Photosynthetic responses to dehydration were examined by the simulataneous measurement of O2 evolution and chlorophyll (Chl) fluorescence in green pepper leaves. Dehydration was induced by immersing the plant roots directly in the Hoagland solution containing varying concentration (2-30%) of polyethylene glycol(PEG-6000) . Water potential of the leaf was decreased time-and concentation -dependently by PEG-treatment. The decrease in water potential of leaf was correlated with the decrease in both the maximal photosynthesis (Pmax) and quantum yield of O2 evolution, but Pmax dropped more rapidly than quantum yield at all water deficit conditions tested. However, Chl fluorescence parameters were not affected much. Dehydration did not change the initial fluorescence (Fo) and maximum photochemical efficiency(Fv/Fm) of photosystem(PS) II. Both the photochemical quenching (qP) and non-photochemical quenching(NPQ) were not changed by dehydration under low PFR(50 $\mu$mols m-2s-1 ). In contrast, under high PFR(270$\mu$mols m-2s-1)qP was slightly decreased while NPQ was greatly increased. The fast induction kinetics of Chl fluroecence showed no change in Chl fluorescence pattern by dehydration at high PFR (640 $\mu$mols m-2s-1 ), but exhibited a significant drop in peak level(Fp)at low PRFR (70$\mu$mols m-2s-1 ). PS I oxidation and reduction kinetics revealed normal reduction but delayed oxidation to P-700+, suggesting no lesionin electron flow from PSII to PSI , but impaired electron transport to NADP+,These results suggest that water stress caused by PEG-treatment results in the reduction of photosynthesis, promarily due to the reducted electron trasport from PSI to NADP+ or hampered subsequent steps involving Calvin Cycle.
A cyanobacteria-like organism Aaryochloris marina performs oxygenic photosynthesis with near far-red light by the use of chlorophyll d. Reaction center chlorophyll (Chl) of Photosystem (PS) II of A. marina was studied by analysis of millisecond-delayed fluorescence. Delayed fluorescence is emitted by Chi d indicating efficient energy transfer between antenna Chi d molecules and the unknown primary electron donor of PS II. P740 a reaction center Chl of PS I of A. marina is shown to give a dimer type cation, and triplet state with a D value of 245xlO$\^$-4/ cm$\^$-l/ in contrast to the 280-290 xlO$\^$-4/cm$\^$-l/ values of P700 suggesting triplet spins interacting at a 5% larger distance in P740 than in P700.
Chlorophyll (Chl) fluorescence imaging was used to investigate the effectiveness of in vivo incorporation methods for two chemicals, 3-(3',4'-dichlorophenyl)-1,1-dimethylurea (DCMU) and methyl viologen (MV) in rice, a monocot, and cucumber, a dicot, leaves. four different methods (vacuum infiltration, floating, transpiration-aided incorporation through petiole and spraying) were compared, and $F_i$ and $F_v$/$F_m$ were chosen for the imaging of the DCMU- and MV-treated leaves, respectively. The effects of the chemicals in plants were generally heterogeneous over the whole leaf area. Moreover, the effectiveness of the treatment of a chemical in plant leaves was dependent on chemical species, plant species, concentration of the chemical, the treatment method, the duration of the treatment, the existence of light and detergent, etc. In conclusion, we suggest that to achieve the proposed effects of chemicals in plants for an actual experiment, these factors must be considered before the chemical treatment, and the best method for the treatment of the chemicals tested was floating and vacuum infiltration in the dicot and the monocot leaves, respectively, as drawn from Chl fluorescence imaging analysis.
