Isolation and purification of Cd-binding high molecular weight protein in rat liver(I) (흰쥐 간에서의 카드뮴 결합 고분자량 단백질의 분리 및 정제(I))

  • We isolated two Cd-binding high molecular weight proteins from intraperitoneally cadmium injected rat liver. Molecular weight of Cd-BP(I) purified from Sephacryl S-100 and DEAE Sepharose column chromatography and Cd-BP(II) purified from DEAE Sepharose column chromatography and Sulphonyl Sepharose column chromatography was 33,000 and 18,400, respectively. Alcohol dehydrogenase and alkaline phosphatase acitivities were not detected from two purified proteins.

Induction of Cd-binding High Molecular Weight Protein [Cd-BP(II)] in Rat Tissues (흰쥐 조직에서의 카드뮴 결합 고분자량 단백질 [Cd-BP(II)]의 유도)

  • The study was carried out on the biochemical characters of Cd-BP(II) after isolation and purification of the protein from the liver of rat ip injection with Cd. A continued study has been doing whether Cd-BP(II) could be induced by Cd or by the other metals such as Zn and Cu. Antisera were made against the antigen of Cd-BP(II) from New Zealand white rabbits. We carried out g-globulin purification, then Ouchterlony test and gel immunodiffusion test. Cd-BP(II) was also found in normal tissues of rat. It was induced up to a considerable level by Cd, whose induced level was higher than that Cu or Zn treatment. The level of induction by Cu or Zn pretreatment plus Cd treatment was lower than that by single treatment of Cu or Zn. Such a result was presumably related to the Cd toxicity.

  • A Cd-binding protein was identified in the renal cytosol of the Antarctic clam Laternula elliptica which naturally contains high concentrations of Cd. The Cd-binding protein showed similar characteristics of metallothionein (MT) in molecular weight (about 10-12 kDa) and low spectral absorbance at 280 nm with relatively high absorbance at 254nm. Results of immuno-histochemical staining suggested that the MT-like Cd-binding protein was mainly located in the epithelial cells of the kidney. The MT-like protein was a major ligand of cytosolic Cd as shown in the elution profiles of chromatography and may play an important role in Cd sequestration and accumulation in L. elliptica kidney. A considerable amount of Cd was also found to be associated with particulate fraction, indicating the sequestration to particulate fraction is as important as binding to the cytosolic MT-like protein in Cd accumulation in the kidney.

  • 본 연구는 중금속에 의한 생체내 독성기전을 연구하고자 흰쥐간의 상층액에서 카드뮴과 잘 결합하는 HMWP들을 분리, 정재하고 나아가 생화학적 특성을 밝히고 이단백질이 카드뮴에 의해 생성되는 것인지 아니면 간의 기존 단백질인지를 밝히고자 함을 연구의 목적으로 하였다. CdCl %2 (3mg/kg body wt.)을 3일간 ip injection시킨 후 흰쥐의 간을 적출하고 균질화하여 원심분리한 crude extract를 직접 Sephacryl S-100에 흡착시켜 10mM phosphate buffer(pH 7.0)으로 용출시켰다. 용출된 fraction tube를 UV spectrometer로 흡광도를 측정하고 atomic absorption spectrophotometer로 카드뮴량을 측정하여 카드뮴량이 높은 분획을 모아서 ion exchange column chromatography(DEAE-Sepharose)에 흡착시켜 염농도 구배로 chromatography를 실시하고, 음이온 교환수지에서 흡착되지 않고 용출된 분획을 ultrafiltration으로 농축시킨 후 S-Sepharose에 흡착시켜 염농도 구배로 chromatography를 실시한 결과 두 종류의 카드뮴 결합 단백질(Cd-BP)를 분리, 정제하였다.

Accumulation and Elimination of Cadmium and Zinc in Littorina brevicula (총알고둥에서 카드뮴과 아연의 축적과 제거)

  • Accumulation, elimination and subcellular distribution of heavy metals in Littorina brevicula exposed to cadmium and zinc separately and concurrently were investigated. When the winkles had been exposed to 400 ㎍/L CdCl₂ and 3000 ㎍/l ZnSO₄ separately for 90 days, each of the metal body burden in the whole sofl parts increased in proportion to time of exposure until 70 days. But it didn't increase after 70 days. But when the winkles had been exposed to cadmium and zinc simultaneously, cadmium body burden decreased but zinc body burden increased as compared to the winkles exposed to each of the metal. We also found that cadmium accumulated in the winkles was not depurated for 42 days, but zinc accumulated in them was depurated. Especially, zinc was depurated faster when they had been exposed to mixture of cadmium and zinc. After the winkles had been exposed to cadmium and zinc separately for 70 days, about 60% cadmium of the total body burden was associated with the soluble fraction, while about 75% zinc of the total body burden was associated with insoluble fraction. And these trends of metal partitioning did not alter when the winkles had been exposed to metal mixture. After the soluble fraction applied to gel-filtration chromatography column, the distribution patterns of cadmium and zinc associated with proteins or ligands were different each other. Most of cadmium (>90%) in the soluble fraction was bound to MBP-1 (Metal-binding protein-1), about 6.5 kDa), while zinc was distributed evenly to HMW (High molecular weight fraction, >60 kDa), MBP-1, MBP-2 (about 5 kDa), LMW (Low molecular weight fraction, <1 kDa).
