Inhibitory Effects of Chlorine Dioxide and a Commercial Chlorine Sanitizer Against Foodborne Pathogens on Lettuce (양상추에 오염된 병원성 미생물에 대한 Chlorine Dioxide 및 상업적 Chlorine 살균소독제의 저해효과 평가)

  • This study compared the effects of chlorine dioxide and a commercial chlorine sanitizer for inhibiting foodborne pathogens, including Salmonella enterica serovar Typhimurium, Listeria monocytogenes, and Escherichia coli O157 : H7, on lettuce leaves. The lettuce samples were inoculated with each cocktail of the three strains, and were then treated with chemical sanitizers [distilled water, 100 ppm commercial chlorine and 50 ppm, 100 ppm, 200 ppm chlorine dioxide ($ClO_2$)] for 1 min, 5 min, and 10 min at room temperature($22{\pm}2^{\circ}C$). Following inoculation of the leaves, initial populations of E. coli O157:H7, L. monocytogenes, and S. Typhimurium were approximately 5.54, 4.47, and 5.12 log CFU/g, respectively these levels were not significantly reduced by the treatment with water,whereas the 100 ppm commercial chlorine sanitizer treatment and $ClO_2$ (at all tested concentrations) were effective at reducing levels of all three pathogens. The treatment of 200 ppm $ClO_2$ for 10 min was most effective at inhibiting the three pathogens, and reduction levels of E. coli O157 : H7, L. monocytogenes, and S. Typhimurium were 2.28, 1.95, 1.76 log, respectively. The inhibitory effect of $ClO_2$ increased with increasing treatment concentration of $ClO_2$, but there was no significant difference by the treatment times. When chemically injured cells of E. coli O157 : H7 and L. monocytogenes and S. Typhimurium were examined by SPRAB and selective overlay methods, respectively, it was observed that the commercial chlorine sanitizer generated greater numbers of injured L. monocytogenes than the $ClO_2$ treatment. From the overall results, $ClO_2$ was more effective at inhibiting pathogenic bacteria compared to the commercial chlorine sanitizer therefore, it has potential to be utilized as an alternative sanitizer to increase the microbial safety of fresh produce.

Relationship between some chemical components in the rice plants and varietal reaction to blast disease (도열병 품종저항성과 도체내 성분과의 관계)

  • An investigation was carried out to know the relationship between resistance of rice varieties to blast disease and the chemical components, especially total nitrogen and total sugars, in the rice plants. The results sic summarized as follow: 1) The nitrogen contents in the resistant variety were less than those of susceptible one, and sugar contents were reversed. Accordingly, the C/N ratio in the resistant variety was higher than that obtained by susceptible one. 2) The free amino acids contents, especially, Glutamine, Valine, Leusine and Iso-leusine, in the resistant varieties were more than those of the susceptible varieties. 3) The starch synthetic activity of rice leaves in the resistant varieties was higher than that of susceptible one in fructose and glucose solutions, but it was reversed in sucrose solution. 4) When more nitrogen was dressed, the total nitrogen content of rice leaves was increased than the ordinary dressing. The rate of increase in nitrogen content in resistant variety was lower than those of the susceptible. The total sugar content of rice plants dressed more nitrogen was decreased at early tillering stage, but increased at maximum tillering stage. It seemed that the rate of increase of total sugar in the resistant variety was higher than those of the susceptible.

Anti-inflammatory Triterpenes from Euonymus alatus Leaves and Twigs on Lipopolysaccharide-activated RAW264.7 Macrophage Cells

  • As a part of ongoing phytochemical research on Euonymus alatus leaves and twigs, we have isolated one new reissantane-type triterpene (1), together with eight known triterpenes, isoarborinol (2), friedelin (3), abruslactone A (4), $3{\beta},22{\alpha}$-dihydroxyolean-12-en-29-oic acid (5), $3{\alpha},22{\alpha}$-dihydroxyolean-12-en-29-oic acid (6), $3{\alpha},22{\beta}$-dihydroxyolean-12-en-29-oic acid (7), $22{\alpha}$-hydroxy-3-oxoolean-12-en-29-oic acid (8), demethylregelin (9). The structure of 1 has been elucidated as 24,24-dimethyl-reissant-7(8),25-dien-$3{\alpha}$-ol, by extensive 1D and 2D spectroscopic methods including $^1H$-NMR, $^{13}C$-NMR, $^1H-^1H$ COSY, HSQC, HMBC and NOESY. Anti-inflammatory activities of the isolated compounds were determined as potential regulating excessive inflammatory responses in RAW264.7 macrophage cells. Particularly, abruslactone A (4) and demethylregelin (9) showed the most potent activity, which effectively reduced the expression of iNOS protein and subsequent nitric oxide production induced by lipopolysaccharide in RAW264.7 cells.

