목적: 종양 내에서 산소공급 부족현상으로 발생하는 저산소증 조직에서 저온온열치료($42^{circ}C$)와 nicotinamide에 의한 perfusion limited 저산소증의 개선 효과를 마우스 종양 모델을 이용하여 종양 내 $[^18F]FMISO$ 섭취변화를 이용하여 증명할 수 있는지 알아보고자 하였다. 대상 및 방법: C3H 마우스에 $[^18F]FMISO$를 정주하고 11개 장기에서 $\%ID/g$을 구하여 biodistribution을 관찰하였다. 또한 같은 마우스에 동종 종양세포인 SCC7을 이식하여 종양모델을 만들고 저온온열치료($42^{circ}C$)와 nico-tinamide를 투여한 마우스와 대조군 마우스에서 $[^18F]FMISO$의 섭취정도 차이를 $\%ID/g$, autoradiography, PET scan을 시행하여 비교하고자 하였다. 결과: 대조군에서 종양의 FMISO의 섭취는 5.1+/-2.28 $\%ID/g$였고, 종양/근육, 종양/혈액의 섭취비는 2.2와 1.8이었다. 실험군에서는 각각 2.4+/-0.64 $\%ID/g$, 1.4와 1.2를 나타내어 대조군보다 유의하게 낮았다(p<0.021). Autoradiography에서 대조군의 종양 내부에 FMISO가 섭취됨을 확인하였고, 저온온열치료와 nico-tinamide를 투여한 실험군에서는 섭취가 감소된 것을 관찰하였다. 결론: C3H 마우스와 동종 종양세포인 SCC-VII을 이용한 종양모델에서 $[^18F]FMISO$가 종양내에 섭취가 되어 자산소증 종양모델로 적절함을 확인하였고, 저온온열치료($42^{circ}C$)와 nicotinamide에 의한 perfusion limited 저산소증 개선효과를 $[^18F]FMISO$의 종양 내 섭취가 감소하는 것을 통하여 확인할 수 있었다.
Fructose-1, 6-diphosphate (FOP), a glycolytic metabolite is reported to ameliorate inflammation and inhibit the nitric oxide production in murine macrophages stimulated with endotoxin. It is also reported that FOP has cytoprotective effects against hypoxia or ischemia/reperfusion injury in brain and heart. In this study, we examined whether FDP has protective effects on UV-induced oxidative damage in skin cell culture system and human skin in vivo. FDP had a protective role in UVB-induced LDH release and ROS accumulation in HaCaT although it did not show direct radical scavenging effect in the experiment using 1, 1-diphenyl-2-picrylhydrazyl (DPPH). FDP also preserved cellular GSH content after UV irradiation in HaCaT and normal human fibroblast culture system. Cellular oxidative stress induces multiple downstream signaling pathways that regulate expression of multiple gene including MMP-1 and collagen, we examined the effects of FDP on UV-induced alteration of these protein expression in fibroblast culture and human skin in vivo. The increased MMP-1 expression in fibroblast and human skin by UV irradiation was significantly decreased by FDP. FDP also prevented the UV-induced decrease of collagen expression in fibroblast and human skin. Moreover, the decreasing the intracellular levels of reducing equivalents in human fibroblast by glutathione (GSH) depletion lowered the UVA dose threshold for reduction of procollagen expression, indicating that the differences of glutathione contents define the susceptibility of fibroblasts towards UV-induced reduction of procollagen expression. FDP also preserved cellular GSH content after UV irradiation, indicating that FDP has protective effects on UV-induced reduction of procollagen expression, which are possibly through maintaining intracellular reducing equivalent. Based on these premises, we examined the effect of daily use of a moisturizer containing FDP on facial wrinkle in comparison with vehicle moisturizer lacking FDP. In the clinical study, FDP significantly decreased facial wrinkle compared with vehicle alone after 6 months of use. Our results suggest that FDP has anti-aging effects in skin by increasing cellular antioxidant system and preventing oxidative signal and inflammatory reaction. Therefore FDP may be useful anti-aging agent for cosmetic purpose.
