Cellulolytic mocrooganisms were isolated from the cellulose-cultural properties. Among them, Aspergillus clavatus with the highest cellulase activity was identified by the morphological characteristics and it's enzyme activities were compared on the various cultural conditions. It was found that the induction of carboxymethylcellulase(CMCase), avicelase and salicinase in CMC liquid media showed the highest enzyme acitivity on five day's cultivation at $30^{\circ}C$ and thereafter their activities were gradually decreased with time. After crude extracellular enzymes precipitated with the 70% saturated ammonium sulfate solution were dialyzed with 20 mM acetate buffer pH 6.0, each crude enzyme was examined. The optimal activities of CMCase and avicelase were both found to be at $50^{\circ}C$ and pH 6.0. Their thermal stability was appeared at the $50^{\circ}C$. CMCase and avicelase had the maxima activities with 1.5% and 2.2% substrate concentration, respectively. The concentration of 5 mM $Mg^{++}$ or $Ca^{++}$ was found to have a maximum cellulase activity and its activity was inhibited with more than 5 mM $Cu^{++}$ and $Zn^{++}$ concentration. Cellulase activity was also inhibited sensitively by the inhibitors such as 2-mercaptoethanol, iodine and sodium azide.
One hundred thirty nine bacteria were isolated from rotten onions collected from main producing districts, Chang-Nyung, Eui-Ryung, and Ham-Yang in Korea. The $18\%$ (25 strains) of bacterial isolates have carboxymethylcellulase (CMCase) activity and the $53\%$ (74 strains) have polygalacturonase (PGase) activity. Thirty one among randomly selected 45 strains of PGase producing bacteria have pathogenicity to onions. The isolates were classified into Pseudomonas sp. (18 strains), Bacillus sp. (11 strains), Yers-inia sp. (7 strains), and others (9 strains) on the basis of FAMEs patterns. Eighteen strains of Pseudomonas sp. were mainly divided into three cluster in the dendrogram and only the two clusters of them showed pathogenicity to onions. CMCase and PGase activities of Pseudomonas sp. weaker than those of Bacillus sp.. However, the pathogenicity of pseudomonas sp. to soften onions was stronger than that of Bacillus sp. Inoculation of $10^{2}$ cfu of Pseudomonas sp. gives rise to softening of onions. Pseudomonas sp. was identified as Pseudomonas gladioli by biochemical and physiological characteristics. P. gladioli is the first reported bacterium as a pathogen of onion in Korea. In low temperature, P. gladioli showed better growth and higher PGase activity than those of Bacillus sp. identified as Bacillus subtilis. And pH 9.0 is optimal pH for PGase activity of B. subtilis while that of P. gladioli is pH $5.0\∼6.0$ which is the acidity of onions. Taken together, P. gladioli may be a main pathogene of onion rot during the cold storage condition.

The involvement of the cell-wall degrading enzymes in Rhizobium has long been an unsolved question about the infection process in the formation of root nodule. To assess the contribution of the cellulase to the nodulation of rhzobia, here we report the production of cellulase from R. meliloti TAL1372 which degrade carboxymethylcellulose (CMC) model substrate with CMC-plate method. We constructed a genomic library by cloning Sau3A-digested genomic DNA from R. meliloti TAL1372 into the BamHI site of the cosmid vector pLAFR3. Out of more than one thousand transductants of E. coli, one clone (pRC8-71) had CM-cellulase activity and contained pLAFR3 cosmid with 30 kb insert of R. meliloti DNA The product of CM-cellulase gene was analyzed by native PAGE. About 45 kD protein was considered to be a product of the gene. Tn5 mutagenesis reveals that the structural gene located in a ca. 3 kb KpnI fragment. The cellulase-minus mutants of R. meliloti TAL1372 were obtained by Tn5 mutagenesis of pRC8-71 and marker exchange techniques. Analyses of the nodulation ability of these Tn5 mutants showed that the CM-cellulase gene of R. meliloti TAL1372 may be involved in early nodulation development on alfalfa (Medicago satiua).

