Effect of Reducing Inflammation of Coptis chinensis Extract -Ceramide Complex through ECS Control in Atopic Dermatitis

  • Objectives: The purpose of this study was to confirm effect of reducing inflammation of Coptis chinensis extract-ceramide complex through the endocannabinoid system (ECS) control in atopic dermatitis. Methods: 8-week-old ICR mice were divided into normal group (Ctrl), lipid barrier elimination group (ADE), palmitoylethanolamide treated group after lipid barrier elimination (PEAT), and Coptis chinensis extract-ceramide complex applied group after lipid barrier elimination (CRA). After inducing atopic dermatitis, cannabinoid receptor (CB) 1, CB2, CD68, p-I𝜅B, iNOS, substance P and serotonin were observed to confirm the regulation of the ECS, macrophage activity and mast cell activity. Results: CB1 and CB2 showed higher positive reactions in the CRA than in the ADE and PEAT. CD68, p-I𝜅B and iNOS showed higher positive reaction in the ADE, PEAT and CRA than in the Ctrl, but the increase in the positive reaction was lower in the CEA compared to the ADE and PEAT. Substance P and serotonin showed higher positive reaction in the ADE, PEAT and CRA than in the Ctrl, but the increase in the positive reaction was lower in the CEA compared to the ADE and PEAT. Conclusions: The effects of Coptis chinensis extract -ceramide complex were confirmed on the regulation of the ECS, macrophage activity and mast cell activity.

Mad2B forms a complex with Cdc20, Cdc27, Rev3 and Rev1 in response to cisplatin-induced DNA damage

  • Mitotic arrest deficient 2 like 2 (Mad2L2, also known as Mad2B), the human homologue of the yeast Rev7 protein, is a regulatory subunit of DNA polymerase ζ that shares high sequence homology with Mad2, the mitotic checkpoint protein. Previously, we demonstrated the involvement of Mad2B in the cisplatin-induced DNA damage response. In this study, we extend our findings to show that Mad2B is recruited to sites of DNA damage in human cancer cells in response to cisplatin treatment. We found that in undamaged cells, Mad2B exists in a complex with Polζ-Rev1 and the APC/C subunit Cdc27. Following cisplatin-induced DNA damage, we observed an increase in the recruitment of Mad2B and Cdc20 (the activators of the APC/C), to the complex. The involvement of Mad2B-Cdc20-APC/C during DNA damage has not been reported before and suggests that the APC/C is activated following cisplatin-induced DNA damage. Using an in vitro ubiquitination assay, our data confirmed Mad2B-dependent activation of APC/C in cisplatin-treated cells. Mad2B may act as an accelerator for APC/C activation during DNA damage response. Our data strongly suggest a role for Mad2B-APC/C-Cdc20 in the ubiquitination of proteins involved in the DNA damage response.

A DPL (DNA/peptide/liposomes) Tripartite Complex Effective for Transfection in Serum

  • A short peptide corresponding to the nuclear localization signal (NLS) of human immunodeficiency virus (HIV)-l Tat protein, Arg-Lys-Lys-Arg-Arg-Gln-Arg-Arg-Arg, was employed to improve the efficiency of cellular uptake of nucleic acids. The peptide was first mixed with a reporter plasmid and then with cationic liposomes to form a tripartite complex of DNA/peptide/liposomes (DPL). Transfection efficiency of the DPL complex was compared with that of the conventional DNA/liposomes (DL) complex. When the DPL complex was formed with various cationic liposomes, DOTAP/DOPE (DP) liposome exhibited superior transfection efficiency to other liposomes tested in vitro. With the inclusion of the peptide, the DPL complex showed much enhanced transfection in various cancer cell lines. Particularly, transfection of the DPL complex in serum increased cellular uptake of a transgene up to 2 fold when compared with that in a serum free condition. Further, when the DPL complex was infused through the ureteric route of a rat, transfection efficiency was shown to be better in reporter gene expression than that obtained with the DL complex. This study shows that the DPL complex that is easy to formulate can be employed for much enhanced cellular uptake of a trans gene.

Synchronization in Complex Systems

  • In this paper, we introduce a complex systems synchronization method using hyper-chaos circuit consist of State-Controlled Cellular Neural Network (SC-CNN). We make a complex systems using SC-CNN with the n-double scroll. A complex system is created by applying identical n-double scroll or non-identical n-double scroll and Chua's oscillator with weak coupled method to each cell. Complex systems synchronization were achieved using GS(Generalized Synchronization) method between the transmitter and receiver about each state variable in the SC-CNN.

  • In this paper we propose how to detect circular objects in the gray scale image and segment them using the discrete Morse theory, which makes it possible to analyze the topology of a digital image, when it is transformed into the data structure of some combinatorial complex. It is possible to get meaningful information about how many connected components and topologically circular shapes are in the image by computing the persistent homology of the filtration using the Morse complex. We obtain a Morse complex by modeling an image as a cubical cellular complex. Each cell in the Morse complex is the critical point at which the topological structure changes in the filtration consisting of the level sets of the image. In this paper, we implement the proposed algorithm of segmenting the circularly shaped objects with a long persistence of homology as well as computing persistent homology along the filtration of the input image and displaying in the form of a persistence diagram.

