Genetic Features of Cheju Horses based on Transferrin Gene Frequency (Transferrin 유전자빈도에 의한 제주마의 유전적 특성)

  • To investigate genetic features and the allele distribution of transferrin gene in three Cheju horse groups (group I, 137 horses of Jeju institute; group II, 107 horses of farms; group III, 89 racing horses) and three foreign breeds(l03 Thoroughbred, 10 Mongolian and 5 American Quarter horses), transferrin gene exons 13, 15, and 16 were analyzed by SSCP. The allele frequencies of transferrin gene of these groups and breeds were used to calculate genetic distances and to test population differentiations. The Fst values were 0.067 between Cheju horse groups I and II, 0.070 between Cheju horse groups I and group III, 0.091 between Cheju horse group I and Mongolian breed, and 0.189 between Cheju horse group I and Thoroughbred breed. Cheju horse group I showed significant population differentiation from other two Cheju horse groups and three foreign breeds while Cheju horse group III showed significant population differentiation only from Cheju horse group I and Thoroughbred breed(p <0.05). Results indicate that three Cheju horse groups showed population differentiation between each other, suggesting genetic heterogeneity of Cheju horses.

Ultrasonographic Evaluation of the Thyroid Glands, Jugular Vein and Carotid Artery in Horse (말에서 갑상선, 경정맥 및 경동맥의 초음파상)

  • Nowadays many kinds of foreign breed horse and Korean native Cheju horse are raised in Korea. Horse has complicated structure and function to maintain normal health, and to overcome severe condition during gallop at full speed. Sometimes the disease take a bad turn becuase of difficulty of correct diagnosis for horse disease. In recent times, horse farm for fattening and reproduction is gradually increasing. However, thoracic disease and abdominal disease are frequently occuring by defect in raising management. But the diagnosis of these kinds of disease is difficult by former diagnostic method. Ultrasonographic diagnostic method is a method to slove the problem. In abroad, nowadays ultrasonography has been reported for circulatory organs and abdomonal organs in horse, cattle, sheep, and dog, and the importance of ultrasonographic diagnosis is recognized in veterinary clinical medicine. In the country, ultrasonography of Korean native goat, cattle and dog was recently reported, but there was no report for the ultrasonography of horse. So, ultrasonogram for the thyroid glands, jugular vein and carotid artery was evaluated in Cheju horse and Thoroughbred horse. Thyroid gland, jugular vein and carotid artery were determined on both sides of neck. The length of right thyroid lobe at the Thoroughbred horse, Thoroughbred foul, Cheju horse and Cheju foul were 54.8, 45.1, 46.1 and 42.2 mm, respectively. The width of right thyroid lobe at the Thoroughbred horse, Thoroughbred foul, Cheju horse and Cheju foul were 18.1, 14.3, 14.6 and 14.2 mm, respectively. The height of right thyroid lobe at the Thoroughbred horse, Thoroughbred foul, Cheju horse and Cheju foul were 27.3, 25.6, 26.2 and 25.5 mm, respectively. Similar ultrasonographic measurements were obtained for the left thyroid gland. The internal diameter of left jugular vein at the Thoroughbred horse, Thoroughbred foul, Cheju horse and Cheju foul were 10.6, 9.7, 10.5 and 9.2 mm, respectively. Similar ultrasonographic measurements were obtained for the right jugular vein. The internal diameter of right carotid artery at the Thoroughbred horse, Thoroughbred foul, Cheju horse and Cheju foul were 9.5, 8.8, 9.1 and 8.5 mm, respectively. Similar ultrasonographic measurements were obtained for the left carotid artery. It is concluded that the ultrasonographic values of this study provides references for the diagnosis of morphologic changes in the thyroid gland, jugular vein and carotid artery in Korean native Cheju horse and Thoroughbred horse.

Sasa quelpaertensis Leaf Extracts Induce Apoptosis in Human Leukemia HL-60 Cells

  • In east Asia, the leaves of various Sasa species have been used in folk medicine for centuries. The effects of the methanolic extract and its subsequent fractions derived from the leaves of Sasa quelpaertensis Nakai on cell proliferation in human leukemia HL-60 cells were evaluated. The ethyl acetate fraction of this extract (ESQL) significantly reduced cell viability in a dose-dependent manner ($0-250\;{\mu}g/mL$). ESQL ($IC_{50}=24.8\;{\mu}g/mL$) exhibited growth inhibition comparable to the main constituent of green tea, epigallocatechin ($IC_{50}=26.2\;{\mu}g/mL$), which was used as a positive control. ESQL treatment induced apoptosis, which was confirmed by the presence of nuclear condensation and annexin V-staining. These results demonstrate that ESQL contains chemopreventive phytochemicals that may be useful in neutraceutical applications.

