The Screening of Hepatic Functional Improvement, Liver Protection and Antifibrotic Effect from Dried Extracts of Concha Cipangopaludinae in Rats (랫드에서 전라 추출물의 간기능 개선, 간보호 및 항섬유화 효과 검색)

  • Oxidative stress and its consequent lipid peroxidation exert harmful effects, which have been currently involved in the generation of carbon tetrachloride (CCl₄)-induced fibrosis(cirrhosis). In this study, it was investigated whether dried extract of 田螺(Concha Cipangopaludinae; CC) has liver functional improvement, antioxidative and antifibrotic effect in rats those were induced liver fibrosis by CCl₄ administration. The female Sprague-Dawley rats were divided into 3 groups(Normal, AC, AC-CC) and were observed in 6 weeks. Except for normal group, liver fibrosis(cirrhosis) in rats were developed by CCl₄ administration(0.8 ㎖/rat/week). And the rats were treated with prepared CC(p. o. 2 ㎖/day/rat). At the time of sacrifice, the liver, kidney and spleen were weighed and the ratio of organ weight/body weight was calculated. The MDA, hyp and biochemical parameters(AST, ALT, ALP, t-bili) were measured in sera and liver tissue of rats. The strong yellow color of urine was observed in all CCl₄-treated group compared with normal group, but jaundice didn't appear in CCl₄-treated group. The mortality of CCl₄-treated group is very low(<13%) during 6 weeks of observation time. The ratio of liver/body as well as the weight of liver in CCl₄-treated rats significantly increased compared with that in normal group(p<0.001). The level of clinical parameters in sera of all liver fibrosis(cirrhosis) developed rats were significantly higher than that of normal group(p<0.001-0.05). Especially the value of BUN, ALP, t-bilirubin in AC-CC group showed 20.9%, 19.6%, 47.9% lower than that in AC group. The content of hyp in CCl₄-treated rats was significantly higher than normal group(p<0.001~<0.05), and showed 12.2% lower value in the AC-CC group than AC group(p<0.05). The production of lipid peroxidation(MDA) in sera and liver tissue significantly increased under the fibrotic(cirrhotic) condition(p<0.001~<0.05). Especially the MDA value of AC-CC group in sera significantly 46.5% decreased compared with that of AC group(p<0.05), and the MDA value of AC-CC in liver tissue showed 21.4% lower than that of AC group. Concha Cipangopaludinae can be improved hepatic function, and maybe have effect of liver protection, antioxidation and antifibrosis.

The Thermodynamics of the Formation of Pyridine-Halogen and Interhalogens Charge Transfer Complexes (피리딘과 할로겐 또는 할로겐간 화합물 사이의 전하이동 착물생성에 관한 열역학적 연구)

  • Ultraviolet spectrophotometric investigations were carried out on the systems of pyridine with iodine, iodine monobromide and iodine monochloride in carbon tetrachloride. The results reveal the formation of the one to one molecular complexes of the type, $C_5H_5N{\cdot}I_2$, $ C_5H_5$N{\cdot}IBr and $ C_5H_5N{\cdot}ICl$. Considering ${\lambda}_max$ according to the formation of charge transfer complexes has the blue shift with the increasing temperatures$25, 40, 60^{\circ}C$ the equilibrium constants K and molar absorptivities $\varepsilon$ of complexes were obtained. From these values, the thermodynamic parameters ${\Delta}H$, ${\Delta}G$ and ${\Delta}S$ for the formation of the above charge transfer complexes were obtained. These results indicate that the relative stabilities of iodine, iodine monobromide and iodine monochloride complexes with pyridine increase in the order, $ C_5H_5N{\cdot}I_2$ < $ C_5H_5N{\cdot}IBr$ <$ C_5H_5N{\cdot}ICl$. This may be a measure of relative acidity of halogen and interhalogen toward pyridine and can be explained by the polarizabilities of electron acceptors and the difference of electronegativities of halogen atoms.

