Effects of Various Thermal Treatments on Physicochemical Properties of Wheat Bran (밀기울에 대한 열처리가 이화학적 특성에 미치는 영향)

  • Various thermal treatments such as toasting, roasting, drum drying, autoclaving+drum drying, microwaving+drum drying and extrusion were attempted to investigate their effects on chemical composition (proximate composition, Klason lignin, neutral sugar, uronic acid, starch, free sugar, dietary fiber, phytic acid, etc) and functional properties (water solubility index, water absorption index, water holding capacity, oil holding capacity, bulk density, swelling, etc) of wheat bran. Thermal treatments on wheat bran increased soluble dietary fiber (SDF) content from 2.7% (raw sample) to 4.6% (toasted), 4.5% (roasted), 4.6% (drum dried), 5.2% (autoclaved), 3.7% (microwaved), 5.6% (extruded). In contrast, total dietary fiber (TDF) content remained nearly constant regardless of thermal treatments, reflecting the concomitant decrease in insoluble dietary fiber (IDF). It was also found that water holding capacity (WHC) increased $20{\sim}75%$ due to thermal treatments employed in this research. Both bulk density and swelling were not directly relevant to WHC, but thier multiplication exhibited good correlation $(R^2=0.94)$ with WHC. Scanning elctron microscopy (SEM) exhibited that structure of raw wheat bran was significantly modified, whose degree and shape well reflected the types of thermal treatments.

  • Electro-electrodialysis of hydriodic acid with HI molality of ca. 9.5 $mol/kg-H_2O$ was examined in the presence of iodine using a commercial cation exchange membrane, CMB, as a separator. For the increase of the selectivity of proton permeation, the membrane was radiation-treated by accelerated electron radiation. The membrane properties (area resistance, ion exchange capacity, water content) of the radiation-treated membranes were measured. The area resistance in 2 $mol/dm^3$ KCl solution, ion exchange capacity and water content of the radiation-treated membranes at each dose rate dad almost the same value as that of the non-treated membrane (original of CMB membrane). Electro-electrodialysis of hydriodic acid with HI molality of ca. 9.5 $mol/kg-H_2O$ was examined at $75^{\circ}C$ with 9.6 $A/dm^2$. The radiation-treated cation exchange membrane by accelerated electron radiation had higher selectivity of the proton permeation by cross-linking structure of polymer than that of the non-treated membrane.

Analysis of Material Characteristics for Blackwares at Excavated from the Bonggok Kiln Site of Jinan-gun (진안 봉곡도요지 출토 흑유자기의 재료학적 특성 분석)

  • The purpose of this study is to identify the material characteristics of blackwares excavated from the Bonggok kiln site in Jinan-gun, Jeollabuk-do Province, South Korea, by scientific analyses and a reproduction experiment. Scientific analyses were conducted to determine the physical properties, chemical composition and for microscopic observation of blackwares. A reproduction experiment was also conducted by producing ceramic samples with black glaze, red ocher and limestone, and then cross sections of the samples were observed. The analysis results showed that all the excavated blackwares have similar properties such as chromaticity, specific gravity, absorption factor and porosity, but there is a difference depending on the crystallization of the ceramics. Thermal analysis showed that the exothermic peak was observed at about $1200^{\circ}C$, and crystal structure analysis indicated the presence of mullite. Hence, it can be concluded that the firing temperature of the excavated blackwares was around $1100-1200^{\circ}C$. Moreover, the glaze present on these blackwares has about 22% higher $Fe_2O_3$ content than those excavated from other places; however, the former has lower CaO content. For the reproduction experiment, samples were made using limestone as a flux and red ocher as a glaze. The results show that the cross section of the glaze layer of the reproduced sample have iron crystals with dendritic structures, similar to those present in the excavated black-wares. It is assumed that such iron crystals were formed during the process of melting and solidification of the iron oxide present in the blackwares.

