Polyamines and Their Metabolites as Diagnostic Markers of Human Diseases

  • Polyamines, putrescine, spermidine and spermine, are ubiquitous in living cells and are essential for eukaryotic cell growth. These polycations interact with negatively charged molecules such as DNA, RNA, acidic proteins and phospholipids and modulate various cellular functions including macromolecular synthesis. Dysregulation of the polyamine pathway leads to pathological conditions including cancer, inflammation, stroke, renal failure and diabetes. Increase in polyamines and polyamine synthesis enzymes is often associated with tumor growth, and urinary and plasma contents of polyamines and their metabolites have been investigated as diagnostic markers for cancers. Of these, diacetylated derivatives of spermidine and spermine are elevated in the urine of cancer patients and present potential markers for early detection. Enhanced catabolism of cellular polyamines by polyamine oxidases (PAO), spermine oxidase (SMO) or acetylpolyamine oxidase (AcPAO), increases cellular oxidative stress and generates hydrogen peroxide and a reactive toxic metabolite, acrolein, which covalently incorporates into lysine residues of cellular proteins. Levels of protein-conjuagated acrolein (PC-Acro) and polyamine oxidizing enzymes were increased in the locus of brain infarction and in plasma in a mouse model of stroke and also in the plasma of stroke patients. When the combined measurements of PC-Acro, interleukin 6 (IL-6), and C-reactive protein (CRP) were evaluated, even silent brain infarction (SBI) was detected with high sensitivity and specificity. Considering that there are no reliable biochemical markers for early stage of stroke, PC-Acro and PAOs present promising markers. Thus the polyamine metabolites in plasma or urine provide useful tools in early diagnosis of cancer and stroke.

Physiological Responses of Oxygen-Tolerant Anaerobic Bifidobacterium longum under Oxygen

  • In order to investigate what kind of response anaerobic bifidobacteria has on oxygen stress, five oxygen-tolerant bifidobacteria were isolated from human fecal samples. All were temporarily identified as Bifidobacterium longum through an analysis of carbohydrate utilization patterns and cellular fatty acid profiles. In the presence of oxygen, the lag phase became extended and the cell growth was suppressed. Bifidobacterial cell was able to remove dissolved oxygen in an early stage of growth and to overcome oxygen stress to a certain extent. The cell became long n size and showed a rough surface containing many nodes which were derived from abnormal or incomplete cell division. Cellular fatty acid profiled changed remarkably under a partially aerobic condition, so that the carbon chain of cellular fatty acid became short. All the dimethyl acetals originated from plasmalogen were reduced, any cyclopropane fatty acid, 9, 10-methyleneoctadecanoic acid ($C_{19:0}cyc9,10$), was increased remarkably. Oxygen stress induced a 5.5 kD protein in B. longum JI 1 of the oxygen-teolerant bifidobacteria, that was named Osp protein, and its N-terminal amino acid sequence was as follows: unknown amino acid-Thr-Gly-Val-Arg-Phe-Ser-Asp-Asp-Glu. Therefore, the oxygen-tolerant bifidobacteria seemed to defend against oxygen stress byincreasing the content of short fatty acid and cyclopropane fatty acid, and induction of an oxygen stress protein, but not the plasmalogen.

Stimulatory Effect of Korean Red-Ginseng Extract on the Proliferation and . Cellular Activity of Lymphocytes (홍삼 추출물의 임파구 증식 및 활성 촉진효과)

