전해산화수로 세척한 절임 배추의 저장중 품질 특성 (Quality Characteristics of Salted Chinese Cabbage Treated with Electrolyzed-Acid Water during Storage)

  • 절임 배추의 저장 기간을 연장하기 위하여 배추를 절인 다음 전해산화수로 세척하고 저장하면서 품질 특성의 변화를 측정한 결과는 다음과 같았다. 즉 절임 배추를 전해산화수로 세척한 처리구가 대조구보다 낮은 pH를 보였으며, 저장 8일까지는 대조구에 비하여 낮은 pH를 나타내었다. 적정산도는 저장기간 동안 0.11-0.15%로 큰 변화를 나타내지 않았으며, 대도구와 처리구간에도 큰 차이가 없었다. 절임 배추를 전해산화수로 세척한 직후에는 대조구에 비하여 총균수가 $10^3\;CFU/mL$ 정도 낮았으나 처리구의 총균수는 저장 6일에 대조구와 비슷하였다. 유산균수도 세척 직후에는 처리구가 대조구보다 낮았으며, 저장 4일에는 대조구와 처리구의 차이가 가장 현저하였다. 저장 2일까지는 국물의 밝은 정도(L)에 있어서 대조구와 처리구간에 차이가 없었으나 그 이후에는 처리구가 대조구보다 높은 값을 나타내었다.

  • $GB_{10}$, chitofarm 및 왕겨숯과 왕겨숯액을 天然物質을 가을 배추 栽培圃場에 種子處理, 土壤處理 및 葉面撤布를 實施하여 收量과 成分分析을 遂行한 實驗成績은 아래와 같다. 1. 배추 生育特性中 株重, 球重, 球長, 球莖, 葉長, 葉幅 및 糖度에 있어서 Chitofarm 等 天然物質處理區가 無處理區에 비하여 大部分 有意差가 있었으나 天然物質 相互間에는 有意差가 없었다. 2. 배추 收量에 있어서는 $GB_{10}$ 土壤處理區+$GB_{10}$ 1% 葉面撤布區가 3.27kg으로 check區 보다 53% 增收되었다. 3. 배추 植物體 成分分析 結果 T-N, $P_2O_5$, $K_2O$, CaO, MgO, $Na_2O$, 및 Mn 等의 含量에서 天然物質 無處理區보다 check區 보다 多量이었으므로 品質向上에 效果的이었다.

Changes in Free Amino Acid, Carotenoid, and Proline Content in Chinese Cabbage (Brassica rapa subsp. Pekinensis) in Response to Drought Stress

  • Chinese cabbage grown during autumn season is confronted with drought conditions for a certain period, especially during the early growth stage. In this study, we investigated the effects of drought stress on plant growth characteristics, as well as free amino acid, carotenoid, and proline in Chinese cabbage. Chinese cabbage seeds (Bulam Plus) were germinated, and all the seedlings were transplanted into plastic containers (28 cm diameter ${\times}$ 22 cm high) containing a commercial growth medium. The soil water content was measured and maintained at 10% for the drought-stressed plants and at 30% for the control plants, for three weeks. The results revealed that plant growth parameters were lower in the drought-stressed plants than in the control plants. The total free amino acid content tended to decrease in both drought-stressed and control plants with time. The total free amino acid content was found to be lower in the drought-stressed plants than in the control plants and the proline content was unaffected. Moreover, at three weeks after treatment, carotenoid content in drought stressed plants was significantly higher than that in the untreated plants. We believe that our study makes a significant contribution to the literature because the effects of drought stress on plant growth parameters, free amino acid, carotenoids, and proline accumulation in autumn growing cultivar of Chinese cabbage have not been widely studied in Korea, and our study provides valuable information in this regard, as Chinese cabbage is consumed throughout the year in Korea.

통배추의 염절임 방법에 따른 특성변화 (Characterization of Chinese Cabbage during Soaking in Sodium Chloride Solution)

  • 김치제조의 산업화로 현장에서 도움이 될 수 있는 배추의 절임조건의 영향을 보고자 절임수의 온도 및 절임 시간, 절임시 적재높이, 세척조건, 탈수조건 등을 달리하여 실험하였다. 절임시간이 길수록 그리고 절임수의 온도가 높을수록 절인 배추의 소금농도가 높아짐을 관찰할 수 있었다. 적재 높이를 고려한 배추의 염도 측정에서는 하단의 염도가 상단보다 더 높게 나타났다. 세척과 탈수를 할수록 염도가 떨어지는 경향을 나타냈다. 높이에 따른 배추의 미세구조 변화에서 절인 배추의 세포조직은 높이에 따라 서로 차이를 나타냈다. 염절임을 한 배추는 세척을 할수록 원래의 상태로 회복되는 경향을 나타내었다.

