Development of an E. coli Expression Cassette for the Efficient Production of a Harmful Protein

  • In order to produce a harmful protein more efficiently, this expression cassette, dubbed pCol-MICT, is directed by the colicin promoter, and was constructed by the insertion of a $rrnBT_1T_2$ fragment of pEXP7, and a MxelnteinCBD fragment of pTXB3, into pSH375. To test whether harmful proteins, including proteolytic enzymes, could be effectively produced by this cassette, the carboxypeptidase (CPase) Taq gene was inserted into the pCol-MICT cassette to yield pCol-CPase Taq-MICT. E coli W3l 10 tells harboring pCol-CPase Taq-MICT produced a large quantity of this enzyme, as much as 47.2 mg of purified from per liter of culture, when cultured in the presence of mitomycin C ($0.4{\mu}g/mL$). This indicates that the colicin promoter-controlled E, coli expression cassette was able to produce almost 8 times of protein than the conventional tar promoter-based system, and that this cassette may be useful in the Synthesis of other harmful proteins.

Structural Analysis of Class I Integron Gene Cassette and Assessment of Genetic Relationships by PFGE of Salmonella enterica Serovar Typhimurium Isolated in Gyeongbuk Area (경북지역에서 분리된 Salmonella enterica Serovar Typhimurium의 Class I Integron Gene Cassette 특성과 PEGE 유형분석)

  • Thirty five Salmonella enterica serovar Typhimurium strains were isolated from diarrheic patients and pigs in Gyeongbuk area from 2003 to 2004. All 35 strains (17 strains from diarrheic patients and 18 from pigs) were resistant to more than one drug and most of strains isolated from pigs were resistant to ampicillin, ohloram-phenicol, streptomycin, sulfamethoxazole-trimethoprime, tetracyclin and nalidixic acid. Each isolate was also screened or the presence of class I, II and III integron gene cassettes. Among 35 strains,3 out of 17 strains isolated from diarrheic patients, carried dhfrX-orfF-aadA2 integron gene cassette and among 18 strains isolated from diseased pigs, 11 strains carried dhfrX-orfF-aadA2 integron gene cassette and 1 strain carried aadA2 integron only. But any class II and class II integron gene cassette were not detected in 35 strains. Thirty five strains were divided by five pulsotypes. Thirty one strains out of thirty five were pulsotype A. Among the remaining 4 strains, one each strain belonged to pulsotype B, C, D and pulsotype E. This data of pulsotypes showed that the widespread of pulsotype A, Salmonella enterica serovar Typhimurium in human and pigs in Gyeongbuk area may have been caused by the dissemination of a few epidemic strains in this area. Thirteen strains contain dhfrX-orfF-aadA2 integron gene cassette showed pulsotype A and one strain contains dhfrX-orfF-aadA2 integron gene cassette showed pulsotype B. One strain contains aadA2 integron showed pulsotype E. But fifteen strains do not contain any integron showed pulsotype A.

Heterologous Expression of Hybrid Type II Polyketide Synthase System in Streptomyces Species

  • Polyketides are an extensive class of secondary metabolites with diverse molecular structures and biological activities. A plasmid-based minimal polyketide synthase (PKS) expression cassette was constructed using a subset of actinorhodin (act) biosynthetic genes (actI-orfl, actI-orf2, actI-orf3, actIII, actⅦ, and actIV) from Streptomyces coelicolor, which specify the construction of an orange-fluorescent anthraquinone product aloesaponarin II, a type II polyketide compound derived from one acetyl coenzyme A and 7 malonyl coenzyme A extender units. This system was designed as an indicator pathway in S. parvulus to generate a hybrid type II polyketide compound via gene-specific replacement. The act ${\beta}-ketoacyl$ synthase unit (actI-orfl and actI-orf2) in the expression cassette was specifically replaced with oxytetracycline ${\beta}-ketoacyl$ synthase otcY-orfl and otcY-orf2). This plasmid-based hybrid PKS cassette generated a novel orange-fluorescent compound structurally different from aloesaponarin II in both S. lividans and S. parvulus. In addition, several additional distinctive blue-fluorescent compounds were detected, when this hybrid PKS cassette was expressed in S. coelicolor B78 (actI-orf2 mutant), implying that the expression of plasmid-based hybrid PKS cassette in Streptomyces species should be an efficient way of generating hybrid type II polyketide compounds.

