Possible Roles of LAMMER Kinase Lkh1 in Fission Yeast by Comparative Proteome Analysis

  • To investigate the possible roles of LAMMER kinase homologue, Lkh1, in Schizosaccharomyces pombe, whole proteins were extracted from wild type and lkh1-deletion mutant cells and subjected to polyacrylamide gel electrophoresis. Differentially expressed proteins were identified by tandem mass spectrometry (MS/MS) and were compared with a protein database. In whole-cell extracts, 10 proteins were up-regulated and 9 proteins were down-regulated in the mutant. In extracellular preparations, 6 proteins were up-regulated in the lkh1+ null mutant and 4 proteins successfully identified: glycolipid anchored surface precursor, $\beta$-glucosidase (Psu1), cell surface protein, glucan 1,3-$\beta$-glucosidase (Bgl2), and exo-1,3 $\beta$-glucanase (Exg1). These results suggest that Lkh1 is involved in regulating cell wall assembly.

Corneal Squamous Cell Carcinoma Presumed to Arise from Corneal Fibrosis in a Shih-Tzu Dog

  • Squamous cell carcinoma (SCC) is a form of neoplasm that origins from the epithelial surface of many organs. Ocular occurrence in small animals is rare, especially in the central cornea without the involvement of limbus or conjunctiva. In the current case, a 10-year-old, spayed female Shih-Tzu was presented with a central corneal mass. Through an ophthalmic examination the sign of corneal scarring around the mass was found. Pink fleshy lesion that protruded outward was removed through superficial keratectomy. The mass with the size of $8.5{\times}6.5$ millimeter was histopathologically diagnosed as corneal SCC infiltrating not only the epithelium, but also the superficial corneal stroma with surrounding superficial lymphoplasmacytic and neutrophilic chronic keratitis. Also, the adjacent, non-affected corneal epithelium was markedly hyperplastic and keratinized; the adjacent stroma was moderately vascularized and fibrotic. The pulse-dose therapy using 1% topical 5-flourouracil were applied for five cycles. The tumor has not recurred through nine months of follow-up time.

  • Primary lymphoma originating from the lacrimal drainage system is a rare disease. Such lymphomas are mostly B-cell in origin and present nonspecific symptoms. The treatment of malignant lymphoma of the lacrimal drainage system is slightly different. We present the case of a 71-year-old woman with a painless mass below the medial canthus. Computed tomography (CT) scan of the orbit revealed a mass invading the right lacrimal sac. An incision biopsy was obtained, and the pathologic findings suggested a diagnosis of primary diffuse large B-cell lymphoma of the lacrimal sac. The patient was treated with chemotherapy and intrathecal methotrexate. After completing eight cycles of chemotherapy, the patient was followed up by a CT scan, which revealed nearly total resolution of an ill-defined enhancing mass. At the time of this case report writing, the patient is in complete remission at six months with no other complications.

  • A 5-year-old Labrador Retriever presented with a subcutaneous mass on the right scapular region with localized calor. On magnetic resonance imaging (MRI), a heterogeneously hypointense mass was seen on T1-weighted images (WI) and hyperintense on T2WI and fat-suppressed T2WI with marginal contrast enhancement on postcontrast T1WI and invasion into the surrounding tissue. Histopathological examination of the mass confirmed subcutaneous mast cell tumor (MCT) infiltrating to the adjacent subcutaneous fat. This is the first report to describe the MRI findings of subcutaneous MCT in veterinary medicine. This study suggests that MRI may help evaluate the extent of invasion of subcutaneous MCT.

  • A model for predicting potentials of land erosion and deposition over a natural basin was developed based on the mass balance principle. The program was developed based on sediment mass balance principle for each cell in a GIS. Sediment yield from a cell was estimated with RUSLE. The outflow sediment from a cell was calculated by multiplying the sediment yield of the cell by the sediment delivery ratio (SDR) of the cell. The outflow sediment from the upstream cell becomes the incoming sediment of the downstream cell. Therefore the erosion and deposition potential of each cell could be determined from the sediment mass balance i.e., the difference between the incoming and outflow of sediments of each cell. The developed model was validated by comparing the predicted sediment yields for three basins with measured data.

  • A simplified resistor network model for electrical and mass transport in anode-supported planar solid oxide fuel cell (SOFC) was constructed in order to investigate the effect of interconnect rib geometry on the cell performance. For accurate potential calculation, activation and concentration over-potentials at the electrode/electrolyte interfaces were fully considered in this calculation. When contact resistance was not considered, the optimum interconnect rib length were calculated to be $0.1{\sim}0.2$ mm for 2 mm half unit cell for given operation conditions and properties. However, with realistic contact resistance, the interconnect rib length should be increased to provide larger contact area and thus to obtain better performance.