교배육종 및 소포자 배양에 의한 뿌리혹병 race4 저항성 배추 계통 육성 (Development of clubroot race4 resistant inbreds using conventional breeding and microspore culture method in Chinese cabbage)

  • To develop clubroot resistant Chinese cabbage inbreds, IT 033820, a clubroot resistant turnip, was cross pollinated with a Chinese cabbage inbred of BP 079. From 2005, conventional breeding and microspore culture method performed using these F1 plants as parental materials. In 2007, conventional breeding method resulted in 21 F3 inbreds. After inoculation of clubroot race 4, one inbred showing 83% resistant was selected and registered as 'onkyo 20036ho' in 2008. From 2005, we scanned hybrid cultivars using micro spore culture and developed many doubled haploid (DH) lines in Chinese cabbage. Using Chinese cultivar of 'oong-baek 2ho' we developed 26 DH inbreds in 2007. After inoculation of clubroot race 4, one DH inbred showing 77% resistant and yellow inner leaf color was selected and registered as 'onkyo 20034'in 2008. We found conventional breeding method was effective using introduced germplasm showing low germination. However, when using hybrid cultivar as starting material, microspore culture method was powerful for developing various inbred in short time.

제주도 화산회토양의 배추와 양배추에 대한 질소의 시비추천식 설정 (Recommendation of Optimum Amount of Fertilizer Nitrogen Based on Soil Organic Matter for Chinese Cabbage and Cabbage in Volcanic Ash Soils of Cheju Island)

  • 현재 채소류에 대한 질소시비량 추천식은 육지부토양에서 시험한결과를 이용하여 설정된 것으로 이를 화산회토양에서도 활용하는 문제가 있어 이를 검토하기위하여 제주도 토양을 중심으로 1999년에는 pot와 2000년에는 포장에서 몇개의 질소시비량수준을 두고 배추를 재배한 시험결과는 다음과 같다. 1. Pot와 포장시험에서 배추의 질소 최고시비량은 $294{\sim}331kg\;ha^{-1}$이었고 토양의 유기물함량과 최고시비량과의 관계를 이용하여 화산회토양에 알맞는 새로 질소시 비량 추천식을 설정하였다. 2. 배추의 질소시비량시험을 통한 화산회토의 실제 질소시비적량은 기존의 추천식을 이용한 시비량과는 큰차이를 보였고 개선된 질소시비 추천식으로부터 산출된 질소 시비량과는 근소한 차이를 보였다. 3. 수화기 T-N 함량 및 질소흡수량은 질소시비량이 증가할수록 많았으나 질소이용 율은 질소시비량이 증가할수록 낮아지는 경향이었다. 4. 화산회토양에서 재배하는 양배추 대한 토양유기물함량에 따른 질소 시비량은 배추대비 질소흡수량을 흡수량비교 계수로 하고 이를 이용하여 양배추의 새로운 질소시비 추천식을 도출하였다.

Utilization of the bar gene to develop an efficient method for detection of the pollen-mediated gene flow in Chinese cabbage (Brassica rapa spp. pekinensis)

  • To develop an efficient screening method for detection of the transgene in Chinese cabbage (Brassica rapa spp. pekinensis) utilizing Basta spray, optimal conditions for Basta application were examined in this study. Two transgenic Chinese cabbage lines were obtained through Agrobacterium-mediated transformation and used as transgenic positive controls in the Basta screening experiment. Differential concentrations of glufosinate-ammonium were sprayed into three different growth stages of 12 commercial Chinese cabbage cultivars. The results showed that no plants could survive higher than 0.05% glufosinate-ammonium, and plants at the 2-3 leaf stage were most vulnerable to glufosinate-ammonium. On the other hand, no damage was observed in the transgenic control plants. Reliability of the Basta spray method was proven by showing perfect co-segregation of the tolerance to glufosinate-ammonium and the presence of the bar gene in T1 segregating populations of the transgenic lines, as revealed by both PCR and Southern blot analyses. Using the developed Basta screening method, we tried to investigate the transgene flow through pollen dispersal, but failed to detect any transgene-containing non-transgenic Chinese cabbages whose parents had been planted adjacent to transgenic Chinese cabbages in field conditions. However, the transgene was successfully detected using Basta spray from the non-transgenic plants bearing the transgene introduced by hand-pollination. Since the Basta spray method developed in this study is easy to apply and economical, it will be a valuable tool for understanding the mechanism of gene flow through pollen transfer and for establishing a biosafety test protocol for genetically modified (GM) Chinese cabbage cultivars.

