Fermented Ginseng with Bifidobacterium Inhibits Angiogenesis of Human Umbilical Endothelial Cells in vitro and in vivo

  • Ginseng is a widely-used alternative medicine for the treatment of cancer, diabetes, and cardiovascular diseases. Active components of P. ginseng, absorbed through gastrointestinal tract are the fermented ginsenosides by intestinal microorganisms. In the present study, we investigated the inhibitory effects of fermented ginseng with bifidobacterium (FGb) on the angiogenesis by analyzing in vitro tube formation and invasion assay using human umbilical vein endothelial cells (HUVECs), and in vivo angiogenesis using chick chorioallantoic membrane (CAM) assay. Treatment with FGb inhibited tube-like structure formation in a concentration-dependent manner. In addition, FGb significantly suppressed HUVEC invasion through Matrigel. Moreover, FGb dosedependently inhibited VEGF-induced angiogenesis in a CAM assay. These results suggest that FGb is a valuable anti-angiogenic remedy.

  • Propolis, obtained from honeybee hives, has been used in Oriental folk medicine as an anti-inflammatory, anti-carcinogenic, and immunomodulatory agent. There is considerable evidence suggesting that angiogenesis and chronic inflammation are codependent. Blockage of angiogenesis results in an anti-inflammatory effect. Ethanol (EEP) and ether extracts of propolis (REP), and caffeic acid phenethyl ester (CAPE), an active component of propolis, were examined for their anti-angiogenic activities using the chick embryo chorioallantoic membrane (CAM), and the calf pulmonary arterial endothelial (CPAE) cell proliferation, assays. The presence of EEP, REP and CAPE inhibited angiogenesis in the CAM assay and the proliferation of CPAE cells. The results suggest that anti-angiogenic activities of EEP, REP and CAPE are also responsible for their anti-inflammatory effect.

  • Angiogenesis refers to the formation of new capillary blood vessels, or neovascularization occurring under various physical conditions, such as development of the embryo, formation of corpus luteum, wound healing and pathological conditions including tumor growth and metastases, hemangiomas, diabetic retinopathy, rheumatoid arthritis. There are many evidences that angiogenesis is important for the progressive growth of solid tumors and also permits the shedding of metastatic tumors from the primary site. Thus, treatment of angiogenesis inhibitors might be a novel strategy for tumor growth inhibition. Normal vascular endothelial cells are in a state of differentiation and angiogenic endothelial cells migrate and proliferate, and they subsequently differentiate into vessel-forming quiescent phenotype cells, Therfore, it was speculated that a modifier of cell differentiation could also affect angiogenesis. In order to identify new antiangiogenic factors, the research was conducted to estimate the inhibitory activities of cell differentiation agents by means of chick embryo chorioallantoic membrane(CAM) assay. Hence, we have established the CAM assay for the screening of antiangiogenic agents. Using the CAM assay, we found that ursolic acid, a tumor cell differentiation-inducing agent, showed a markedly inhibitory effect on chick embryonic angiogenesis.

  • Dongchunghacho (Dong-Chong-Xia-Cho in Chinese) is one of entomogenous fungi that grow as parasites mainly to pupae or larvae. It includes many different genera such as Cordyceps, Paecilomyces, Torrubiella and Podonectria. The ethanolic extract of Cordyceps scarabaeicola, prepared from its fruiting bodies, showed significant inhibitory activity on angiogensis, which was detected by chick embryo chorioallantoic membrane (CAM) assay. The ethanolic extract of media-cultured Paecilomyces japonica also showed significant anti-angiogenic activity in CAM assay. (omitted)

  • Human milk was examined for antiangiogenic activities by using the chick embryo chorioallantoic membrane (CAM) assay and endothelial cell growth. The low molecular weight (20 KD>$\sim$ ) fraction of human milk stimulated the angiogenesis and increased the endothelial cell growth. These results suggest that the increase of angiogenesis and endothelial cell growth might be attributed to several growth factors and/or angiogenic factors in low molecular fraction (20 KD>$\sim$) of human milk.

  • Angiogenesis, the formation of new blood vessels, is considered to be involved in many pathological symptoms such as diabetic retinopathy, arthritis, inflammation and solid tumour. In particular, it is thought that angiogenesis is critical for development and growth of solid tumour. Recent study shows that there is a highly significant association of microvessel density with overall survival and relapse-free survival in patients with breast tumour In this study, the inhibition effect of angiogenesis of traditional herbs used for the treatment of cancer was examined. It was found out that the extract of Agaricus blazei by boiling water is a possible inhibitor of angiogenesis. It inhibited normal developmental angiogenesis In the chorioallantoic membrane of chick embryos and also inhibited capillary-like tube formation by endotherial cells on matrigel in vitro. These results suggest that Agaricus blazei can be a potent angiogenesis inhibitor.

  • In the previous study, we reported that 2'-hydroxy-4'-methxoychalcone, synthetic chalcone inhibited PGE2 production in TPA- stimulated rat peritoneal macrophages by inhibiting the induction of COX-2 protein. The present study was carried out to clarify whether 2'-hydroxy-4'-methoxychalcone inhibit angiogenesis by the experimental methods in vitro and in vivo. 2'-Hydroxy-4'-methoxychalcone decreased angiogenesis of both chick embryos in the chorioallantoic membrane assay and basic fibroblast growth factor-induced vessel formation inthe mouse Martigel plug assay.   • Gardenia jasminoides Ellis has been used in traditional medicine for the treatment of inflammation, jaundice, headache, fever and hypertension. The 70% ethanolic extract of gardenia fruit showed strong anti-angiogenic activity in the chick embryo chorioallantoic membrane (CAM) assay. Among hexane, ethyl acetate, n-butanol and aqueous fractions prepared from the 70% ethanolic extract, the n-butanol fraction was found to be most effective in the CAM assay. (omitted)

  • The angiogenic activity of Aloe vera (Aloe baradensis), known as a good healing plant, was investigated. We have extracted and fractionated dichloromethane extract (G1M1D1) and methanol soluble fraction of dichloromethane extract (G1M1D1M1) which contain low-molecular weight substances of Aloe vera gel. G1M1D1 and G1D1M1 fractions induced a radially arranged, spoke-sheel-like vasculature in chick embryo chorioallantoic membrane (CAM) assay. The angiogenic activity was dose-dependent and the angiogenic pattern in the CAM assay. The angiogenic activity was dose-dependent and the angiogenic pattern in the CAM assay was very similar to that of phorbol 12-myristate-13-acetate (PMA) used as a positive control. The modified CAM assay, e simple and accurate quantitating method, was used to quntitate the angiogenic activity of G1D1M1 fraction. Application of G1M1D1M1 fraction ($100\mug/egg$) resulted in much more intense angiogenesis than in control while slightly less intense angiogenesis than in PMA (100 ng/egg).
