The Expression Patterns of Cdc25A, Cdc25B, Sox2 and Mnb in Central Nervous System in Early Chicken Embryos

  • The sense and antisense digoxigenin-labeled RNA probes of four genes, Cdc25A, Cdc25B, Sox2 and Mnb, were produced by using SP6 and T7 RNA polymerases, respectively, and in vitro transcription. Expression patterns of the four genes were detected by in situ hybridization in HH (Hamburger and Hamilton) stage 10 chick embryos. In general, expression patterns of the four genes were similar. mRNA of the four genes was mostly restricted to the entire CNS (central nervous system). All were confined to an identical region, neural tube, neural groove and caudal neural plate, corresponding to the notochord or spinal cord, but there was some distinction in specific region or in concentration, for example in somites. The overlap in expression at the same developmental stage in the CNS suggests that the four genes may be functional similar or related in CNS development. Expression patterns of the four genes support specific roles of these regulators in the developing CNS.

An Efficient Disease Inspection Model for Untrained Crops Using VGG16 (VGG16을 활용한 미학습 농작물의 효율적인 질병 진단 모델)

  • Early detection and classification of crop diseases play significant role to help farmers to reduce disease spread and to increase agricultural productivity. Recently, many researchers have used deep learning techniques like convolutional neural network (CNN) classifier for crop disease inspection with dataset of crop leaf images (e.g., PlantVillage dataset). These researches present over 90% of classification accuracy for crop diseases, but they have ability to detect only the pre-trained diseases. This paper proposes an efficient disease inspection CNN model for new crops not used in the pre-trained model. First, we present a benchmark crop disease classifier (CDC) for the crops in PlantVillage dataset using VGG16. Then we build a modified crop disease classifier (mCDC) to inspect diseases for untrained crops. The performance evaluation results show that the proposed model outperforms the benchmark classifier.

The Suitability of the CDC Field Triage for Korean Trauma Care

  • Purpose: Accurate and appropriate prehospital field triage is essential for a trauma system. The Korean trauma system (established in 2014) uses the trauma field triage algorithm of the United States Centers for Disease Control and Prevention (CDC). This study evaluated the suitability of the CDC field triage criteria for major trauma cases (injury severity score >15) in Korea. Methods: This retrospective cohort study evaluated trauma patients who presented at the authors' regional trauma center from January 1 to May 7, 2017. The undertriage and overtriage rates of each CDC field triage step were calculated. Receiver operating characteristic curves were constructed, and the area under the curve (AUC) was evaluated for each step. Results: Among the 1,009 enrolled patients, 168 (16.7%) had major trauma. The undertriage/overtriage rates of each step (steps I, II, III, and IV) of CDC field triage were 9.2%/47.4%, 6.3%/50.8%, 4.5%/59.4%, and 5.3%/78.9%, respectively. The AUC values of each CDC triage step were 0.722, 0.783, 0.791, and 0.615, respectively. The AUC values of the separate components of each step (physiologic criteria, anatomic criteria, mechanism-of-injury criteria, and special considerations) were 0.722, 0.648, 0.647, and 0.456, respectively. Conclusions: The CDC field triage system is acceptable, but not ideal, for Korean trauma care. If we follow the protocol, it would be preferable to omit step IV. The Korean Triage and Acuity Scale may be a good indicator for in-hospital triage. However, a new triage protocol that is simple to estimate on-scene while having good performance should be developed.

Status of Health Education by Elementary School Nurses (초등학교 보건교사의 보건교육 실태)

  • Objectives: To investigate the status of health education in elementary schools. Methods: 620 school nurses were surveyed by questionnaires from September to December in 2003. Results: 1) Among the school nurses under inquiry of planning of health education, 3.3% and 9.3% of them did not prepare for teaching plan. 2) The average time for health education by a school nurse was 96.8 hours a year, and handouts for health education were distributed 10.6 times. Among the contents of health education, sex education took the largest portion of health education with 24.6 hours a year. 3) With regard to the contents of health education covered by school nurses, sex education ranked first with 90.2%, next came drug abuse with 78.4%, dental health, CDC, disease control, healthy life and smoking, body structure and function and growth and development safety, alcohol, nutrition and environmental health followed them. 4) The main contents of education were CDC, dental health sex, healthy life and disease control for 1st, 2nd and 3rd grade students, sex, CDC, disease control and safety for 4th grade students, and sex, CDC, drug abuse and smoking for 5th and 6th grade students. 5) 72.6% of school nurses used class room for health education, 20.0% and 7.4% of them used grade and others such as broadcast, respectively. 6) 42.1% of school nurses used blackboard, 37.0% and 18.6% of them used visual media and handout as a teaching aids for health education. 7) 31.6% of school nurses replied that education time was insufficient 9.5% and 15.9% of them replied the contents of health education were inadequate and methods of health education were inappropriate, respectively. Conclusions: For the successful school health education, it would be in need of sufficient time for health education by opening health education course and of modify the various working conditions of school nurses, and those of effective educational materials and media for health education.

