Optimum Enzyme Mixture of Cellulase, Hemicellulase, and Xylanase for Production of Water-Soluble Carbohydrates from Rice Straw (볏짚 유래 수용성 탄수화물 생산에 있어 cellulase, hemicellulase 및 xylanase 최적혼합조건)

  • This study was conducted to investigate the production of water-soluble carbohydrates (WSCs) by treatment of different amounts of rice straw with cellulase, hemicellulase, and xylanase. Treatment of high amounts of rice straw (100 g/l) with cellulase and hemicellulase resulted in similar production of WSCs. Reducing the amount of rice straw to 50 g/l decreased the production of WSCs by hemicellulase but had no effect on WSC production by cellulase. The interaction among rice straw amounts, and hemicellulase and xylanase activities was investigated using a Box Behnken design and a response surface model. An interaction was found only between hemicellulase and xylanase. An enzyme mixture consisting of 0.55 mg/ml of hemicellulase and 0.65 mg/ml of xylanase generated the highest amounts of WSCs, regardless of the amount of rice straw provided. Therefore, the activity of cellulase was higher than that of either hemicellulose or xylanase for WSC production from rice straw. The interaction observed for hemicellulase and xylanase indicates that a combined enzyme treatment could improve the production of WSCs from rice straw.

Studies on the Enzyme Activities of Rhizopus distributed in South Korea(1) - On the amylase, protease and cellulase activities- (한국산 Rhizopus의 효소활성에 관한 연구 (第 1 報) - Amylase, protease 및 cellulase 활성에 관하여-)

  • Enzyme activities, such as glucoamylase dextrinogenic amylase, cellulase, acid protase and neutral protease, of Rhizopus isolated from various substrates collected throughout South Korea are measured, and their enzyme activities are surveyed from taxonomical, ecological and physiological viewpoint. Effect of carbon sources and phytohormones on the amylalse production of Rhizopus are also measured. Among the 735 strains of Phizopus isolated, strain number 587 exhibiting most prominent dextrinogenic amylase and netral protease activity is selected as the best strain, and the strain number 673, 108, 329, 165 and 728 are seleted for their predominant cellulase, acid protease, glucoamylase, dextrinogenic amylase and neutral protease activities, respectively. R.acidus and R.nigricans which exhibited relatively higher callulalse activity, showed lower activities for both amylase. R.tritici exhibited higher protease activity. The relations between activities and various substrates of wild strains are not outstnading difference, although the strains isolated from inland region exhibited more or less higher amylase and cellulase activities, than those of coast region, generally. Lactose and dextrin are most effective carbon sources for glucoamylase and dextrinogenic amylase production of the Rhizopus niveus, respectively. Although all phytohormones tested are effective for production of amylase by the Rhizopus strains, except nicotinamide for glucoamylase production, biotin and ascorbate are most effective for dextrinogenic amylase and glucoamylase production, respectively.

Acanthophysium sp. KMF001, a New Strain with High Cellulase Activity

  • Cellulase is an eco-friendly biocatalyst, and its demand is growing in many industrial applications such as food, textile, paper, and bioenergy. Strains with a high cellulase activities are the starting point for the economic production of cellulase. In a previous study, Acanthophysium sp. KMF001 with high cellulase production ability was selected among 54 wood-rotting fungi. In this study, we evaluated the cellulase productivity of Acanthophysium sp. KMF001 quantitatively and analyzed its taxonomic location using a genetic method. Acanthophysium sp. KMF001 showed high cellulase productivity similar to that of Acanthophysium bisporum and was much better than A. bisporum in specific enzyme activity. The 28S rRNA sequence of Acanthophysium sp. KMF001 was similar to that of Acanthophysium lividocaeruleum MB1825, with 98.40% homology. Phylogenetic analysis suggested that Acanthophysium sp. KMF001 is a new strain. In this study, we propose a new strain with high cellulase productivity.

  • A bacterium producing the extracellular cellulases was isolated from soil and has been identified as Bacillus sp. The isolate, named Bacillus sp. 79-23, was shown to be very similar to B. subtilis on the basis of its biochemical properties. The carboxymethyl cellulase (CMCase) of culture supernatant was most active at 60$\circ$C and pH 6.0, and retained 90% of its maximum activity at pH 7.0. The additional carbon sources affected the CMCase productivity than nitrogen sources in the culture medium. The carbon sources including wheat bran, rice straw, maltose and glucose increased the enzyme productivty. Especially, the maximum CMCase production was 5.2 units/ml in LB medium supplemented with 3% (w/v) wheat bran, which was 13-folds more than that in LB medium. It was found that the enzyme production was in association with the growth of Bacillius sp. 79-23. But, whean bran did not affect the growth of isolate, suggesting that increasement of CMCase production was owing to the induction of CMCase biosynthesis by wheat bran. In addition, both water-soluble and insoluble components of wheat bran was involved in induction of CMCase biosynthesis.