The deteriorative effect of ultraviolet-B(UV-B) radiation on photosynthesis was assessed by the simultaneous measurement of O$_2$ evolution and chlorophyll(Chl) fluorescence in green pepper. UV-B was given at the intensity of 1 W$.$m$\^$-2/, a dosage often encountered in urban area of Seoul in Korea, to detached leaves. Both Pmax and quantum yield of O$_2$ evolution was rapidly decreased, in a parallel phase, with increasing time of UV-B treatment. Chl fluorescence parameters were also significantly affected. Fo was increased while both Fm and Fv were decreased. Photochemical efficiency of PSII(Fv/Fm) was also declined, although to a lesser extent than Pmax. Both qP and NPQ were decreased similarly with increasing time of UV-B treatment. However, PS I remained stable. The addition of lincomycin prior to UV-B treatment accelerated the decline in Fv/Fm to some extent, suggesting that D1 protein turnover may play a role in overcoming the harmful effect of UV-B. The amount of photosynthetic pigments was less affected than photosynthetic response in showing decline in Chl a and carotenoids after 24 h-treatment. Presumptive flavonoid contents, measured by changes in absorbance at 270 nm , 300 nm and 330nm, were all increased by roughly 50% after 8 h-treatment. Among antioxidant enzymes, activities of catalase and peroxidase were steadily increased until 12h of UV-B treatment whereas ascorbate perxidase, dehydroascorvate reductase and glutathione reductase did not show any significant change. The results indicate that deteriorative effect of UV-B on photosynthesis precedes the protection exerted by pigment synthesis and antioxidant enzymes.
This study was conducted to investigate the changes of chlorophyll contents and chlorophyll fluorescence in barley(Hordeum vulgare L.) 7 day old seedling treated with 0.2M, 0.4M, 0.6M, 0.8M, and 1.0M NaCI concentration containing Hepes buffer(pH 7.5). Barley was affected by NaCI treatment. The chlorophyll a, b and carotenoid of barley decreased with an increase in NaCI concentration. However, chlorophyll a, b and carotenoid of barley were not greatly influenced by o.8M and 1.0M NaCl. Fv, Fv/Fm and qP were gradually decreased by higher concentration of NaCI. qP, qNP, qR and qE were gradually decreased by 6hr. During barley chloroplast was development NaCI affected chlorophyll synthesis than photosynthetic activity. Whereas barley seedling leaves were more influenced photosynthetic activity than chlorophyll contents by NaCI.
Spatial and temporal aspects of toxic dinoflagellate Cochlodinium.p blooms and consequences of physical features in complex coastal ecosystems, off the southern Korean coast, have been investigated using data obtained from SeaWiFS and AVHRR as well as in-situ observations. Hydrographic parameters measured using CTD sensors were used to elucidate physical factors affecting the spatial distribution and abundance of Cochlodinium.p blooms. The results show spatial and temporal variations of chlorophyll-a (Chl-a) and sea surface temperature (SST) and reveal significant information about Cochlodinium.p blooms and process underlying their evolution. Satellitederived Chl-a estimates appear to be potential in explicating the evolution, movement and distribution of Cochlodinium.p blooms in the enclosed bays of the South Sea. The existence of thromohaline waters offshore provide favorable conditions for the rapid growth and subsequent southward initiation of Cochlodinium.p blooms that are influenced to flow on the offshore branch (OB) during September. It was observed that there was a significant variation in the sun-induced chlorophyll-a fluorescence signal in the remote sensing fluorescence spectra and its high-intensity was recognized during the period of exponential growth and physical transport. Satellite-derived Chl-a concentration during September 1999 ranged between $360mg/m^3$ inside the Jin-hae and adjacent Bays and $1-6mg/m^3$ in offshore waters, with varying Cochlodinium.p abundances 1500 to 26000 cells $ml^{-1}.$ The closely spaced CTD surveys and satellite-derived SST give a complete overview on the initiation of Cochlodinium.p blooms in hydrodynamically active regions of the offshore southern East Sea by the influence of Tsushima Warm Current (TWC).