Objective: The effects of Pleurotus ostreatus on the feed utilization of broad bean stalks (BBS), rape straw (RS), paddy straw (PS), and corn stalk (CS) was examined. Methods: The four roughages were co-cultured with Pleurotus ostreatus. The chemical composition; enzyme activities of laccase, carboxymethylcellulase (CMCase) and xylanase; carbohydrate and protein fractions (based on The Cornell Net Carbohydrate and Protein System [CNCPS]) were assessed at different days after inoculation (7, 14, 21, 28 d) and un-inoculated roughages (control, 0 d). The digestibility of nutrient components and the gas production of roughage with various incubation times were monitored at 0, 2, 4, 6, 9, 12, 24, 36, 48, 60, and 72 h using an in vitro ruminal fermentation method. Results: A higher CMCase activity (0.1039 U/mL) and earlier time to peak (14 d) were detected in Pleurotus ostreatus cultured with CS (p<0.05). Significantly, the incubation length-dependent responses of cumulative gas production were observed from 24 to 72 hours post fermentation (p<0.05), and these incubation length-dependent effects on cumulative gas production of PS and CS appeared earlier (24 h) for PS and CS than those (48 h) for BBS and RS (p<0.05). The fast-degradable carbohydrate (CA) content for all four roughages significantly increased over time (p<0.05). Nonetheless, increased degradation efficiency for CA treated with Pleurotus ostreatus was detected at both 21 and 28 days of incubation (p<0.05). With the exception of PS (p<0.05), there were no significant difference among the roughages (p>0.05) in slowly-degradable carbohydrate (CB2) at different incubation times (p<0.05). Conclusion: Assessment of the alterations in chemical composition, CNCPS system fractions, and the fermentation kinetics after biological pretreatment may yield a valuable database for evaluating the biological pretreatment of Pleurotus ostreatus in ruminant feed.
In order to know the effects of Garlic, Scallion, Flavonoid, Urushiol, Anthocyanidin and Bio-MOS?? on pathogenic microbes and rumen anaerobic microbes, the growth rate of pathogens (including Escherichia coli O157, Salmonella paratyphi, Listeria monocytogenes and Staphylococcus aureus) and in vitro rumen microbial growth, gas production, ammonia concentration, carboxymethylcellulase(CMCase) activity, and microbial populations were investigated.The growth of pathogens was inhibited by supplementation of 0.1% Flavonoid, Scallion or Bio-MOS?? as biological active materials. And Scallion and Flavonoid had powerful antimicrobial properties on the pathogens applied in paper disc method.Although few effects by biological active materials disappeared in rumen fermentation in vitro, CMCase activity removed with supplementation of 1% of Flavonoid which had antimicrobial property in paper disc method. Scallion, having powerful antimicrobial property on pathogens and no inhibiting on rumen fermentation, might be a source in development of natural antimicrobial agent for ruminants.
$\beta$-Glucosidase of Cellulomonas sp. CS1-1 in cellular compartment was localized with cell-bound form while Avicelase and carboxymethylcellulase (CMCase) were appeared with extracellular enzyme. Cell growth on cellulose or CMC minimal broth was increased by glucose addition. $\beta$-Glucosidase production on cellobiose or CMC minimal broth was repressed by the addition of glucose. However, on CMC minimal broth, the enzyme production was specially stimulated by cellobiose addition. $\beta$-Glucosidase production was also induced by CMC, starcth and maltose compared with glycerol, arabinose, xylose and trehalose. From the above results, it was concluded that glucose effect on $\beta$-glucosidase biosynthesis showed catabolite repression, but enzyme production was induced by cellobiose, CMC, and starch, indicating that $\beta$-glucosidase is inducible enzyme. Yeast extract stimulated $\beta$-glucosidase production more than peptone and ammonium sulfate. $\beta$-Glucosidase activity was increased with 50mM MgCl$_2$in 10mM potassium phosphate buffer (pH 7.0). Optimum conditions for enzyme activities were pH 6.0 and 42$^{\circ}C$, Km value of $\beta$-glucosidase for p-nitrophenyl-$\beta$-D-glucosidase was 0.256mM and Ki for $\beta$-D(+)-glucose was 9.0mM.