  • For the preparation of fermented feed with Lyophyllum decastes, optimum cultural conditions for the production of cellulase were $30^{\circ}C$, pH 6.0, 60-70% moisture content and the cultural of 15 days. Among the submaterial added, 30-40% of rice bran and 0.72% of $(NH_4)_2PHO_4$ were effective for the cellulase production and its production increased when rice straw treated with 4% alkaline peroxides. Solid state fermentation of rice straw with Lyophyllum decastes for 40 days removed 19.9% of lignin, and increased the total nitrogen content to 1.6% from 1.1%. As the fermentation proceeded, the in vitro dry matter digestibility of fermented feed was increased and it increased 21.1% after 35 days.

  • The feasibility of enzymatic hydrolysis of cellulosic materials is dependent on the cost of cellulase, which is strongly influenced by the selection of proper carbon source in the cellulase production medium. When solka floc was used as a carbon source for the production of cellulase by Trichoderma reesei Rut C-30, a maximum of 53.2 U/ml of CMCase activity (4.8 U/ml of FPase activity) was obtained with a concentration of 1 % of solka floc. The cellulase activity decreased to 50% in the presence of 0.5% of glucose in the medium. The production of cellulase was considerably enhanced when solka floc and wheat bran were used together as a carbon source. A medium which contained 1 % of solka floc and 3 % of wheat bran yielded highest cellulase activity: CMCase activity of 76 U/ml and FPase activity of 12.5 U/ml.

Production of Thermostable $\alpha$-Amylase and Cellulase from Cellulomonas sp.

  • A bacterium, isolated from rabbit's waste and identified as Cellulomonas sp., had cellulase and thermostable $\alpha$-amylase activity when grown on wheat bran. Maximum activity of thermostable $\alpha$-amylase was obtained by adding $3\%$ soluble starch. However, soybean oil (1 ml $1^{-1}$) could increase the production of $\alpha$-amylase and cellulase in 'wheat bran. The $\alpha$-amylase was characterized by making a . demonstration of optimum activity at $90^{\circ}C$ and pH 6- 9, with soluble starch as a substrate. The effect of ions on the activity and the stability of this enzyme were investigated. This strain secreted carboxymethyl cellulase (CMCase), cellobiase ($\beta$­glucosidase), and filter paperase (Fpase) during growth on wheat bran. Carboxymethy1cellulase, cellobiase, and Fpase activities had pH optima of 6, 5.5, and 6, respectively. CMCase and cellobiase activities both had an optimum temperature of $50^{\circ}C$, whereas Fpase had an optimum temperature of $45^{\circ}C$.

  • In order to utilize the agricultural waste products, two strains of mold producing powerful cellulolytic enzyme were sereened from various soils, composts, rotten wood and others. The optimum condition of cellulase production was studied. The results obtained were summarized as follows. 1. Two strains of mold which showed remarkably high cellulolytic activity were identified as Aspergillus niger-SM 6 and Trichoderma viride-SM 10. 2. The highest cellulase production was obtained at pH 5.0-6.0 in 5 days. 3. Cellulase production in strain Aspergillus niger-SM 6 increased with the addition of C.M.C., $(NH_4)_2SO_4$, C.S.L., orange peel powder and rice hull. The rice hull, treated with 3N NaOH at $120^{\circ}C$ for 15 min. and neutralized with various acids, was used. Up to 50% of wheat bran could be substituted by the treated rice hull without any decrease of cellulase activity. 4. In the strain of Trichoderma viride-SM 10, cellulase production increased with the addition of C.M.C., $NH_4NO_3$, Vitamin-free casamino acid and orange peel powder, while the other carbon, nitrogen, phosphate sources, natural nutrients and organic substances gave no remarkable effect.

Optimization of Cellulase Production in Batch Fermentation by Trichoderma reesei

  • Maximum cellulase production was sought by comparing the activities of the cellulases produced by different Trichoderma reesei strains and Aspergillus niger. Trichoderma reesei Rut-C30 showed higher cellulase activity than other Trichoderma reesei stains and Aspergillus niger that was isolated from soil. By optimizing the cultivation conditions during shake flask culture, higher cellulase production could be achieved. The FP(filter paper) activity of 3.7U/ml and CMCase (Carboxymethylcellulase) activity of 60U/ml were obtained from shake flask culture. When it was grown in 2.5L fermentor, where pH and DO levels are controlled, the enzyme activities were 133.35U/ml (CMCase) and 11.67U/ml(FP), respectively. Ammonium sulfate precipitation method was used to recover enzymes from fermentation broth. The dried cellulase powder showed 3074.9U/g of CMCase activity and 166.7U/g of FP activity with 83.5% CMCase recovery.