Several ocean color algorithms have been developed for GOCI (Geostationary Ocean Color Imager) using in-situ bio-optical data sets. These data sets collected around the Korean Peninsula between 1998 and 2009 include chlorophyll-a concentration (Chl-a), suspended sediment concentration (SS), absorption coefficient of dissolved organic matter ($a_{dom}$), and remote sensing reflectance ($R_{rs}$) obtained from 1348 points. The GOCI Chl-a algorithm was developed using a 4-band remote sensing reflectance ratio that account for the influence of suspended sediment and dissolved organic matter. The GOCI Chl-a algorithm reproduced in-situ chlorophyll concentration better than the other algorithms. In the SeaWiFS images, this algorithm reduced an average error of 46 % in chlorophyll concentration retrieved by standard chlorophyll algorithms of SeaWiFS. For the GOCI SS algorithm, a single band was used (Ahn et al., 2001) instead of a band ratio that is commonly used in chlorophyll algorithms. The GOCI $a_{dom}$ algorithm was derived from the relationship between remote sensing reflectance band ratio ($R_{rs}(412)/R_{rs}(555)$) and $a_{dom}(\lambda)$). The GOCI Chl-a fluorescence and GOCI red tide algorithms were developed by Ahn and Shanmugam (2007) and Ahn and Shanmugam (2006), respectively. If the launch of GOCI in June 2010 is successful, then the developed algorithms will be analyzed in the GOCI CAL/VAL processes, and improved by incorporating more data sets of the ocean optical properties data that will be obtained from waters around the Korean Peninsula.
Mercury-induced changes in Chl a fluorescence induction kinetics of scratched barley leaf segments were dependent on the presence of light. By the treatment of 50$\mu$M HgCl2 under light condition, Fm and Fp were decreased. However, they were not significantly reduced under dark condition even after 2 h of mercury treatment. Under dark condition the decrease in variable fluorescence (Fv) after P transient was blocked within 20 min of the treatment. The analysis of fast fluorescence rise curve suggests that the inhibitory site of mercury under both light and dark conditions is not at QB binding site and the inhibition does not involve the increase in inactive PSII centers. Under light condition the decrease in Fp was partially recovered by addition of 50 $\mu$M NH2OH. These results suggest that a major inhibitory site of mercury under dark condition is at the reducing side of PSII and the site under light condition is at the oxidizing side of PSII possibly in addition to the one under dark condition. Under both light and dark conditions, energy-dependent quenching(qE) was alomost completely repressed within 20 min of mercury treatment and noticible change in Fo was not observed. The qE repression is probably due to the blockage of transthylakoid ΔpH formation.
In order to determine the relationships between the lea(-burning disease and the light harvesting chlorophyll-protein (LHCP) complex in Panax ginseng C. A. Meyer, we investigated the chlorophyll-protein (CP) complex of the thylakoid membrane and its characteristics. In P. ginseng four Cp-complex bands determined by non-denaturing SDS-PAGE were identified CP I'(containing reaction center of photosystem I and LHCP I antennae), CP I (reaction center of photosystem I) LHCP II** (oligoform of LHCP II), and LHCP II (photosystem II antennae, CP 26 and CP 29) by Bassis and Dunahay's procedures. Under our experimental condition, the CP I band was only observed in P. ginseng and the band intensity of LHCP II** in P ginseng was higher than in spinach and soybean. There were differences in the absorption and fluorescence spectra and chlorophyll a/b ratio of the CP-complex bands between P. ginseng and other Plants. The Polypeptidr content of P. ginseng thylakoid was lower than in spinach and soybean thylakoid, and the Polypeptide profiles of P. ginseng was low band intensity, especially about 29-35 kD, 55 kD, and 60 kD, compared to spinach and soybean. The inhibitory effects of 2,5-dimethylfuran, specific singlet oxygen ($^1O_2$) quencher, showed that singlet oxygen destroyed 60% of chl.a, 90% of chl.b and 70% of carotenoid in bleaching P. ginseng with leaf-burning disease.

The occurrence of harmful algae on the coast of Korea has been a cause of damage to the aquaculture industry and deterioration of the coastal ecosystem environment. A method is required to predict their outbreak in real-time at the site. Therefore, this study attempted to develop a small hybrid optical sensor and real-time monitoring system based on LiDAR that can be used in the field and laboratory and can be applied to various platforms. FMS-L specifically suggested the amount of Chlorophyll a (Chl a) in the sample by measuring and analyzing the fluorescence emitted by the irradiating light. The accuracy of FMS-L was verified by measuring the concentrations of standard Chlorophyll a substances and Margalfidinium polykirkoids. In addition, the precision was verified by comparing the measurement results of FMS-L using commercial equipment Phyto-PAM-II. This equipment is compact and easy to move. Therefore, it can be easily applied to field surveys, allows short time measurements (10 s), and can be applied at a distance of 10 m from the measurement site.