  • The cultivation of ginseng plant (Panax ginseng C.A. Meyer) in Korea as an eminent medicinal herb may be traced far back in history. However, the practices in cultivation have not much improved in terms of efficiency and scientific farming. In the present study some experiments were undertaken for the search of the soil and nutrition conditions, because of the nutritional requirement of ginseng plant shaws quite unique compared with other crops. In both the seed bed and the field 'Yakto' has been traditionally employed or the prime source of nutrition of the crop. Yakto is a complex matter prepared from raw foliage of the broad-leaved trees as the main portion with the admixture of a variety of organic nitrogen source through fermentative processes. The composition of Yakto may be classified coarsely into the decomposed and undecomposed substances, the former being further fractionated according their solubilities, comprising also various colloidal matters whose composition and structure are yet to be known. The Yakto-fractions were subjected to analyze for search of its nature and coarse composition in terms of the distribution of nitrogen, contents of organic functional groups such as -COOH, phenolic-OH, alcholic-OH and methoxyl and hydrolysable sugars. Furthermore, absorption-spectra of each fraction were determined in visible and infrared region and compared the results each other.

Effects of Insecticides on Enzyme Activities in Soil Environment (살충제(殺蟲劑)가 토양환경중(土壤環境中) 효소활성(酵素活性)에 미치는 영향(影響))

  • The effects of insecticides on biochemical precesses in soil were studied by determining the effects of the chemical structure of each insecticides on enzyme activities, pesticide residue and total number of bacteria revealed when soil treated with urea was incubated at $28{\pm}1^{\circ}$ for 56 days. The inhibition effects of insectides on enzyme activites in soil decreased in the order: dithiophosphoric acid > thiophosphhoric acid > phosphoric acid > carbamate insecticides for urease and phosphatase, thiophosphoric acid > dithiophosphoric acid > phosphoric acid > carbamate insecticides for L-glutaminase and protease. The inhibition effects of organophophorus insecticides on enzyme activities in soil were maintained longer than those of carbamate insecticides. Carbamate insecticides increased the activities of protease and L-glutaminase at 56 days. When insecticides were treated in soil together with urea, the degradation of insecticides was accelerated. By treatment of insecticides, the total number of bacteria was decreased at the early stage of treatment but thereafter increased according to phosphoric acid and carbamate insecticides.

  • The Prussian Blue Analogue(PBA) has three dimensional structure and the metal - organic framework material, and it has a variety configurations depending on the type of organic ligands. PBA has been receving an attention in the fields of biosensors, optical, catalytic, and hydrogen storage device. Also, it is an environmental friendly substance with a chemical stability. In addition, PBA is widely used in the filed of adsorption art since we can adjust the size of the fine pores. In this study, we synthesized $Mn_3[Fe(CN)_6]_2$, an organometallic framework chains by using a hydrothermal synthesis method. We used $K_4[Fe(CN)_6]$ and $MnCl_2$ as precursors. We also produced a manganese iron oxide, by baking the synthesized material. The effect of the size and shape of the particles was examined by controling pH of the precursor solution, the molar concentration of the precursor, and reaction time as the experimental variables. Synthesized absorbent was analyzed by XRD, SEM, FT-IR, UV-Vis, and TG / DTA to evaluate the adsorption properties of several dyes.

Purification and Structure of Antioxidative Substance Derived from Tetraselmis suecica (Tetraselmis suecica유래의 항산화성 물질의 정제 및 구조)

  • Tetraselmis suecica (T. suecica) of Prasinophyta was selected because the growth rate is comparatively higher and the culturing is also easy. In order to investigate antioxidative activity, the soluble elements of T. suecica were fractionated using water and organic solvents such as methanol, hexane, chloroform, ethylacetate and butanol. The chloroform fraction of T. suecica showed strong antioxidative activity. The potential antioxidative activity was detected in hexane: ethylacetate (1:5) once used the fractions by different mixtures of organic solvents. This fraction was further purified by preparative thin layer chromatography (PTLC) and repeated reverse-phase HPLC. On the basis of chemical and spectroscopic evidences obtained by UV, FT-IR, FAB-MS and NMR, the compound purified from T. suecica was identified as pheophorbide-a. The antioxidative activity of the compound was comparable to that of $\alpha$-tocopherol and could be act as an antioxidant in foods.

