본 연구에서는 황벽나무, 두충나무 등을 포함하는 복합수목추출물의 항균활성 효과 및 안전성에 대해 검토하였다. 항균활성은 피부상재균이며 기회 병원성균인 Staphylococcus aureus, Pseudomonas aeruginosa, Escherichia coli 및 Candida albicans에 대해 disc 확산법으로 실험되었다. 안전성 시험으로는 단회 경구투여 독성시험, 단회투여 흡입독성 시험, 반복투여 흡입독성시험이 실시되었다. 항균력효과시험 결과 추출물은 Staphylococcus aureus, Pseudomonas aeruginosa, Candida albicans에 대해 우수한 항균활성을 보여주었으나, Escherichia coli에서는 항균활성이 없었다. 복합수목추출물의 단회 경구투여 독성시험, 단회투여 흡입독성시험 및 반복투여 흡입독성시험의 결과 독성은 관찰되지 않았다. 따라서 황벽나무, 두충나무 등을 포함한 복합수목추출물은 천연 항살균제로서 상업화 가능성이 매우 높음을 알 수 있었다.
KH-502 (Flupyrazofos), a new organophosphorus insecticide synthesized by Korea Re-search Institute of Chemical Technology, was found to be effective against diamond-back moth(Plutella xylostella). This study was carried out to determine the acute toxicity of KH-502 in Sprague-Dawley rats and ICR mice. The test article was orally or dermally administered to the animals. Death, tremors, salivation, lacrimation, abnormal gait and corneal opacity were observed. Decrease in body weight gain was observed in all treatment groups. At necropsy, dark red coloration of lung, enlargement of adrenal glands and atrophy of spleen were observed. The oral $LD_{50}$ value was 372 mg/kg in male rats, 605 mg/kg in female rats, 186 mg/kg in male mice, and 115 mg/kg in female mice. And the dermal $LD_{50}$ was 4086 mg/kg in male and 3881 mg/kg in female rats.
The acute oral LD5O toxicity values of isazofos, pyraclofos, diazinon and methomyl were determined for Japanese quail based on OECD guideline. The $LD_{50}$ of isazofos, pyraclofos and diazinon was 16.26 mg/kg, and 7.11mg/kg body weight In female respectively. And the $LD_{50}$ of each chemical in male was 21.44, 35.64, 8.28 mg/kg body weight respectively. Diazinon was the most susceptible compounds to Japanese quail in both sexes. The $LD_{50}$ of methomyl was 21.24 mg/kg body weights in female, and 28.28 mg/kg body weight in male respectively. Diazinon, isazofos and methomyl were more toxic In the female than male. The symptoms of poisoning were similar in quails administrated with each chemicals. The clinical sign in Japanese quail were ataxia, salivation, diarrhea, ruffled feather and convulsion at dead point. There were severe hemorrhage and catarrhal inflammation from duodenum to ileum In all compounds. In Japanese quail treated with organophosphorus and carbamate compounds, brain acetylcholinesterase was inhibited by 88-96. The recovery was not observed after 5 h in sublethal dose.
This study assessed the contribution of heavy metals to total toxicity as well as the presence other toxic compounds before and after adding the chemical P to concurrently conducted bioassay tests of Daphnia magna and P. Phosphoreum. The following conclusions were drawn from this study : With excessive EDTA dosage, a toxicity reduction in Microtox would occur due to a metal-comples being formed. Microtox was far less sensitive than D. magna to heavy metal toxicity, but extended exposure time and reagent could increase the sensitivity.
To screen biologically active components of the higher fungi of Korea, the carpophores of Auricularia polytricha, a well-known edible mushroom, were extracted with 0.14M NaCl solution. The extractive was successively fractionated by adding ammonium sulfate in various amounts, the respective precipitates being weparated by centrifugation, dialyzed and freeze-dried. When a dose of 60mg/kg of each was, i.p., injected into ICR mice, the fraction shich was precopitated at 20% (NH/sub 4/)/sub 2/SO/sub 4/ showed the highest toxicity, killing seven mice within two days. The fraction obtained at 40% (NH/sub 4/)/sub 2/SO/sub 4/ showed the second highest toxicity. The two fractions were named auratoxin I and II after the genus name. The symptoms of the intoxication were convulsion during the first 30 minutes after the injection, then sleeping within an hour, and tremor, lacrimation, nasal and ophthalmic bleeding, congestion and death in 24 hours. Particularly the spleen of the mice was fonud to be enlarged remarkably. The chemical analysis of the toxins showed that auratoxin I consisted of 4.4% protein and 84.5% polysaccharide and that auratoxin II 35.8% protein and 48.0% polysaccharide.
Occupational exposure to wollastonite can occur during its mining, processing and using. Wollastonite was analyzed for its physicochemical properties by transmission electron microscope and scanning electron microscope equipped with energy dispersive X-ray spectrometer. Wollastonite fibers were 3.3${\mu}m$ in diameter(av.) and were 30.5${\mu}m$ in length(av.). The atomic composition of wollastonite was Si 56.41% and Ca 43.59%. Respiratory toxicity of wollastonite has been studied in term sequential in Sprague-Dawely rats. UICC chrysotile (average diameter 0.03${\mu}m$, average length 2.93${\mu}m$) was applied as the positive control. The effects of 2mg wollastonite on respiratory system and pathological changes were evaluated after 1, 4, 8, 12 weeks instilled into rat lungs. Inflammation response broke out from 1 week after instilled with wollastonite and the pathological examination further showed increased legions of granulomatous inflammation after 4 weeks, but decreased granulomatous inflammation after 8 weeks, whereas chrysotile induced progressive interstitial granulomatous inflammation and fibrosis as a function of time.