Background: The discovery that microRNAs (miRNAs) regulate proliferation, invasion and metastasis provides a principal molecular basis of tumor heterogeneity. Microvessel distribution is an important characteristic of solid tumors, with significant hypoxia occurring in the center of tumors with low blood flow. The distribution of miR-374a in breast tumors was examined as a factor likely to be important in breast cancer progression. Methods: Breast tissue samples from 40 patients with breast cancer were classified into two groups: a highly invasive and metastatic group (HIMG) and a low-invasive and metastatic Group (LIMG). Samples were collected from the center and edge of each tumor. In each group, six specimens were examined by microRNA array, and the remaining 14 specimens were used for real-time RT-qPCR, Western blot and immunohistochemical analyses. Correlation analysis was performed for the miRNAs and target proteins. Follow-up was carried out during 28 months to 68 months after surgery, and survival data were analyzed. Results: In the LIMG, the relative content of miR-374a was lower in the center of the tumor than at its edge; in the HIMG, it was lower at the edge of the tumor, and miR-374a levels were lower in breast cancer tissues than in normal tissues. There was no difference between VEGF-A and VCAM-1 mRNA levels at the edge and center of the tumor; however, we observed a significant difference between VEGF-A and VCAM-1 protein expression levels in these two regions. There was a negative correlation between miR-374a and target protein levels. The microvessel density (MVD) was lower in the center of the tumor than at its edge in HIMG, but the LIMG vessels were uniformly distributed. There was a significant positive correlation between MVD and the number of lymph node metastases (Pearson correlation, r=0.912, P<0.01). The median follow-up time was 48.5 months. LIMG had higher rate of disease-free survival (100%, P=0.013) and longer median survival time (66 months) than HIMG, which had a lower rate of 75% and shorter median survival time (54 months). Conclusions: Our data demonstrated miR-374a to be differentially distributed in breast cancer; VEGF-A and VCAM-1 mRNA had coincident distribution, and the distribution of teh respective proteins was uneven and opposite to that for the miR-374a. These data might explain the differences in the distribution of MVD in breast cancer and variation in breast cancer prognosis.
Our previous proteomic study demonstrated that oxidative stress and antioxidant delphinidin regulated the cellular level of $p27^{kip1}$ (referred to as p27) as well as some heat shock proteins in human colon cancer HT 29 cells. Current study was conducted to validate and confirm the regulation of these proteins using both in vitro and in vivo systems. The level of p27 was decreased by hydrogen peroxide in a dose-dependent manner in human colon carcinoma HCT 116 (p53-positive) cells while it was increased upon exposure to hydrogen peroxide in HT 29 (p53-negative) cells. However, high concentration of hydrogen peroxide (100 ${\mu}M)$ downregulated p27 in both cell lines, but delphindin, one of antioxidative anthocyanins, enhanced the level of p27 suppressed by 100 ${\mu}M$ hydrogen peroxide. ICR mice were injected with varying concentrations of hydrogen peroxide, delphinidin and both. Western blot analysis for the mouse large intestinal tissue showed that the expression of p27 was upregulated by 25 mg/kg BW hydrogen peroxide. To investigate the association of p27 regulation with hypoxia-inducible factor 1-beta (HIF-$1{\beta}$), the level of p27 was analyzed in wild-type mouse hepatoma hepa1c1c7 and Aryl Hydrocarbon Nuclear Translocator (arnt, HIF-$1{\beta}$)-defective mutant BPRc1 cells in the absence and presence of hydrogen peroxide and delphinidin. While the level of p27 was responsive to hydrogen peroxide and delphinidin, it remained unchanged in BPRc1, suggesting that the regulation of p27 requires functional HIF-$1{\beta}$. We also found that hydrogen peroxide and delphinidin affected PI3K/Akt/mTOR signaling pathway which is one of upstream regulators of HIFs. In conclusion, hydrogen peroxide and antioxidant delphinidin seem to regulate intracellular level of p27 through regulating HIF-1 level which is, in turn, governed by its upstream regulators comprising of PI3K/Akt/mTOR signaling pathway. The results should also encourage further study for the potential of p27 as a biomarker for intracellular oxidative or antioxidant status.