Molecular Characterization of a thiJ-like Gene in Chinese Cabbage

  • A cDNA clone for a salicylic acid-induced gene in Chinese cabbage (Brassica rapa subsp. pekinensis) was isolated and characterized. The cabbage gene encoding a protein of 392 amino acids contained a tandem array of two thiJ-like sequences. ThiJ is a thiamin biosynthesis enzyme that catalyzes the phosphorylation of hydroxymethylpyrimidine (HMP) to HMP monophosphate. Although the cabbage gene shows a similarity to bacterial thiJ genes, it also shares a similarity with the human DJ-1, a multifunctional protein that is involved in transcription regulation, male fertility, and parkinsonism. The cabbage thiJ-like gene is strongly induced by salicylic acid and a nonhost pathogen, Pseudomonas syringae pv. tomato, which elicits a hypersensitive response in Chinese cabbage. Treatment of the cabbage leaves with BTH, methyl jasmonate, or ethephon showed that the cabbage thiJ-like gene expression is also strongly induced by BTH, but not by methyl jasmonate or ethylene. This indicates that the cabbage gene is activated via a salicylic acid-dependent signaling pathway. Examination of the tissue-specific expression revealed that the induction of the cabbage gene expression by BTH occurs in the leaf, stem, and floral tissues but not in the root.

Nutritional value and in situ degradability of fruit-vegetable byproducts and their feeding effects on performance of growing Hanwoo steers

  • Objective: This study was conducted to evaluate nutritional value and in situ degradability of fruit-vegetable byproducts and their feeding effects on performance of growing Hanwoo steers. Methods: Nutritional value and in situ degradability of cabbage, Chinese cabbage and fruit-vegetable byproducts were assessed. In vivo feeding trial was also performed for 12 weeks. Thirty-six growing steers were randomly allocated into three groups according to body weight (BW) and age in 12 pens (4 replications/treatment) and assigned to one of the three dietary treatments: control (byproduct 0%), FV-B (fruit-vegetable byproduct 20%), and CA-B (cabbage peel 15% plus Chinese cabbage peel 15%, total byproduct 30%). Results: The crude protein contents of cabbage, Chinese cabbage and fruit-vegetable byproducts were 18.69%, 20.20%, and 10.07%, respectively. Concentrations of neutral detergent fiber (NDF) were higher in cabbage (22.31%) and Chinese cabbage (28.83%) than fruit-vegetable (13.94%). Higher concentrations of non-fiber carbohydrate were observed for fruit-vegetable (66.72%) than cabbage (44.93%) and Chinese cabbage byproducts (24.69%). The effective degradability (ED) of both dry matter (DM) and NDF for fruit-vegetable byproduct (DM, 84.69%; NDF, 85.62%) was higher (p<0.05) than cabbage (DM, 68.47%; NDF, 55.97%) and Chinese cabbage byproducts (DM, 68.09%; NDF, 54.22%). The DM intake was not different among treatments because the amount of feed was kept constant according to the BW of growing steers to prevent overweight during the growing period. The average daily gain during the whole experimental period was not different among treatments (1.26, 1.25, and 1.34 kg/d for control, FV-B, and CA-B). The ED of both DM and NDF degradability of the total mixed ration (TMR) diets were very similar among treatments. Feed conversion ratio during the whole period showed no significant difference among treatments. Conclusion: This study demonstrates that fruit-vegetable and cabbage byproducts up to 20% and 30% (as fed basis), respectively can be included in TMR diets for growing beef cattle.

저장 조건에 따른 월동배추의 품질 변화 (Quality Changes in Winter Chinese Cabbage with Various Storage Methods)

  • 배추의 V/H ratio는 PE film으로 포장한 배추가 포장하지 않은 control에 비해 더 높게 유지되고 있으며, 속잎이 겉잎에 비해 V/H ratio가 더 높게 유지되었다. Pouch내의 $CO_2$의 농도는 처리구 모두 15일 정도까지는 $CO_2$량이 급격히 증가하고 그 이후부터는 거의 일정한 경향을 보였다. pouch내의 $O_2$ 농도는 저장기간이 길어짐에 따라 15일까지 급격히 감소하다가 그 후에는 변화량이 적었다. $O_2$의 농도는 $CO_2$와 반대로 film의 두께가 얇을수록 pouch 내의 $O_2$ 농도가 높았으며, 0.1mm film의 경우 $O_2$의 농도가 5% 미만으로 상당히 적었다. 무처리의 경우 수분이 빠져나가 많이 시들었지만 조직은 PE film포장 시료에 비해 많이 부패하지 않았기 때문에 ethanol 생성량도 많지 않았으며, 0.03mm film의 경우도 다른 film 포장 처리구에 비해서 ethanol 축적량이 많지 않았다. 그러나 0.06mm와 0.1mm film으로 포장한 배추는 저장 30일 이후부터 ethanol 생성량이 급격히 증가하였다. 저온에서 저장한 배추의 유리당 함량을 HPLC로 분석한 결과, 저장 0일에서 15일 사이에 당이 급격히 감소하고 그 이후에는 변화가 적었다. 배추의 유기산 함량은 control의 경우 전체적으로 유기산의 감소량이 가장 적었으며, film별로는 두께가 두꺼울수록 감소량이 많았다. Lactic acid와 acetic acid는 매우 소량 존재하였으며, lactic acid의 경우에는 저장 0일째 보다 저장기간이 길어질수록 약간 증가하였다.