  • The Golgi complex plays a central role in protein secretion by regulating cargo sorting and trafficking. As these processes are of functional importance to cell polarity, motility, growth, and division, there is considerable interest in achieving a comprehensive understanding of Golgi complex biology. However, the unique stack structure of this organelle has been a major hurdle to our understanding of how proteins are secreted through the Golgi apparatus. Herein, we summarize available relevant research to gain an understanding of protein secretion via the Golgi complex. This includes the molecular mechanisms of intra-Golgi trafficking and cargo export in the trans-Golgi network. Moreover, we review recent insights on signaling pathways regulated by the Golgi complex and their physiological significance.

A Splice Variant of the C2H2-Type Zinc Finger Protein, ZNF268s, Regulates NF-κB Activation by TNF-α

  • $I{\kappa}B$ kinase (IKK), the pivotal kinase in signal-dependent activation of nuclear factor-${\kappa}B$ (NF-${\kappa}B$), is composed of multiple protein components, including IKK ${\alpha}/{\beta}/{\gamma}$ core subunits. To investigate the regulation of the IKK complex, we immunoaffinity purified the IKK complex, and by MALDI-TOF mass spectrometry identified a splice variant of zinc finger protein 268 (ZNF268) as a novel IKKinteracting protein. Both the full-length and the spliced form of the ZNF268 protein were detected in a variety of mammalian tissues and cell lines. The genes were cloned and expressed by in vitro transcription/translation. Several deletion derivatives, such as KRAB domain (KRAB) on its own, the KRAB/spacer/4-zinc fingers (zF4), and the spacer/4-zinc fingers (zS4), were ectopically expressed in mammalian cells and exhibited had different subcellular locations. The KRAB-containing mutants were restricted to the nucleus, while zS4 was localized in the cytosol. TNF-${\alpha}$-induced NF-${\kappa}B$ activation was examined using these mutants and only zS4 was found to stimulate activation. Collectively, the results indicate that a spliced form of ZNF268 lacking the KRAB domain is located in the cytosol, where it seems to play a role in TNF-${\alpha}$-induced NF-${\kappa}B$ activation by interacting with the IKK complex.

Blind Channel Estimation based on Hadamard Matrix Interstream Transmission for Multi-Cell MIMO Networks (다중 셀 MIMO 네트워크를 위한 Hadamard 행렬 Interstream 전송 기반 Blind 채널 추정)

  • In this paper, we introduce a Hadamard matrix interstream transmission based blind channel estimation for multi-cells multiple-input and multiple-output (MIMO) networks. The proposed scheme is based on a network with mobile stations (MS) which are deployed with multi cells. We assume that the MS have the signals from both cells. The signal from near cell are considered as desired signal and the signals from the other cells are interference signal. Since the channel is blind, so that we transmit Hadamard matrix pattern pilot stream to estimate the channel; that gives easier and fast channel estimation for large scale MIMO channel. The computation of Hadamard based system takes only complex additions, and thus the complexity of which is much lower than the scheme with Fourier transform since complex multiplications are not needed. The numerical analysis will give perfection of proposed channel estimation.

  • The human visual cortex has the characteristic that reacts sensitively to stimuli with special directional or temporal frequency changes while it is insensitive to selective stimuli of spatial phases. In this paper we implemented the model of complex cell using an image estimation iterative algorithm by Gabor wavelet transform. The performance of implemented model evaluated the consistency between the physiological experimental results in related papers. The implemented model is limited in the complete model of the receptive field in the retina where simple cells and complex cells are distributed together. But the implemented model express the reaction of the complex cells from the point of view of the detection of corners and edges.

Review on Korean Herbal Medicine Complex Prescriptions and Single Herbal Medicines for Osteoporosis - Focused on Domestic Research Published Since 2013 (골다공증의 한방 복합 처방 및 단일 본초에 대한 국내 연구 고찰 - 2013년 이후 발표된 국내 연구를 중심으로)

  • The purpose of this study was to investigate the effect of single herbal medicines and Korean herbal medicine complex prescriptions for osteoporosis in Korea. We targeted studies published in domestic journals from January 1, 2013 to April 1, 2021. For domestic databases, RISS, NDSL, OASIS, DBpia, KISS, KISTI, and Korean Traditional Knowledge Portal were used. Words such as "osteoporosis", "herbal medicine", "herbal combination prescription" and "herbal" were combined and searched. A total of 32 studies were selected. Among them, 8 studies were conducted exclusively for animal studies, 18 studies were conducted on cell experiments, and 6 studies were conducted on both animal and cell experiments. There were 18 studies using a single herb and 14 studies using a combination of Herbal medicine. As a result of the investigation of a single herb, it was found that 補陽藥 was most widely used, and it was found that it was mainly used in the herbal combination prescription, mainly for 補陽藥, 補氣藥, and 補陰藥. Studies in animal experiments revealed significant results mainly in bone mineral density, serological indicators, and morphological indicators of bone tissue. In the study of cell experiments, the effects were found in factors related to the differentiation of osteoclasts and inhibitors of inflammation. Through the results of this study, we are able to confirm the effective single herbal medicines and herbal medicine complex prescriptions for the treatment of osteoporosis, and it is expected that they will contribute to clinical treatment.
