. A. crispa and A. pusilla plants grown in 200 PPFD were investigated to have low chlorophyll contents. Meanwhile, chlorophyll fluorescence parameters differed based on light levels. In A. crispa, the Fv/Fm (0.77), DIo/RC (0.47) and Fm/Fo (4.77) parameters tended to be poor at 200 PPFD compared to those at other light intensities. Similarly, the DIo/RC, Fm/Fo, and Pi_Abs parameters of A. pusilla plant (200 PPFD) are 0.45, 4.48 and 2.42, respectively, which can be considered stress. The analysis of fluorescence in A. japonica showed that all parameters except ETo/RC had similar levels regardless of light intensity. The ETo/RC parameter was 0.49 and 0.72 in the control plants and plants 200 PPFD, respectively, which was lower than those in plants at other light intensities. Therefore, it seems that the relatively high light intensity acted as a stressor for Ardisia plants.

Photosynthetic Characteristics of Polyvinylalcohol-Immobilized Spinach Chloroplasts (Polyvinylalcohol에 고정화한 시금치 엽록체의 광합성특성에 대한 연구)

  • Photoxynthetic properties of polyvinylalcohol (PVA)-immobilized chloroplast especially regarded to stability of photosynthetic electron transport and the fluorescence induction pattern were studied. When isolated spinach chloroplasts were immobilized with PVA, it showed good preservation of photosynthetic electron transport activity, especially PS II activity, during storage at -15$^{\circ}C$, 4$^{\circ}C$ and 2$0^{\circ}C$. And immobilized chloroplasts revealed similar thermostability of whole chain electron transport to free chloroplsts. And the absorption peak of red band of chloroplasts showed the blue-shift of 2-4 nm after immobilization. Fv/Fm ratio of chlorophyll fluorescence slightly decreased after immobilization. White light pulse after continuous light do not induced the additional fluorescence rise. This means chlorophyll fluorescence at room temperature reached to Fmax under continuous light in the immobilized chloroplasts. It seems that PVA may be a good candidate for immobilization matrix for the preservation of photosynthetic function of thylakoids and for the continuous use of chloroplast membranes of higher plants for solar energy storage and conversion.

  • As cool-season plant, Panax ginseng C. A. Meyer is planted under shade-installation with tall front and low rear. However, at different planting positions, distinct differences come out because ginseng grows at the same position within 3~5 years and the growth circumstance changes a lot by the shade-installation. So, in this study, changes of temperature, photosynthesis and chlorophyll fluorescence with varieties of shading material and planting position were investigated. Light transmittances by polyethylene shade net and silver-coated shading plate as planting materials were measured according to different planting positions. Photosynthetic rate and chlorophyll fluorescence were measured by LI-6400-40 (Li-Cor). According to different planting positions, light intensity was higher in silver-coated shading plate than in polyethylene shade net, and higher at front than rear. Also, photosynthetic rate showed the same tendency, which had a positive correlation to light intensity. But this treatment caused a lower Fo compared with polyethylene shade net because of the stress by light and temperature. Also, Fv/Fm and ETR were higher in silver-coated shading plate. Fo was similar at front and rear according to silver-coated shading plate and ETR was higher at front.

Chlorophyll a Fluorescence Parameters of Hulled and Hull-less Barley (Hordeum vulgare L.) DH Lines Inoculated with Fusarium culmorum

