The Cytotoxicity of Eutigosides from Eurya emarginata Against HL-60 Promyelocytic Leukemia Cells

  • Two phenolic glucosides, eutigoside Band eutigoside C were isolated from the fresh leaves of Eurya emarginata. These two phenolic glucosides exerted a significant inhibitory effect on the growth of HL-60 promyelocytic leukemia cells. Furthermore, when the HL-60 cells were treated with eutigoside C, several apoptotic characteristics such as DNA fragmentation, morphologic changes, and increase of the population of sub-G1 hypodiploid cells were observed. In order to understand the mechanism of apoptosis induction by eutigoside C, we examined the changes of Bcl-2 and Bax expression levels. The eutigoside C reduced BcI-2 protein and mRNA levels, but slightly increased Bax protein and mRNA levels in a time-dependent manner. When we examined the activation of caspase-3, an effector of apoptosis, the eutigoside C increased the expression of active form (19-kDa) of caspase-3 and the increase of their activities was demonstrated by the cleavage of poly (ADP-ribose) polymerase, a substrate of caspase-3, to 85-kDa. The results suggest that the inhibitory effect of eutigoside C from E. emarginata on the growth of HL-60 appears to arise from the induction of apoptosis via the down-regulation of BcI-2 and the activation of caspase.

  • Escherichia Coli ornithine transcarbamylase is the enzyme which catalyzes the L-citrulline biosynthesis from L-ornithine and carbamyl phosphate. To facilitate the purification of enzyme which will be used for many biochemical studies such as structure and function relationships and catalytic mechanisms, the cloning and expression of E. coli argI gene for ornithine transcarbamylase was conducted. argI was amplified from genomic DNA of E. coli strain of $DH5{\alpha}$, by polymerization chain reaction (PCR) method. The amplified argI gene was ligated to the prokaryotic expression vector pKK223-3 and used for transformation of E. coli TB2 which was deficient of ornithine transcarbamylase. The over-produced enzyme by the tnansformant was purified by ammonium sulfate fractionation, heat denaturation and affinity chromatography. The result of SDS denaturation gel electrophoresis for the purified enzyme showed a single band of about 38 kDa of ornithine transcarbamylase. Kinetic data for the expressed enzyme gave almost the s?????? values as those of the wild type enzyme. The $k_{cat}$, of the enzyme was $1.0{\times}10^5min^{-1}$, and $K_ms$ for ornithine and carbamyl phosphate were 0.35 mM and 0.06 mM, respectively.

Anti-inflammatory Effect of Unripe Fruit of Citrus grandis Osbeck in RAW 264.7 and HaCaT Cells (RAW 264.7 및 HaCaT Cell에서 당유자 미숙과의 염증억제 효과)

  • We investigated the anti-inflammatory activities of unripe fruit of Citrus grandis Osbeck growing at Jeju Island, through the evaluation of their inhibitory effect on the production of inflammatory markers (IL-6, iNOS, COX, TARC and MDC) in RAW264.7 murine macrophage cells and HaCaT human keratinocyte cells. Among the sequential solvent fractions obtained from crude extract, hexane and chloroform $(CHCI_3)$ fractions showed potential inhibitory activity on the mRNA expressions of IL-6, iNOS and COX-2 at the concentration of $100\;{\mu}g/ml$ in RAW264.7 cells. Also, EtOAc fraction showed inhibitory activity on the thymus and activation-regulated chemokine (TARC)/CCL17 and macrophage-derived chemokine (MDC)/CCL22 at the concentration of $50\;{\mu}g/ml$ in HaCaT cells. These results suggest that the unripe fruit of C. grandis may have anti-inflammatory activity through the suppression of inflammatory markers (IL-6, iNOS, COX, TARC and MDC).

The Reverse Effects by Electroacupunture on Jun Chung (GV-26) in Dogs (개에서 정중(GV-26) 혈위 전침이 마취 회복에 미치는 영향)