Comparison of Cuticular Hydrocarbons of Different Developmental Stages of the Spot Clothing Wax Cicada, Lycorma delicatula (Hemiptera: Fulgoridae) (꽃매미(Lycorma delicatula)의 발육단계별 표피탄화수소 비교)

  • Aliphatic cuticular hydrocarbons (CHCs) of different developmental stages of the spot clothing wax cicada, Lycorma delicatula (Hemiptera: Fulgoridae) were analyzed using GC and GC-MS. The numbers of carbons in the major CHCs of each developmental stage 32, 33, 28, 38, 37 in the egg, 1st, 2nd, 3rd, and 4th instar nymphal stages, and adults, respectively. The cuticle of Lycorma delicatula contains mainly methyl-branched 9-methylheptacosane (15.11%) in the egg stage, and a high proportion of n-heptacosane in nymphal stages (15.75, 22.42, 25.04, and 23.11 % in the 1st, 2nd, 3rd and 4th instars, respectively). In contrast, male and female adults had high proportions of n-nonacosane (13.42 and 16.55%). The chemical constituents of CHCs were classified into five groups (n-alkanes, monomethylalkanes, dimethylalkanes, trimethylalkanes, olefins) and group profiles of each developmental stage were compared. Egg surface was composed mainly monomethylalkanes (45.39%), a saturated hydrocarbon. Nymph CHCs consisted primarily of n-alkanes (37.63 to 46.12%). There was a difference between adult male and female CHCs. However, both contained n-alkanes and monomethylalkanes. CHCs with trimethyl or double bonded structure were rare in all stages.

Fabrication, Estimation and Trypsin Digestion Experiment of the Thermally Isolated Micro Teactor for Bio-chemical Reaction

  • This paper describes design, fabrication, and application of the silicon based temperature controllable micro reactor. In order to achieve fast temperature variation and low energy consumption, reaction chamber of the micro reactor was thermally isolated by etching the highly conductive silicon around the reaction chamber. Compared with the model not having thermally isolated structure, the thermally isolated micro reactor showed enhanced thermal performances such as fast temperature variation and low energy consumption. The performance enhancements of the micro reactor due to etched holes were verified by thermal experiment and numerical analysis. Regarding to 42 percents reduction of the thermal mass achieved by the etched holes, approximately 4 times faster thermal variation and 5 times smaller energy consumption were acquired. The total size of the fabricated micro reactor was $37{\times}30{\times}1mm^{3}$. Microchannel and reaction chamber were formed on the silicon substrate. The openings of channel and chamber were covered by the glass substrate. The Pt electrodes for heater and sensor are fabricated on the backside of silicon substrate below the reaction chamber. The dimension of channel cross section was $200{\times}100{\mu}m^{2}$. The volume of reaction chamber was $4{\mu}l$. The temperature of the micro reactor was controlled and measured simultaneously with NI DAQ PCI-MIO-16E-l board and LabVIEW program. Finally, the fabricated micro reactor and the temperature control system were applied to the thermal denaturation and the trypsin digestion of protein. BSA(bovine serum albumin) was chosen for the test sample. It was successfully shown that BSA was successfully denatured at $75^{\circ}C$ for 1 min and digested by trypsin at $37^{\circ}C$ for 10 min.

A Study of Electrical Anisotropy of n-type a-plane GaN films grown on $\gamma$-plane Sapphire Substrates ($\gamma$-plane 사파이어 기판 위에 성장한 무분극 ${alpha}$-plane GaN 층의 전기적 비등방성 연구)

  • We report on the electrical properties of Ti/Al/Ni/Au (20 nm/ 150 nm/ 30 nm/ 100 nm) Ohmic contacts and the anisotropic conductivity of n-type ${\alpha}$-plane ([11-20]) GaN grown on $\gamma$-plane ([1-102]) sapphire substrates. The Ti/Al/Ni/Au Ohmic contacts and their sheet resistances are characterized by using the transfer length method (TLM) as a function of azimuthal angles. It is found that the specific contact resistance does not depend on the axis orientation and there are significant electrical anisotropy in ${\alpha}$-plane GaN films on $\gamma$-plane sapphire substrates, and the sheet resistance varies with azimuthal angles. The sheet resistance values in the direction parallel to m-axis [1-100] are 25% ~ 75% lower than those parallel to c-axis [0001] directions. Thus, Basal stacking faults (BSFs) are offered as a feasible source of the anisotropic mobility in defected m-axis direction because the band-edge discontinuities owing to the differential band gap structure.