  • Barley worldwide is affected seriously by Fusarium seedling blight (FSB) and Fusarium head blight (FHB) diseases caused by the Fusarium species. The objective of this study was to facilitate the resistance of hulled and hull-less barley at different growth stages to F. culmorum according to direct parameters: disease rating (DR), fresh weight of leaves and roots, kernel weight per spike, kernel number per spike, plump kernels, and indirect parameters - chlorophyll a fluorescence (CF). Plate assay, greenhouse and field tests were performed on 30 spring barley doubled haploid (DH) lines and their parents infected with Fusarium culmorum. Direct parameters proved that hulled genotypes show less symptoms. Most studied chlorophyll a fluorescence (CF) parameters (apart from DIo/CS - amount of energy dissipated from PSII for laboratory test, TRo/CS - amount of excitation energy trapped in PSII reaction centers, ETo/CS - amount of energy used for electron transport and RC/CS - number of active reaction centres in the state of fully reduced PSII reaction center in field experiment) were significantly affected by F. culmorum infection. In all experiments, hulled genotypes had higher values of CF parameters compared to hull-less ones. Significant correlations were detected between direct and indirect parameters and also between various environments. It was revealed that ABS/CS, TRo/CS, and RC/CS have significant positive correlation in greenhouse test and field experiment. Significant correlations suggest the possibility of applying the CF parameters in selection of barley DH lines resistant to F. culmorum infection.

  • The occurrence of harmful algae on the coast of Korea has been a cause of damage to the aquaculture industry and deterioration of the coastal ecosystem environment. A method is required to predict their outbreak in real-time at the site. Therefore, this study attempted to develop a small hybrid optical sensor and real-time monitoring system based on LiDAR that can be used in the field and laboratory and can be applied to various platforms. FMS-L specifically suggested the amount of Chlorophyll a (Chl a) in the sample by measuring and analyzing the fluorescence emitted by the irradiating light. The accuracy of FMS-L was verified by measuring the concentrations of standard Chlorophyll a substances and Margalfidinium polykirkoids. In addition, the precision was verified by comparing the measurement results of FMS-L using commercial equipment Phyto-PAM-II. This equipment is compact and easy to move. Therefore, it can be easily applied to field surveys, allows short time measurements (10 s), and can be applied at a distance of 10 m from the measurement site.

Chlorophyll Fluorescence and Growth Response of Three South Korea Native Fern Species under In-door Light Intensity

  • This study investigated the growth of native ferns under indoor light intensities to identify the introduction possibility as in-door ornamental plants. Three evergreen perennial fern species used in this experiment were Coniogramme japonica (Thunb.) Diels, Woodwardia japonica (L. f.) Sm., and Cyclosorus acuminatus (Houtt.) Nakai ex H. Itô. The light intensities were adjusted to 10, 50, 100 and 200 PPFD (µmol·m-2·s-1) based on the measurement of the various indoor light quantities. The experiment was conducted for a total of 8 weeks, and the light period (12/12h), temperature (25±1℃), and humidity (55±3%) were maintained during the experiment. The control plant group was grown in glass greenhouse for the same period. As the result of the study, in door C. japonica showed better growth under light intensities of 100, 200 PPFD. However, withering of the plants were observed under all light intensities except the control, which causes an ornamental value decrease. This seems to be related to the increase of DIo/RC value in chlorophyll fluorescence parameters. In the W. japonica growth data, the plant height was not significantly different from the control but, the leaf number decreased more than a two-fold. Also, the formed leaves turned brown and showed a poor growth and SPAD value at 200 PPFD had decreased significantly. Growth data of C. acuminatus was not significantly different with the control at all light intensities however, withering was observed at 100 and 200 PPFD. In chlorophyll fluorescence parameters, significant decrease in Pi_Abs and increase in DIo/RC value at 200 PPFD impose that stress caused by the intense light might be the reason of the withering of the plants.

  • The absorbance and fluorescence yields of chl-a vs. concentration of n-prOH in diethyl ether, benzene and iso-octane were shown the characteristic point which chl-a structures are changed to monomer by the solvation of oligomer, and the spectral differences of fluorescence excitation between oligomer and monomer were identified by fluorimetry. All the maximum wavelength of absorbance, fluorescence excitation and fluorescence emission were shifted to longer wavelength. The ratios of soret/red band were depended on the band intensions and the polarities of solution in organic solvents mixed with n-prOH.