  • This study was performed to investigate the reverse effects of Jen Chung(Ren Thong, Shui Gou, GV-26) electroacupuncture after tiletamime-zolazepam administration in dogs. Seven healthy dogs ranging in weigh from 3.5 to 6.5 kg were used in this experiment. The treatment group was electrostimulated to Jen Chung(+) and Su Liao(Shan Gen, GV-25, -) for 20 minutes after 10 minutes of anesthesia. The control group I was anesthetized with tiletamine-zolazepam. The control group II was electrostimulated to nonacupuncture point for 20 minutes after 10 minutes of anesthesia. Various parameters were evaluated including the onset and recovery time of anesthesia, heart rates, body temperature, respiratory rates and electrocardiogram. The recovery time of the treatment group was shorter than that of the control group I and the control group II(p < 0.05). The treatment group had an increase in the heart rate from 20 minutes to 30 minutes after administration of tiletamime-zolazepam compared to the control group I and control group II(p < 0.05). The decrease in the body temperature was observed in all groups, but the body temperature of the treatment group was higher than the body temperature of the other groups. Respiratory rates gradually increased in all groups, but the treatment group had an increase in the respiratory rates from 20 minutes to 30 minutes after administration of tiletamime-zolazepam compared to the other groups. In the three groups, there was no specific finding on the electrocardiogram. In the result, the electroacupuncture on Jen Chung in dogs was effective for the reversed effects the tiletamine-zolazepam anesthesia 20 minutes after induction.

Effect of Green Tea Extract on Healing of Contaminated Wound in Dogs (개의 피부 감염창 치유에 대한 녹차 추출물의 국소도포 효과)

  • The purpose of this study is to assess healing effect of the green tea on Staphylococcus contaminated wound. On the back of 7 dogs, 6 full-thickness skin wounds($2cm{\times}2cm$) were made and Staphylococcus intermedius was inoculated. Each wound was applied with the 2%(experimental group I), 1%(experimental group II) extract of green tea and normal saline(control group I) after inoculation. The wound contraction rate of the experimental group I and II was higher than that of the control group in entire period. And from the $6^{th}\;to\;22^{nd}$ day, the wound contraction rate of the experimental group I and II was significantly higher than that of the control group(p<0.05). The occupation rate of wound healing in the experimental group I was higher than that of the control group from $0\;to\;9^{th}$ day except from $1^{st}\;to\;2^{nd}$ day and also higher than that of the experimental group II from $0\;to\;3^{rd}$ day except from $1^{st}\;to\;2^{nd}$ day. And the occupation rate of wound healing in the experimental group II was higher than that of the control group from $1^{st}\;to\;8^{th}$ day except from $2^{nd}\;to\;3^{rd}$ day. According to concentration of green tea extract increased, the wound contracted more vigorously in earlier period. The tensile strength of the experimental group I and II was significantly higher than that of the control group(p<0.05). And that of the experimental group I was also higher than in the experimental group II(p<0.05). The number of S. intermedius in the experimental group I and II was decreased more rapidly than that of the control group. From the $4^{th}$ to the $11^{th}$ day, the number of S. intermedius in the experimental group I was significantly lower than that of the experimental group II and the control group. And from the $4^{th}$ to the $7^{th}$ day, the number of S. intermedius in the experimental group was significantly lower than that of the control group. The histopathological findings of the experimental group I and II were similar to those of the control group on the $1^{st}\;and\;4^{th}$ day. On $14^{th}$ day, reepithelialization was completed in the experimental group I and II except the control group. Higher wound contraction rate, better bacteriocidal action, higher tensile strength and better histopathological findings were observed in the experimental group I and II than in the control group. These results suggest that the topical application of green tea extract can promote contaminated wound healing in dogs.

  • Impedance tomography (EIT) is a relatively new imaging modality in which the internal impedivity distribution is reconstructed based on the known sets of injected currents through the electrodes and induced voltages on the surface of the object.   • Angiotensin I-converting enzyme (ACE) inhibitory activities of elk antler hydrolysates prepared with three kinds of proteases, pepsin, trypsin and $\alpha-chymotrypsin$, were investigated. The ACE inhibitory activity of the pepsinolytic hydrolysate was the highest with an $IC_{50}$ value of $9.3\mu g/mL.$ In addition, three kinds of pepsinolytic hydrolysates with relatively high molecular weights (over 10,000 Da), medium molecular weights (5,000 to 10,000 Da), and low molecular weights (below 5,000 Da) were fractionated using an ultrafiltration membrane system. The below 5,000 Da hydrolysate exhibited the highest ACE inhibitory activity. These results indicate that the pepsinolytic hydrolysates of elk velvet antler could be a good source of peptides with ACE inhibitory activity.

  • The present study was conducted to investigate the incidence of equine herpesvirus 1(EHV-1) infection in Korean horses by cell EUSA and serum neutralization test A significant level of antibodies to EHV-1 was detected from 539(82.8%) by cell EUSA and from 524 horses(80.5%) by SNT indicating that EHV-1 infection in Korean horses are widely spreaded throughout the country.