본 연구는 연안 환경오염 및 양식용수의 이용과정에서 파생될 수 있는 철 오염이 넙치의 생리적 변화에 미치는 영향을 파악하기 위하여 12주 동안의 만성적 조건하에서 그들의 조직, 대사 및 생존의 변화를 검토하였다. 철에 노출시킨 넙치 아가미의 뚜렷한 조직학적 변화양상은 0.93mg/$\ell$ 이상의 농도에서 2주 후부터 조사개체 중에 $50\%$이상이 나타났다. 이들의 변화양상은 새판의 만곡, 새판 말단부의 곤봉-화, 상피세포의 파괴, 상피층의 박리현상, 괴사 및 새판의 융합 등으로 나타났으며, 이 같은 손상정도는 철 농도가 증가함에 따라 심하게 나타났다. 산소소비는 철 농도 0.42mg/$\ell$ 이하에서는 유의한 감소가 인정되지 않았으나, 0.93 mg/$\ell$ 농도에서는 실험 2주 후부터 유의한 감소가 관찰되었고, 12주 후의 0.93, 4.89 및 9.78mg/$\ell$농도에서 각각 37.1, 43.5 및 $53.2\%$가 감소하였다. 생존율은 0.42mg/$\ell$이하의 철 농도에서 $100\%$가 관찰되었고, 0.93mg/$\ell$농도에서는 $92.5\%$의 생존율을 보였으나, 유의한 감소는 철 농도 4.89mg/$\ell$에서 실험 5주 후, 9.78mg/$\ell$농도에서는 4주 후부터 관찰되었다. 이상의 결과는 연안해역 및 양식장 등에 0.93 mg/$\ell$ 이상의 철 농도가 존재할 경우, 넙치는 아가의 손상 및 대사활동의 저하 등으로 인해 성장에도 악영향을 받을 수 있을 것으로 사료된다.
마우스 종양인 SCCVll과 RIF-1에서 선택적 저산소세포 치사제인 SR 4233 (tirapazamine)과 분할 방사선조사를 병용하면 저산소분획이 큰 종양에서 방사선 감수성이 더욱 증강될 것인가를 확인하기 위하여 본 연구를 실시하였다. 종양세포 이식이전에 방사선조사를 받은 정상조직에서 성장한 종양의 저산소세포 분획이 크다는 사실을 이용하여 저산소세포분획이 큰 종양 모델을 수립 사용하였다. 일정 용적의(100$mm^{3}$) 종양에 대하여 1회 2 Gy를 1일 2회씩 6회 또는 1회 2.5 Gy를 1일 2회씩 8회 조사하는 등의 분할조사법을 사용하여 방사선조사를 시행하였다. SR 4233를 병용한 경우에는 매회 방사선조사 30분 전에 0.08 mmol/kg를 복강내 주사하였다. 정상조직에서 성장한 종양에 비하여 방사선 조사를 받은 조직에서는 성장한 종양은 분할 방사선조사에 대하여 약 5배의 내성을 보였으나 SR 4233 분할 치료에 대하여는 약 5배의 감수성을 나타내었다. 정상 종양 및 저산소세포 분획이 큰 종양의 방사선 감수성은 공통적으로 SR 4233의 병용후 증강되었으며 저산소세포 분획이 큰 종양에서의 증강율이 정상종양에서의 증강율과 동일하거나 더욱 높았다. 따라서 선택적 저산소세포 치사제인 SR 4233과 분할 방사선조사를 병용할 경우 저산소분획이 큰 종양에서 치사효과가 더욱 증강된 사실을 확인하였다. 이러한 결과는 높은 저산소세포 분획에 의한 방사선내성을 보이는 종양중 일부는 저산소세포치사제와 방사선치료를 병용함으로써 국소적 종양관해 실패의 요인인 저산소 세포를 극복하는 임상 적용 가능성을 시사한다.