Spindle Cell Tumor in a Syrian Hamster (햄스터 spindle cell tumor 증례보고)

  • A case of spindle cell tumor was presented in a 16-month-old, female syrian hamster. In the left chest area, a 3cm firm elevated recurrent mass was found, surgically removed, and submitted to the Department of Veterinary Pathology, Seoul National University for diagnosis. The mass was soft to firm and tan on sectioning, and contained hemorrhagic area. Histologically, the tumor was composed of sheets of interlacing bundles of spindle-shaped cells with moderate amount of cytoplasm and oval to fusiform nuclei. They were plemorphic and contained 1 to 3 prominent nucleoli. Based on the gross and histological findings, the tumor was diagnosed as a subcutaneous spindle cell tumor. However, the exact origin of neoplastic cells remained undetermined.

Intradural Extramedullary and Subcutaneous Tumors in Neonate : Atypical Myxoid Spindle Cell Neoplasm

  • Tumors of the central nervous system are common in the pediatric population and constitute the second most prevalent tumor type in children. Within this group, spinal cord tumors are relatively rare and account for 1 to 10% of all pediatric central nervous system tumors. We describe a very rare case of an intradural extramedullary spinal cord tumor with a subcutaneous mass and discuss its clinical presentation, pathogenesis, and treatment. A male infant was delivered normally, with uneventful development. At 16 days post-delivery, his family took him to a pediatrician because of a mass on his upper back. Magnetic resonance imaging of the thoracic spine revealed a well-demarcated soft-tissue mass with central cystic change or necrosis at the subcutaneous layer of the posterior back (T2-7 level). Another mass was found with a fat component at the spinal canal of the T1-3 level, which was intradural extramedullary space. After six weeks, the spinal cord tumor and subcutaneous mass were grossly total resected; pathologic findings indicated an atypical myxoid spindle cell neoplasm, possibly nerve sheath in type. The final diagnosis of the mass was an atypical myxoid spindle cell neoplasm. The postoperative course was uneventful, and the patient was discharged after nine days without any neurological deficit. We report a rare case of an intradural extramedullary spinal tumor with subcutaneous mass in a neonate. It is necessary to monitor the patient's status by examining consecutive radiologic images, and the symptoms and neurological changes should be observed strictly during long-term follow-up.

  • Acinic cell carcinoma is a slow-growing solid neoplasm of salivary gland. Although their cytological and histological finding is bland-looking, their biological behavior is unpredictable. We experienced two cases of acinic cell carcinoma of the salivary gland diagnosed by fine needle aspiration biopsy and confirmed by tissue examination. They showed different clinical courses. We compared their cytologic and histologic findings. The first case was a right preauricular mass in a 58 year-old female of 3 years duration. The cytologic smear revealed sheets or small clusters of monotonous cells mimicking normal serous acinar cells with little cellular pleomorphism. She underwent superficial parotid lobectomy. The tumor was a well demarcated 1.5cm sized nodular mass without infiltration into surrounding parenchyme. The second case was a left submandibular mass in a 23 year-old male of 4 years duration. The smear showed more severe pleomorphism of the tumor cells than those of previous case. Excisional biopsy was done. The excised tumor was $5.5{\times}3.5{\times}3cm$ sized multilobulated solid mass with invasion into surrounding parenchyme. The tumor recurred after 20months, thus total excision of the mass and modified radical neck dissection was carried out. From the above findings, cytologic atypism, infiltrative growth pattern and type of initial therapy may be correlated with biologic behavior.

Effects of Ovariectomy on Insulin Resistance and β-Cell Function and Mass

  • The prevalence of type-2 diabetes increases remarkably in post-menopausal women, possibly because insulin secretion fails to compensate for the insulin resistance induced in various tissues by estrogen insufficiency. However, this has not been fully defined. Therefore, the present study investigated whether an ovariectomy (OVX) would increase insulin resistance and decrease the $\beta$-cell function and mass in female rats with and without a $90\%$ pancreatectomy (Px). Female rats aged 15 weeks were divided into four groups: 1) OVX + Px, 2) SOVX (sham operation of OVX) + Px, 3) OVX + SPx (sham operation of Px), and 4) SOVX + SPx, and given a $30\%$ fat diet for 8 weeks. At the end of the experimental period, the islet function and insulin resistance were determined using a hyperglycemic clamp and a euglycemic hyperinsulinemic clamp, respectively. The OVX only increased the body weight in the SPx rats, which was partially related to the food intake. Yet, the OVX did increase the peripheral insulin resistance, while the Px increased this resistance further. The OVX and Px both exacerbated the islet function, as measured by the insulin secretion pattern, while delaying and decreasing the first-phase insulin secretion. The OVX only decreased the proliferation of $\beta$-cells in the Px rats, while increasing apoptosis in both the Px and SPx rats. As a result, the OVX decreased the $\beta$-cell mass in the Px rats, but increased the mass in the SPx rats. In conclusion, an OVX was found to accelerate the development and progression of diabetes by increasing the insulin resistance and decreasing the $\beta$-cell mass. Therefore, menopause can be a risk factor for type-2 diabetes, mainly due to a deceased proliferation of $\beta$-cells.