Effect of sodium nitrite on Trichomonas vaginalis (아질산나트륨(sodium nitrite)이 질편모충 증식과 미세구조에 미치는 영향)

  • We have investigated the action of sodium nitrite on the growth and morphologic changes of T uaginolis and on the treatment of subcutaneous abscess by trichomonad in mice. Sodium nitrite inhibited the growth of metronidazole-sensitive KT9 isolate and metronidazole-resistant CDC85 strain of T vcsinalis as concentration of 6 mM and 10 mM respectively Intraperitoneal injection of sodium nitrite (70 ㎍, 100 ㎍, 130 ㎍/g body weight) did not reduce the size of abscess produced by subcutaneous inoculation of T uasinnlis in mice. T uosinnlis, treated with sodium nitrite at concentration giving about 50% inhibition of growth, showed fissures, many vacuoles and electron-translucent zone in the cytoplasm by transmission electron microscopy. In the case of CDC85 treated with 9 mM sodium nitrite, hydrogenosomal matrical change , destruction of hydrogenosomal membrane, autophagic vacuoles, disappearance of Golgi complex and polysome were notably observed . With above results, it is assumed that sodium nitrite inhibits the growth of metronidazole-sensitive and - resistant strains of T. ucsinalis and induces the morphological changes of 7 uusinalis although it does not affect in reducing of abscess size by vagiginalis in mice.

  • 현대 전력산업은 IEC61850의 표준화작업이 한창 진행중이다. IEC61850-7-4는 전력설비 요소마다 Logical Node를 선언해 놓았다. 현재의 Logical Node는 보호, 제어, 인터페이스, 기록, 자동, 측정, 센서 & 모니터링, 변전기기 등의 그룹으로 나뉘어져 있다. 그러나 본 논문에서 대상으로 하는 GIS 예방진단 시스템에 관한 Logical Node와 공통 데이터 클레스(CDC: Common Data Class)가 명확히 선언되지 않았다. 1993년 IEC61850의 객체모델이 제안될 때부터 변전소의 다양한 설비를 완벽하게 객체화 할 수 있는가에 대한 의문이 있었으며 현재에서 이런 의문점을 개선하기 위하여 TC57위원회에서는 지속적인 모델 수정 및 정비 작업을 수행하였다. 실제로 현재에도 전력품질에 대한 logical node가 'Q'그룹으로 새롭게 제시되고 있고 풍력발전이나 분산전원에 대한 logical node가 추가되거나 기존의 IEC61850-7-4의 내용이 수정되고 있는 실정이다. 본 연구는 이와 같이 변전설비 자동화에 필요하지만 IEC TC57에서 표준화되지 않은 온라인 GIS 예방진단용 설비에 대한 표준모델을 제시하였고 모델에 대한 logical node나 CDC를 구성하였다.

Pre-immigration Screening for Tuberculosis in South Korea: A Comparison of Smear- and Culture-Based Protocols

  • Background: Tuberculosis (TB) is the most important disease screened for upon patient history review during preimmigration medical examinations as performed in South Korea in prospective immigrants to certain Western countries. In 2007, the U.S. Centers for Disease Control and Prevention (CDC) changed the TB screening protocol from a smear-based test to the complete Culture and Directly Observed Therapy Tuberculosis Technical Instructions (CDOT TB TI) for reducing the incidence of TB in foreign-born immigrants. Methods: This study evaluated the effect of the revised (as compared with the old) protocol in South Korea. Results: Of the 40,558 visa applicants, 365 exhibited chest radiographic results suggestive of active or inactive TB, and 351 underwent sputum tests (acid-fast bacilli smear and Mycobacterium tuberculosis culture). To this end, using the CDOT TB TI, 36 subjects (88.8 per $10^5$ of the population) were found to have TB, compared with only seven using the older U.S. CDC technical instruction (TI) (p<0.001). In addition, there were six drug-resistant cases which were identified (16.7 per $10^5$ of the population), two of whom had multidrug-resistance (5.6 per $10^5$ of the population). Conclusion: The culture-based 2007 TI identified a great deal of TB cases current to the individuals tested, as compared to older U.S. CDC TI.