고도병(High altitude disease)의 위험에도 불구하고, 고냉지에서 한우를 사육할 경우, 감염이나 스트레스 위험이 낮아서 한우의 육질을 개선하는데 도움이 된다고 한다. 하지만 어떤 고도가 고도병의 위험성이 없이 최적의 사육환경을 제공하는지에 대한 연구가 부족한 실정이다. 따라서 본 연구에서는 3가지 다른 고도(200m, 400m and 800m)에서 사육되고 있는 한우를 대상으로 혈액 검사, 혈청 생화학 검사 및 스트레스와 관련된 호르몬 검사를 실시하였다. 본 연구에서 적혈구 수치와 혈색소의 농도는 800m고지에서 사육된 한우에서 가장 높았고, 200m고지에서 사육된 한우에서 가장 낮게 나타났다. 이러한 결과는 저산소증에 따른 골수와 적혈구 신생 자극 반응에 기인한 것으로 보인다. 혈중의 AST, BUN과 cortisol농도는 800m고지에서 사육된 한우에서 가장 낮게 측정되었으며, 전반적인 건강상태도 200m지대에서 사육된 한우보다 800m지대에 사육된 한우 집단이 건강 상태가 양호한 것으로 나타났다. 이 결과는 한우의 건강에 영향을 주는 스트레스 요인이 800m고지에서 최소로 나타남을 의미하는 것 같다.
한국 연안 해역의 와편모조류 시스트의 연구 현황과 특성을 파악하기 위해 한국 연안과 내만은 물론 제주도 남부와 황해 일부해역을 포함한 동중국해를 대상으로 수행한 45편의 학술연구논문을 분석하였다. 결과 한국의 와편모조류 시스트는 1980년대 후반부터 연구가 시작되었고, 대상 해역은 적조발생 등 부영양화 해역이나 연안개발이 이루어지는 해역에 집중되었으며, 동해를 대상으로 한 연구결과는 없었다. 연구 내용도 단순한 출현 종 분석에서 부영양화 진행 등 해양환경의 변화과정 추적, 적조 발생에서 종자군으로서 역할, 세디맨트 트랩을 이용한 시스트의 형성과 변화양상 추적, 수소이온 농도 등 해양환경 변화에 따른 형태변화, 실험실에서 특정 종의 발아 특성 등 국제적 수준의 다양한 연구가 진행되고 있었다. 출현종이나 출현 세포밀도 역시 해역에 따라 매우 다양한 특성을 보인다. 그러나 한국의 와편모조류 시스트 연구는 아직 체계적 연구가 이루어지지 못하고 있고, 단편적이면서 개별적 내용으로 매우 제한적인 연구성격을 나타내었다. 특정 해역에 편중된 연구결과와 특정 연구 연구 집단에 의해 산발적인 연구가 수행되는 경향을 나타내었다. 해양에서 표층퇴적물의 와편모조류 시스트는 표영 환경의 누적지표를 나타내는 것으로 해양환경 변화를 효율적으로 추적하는 데 유용한 지표가 되고 있음을 고려할 때, 국내에서도 조직적인 인력양성과 함께 해양생태계의 구성인자로서 필수 해양생물 분야로 취급하는 연계적인 연구 추진이 절실하게 요구된다.