Correlation Analysis between Growth and Environmental Characteristics in Abeliophyllum distichum Habitats (미선나무 자생지의 생육특성과 환경특성간의 상관분석)

  • The purpose of this study is to propose the raw data of conservation and restoration by analyzing the environmental and growth characteristics in Abeliophyllum distichum habitats. The types of soil were sandy loam, loam and silty clay loam and the correlation on chemical characteristics of soil appeared that T-N and C.E.C were highly correlative. In case of growth characteristics, height and diameter in 1st site, crown in 2nd site, number of leaves in 5th site and number of seeds in 4th site were better than other sites. In the results of correlation on growth characteristics, height and diameter were highly correlative. In case of growth and chemical characteristics, EC and height were highly correlative. In the results of regression analysis, R-square of height and EC was some 70.4% and that of number of leaves and T-N was some 70.2%. To conserve and restore Abeliophyllum distichum habitats, there were demanded that the accurate environmental analysis that considered such diverse factors as light intensity, temperature, genetic quality and so forth.

Isolated and Identification of Biological Activity Compounds from Leaves and Stem of Paeonia lactiflora Pallas (작약 잎과 줄기의 생리활성 물질 분리 및 동정)

  • This study was conducted to identify physiologically active substances from leaves and stems of peony plant. MeOH extracts and column chromatography were employed to isolate active compounds and chemical structure were identified by IR, UV, Mass and NMR. The results obtained can be summerized as followings : Chemical structure of compound 1 was identified as oleanolic acid (white color form) of triterpenoid group, which is firstly identified from the above part of peony. Compound 2 was identified as kaempferol (yellow needle form) of flavonoid group, which was firstly identified from the root, leaf and stem of peony. Compound 3 was identified as methyl gallate (white power form) of phenol group, which was firstly identified from the above part of peony. Compound 4 was identified as astragalin (bright yellow needle form) of flavonoid group, that was firstly identified from the leaf and stem of peony. Compound 5 was identified as paeoniflorin (white color form) of monoterpene group, that was firstly identified from the above part of peony.

Enhanced oxidative stability of meat by including tannin-rich leaves of woody plants in goat diet

  • Objective: The aim of this study was to evaluate the effect of dietary incorporation of tanninrich woody species on meat oxidative stability, carcass traits and meat quality in goats. Methods: Two tannin-rich species were tested using a three-treatments feeding trial, where treatments consisted of: Larrea divaricata and Acacia aroma both at 12.5% in dry matter basis of the diet and a control diet (alfalfa hay). All feeding diets were iso-protein and iso-energy. Carcass conformation, carcass compactness, carcass fatness and subcutaneous fat deposition were evaluated. Intake, liveweigh, Longissimus thoracis et lumborum muscles of goats were analyzed in order to evaluate quality parameters such as pH value, instrumental color evaluation, water holding capacity, total phenolic content, antioxidant activity, meat oxidative stability and fatty acid profiles in meat. Results: Feed intake, liveweight gain, carcass, and meat traits did not differ among treatments. Changes in meat lipid profile among treatments were observed for oleic and elaidic acid contents. Meat total phenolic content and antioxidant activity did not differ among treatments; although, meat oxidative status after storage at room temperature, as well as under refrigerated and frozen conditions were different between control and both supplemented groups. Conclusion: The inclusion of Acacia aroma and Larrea divaricata leaves in goat diet enhanced meat oxidative stability. Modulation of the ruminal biohydrogenation of fatty acids produced by condensed tannins of these plant species need to be further investigated.

Separation and Characteristics of Essential Oil from Dendropanax morbiferus (황칠나무 정유의 분리 및 특성)

  • Essential oil was separated from Dendropanax morbiferus (DM) by means of hot water extraction and supercritical extraction, respectively, and the separated essential oil was analyzed qualitatively and quantitatively, and the antioxidant effect of essential oil was investigated. In addition, yields of essential oil and Caryophyllene according to the drying and extraction methods for each part of DM were obtained and compared, respectively. The yield of essential oil was found to be high in the order of bark > leaves > twigs > limb. When the freeze-dried DM leaves were supercritically extracted, the yields of essential oil and Caryophyllene were 23.2 g/kg DM and 429.6 mg Caryophyllene/kg DM, respectively. The essential oil of DM showed an antioxidant effect even at a low concentration, and the concentration of the essential oil of SC50, which means 50% of 2,2-diphenyl-1-picrylhydrazyl (DPPH) radical scavenging activity, was about 0.34%.