Development of Automatic Well-plate Changing Robot System for Genome Project (유전체 연구를 위한 Well-plate 자동 교환 시스템의 개발)

  • In this study, the automatic system exchanging well-plates was developed as a basic stage of the genome project. The developed system consisted of the plate fixing well-plates, the well-plate cassette, the head to move a well-plate from the well-plate cassette to the plate fixing well-plates before genome work or from the plate to the cassette after the work, the manipulator to move the head on the X, Y and Z axes and the control system. The performance test to exchange well-plates with the robotic system developed was carried out. The time to set an well-plate from the well-plate cassette onto the board fixing well-plates was 55 seconds and the time for 9 ones was 8 minutes and 15 seconds. It took 57 seconds to move a well-plate from the board to the cassette and 8 minutes and 33 seconds for 9 ones.

  • Flow field in a roller conveyor system, induced from the movement of a cassette in which glasses for flat panel display are loaded, is numerically studied in this paper. Contamination on the glass surface by dust particles produced from mechanical friction between roller and cassette is predicted from the analysis results of flow fields. Results show that a large swirl flow is formed under the moving cassette with constant speed. This swirl flow is confined only under the cassette because two main streams from the backward and the fan filter unit on the top ceiling are sufficiently strong. Therefore, it can be said that possibility of the contamination by the particles originated from the friction is relatively low. It is also revealed that flow direction between glass plates is changed according to the speed of the cassette movement due to the shear force of glass plates.

  • In this paper is studied a method of interfacing microcomputer system with an ordinary cassette tape recorder The use of a parallel interface adapter has made it possible not only to construct a byte in software method but also to control noter operation of the recorder. Considerable improvement of the data handling speed is accomplished in spite of a relatively simplified network configurations.

  • In the drawing from round billet to non-circular section there are two different processes through solid hole die(HD) and the other cassette roller dies(CRD). The CRD process has several cassette type rollers and a billet is able to move through the given gaps between two profiled rollers. The objective of this study is based on the analysis and evaluation of two aforementioned processes using experiments and finite element simulation. In order to simulate the multi-stage drawing process from circular sectioned billet to rounded square section, the finite element analysis is applied to the process using a commercially available DEFORM-3D code. Two types of experimental drawing tests through designed and manufactured dies for pure copper and aluminum alloy are carried out at room temperature. The analysis included comparison of material properties before and after drawing of each process and also provide some useful information by a FEM simulation.

  • Computed Radiography(CR) is a relatively new technology that relies on an image plate(IP) as an alternate x-ray sensor to screen/film. Standard CR cassettes do not have lead foil behind the IP to control scatter radiation. The result of this study indicate that such control is needed. In most screen/film cassettes, that lines the rear of the cassette eliminates back scatter radiation. This study was performed to Investigate on the effects of back scatter in CR images by size of exposure field, distance between the CR cassette and the wall of radiography room. 1. It showed artifacts from hinges and clips located on the back of CR cassette by back scatter radiation. 2. The greater effects of back scatter radiation in CR images was attributed to the greater size of exposure field and the longer distance between the CR cassette and the wall of radiography room.

Infection Control of Computed Radiography Portable in Radiology (영상의학과 이동촬영장비의 감염 관리)

  • The purpose of this study is to prevent infection in the hospital by computed radiography portable and to provide basic data on infection-related education by investigating bacterial contamination level of computed radiography portable equipment using IP cassette. The results suggest that IP cassette No. 1 is infected with CNS and VRE, no. 2 with CNS, No. 3 with CNS and Pseudomonas aeruginosa, No. 4 with CNS, No. 5 with CNS and Bacillus sp., and No. 6 with enterococcus faecium. Enterococcus faecium and bacillus sp. were detected from the IP reader and Acinetobacter baumannii was detected on the mobile handle; Bacillus sp. on the control buttons, CNS and Bacillus sp. from the irradiation control handle, Acinetobacter baumannii on the x-ray generation switch, and CNS on the barcode scanner. In addition, Bacillus sp. Acinetobacter baumannii was found on the IP cassette mobile table and CNS and bacillus sp. were found on the lead apron. Acinetobacter baumannii and CNS were detected from the medical gloves worn by a radiological technologist during radiography. This suggests that IP cassette should be sterilized after use as it can hand over bacteria to IP reader and IP mobile table. Medical gloves that are in direct contact with patients should also be replaced after using them once and other supplies such as x-ray generation switch and lead apron should thoroughly be sterilized to prevent infection due to radiography as they are in a lot of contact with patients.