Esophageal Rupture Due to Diving in Shallow Waters

  • Delayed esophageal rupture due to blunt injury is not new. However, rupture due to suspected barotrauma is very rare. We describe a case of esophageal rupture in a male 24-year-old patient after diving in shallow waters. The patient was quadriplegic and could not experience the typical chest pain related to rupture and resulting mediastinitis. The rupture was discovered 4 days after emergency decompressive laminectomy and fusion for his cervical spine. The rupture was evidently caused by barotrauma and was discovered four days after admission. He underwent primary closure and pericardial flap as a life-saving procedure.

  • Enzyme-linked Immuno sorbent Assay (ELISA) for the serological diagnosis of Brucella abortus was developed and compared with plate agglutination test. Cell wall antigen was extracted from Brucella abortus 1119-3 by sonication and with a sodium deoxychlate solution. Optimum protein concentration of coating antigen were $0.4{\mu}g/100{\mu}{\ell}$ protein on each microtiter plate well. Horse radish peroxidase (HRP) labeled protein-G was used as a tracer of reacted antibodies. ELISA confirmed the agreeable results of 40 cases out of 43 cases by plate aggulutination test. ELISA diagnosed positive cases(10 out of 12) and negative cases (1 out of 12) with dubious sera by plate agglutination test. From this results ELISA could be used for the early diagnostic tools of Brucellosis in cattle.

  • Although hemothorax and pneumothorax are common complications seen in rib fractures, focal extrapleural hematoma is quite rare. We report a 63-year-old female patient that developed large focal extrapleural hematoma after falling off a second floor veranda. The patient had sustained 3, 4, 5th costal cartilage rib fractures and a sternum fracture. She had developed suspected empyema with loculations with small amount of hemothorax. She underwent a planned early decortication/adhesiolysis by video assisted thoracoscopic surgery at the 12th post-trauma day due to failed drainage. Unexpectedly, she had no adhesions or any significant retained hematoma mimicking a mass, but was found with the focal extrapleural chest wall hematoma. She was discharged on postoperative 46th day for other reasons and is doing fine today.

  • The conditions required for plant transformation through the electroporation system and for the electrofusion of the prtoplasts were investigated for geranium (Pelargonium zonale hybrids). The optimum condition for electroporation was 1.77 kV/cm for $40\;{\mu}sec$ under which 70% of the protoplasts were viable and 58% of the viable protoplasts were stained with methylene blue. The pBin19 DNA plasmid used as a carrier vector was isolated from E.coli $DH5{\alpha}$ strain, purified, identified by the electrophoresis on agarose gel and electroporated into the protoplasts. The KM8 liquid medium gave better cell division than any other media. One MHz of AC frequency with 40 V/cm of amplitude for 15 sec followed by 0.5 kV/cm of DC amplitude for $60\;{\mu}sec$ was most efficient for the electrofusion of protoplasts.

Prevalence and Related Factors of Dementia in Elderly People in Jeju Island (제주도 노인들의 치매유병률과 관련요인)

  • As the population ages, health problems of the elderly, including functional disorders and cognitive impairment, have become some of the most important social issues in Korea. The purpose of this study is to discover the rate of prevalence and factors related to dementia among the elderly in Jeju Province. 2405 elderly in Jeju Province were interviewed, using a questionnaire consisting of sociodemographic characteristics: health behavior, quality of life, and MMSE-K. Prevalence of dementia was 9.4%(11.7% of females, 4.1% of males). Factors related to dementia, analyzed by multiple logistic regression, were age, working status, subjective health status, social support, and subjective economic status. In community health care for the elderly, factors relating to dementia have to be considered. When planning community health care, priority should be given to the elderly who: need care but live alone; lack social support; have a low standard of living; believe they are not in a good state of health; or whose life satisfaction is low.

Studies on the Floristic Composition and Succession of the Shrub Communities at the Summit of Mt. Halla, Cheju Island, Korea

  • Empetrum nigrum dominant community or Rhododendron mucronulatum and R. yedoense var. poukhanense dominant shrub community are developing above l,500m on Mt. Halla in Cheju Island, Korea. These communities were formed as a result of the forest of Abies koreana regression by grazing or erosion over a long period of time. This study was conducted using the phytosociological method and it clarified the floristic composition of the community in Cheju Island by comparing with a similar community in Japan. Also this study interpreted the development data of these communities and considered community succession. As a result of our interpretation, shrub community of Cheju Island is recognized as two associations, Festuco obinae-Empetretum nigrum ass. nov and Rhododendretum mucronulatum ass. nov. Each association of Cheju Island is composed of endemic species of Cheju Island. related to the Korean peninsula and to Japan. The two associations are divided into 8 sub-units but each community development has been created with longtime grazing and prescribed fire. Maybe the location stability by stoppage of interference and grazing will process community transition Quickly. Comparing the shrub communities of Cheju Island with Maianthemo-Rhododendretum in Kyushu, Japan, which is close geographically, both areas have the species of Miscanthetea sinesis and Ericaceae in common. But Cheju Island has more abundant species and has higher ratio of appearance of alpine plants. The shrub communities of Cheju Island and Kyushu. Japan are identified as having a different rank over Alliance.