This study was conducted to investigate the effects of supplementation of synbiotics manufactured with anaerobic bacteria, yeast and mold on preservation of total mixed ration (TMR) by exposing days. Eight treatments were composed of untreated synbiotics(US), bacterial synbiotics (BS), yeasty synbiotics (YS), moldy synbiotics (MS), bacterial and mouldy synbiotics (BMS), yeasty and moldy synbiotics (YMS), bacterial and yeasty synbiotics (BYS), and bacterial, yeasty and moldy synbiotics (BYMS). After 7 days of anaerobic fermentation, fermented-TMRs were exposed to the air during 1, 3, 5, 7, 14 and 21 days. One hundred forty four (8 treatments${\times}$6 days${\times}$3 replications) fermented-TMRs were manufactured by vinyl bag ($43\;cm{\times}58\;cm$). Although no significant differences in the activities of carboxymethylcellulase, xylanase and amylase were observed among treatments, theirs acivities were seemed to increase by treatment of BYS or YMS containing yeast. Total bacterial and mold counts also decreased in the treatments containing yeast. Potential pathogenic bacteria were less detected in BYS and BMYS for E. coli, BMYS and YS for Salmonella, and BMS and BMYS for Shigella than those of the other treatments, MS was, however, contaminated easier than US by pathogenic bacteria. From above results, synbiotics containing facultative anaerobic yeast have effects for preservation of TMR fermented anaerobically. Particularly, BMYS treatment having good results in nutrient contents, dry matter loss and pathogenic bacteria amounts was a resonable synbiotics for preservation of the fermented-TMR.
This study was conducted to determine effects of supplementation levels of aqueous direct-fed microbials (DFM; Bacillus spp.) to TMR(exp. 1.) and aqueous DFM addition under the various ratios of starch and cellulose(exp. 2.) on ruminal fermentation and fibrolytic enzyme activity. In experiment 1, ruminal fluids taken from rumen-cannulated Holstein cows were incubated during 24 hr by using TMR as substrates. Aqueous DFM was applied at a rate of 0, 0.025 and 0.05%, respectively. The pH of 0.025% treatment was not significantly different from that of control at 6 and 9 hr, but it was significantly lower (P<0.05) than 0.05% treatment. Concentrations of ammonia-N and VFAs were not affected by supplementing aqueous DFM. The A:P ratio of 0.05% treatment was significantly increased(P<0.05) by supplementation of aqueous DFM as compared with that of control at 24 hr. Although overall fibrolytic enzyme activities were not significantly affected by supplementing aqueous DFM, CMCase(carboxymethylcellulase) activity showed significant increase(P<0.05) compared to control at 6hr. However, the xylanase activity of 0.05% treatment significantly decreased(P<0.05) at 12 hr due to the application of aqueous DFM. There was no significant difference for in vitro dry matter disappearance among treatments. In experiment 2, ruminal fluids were incubated under the condition of various ratios of starch to cellulose(90:10, 70:30, 50:50, 30:70 and 10:90) with or without aqueous DFM(0.025%). Ruminal pH was unaffected by the addition of aqueous DFM, however, as increased level of starch, ruminal pH partially showed significant decrease(P<0.05). Ammonia-N concentration was not affected by aqueous DFM and ratio of starch and cellulose. On 9 hr incubation, DFM addition at a ratio of 70:30 showed significantly (P<0.05) lower value of ammonia-N(35.65 mg/dL) than that(65.05 mg/dL) of control. Concentrations of VFAs were significantly increased(P<0.05) by aqueous DFM addition compared with control at the same ratio on 6 hr incubation. The overall CMCase activity was not affected by aqueous DFM addition. However, the xylanase activity by aqueous DFM partially showed significant differences at the ratios of 90:10, 30:70 and 10:90. Our results indicated that supplementation of aqueous DFM did not significantly improve in vitro fermentation and fibrolytic enzyme activity. In addition, the DFM utilized in this study did not show consistent results by having various effects on ruminal fermentation under different feeding regimens.