Hypoxia-inducible factor-$1{\alpha}$ ($HIF-1{\alpha}$) mediates tumor cell adaptation to hypoxic conditions and is a potentially important anticancer therapeutic target. We previously developed a method for synthesizing a benzofuran-based natural product, (R)-(-)-moracin-O, and obtained a novel potent analog, MO-460 that suppresses the accumulation of $HIF-1{\alpha}$ in Hep3B cells. However, the molecular target and underlying mechanism of action of MO-460 remained unclear. In the current study, we identified heterogeneous nuclear ribonucleoprotein A2B1 (hnRNPA2B1) as a molecular target of MO-460. MO-460 inhibits the initiation of $HIF-1{\alpha}$ translation by binding to the C-terminal glycinerich domain of hnRNPA2B1 and inhibiting its subsequent binding to the 3'-untranslated region of $HIF-1{\alpha}$ mRNA. Moreover, MO-460 suppresses $HIF-1{\alpha}$ protein synthesis under hypoxic conditions and induces the accumulation of stress granules. The data provided here suggest that hnRNPA2B1 serves as a crucial molecular target in hypoxiainduced tumor survival and thus offer an avenue for the development of novel anticancer therapies.
A high salt diet contributes to kidney damage by causing hypoxia and oxidative stress. Recently, an increase in dietary salt has been reported to induce an inflammatory phenotype in immune cells, further contributing to kidney damage. However, studies on the exact mechanism and role of a high salt diet on the inflammatory response in the kidneys are still insufficient. In this study, a cisplatin-induced acute kidney injury model using C57BL/6 mice was used to analyze the effect of salt intake on kidney injury. Results showed that high salt administration aggravated kidney edema in mice induced by treatment with cisplatin. Moreover, the indicators of kidney and liver function impairment were significantly increased in the group cotreated with high salt compared with that treated with cisplatin alone. Furthermore, the exacerbation of kidney damage by high salt administration was also associated with a decrease in the number of cells in the immune regulatory system. Additionally, high salt administration further decreased renal perfusion functions along with increased cisplatin-induced damage to proximal tubules. This was accompanied by increased expression of T cell immunoglobulin, mucin domain 1 (a biomarker of kidney injury), and Bax (a pro-apoptotic factor). Moreover, cisplatin-induced expression of proinflammatory mediators and cytokines, including cyclooxygenase-2 and tumor necrosis factor-α in kidney tissue, was further increased by high salt intake. Therefore, these results indicate that the kidney's inflammatory response by high salt treatment can further promote kidney damage caused by various pathological factors.

제 15 조 (계약 해지)
회원이 이용계약을 해지하고자 하는 때에는 [가입해지] 메뉴를 이용해 직접 해지해야 합니다.
제 16 조 (서비스 이용제한)
① 당 사이트는 회원이 서비스 이용내용에 있어서 본 약관 제 11조 내용을 위반하거나, 다음 각 호에 해당하는
경우 서비스 이용을 제한할 수 있습니다.
- 2년 이상 서비스를 이용한 적이 없는 경우
- 기타 정상적인 서비스 운영에 방해가 될 경우
② 상기 이용제한 규정에 따라 서비스를 이용하는 회원에게 서비스 이용에 대하여 별도 공지 없이 서비스 이용의
일시정지, 이용계약 해지 할 수 있습니다.
제 17 조 (전자우편주소 수집 금지)
회원은 전자우편주소 추출기 등을 이용하여 전자우편주소를 수집 또는 제3자에게 제공할 수 없습니다.
제 6 장 손해배상 및 기타사항
제 18 조 (손해배상)
당 사이트는 무료로 제공되는 서비스와 관련하여 회원에게 어떠한 손해가 발생하더라도 당 사이트가 고의 또는 과실로 인한 손해발생을 제외하고는 이에 대하여 책임을 부담하지 아니합니다.
제 19 조 (관할 법원)
서비스 이용으로 발생한 분쟁에 대해 소송이 제기되는 경우 민사 소송법상의 관할 법원에 제기합니다.
[부 칙]
1. (시행일) 이 약관은 2016년 9월 5일부터 적용되며, 종전 약관은 본 약관으로 대체되며, 개정된 약관의 적용일 이전 가입자도 개정된 약관의 적용을 받습니다.
