Epidemiological and laboratory studies provide insight into the anti-carcinogenic potential of garlic and its constituent compounds. Garlic is appealing as an anti-carcinogenic agent due to its ability to induce apoptosis in vitro. Diallyl disulfide (DADS) is one of the major components of garlic that used to determine inhibition of cell proliferation and induced apoptosis in human colon cell lines. In this study, human colorectal cancer cell lines (LOVO, HCT-116, SW-480) were exposed to DADS. The inhibitory effects of DADS dose level more than $50\;{\mu}M$ in the cell viability of all cell lines. Cell growth activity inhibits of human colon cancer cell lines. The inhibitory effects of DADS dose level more than $25\~50\;{\mu}M$ in the cell growth using MTT assay. We found that DADS may have the apoptosis action (chromatin condensation, DNA fragmentation) using DAPI staining and increased the expression of caspase-3 at the dose level more than $100\;{\mu}M$, decreased the expression level of $\beta-catenin$ at dose dependent in the western blotting. We suggest that DADS may have a potential candidate as cancer chemopreventive agents.
The water extract of Boyanghwanoh-tang has been used for treatment of ischemic vascular disease in oriental medicine. However, little is known about the mechanism by which the water extract of Boyanghwanoh-tang rescues cells from these damages. Therefore, this study was designed to evaluate the protective effects of Boyanghwanoh-tang on zinc-mediated cytotoxicity in H9c2 cardiomyoblast cells. This study demonstrates that, after treatment of H9c2 cells with zinc, there was a decrease in cell viability in a dose dependent manner, and there was a chromatin condensation. Zinc induced the change of cell morphology. In addition, zinc induced mitochondrial dysfunction. Zinc-induced H9c2 cell death was remarkably prevented by the pretreatment of Boyanghwanoh-tang consistently with increase of the peroxoredoxin 1, 2, 3, 5, and 6 expression. Taken together, the results suggest that zinc induced severe cell death in H9c2 cardiomyoblast cells, and that protective effects of Boyanghwanoh-tang against oxidative injuries are achieved through regulation of peroxiredoin expression.

The effects of ursodeoxycholic acid (UDCA) and its novel derivative, named as HS-1030, on the proliferation of HepG2, human hepatocellular carcinoma cells were investigated. Whereas UDCA had no significant effect in a concentration range we have tested, HS-1030 inhibited the proliferation of HepG2 cells in a concentration dependent manner. Surprisingly, HS-1030 had no effect on the proliferation of Human Chang liver cell which is a normal liver cell line. We also found that proliferation-inhibitory effect of HS-1030 was due to the induction of apoptosis of HepG2 cells, which was confirmed by observing the internucleosomal DNA fragmentation and morphological changes (ie., cell shrinkage, nuclear condensation and the formation of apoptotic bodies). These results suggest that HS-1030 may be a good candidate as a drug for the treatment of liver cancer.

The present study was performed to observe the role of protein kinase C (PKC) inhibitors (H-7, staurosporine) and daunorubicin in the cell death process of MCF7 cells; and examined whether or not the type of induced cell death was apoptosis. The usefulness of the combined therapy of PKC inhibitors and daunorubicin to improve the adverse effect of daunorubicin was also investigated. Cell death was induced by treatment with PKC inhibitors or daunorubicin. Characteristic morphologic features of cell shrinkage, chromatic condensation, and cytoplasmic vacuolization were observed. These treatments also stimulated the cleavage of poly-(ADP-ribose) polymerase (PARP), an early event in apoptosis. With slight differences in the percentage of apoptosis-induced cells, staurosporine, H-7 and daunorubicin effectively induced apoptosis in MCF7 cells. Furthermore, combined treatment of PKC inhibitors and daunorubicin significantly drove the cells into an apoptotic state. Hence, our results revealed the possible therapeutic value of combined therapy for the prevention of drug resistance and adverse side effects.
본 연구에서는 free radicals의 산화적인 손상에 의한 세포사멸에 있어서 녹차성분의 하나인 (-)epigallocatechin gallate의 억제효과를 규명하였다. 우선 radicals 소거작용에 있어서 (-)epigallocatechin gallate는 탁월한 항산화력을 발휘하였다. 혈관손상과 직결되는 혈관내피세포를 이용하여 hydroxyl radical의 $H_2O$$_2$에 의한 산화적인 손상을 유발시켜 세포생존율을 조사하였는데, (-)epigallocatechin gallate는 100 $\mu\textrm{m}$ 이하의 농도에서는 그 자체 독성이 없었고 $H_2O$$_2$의 산화적 독성효과를 경감시키는 것으로 나타났다. 그러나 flavone인 apigenin은 고농도에서 독성을 가지며 radical 소거활성이 미약하고 $H_2O$$_2$의 산화독성은 경감시키지 못하였다. 다양한 세포사멸 검출법을 이용하여 세포 및 세포핵의 형태학적 양상을 조사한 결과, 0.25mM $H_2O$$_2$에 의한 24시간 이내의 세포죽음은 세포사멸현상에 의하여 초래되었다. 그러나 이러한 세포사멸과정을 겪고 있는 혈관내피세포에 50 $\mu\textrm{m}$ (-)epigallocatechin gallate를 처리한 경우에 세포핵의 응축이나 DNA fragmentation은 사라지고 세포사멸작용을 억제시키는 효과를 보여주었다. 예상한 바와 같이 apigenin의 flavone은 세포사멸 억제효과를 나타내지 못하였다. (-)Epigallocatechin gallate는 녹차에 함유된 catechins의 하나로서 free radicals의 산화적 손상에 의한 세포사멸에 있어서 탁월한 방어적 인 세포생리학적인 기능을 지니고 있으며, 혈관노화 및 혈관손상과 함께 유발되는 세포사멸성 심혈관질환의 예방과 치료에 기능성 식품 신소재로서 활용될 수 있으리라 본다.