Pulmonary toxicity of talc containing tremolite asbestos (TCT) has been studied in term sequential in Sprague-Dawely rats. UICC chrysotile(average diameter 0.03${\mu}m$, average length 2.93${\mu}m$) was applied as the positive control. TCT was analyzed for its physicochemical properties by transmission electron microscope equipped with energy dispersive X-ray spectrometer(TEM-EDS). The effects of 2mg TCT(talc average diameter 9.7${\pm}$8.4${\mu}m$; tremolite average diameter 1.6${\pm}$1.6${\mu}m$, average length 10.8${\pm}$7.0${\mu}m$) on pathological changes were evaluated after 1, 8 weeks instilled into rat lungs. 2mg Chrysotile continuously affected lung pathological changes. Inflammation and granuloma response broke out from 1 week after instilled with chrysotile and the pathological examination further showed increased legions of lung after 8 weeks. But TCT did not showed lung pathological changes. The biopersistence of TCT and chrysotile was evaluated by TEM- EDS. Whereas chrysotile continuously have retained to 8 weeks instilled into rat lungs, talc of TCT showed statistically significant decrease of diameter from 1 weeks and statistically significant change in Si atomic % compositions at 8 weeks instilled into rat lungs. Physicochemical properties of tremolite of TCT were not affected until 8 weeks instilled into rat lungs. This study showed that the durability of TCT in the lungs is much weaker than chrysotile.
동물실험에 의존하는 전통적인 위해성평가 방법의 한계가 명확하게 드러나면서, 동물실험이 아닌 최첨단 기술을 활용한 화학물질 위해성평가 방법론들이 연구되고 있다. 이는 곧 신규접근법(New Approach Methodologies, NAMs)이라는 개념으로 구체화되었으며 차세대 위해성평가(Next generation risk assessment, NGRA) 프레임워크로 구현되었다. 하지만 신규접근법 데이터는 신뢰도 문제로 인해 극히 제한적으로 화학물질의 규제 의사결정과정에 활용되고있다. 이에 규제 공동협력기관 APCRA 이니셔티브는 화학물질 위해성평가에 신규접근법 데이터의 활용을 증진시키기 위해 여러 사례연구를 수행하였으며, 이를 바탕으로 차세대 위해성평가의 전망에 대해 논의한 바 있다. 따라서 본 총설에는 APCRA 이니셔티브가 수행한 사례연구 두 편을 중심으로 신규접근법을 활용한 차세대 위해성평가의 개념과 전망을 제시하였다.
Objectives: The present study was conducted in order to investigate the reproductive toxicity in rats exposed to 1,4-dichlorobutane. Methods: The test chemical was administered orally at 0, 8.3, 50 and 300 mg/kg/day. Males were administered daily for 10 weeks prior to the mating period. Females were administered from between two weeks before mating to the 21stday of lactation. Results: In both sexes, a decrease in body weight and an increase in the weights of the liver and kidneys were observed. In males, discoloration of the liver, hepatocyte hypertrophy and mineralization in the kidneys were observed. In females, animal deaths, dystocia and pup deaths due to maternal dysfunction were observed. In F1 animals of both sexes, a decrease in body weight was observed at 300 mg/kg/day. An increase in the weights of the liver in both sexes, mineralization in the kidneys of males, animal deaths, hepatocyte hypertrophy and pup deaths due to maternal dysfunction were observed at 50 mg/kg/day. Mineralization in the kidneys of males was observed at 8.3 mg/kg/day. Therefore, the no-observed-adverse-effect levels (NOAELs) of 1,4- dichlorobutane were considered to be under 8.3 mg/kg/day for males, 8.3 mg/kg/day for females, more than 300 mg/kg/day for fertility in both sexes, 8.3 mg/kg/day for maternal functions and 50 mg/kg/day for F1 offspring. The absolute toxic dose was believed to be 8.3 mg/kg/day for males, 50 mg/kg/day for females, 50 mg/kg/day for maternal functions and 300 mg/kg/day for F1 offspring. However NOAEL for fertility could not be determined since there were no treatment-related changes. Conclusions: Under the present experimental conditions, 1,4-dichlorobutane is a Category 1B Reproductive Toxicant (presumed human reproductive or developmental toxicant).
The purpose of this study is to investigate toxic effects of iso-butylalcohol (iBA) in Sprague-Dawley (SD) rats under the exposure of 6 hours a day, 5 days a week for 13 weeks by inhalation, and to evaluate the occupational safety of iBA in comparison with the permissible exposure level (PEL) stipulated by the Occupational Safety and Health Administration (OSHA). iBA did not induce any abnormal changes from the aspects of clinical signs, feed consumption, ophthalmic test, urinalysis, hematology and blood chemistry during and at the terminal of the inhalation toxicity tests. We did not find any abnormal findings in the gross and microscopic observations due to the inhalation of iBA. There was no alteration in relative organ weights by the inhalation of iBA. No observed adverse effect level (NOAEL) of iBA was considered to be more than 3,000 ppm in rats under the inhalation of 6 hours a day, 5 days a week for 13 weeks. Fifty ppm of iBA, the PEL regulated by OSHA, is too conservative for working places. As iBA showed no abnormal observations in all the experimental parameters at any concentration under this experimental condition, we suggest that 150 ppm is safe enough for the PEL of iBA in the working areas, even taking into onsideration that OSHA lowered the PEL to 50 ppm for fear of the probable risk of its skin irritation.