Path-Coefficient Aanalysis of Yield-Characters in Tobacco (연초 수량 및 수량관련형질들의 경로분석)

  • The study was intended to clarify the direct and indirect effects of agronomic and chemical characters influencing upon yield in tobacco breeding. The eight parents and a set of 28 crosses of F$_1$'s and F$_2$'s were used as materials, and planted on two different cultivated systems, i.e., oriental's and burley systems, during 1986 to 1988 at Daegu Experiment Station. Four characters which were leaf length, leaf width, days to flowering and number of leaves per plant influenced the highly direct effects upon the leaf yield, but five chemical components were expressed as low effects. In indirect effects, leaf length, days to flowering and total nitrogen had the high indirect effects influencing upon leaf yield via the other characters. Accordingly, the results were discussed with a conclusion that the selection should be based on the data from leaf length, days to flowering, leaf width and number of leaves per plant.

Physicochemical Characteristics of Powder from Hot Air and Freeze Dried Leaves and Roots of Acorous calamus L. (열풍건조와 동결건조에 따른 수창포(Acorous calamus L.) 분말의 부위별 이화학적 특성)

  • The physical and chemical characteristics of powders from hot air and freeze dried leaves and roots of Acorous calamus L. were investigated. Two parts, upper and lower, of leaves, and two kinds of roots, 4 and 6 years old, were dried at 30 and $40^{\circ}C$, freeze-dried, and ground to make powder. Contents of moisture, ash, crude fat, and crude protein in freeze dried powder of upper leaf were 4.87%, 6.73%, 2.22%, and 3.57%, respectively. Water absorption index (WAI) and water solubility index (WSI) in freeze dried powder of lower and upper leaves were 8.476 and 0.077 g/mL. Contents of chlorophyll a, chlorophyll b and total chlorophyll in freeze dried powder of upper leaves were 12.18, 16.86, and 29.11 mg/100 g, respectively. Contents of total and reducing sugar in freeze dried powder of 4 and 6 years roots were $111.89{\sim}119.21$ ppm and $5.02{\sim}5.22$ ppm, $109.92{\sim}114.65$ ppm and $5.21{\sim}5.32ppm$. Contents of total polyphenols and flavonoids in freeze dried powder of upper leaf were 125.02 and $21.02{\mu}g/mg$, respectively.

Effect of Lime Materials Application on Reducing Injury of Simulated Acid Rain in Soybean (콩의 산성비 피해경감을 위한 석회물질의 시용효과)

  • This experiment was conducted to find out the effects of lime materials application on reducing injury of simulated acid rain(SAR) in soybean grown in pot contained with sandy loam. Six treatments including control, slaked lime(SL), 1% and 2% lime water(LW) and composite treatments with SL+LW were applied. Slaked lime was applied to soil in pot before planting, and lime water was applied to leaves a day prior to the spray of SAR(pH 2.7) and normal rain(pH 6.0), and these were sprayed at 2-day intervals. Growth, yield and yield components, foliar injury rate, chrolophyll content and photosynthetic activity in leaves, content of mineral nutrients in plant and soil chemical properties were analyzed and investigated. These results obtained are summarized as follows : Seed yield of all lime treatment was reduced by SAR compared with control. But seed yield of all lime treatment was increased with treatment of lime material in soil and on leaves. After 15 and 45 times spray of SAR, all lime treatments were effective in injury reducing visible injury of leaves compared with none treatment. Chlorophyll content in leaves was highest in plants treated with slaked lime+ 1% lime water and photosynthetic activity was highest with treatment of slaked lime. Concentration of total nitrogen, phosphate, and sulfur in soybean plant were increased by the spray of SAR. Concentration of total nitrogen, potassium and calcium in soybean plant were increased with treatment of slaked lime into soil. By treatments of SAR, soil pH was decreased, and total nitrogen and sulfur concentration in soil were increased. However, available phosphate and exchangeable cations in soil such as calcium, magnesium and potassium were reduced. Soil pH, calcium and silicate concentration were increased with treatment of slaked lime into soil.