본 연구는 MDA-MB-231 세포에서 resveratrol의 apoptosis 유발 효과에 대해 알아보기 위해 연구되었다. Cell viability 결과 농도 유의적으로 감소하였으며, DAPI stain에서는 농도 의존적으로 chromatin condensation이 증가하는 것을 확인하였다. Resveratrol은 p53, cleaved-caspase-3, cleaved-caspase-9의 발현을 증가시켰지만, PI3K/Akt의 발현은 시간 의존적으로 감소시켰다. In vivo 실험에서는 resveratrol의 종양 억제 효과를 확인하였다. 50 mg/kg 투여한 군에서 종양의 크기가 대조군에 비해 감소하였으며, TUNEL assay를 통해 apoptosis cell을 관찰한 결과 50 mg/kg 투여한 군에서 많이 관찰되었다. 면역조직화학 염색을 통해 50 mg/kg 투여한 군에서 p53, cytochrome-C, cleaved-caspase-3의 발현이 증가하는 것을 확인하였다. 본 연구의 결과를 종합하여 봤을 때 resveratrol이 MDA-MB-231 세포에 apoptosis를 유발하는데 효과가 있는 것으로 사료된다.
와송 유래 에틸아세테이트(EtOAc) 분획물의 인체 폐암세포 A549에 대한 항암활성을 확인하기 위하여 본 연구를 수행하였다. 폐암 세포에 대한 세포 생존율을 측정하기 위하여 MTS assay를 수행한 결과, 농도 의존적으로 폐암세포 성장 억제효과를 보였다. 세포사멸 유도능을 확인하기 위하여 DAPI 핵염색을 통한 직접 육안관찰을 수행한 결과, EtOAc 분획물을 처리한 군에서 핵내 염색질 응축등의 세포사멸 지표가 관찰되었고, Annexin V-FITC를 이용하여 세포막에 노출된 phosphatidylinositol (PS)를 검출한 결과, 농도 의존적으로 초기 세포사멸 및 후기 세포사멸이 증가하였다. 세포사멸의 또다른 지표인 세포주기 억제능을 확인하기 위하여 G2/M기 관련 유전자인 CDK1, 4, cyclin B1, D1의 mRNA 발현정도를 RT-PCR을 이용하여 확인한 결과, 농도의존적으로 mRNA의 발현량이 현저히 감소하였으며, 세포사멸의 직접적 신호전달 표적 단백질인 p53, Bax, Bcl-2 및 pro-caspase-3등의 발현정도를 확인한 결과, p53과 Bax 단백질의 발현은 농도의존적으로 증가하였고, Bcl-2와 pro-caspase-3 단백질의 발현은 시간 및 농도의존적으로 감소하였다.
A three dimensional numerical model to predict the flow and transport of mixtures and also the electrochemical reactions in polymer electrolyte membrane (PEM) fuel cells is developed. The numerical computation is base on vorticity- velocity method. Governing equations for the flow and transport of mixtures are coupled with the equations for electrochemical reactions and are solved simultaneously including production and condensation of vapor. Fuel cell performance predicted by this calculation is compared with the experimental results and resonable agreements are achieved.
Objectives : This study was designed to investigate the protective mechanisms of Palmul-tang on hypoxia-induced cytotoxicity in H9c2 cardiomyoblast cells. Methods : In this study, we used H9c2 cells. Cells were subjected to hypoxia in the absence and presence of $1000\mu\textrm{g}/ml$ Palmul-tang for 24 hrs. Cells were treated with various concentrations of Palmul-tang for 24 hrs. Cell viability was measured by MTT assay. Hypoxia markedly decreased the viability of H9c2 cells, which was characterized with apparent apoptotic features such as chromatin condensation as well as fragmentation of genomic DNA and nuclei. Results : Palmul-tang significantly reduced hypoxia-induced cell death and apoptotic characteristics. Also, Palmul-tang prevented mitochondrial dysfunction including the disruption of mitochondrial membrane permeability transition (MPT) and an increase in expression of apoptogenic Bcl-2 proteins in hypoxia-H9c2 cells. Conclusions; This study suggests that the protective effects of Palmul-tang against hypoxic damages may be mediated by the modulation of Bcl-2, Bax expression.
Fungal metabolite, gliotoxin is an epipolythiodioxopiperazin (ETP) class and has various roles including immunomodulatory and apoptotic effects. This study was designed to evaluate the mechanism by which gliotoxin exerts the apoptosis on human promyelocytic leukemic HL-60 cells. Herein, we demonstrated that the gliotoxin decreased the cell viability in a time-dependent manner Gliotoxin-induced cell death was confirmed us apoptosis characterized by chromatin condensation and ladder-pattern fragmentation of genomic DNA. Gliotoxin increased the catalytic activities of caspase-3 and caspase-9. Activation of caspase-3 was further confirmed by degradation of procaspase-3 and poly(ADP-ribose) polymerase (PARP) by gliotoxin in HL-60 cells. Furthermore, gliotoxin induced the changes of mitochondrial transmembrane potential (MTP). Antioxidants, including GSH and NAC, markedly inhibited apoptosis with conistent suppression of enzymatic activity of caspase-3, caspase-9, and MTP loss in gliotoxin-treated cells. Taken together, we suggest that gliotoxin function as an oxidant and ploys proapoptotic roles in HL-60 cells via activation of intrinsic caspase cascades as well as mitochondrial dysfunction.