  • The effect of Korean red-ginseng extract on the proliferation and cellular activity of mouse B and T lymphocytes was examined in vitro. Both water and ethanol extract from red-ginseng increased the growth of normal B and T lymphocytes 1.5∼2.5-folds. Saponin and polysaccharide fractions from ginseng extract also stimulated the proliferation of normal lymphocytes much higher than several well-known immunostimulators. B and T lymphoma cell lines responded to the ginseng extract and fractions by growth, too, while non-lymphoid cell lines did not. Immunoglobulin production of unprimed B-lymphocytes was little affected by the ginseng extract and fractions, though the ethanol extract slightly enhanced Ini, production of B-lymphocytes. When cytolytic activity of T lymphocytes against tumor tells was induced in vitro, both of the saponin and polysaccharide fractions and the ginseng ethanol extract increased the cellular activity of cytotoxic T lymphocytes 4-5-folds, while the ginseng water extract did not. Especially, the saponin fraction exhibited 10-times higher stimulatory effect on the cytolytlc activity of cytotoxic T cells than the ethanol extract and the pclysaccharide fraction did. These results suggest that Korean red-ginseng contain potent immunomodulating components to stimulate the proliferation of B and T lymphocytes and the cellular activity of cytotoxic T lymphocytes.

Growth Damage and Alteration of Cellular Tissue of Barley Infected by Barley yellow mosaic virus (보리호위축병 (Barley yellow mosaic virus)에 의한 보리의 생육 피해 및 세포학적 변화)

  • The damage of plant growth and alteration of cellular tissues of barley infected by Barley yellow mosaic virus (BaYMV) was explored. The infected plots significantly damaged in all of measured factors by the disease. In severely diseased plant, the viral infection affected on plant growth like as shorten culm length about 25cm, 36% constrained ratio, comparing to healthy. The yield decreased over 70% in diseased plots by fewer numbers of spike and kernel per square meter and spike, respectively. BaYMV constructed typical inclusion body like a pinwheel type inside barley leaves, and the infection affected on cellular elongation or growth not cell division in examined three parts as stem, neck of panicle and node, related to dwarfness of infected barley. The stem tissues were most severely affected on cell growth as restrained epidermis cell length in diameter and vascular bundle size. In neck of panicle tissues, distribution and size of tissues of fiber and cortex parts, respectively, showed differences between healthy and infected plants. In node part, healthy plant showed bigger tissue size as 1.5 times than infected plant. Theses results suggest that BaYMV infection could affect on the cell growth not cell division, and which resulted shorten culm length in plant growth and decreased yield, finally.

Surface Morphologies and Internal Fine Structures of Bast Fibers

  • Fiber surface morphologies and associated internal structures are closely related to its properties. Unlike other fibers including cotton, bast fibers possess transverse nodes and fissures in cross-sectional and longitudinal directions. Their morphologies and associated internal structures were anatomically examined under the scanning electron microscope. The results showed that the morphologies of the nodes and the fissures of bast fibers varied depending on the construction of the inner fibril cellular layers. The transverse nodes and fissures were formed by the folding and spiralling of the cellular layers during plant growth. The dimensions of nodes and fissures were determined by the dislocations of the cellular layers. There were also many longitudinal fissures in bast fibers. Some deep longitudinal fissures even opened the fiber lumen for a short way along the fiber. In addition, the lumen channel of the bast fibers could be disturbed or disrupted by the nodes and the spi­rals of the internal cellular layers. The existence of the transverse nodes and fissures in the bast fibers could degrade the fiber mechanical properties, whereas the longitudinal fissures may contribute to the very rapid moisture absorption and desorption.

A Study on the Distribution and the Effect of Torreya Fruit Extract on Cellular Slime Molds in Torreya Forest of Cheju Island (제주도 비자림에서의 세포성 점균의 분포 및 비자열매 추출액의 성장 효과에 관한 연구)