Activation of Bovine Oocytes by Combined Treatment with Ionomycin and cdc2 Kinase Inhibitor

  • The success of nuclear transplantation with mammalian oocytes depends critically on the potential of oocytes activation, which mainly caused to prevent the re-accumulation of maturation promoting factor (MPF). This study was conducted to compare the effect of combined treatment of lonomycin with a Hl-histone kinase inhibitor (dimethylaminopurine, DMAP) or cdc2 kinase inhibitor (sodium pyrophosphate, SPP) on activation of bovine oocytes. In vitro matured bovine oocytes with the first polar body (PB) and dense cytoplasm were assigned to 3 experimental groups. For activation treatment, oocytcs were exposed to 5 $\mu$M lonomycin for 5 min (Group 1), and followed by 1.9 mM dimethylaminopurine (DMAP) for 3 h (Group 2) or followed by 2 mM sodium pyrophosphate (SPP) for 3 h (Group 3). The activation effects in the three treatments and the control group (untreated) were judged by the extrusion of the second PB and formation of a pronucleus (PN). Differences among groups were analysed using one-way ANOVA after arc-sine transformation of proportional data. All three treatments led to high activation rates (90% to 95%), with significant difference from the control. However, the extrusion of the second PB and the rate of PN formation differed remarkably among treatments. In Group I and 3, about 95% of the oocytes had extruded the second polar body, but one PN had formed in a higher proportion of oocytes in Group 3 than in Group 1 (90% vs. 5%). In experiment 2, the rates of cleavage and development into blastocysts in Group 1 were significantly lower than those of Group 2 and 3 (8.7% and 0% vs. 50.5% and 11.6%, and 44.6% and 7.2%, respectively, P<0.05). In experiment 3, ~80% of parthenotes in Group 1 were developed with haploid chromosomal sets. However, when ionomycin was followed immediately by DMAP (Group 2). only 20% of parthenotes were haploid. In Group 3, combined treatment with ionomycin and SPP, the appearance of abnormal chromosomal tracts was significantly (P〈0.05) reduced and the proportion of haploid parthenotes was increased to 85% (17/20) than in Group 2. These results demonstrate that SPP acted as a cdc2 kinase inhibitor and formed the haploidy in oocyte activation. Thus, the present study suggests that cdc2 kinase inhibitor, such as sodium pyrophosphate, may have an effective role in oocyte activation for the production of cloned embryos/animals by nuclear transplantation.

Anti-oxidant, Anti-inflammatory and Anti-cancer Effect of Methanol Extract of Pogostemon cablin (광곽향 메탄올 추출물의 항산화, 항염증 및 암세포 증식 억제 효과)

  • In the present study, the substance that show anti-proliferation of cancer cells as well as anti-oxidant and anti-inflammatory effect was searched. As a results, the methanol extract of Pogostemon cablin (P. cablin), is a well-known herb for traditional medicine in Korea and China for treating the digestive disorders, less of appetite, vomiting and diarrhea, inhibited the growth of various cancer cells such as A549, HepG2, MCF7 and HT29 cells. Cytotoxic effect of methanol extraction of P. cablin was excellent in A549 cells. P. cablin extract induced cell cycle arrest at G1 phase of A549 in a dose dependent manner. And it induced phosphorylation of p38 and Cdc25A and reduced expression of Cdc25A, Cdks, Cyclins and phospho-Retinoblastoma (Rb) proteins. Therefore, P. cablin extract seems to act through the p38 - Cdc25A - Cdk - Cyclin - Rb pathway in A549 cells. In addition, P. cablin extract showed anti-oxidant effect by DPPH free radical scavenging assay and anti-inflammation effect by inhibition of lipopolysaccharide (LPS)-induced nitric oxide (NO) in RAW 264.7 cells in a dose-dependent manner. Taken together, these results suggest that P. cablin may be used as not only candidate materials for anti-cancer, anti-inflammatory and anti-oxidant, moreover, it would be possible utilized in various health functional food materials.