  • Dictyostelid cellular slime molds were isolated from soils and harks of the subtropical forest of Torreya nucifera in Cheju island. The results were as follows: Polysphondylium pallidum,Dictyostelium purpureum, D. mucoroides, D. aureo-stipes var. aureo-stipes, D. flavidum, D. miniutum, P.violaceu m, D. monocrhasioides. D. brefeldianum, D. polvcephalum. D. areum var. areum, P. tenuissimum, D. fasciculatum. In this forest, P. pallidum and D. purpureum were occurred dominantly, and D. mucoroides and D. aureo-stipes var, aureo-stipes were the second dominant. It was distinguished that D. purpureum was much more cornmonly found than the other forests. Cellular slime molds from the barks of the tree heights of 1,3 and 5m were occured 7, 3 and 2 species respectively. Torreya fruit extract affected on the growth of D. aureo-stipes var. aureo-stipes and D. flavidum but not on D. purpureum and D. mucoroides.   • Linear stability analysis of radiating counterflow diffusion flames is numerically conducted to examine the instability characteristics of cellular patterns. Lewis number is assumed to be 0.5 to consider diffusional-thermal instability. Near kinetic limit extinction regime, growth rates of disturbances always have real eigen-values and neutral stability condition of planar disturbances perfectly falls into quasi-steady extinction. Cellular instability of disturbance with transverse direction occurs just before steady extinction. However, near radiative limit extinction regime, the eigenvalues are complex and pulsating instability of planar disturbances appears prior to steady extinction. Cellular instability occurs before the onset of planar pulsating instability, which means the extension of flammability.

  • Cellular glass foam (CGF), the reprocessed glass, has a possibility to be used as a medium component in plug culture of horticultural crops due to the its excellent air and water permeability as comparable to perlite. An experiment was conducted to investigate growth of plug seedlings of Lycopersicum esculentum 'Segye' as influenced by irrigation frequency in various medium combinations of CGF (2.0-4.0 mm particle size) and peatmoss. Seeds were sown in 200-cell plug trays, filled with mixtures of CGF and peatmoss either at 33:67 or 25:75 (%. v/v) and were germinated on a fogged propagation bed. The irrigation frequencies used were one, two or three times per every two days. A commercial plug medium (Tosilee medium) was used as the control, and the irrigation frequency in the control was one time per day. Growth of seedlings, and medium pH and EC were measured at 33 days after sowing. The medium composition had little influence on overall growth of seedlings. Irrigation frequency very significant affected number of leaves, leaf area, chlorophyll concentration, fresh and dry weights of shoots and roots, and dry matter. Growth of seedlings was the greatest with the highest irrigation frequency in the 25% CGF+75% peatmoss mixture.

  • Ceramide has become a widely used ingredient in cosmetic and pharmaceutical industries, however, only a few yeast strains were investigated for the synthesis of ceramide and the concentration was very low. Ceramide is not only a core intermediate of sphingolipids but also an important modulator of many cellular events including apoptosis, cell cycle arrest, senescence, differentiation, and stress responses. In this study S.cerevisiae was grown in a batch culture and the cellular content of ceramide was measured at different growth phases. The ceramide content was highest at stationary phase and 2.01 mg ceramide/g cell was obtained.

A Study on the Urban Growth Forecasting for the Seoul Metropolitan Area (서울대도시지역 도시성장 예측에 관한 연구)

  • 도시는 산업혁명 이후 급속히 발달하기 시작한 인간의 대표적인 정주공간으로, 산업활동의 집중, 도시로의 인구유입으로 공간적인 범위가 확대되어 가고 있다. 급속한 도시성장은 환경을 악화시키고 있으며, 이에 따른 온실효과, 대기오염의 증가 등은 심각한 도시문제로 떠오르고 있다. 지속 가능한 도시성장을 위해 도시가 어떻게 성장해 왔는가를 분석하고, 향후의 도시성장을 예측하며, 이에 대한 적절한 대안을 모색하는 것은 도시관리를 위해 매우 중요한 문제이다. 본 연구에서는 서울대도시지역을 대상으로 1957년부터 1997년까지의 도시성장에 관한 자료를 구축하고, 셀룰라 오토마타(Cellular Automata) 개념에 기초하고 있는 도시성장모델(UGM)을 이용하여 1998년부터 2050년까지의 서울대도시지역의 도시성장을 예측하였다.