Effects of Fructus Foeniculi extract on recovering liver function (회향의 간장기능 회복효과)

  • Objectives : Effects of Fructus Foeniculi extract on liver function were investigated in carbon tetrachloride(CCl4) intoxicated rats. Methods : Thirty two male Sprague-Dawley rats with mean weight of $227.28{\pm}7.92g$ were used in these experiments and housed with food and water ad libitum. Fructus Foeniculi extract was administerd at dose 100mg/kg/day and 200mg/kg/day p.o. for 2 weeks after that CCl4 was treated 3 times at dose of 2.5ml/kg, p.o. in alternate day basis. Then serum AFP(${\alpha}$-Fetoprotein), Total protein, Albumin, Triglyceride, Total cholesterol concentrations and ALP (Alkaline phosphatase), AST(Aspartate Aminotransferase), ALT(Alanine Aminotransferase), ${\gamma}$-GT( ${\gamma}$-Glutamyl transferase), LDH(Lactate Dehydrogenase) activities were determined with commercial kit by autoanalyzer. Results : Plasma ${\alpha}$-fetoprotein and total protein concentration showed a tendency to decrease in Fructus Foeniculi extract-treated groups. However, plasma albumin concentration showed no significant differences in all treatment groups. Activity of plasma aspartate aminotransferase and alanine aminotransferase in Fructus Foeniculi extract-treated groups showed a lower value than that of control group. Alkaline phosphatase and lactate dehydrogenase activities showed a tendency to decrease in Fructus Foeniculi treated groups. However, ${\gamma}$-glutamyl transferase activity showed no significant difference in all treated groups. Concentration of plasma triglyceride and total cholesterol showed a high level in CCl4 intoxicated rats but not in Fructus Foeniculi treated groups. Conclusion : Reviewing these experimental results, it appears that Fructus Foeniculi extract have recovering effect against liver injury.

Effects of Herbal-Acupuncture with Inzinsammultang on the Recovery of Liver Injury in Rats (인진삼물탕(茵蔯三物湯) 약침처리(藥針處理)가 백서(白鼠)의 간손상(肝損傷) 회부(回復)에 미치는 영향(影響))

  • These studies were carried out to investigate the effect of Herbal-acupuncture with Inzinsammultang extract on the recovery from liver injury of rats. The liver injury of rats induced with 0.3ml/ea carbon tetrachloride. The Herbal-acupunture with Inzinsam multang extract solution inserted into corresponding focus of Kansu(BL18) in rats. In this study, SD-Rats(Sprague-Dawley) were divided into 4 groups ; Normal group (None treated group), Control-group(The group not treated after $CCI_4$-intoxication), Treated group; Example I-group(Saline-injected groups after $CCl_4$-intoxication) and Example II-group(Inzinsammultang-injected groups after $CCl_4$-intoxication). Through histological observation, Example II-group sho-ws that liver injury is weaker than Control group.(p<0.05) Biochemical assays for each serum enzyme activities of AST, ALT and LDH, levels of albumin, ${\gamma}$-GT, TG, total cholesterol were performed. The results were summarized as follows 1. AST(Aspartate aminotransferase) activity in serum significantly decreased in the Inzinsammultang-injected groups after $CCl_4$-intoxication. Inzinsammultang-injected groups after $CCl_4$-intoxication showed significantly higher AST activities, compared with the Saline-injected groups after $CCl_4$-intoxication.(p<0.05) 2. ALT(Alanine aminotransferase) activity in serum significantly decreased in the Inzinsammultang-injected groups after $CCl_4$-intoxication. The Inzinsammultang-injected groups after $CCl_4$-intoxication showed significantly higher AST activities, compared with the Saline-injected groups after $CCl_4$-intoxication(p<0.05) 3. Level of Albumin in serum significantly increased in the Inzinsarnmultang-injected groups after $CCl_4$-intoxication. 4. LDH(Lactate dehydrogenase) activities in serum significantly decreased in the Inzinsammultang-injected groups after $CCl_4$-intoxication.(p<0.05) 5. ${\gamma}$-GT(Glutamyl transferase) in serum significantly decreased in the Inzinsammultang-injected groups after $CCl_4$-intoxication(p<0.05) 6. As for the TG(Triglyceride) levels in serum, Inzinsammultang-injected groups after $CCl_4$-intoxication is no significant differences compared with the Saline-injected groups after $CCl_4$-intoxication.(p<0.05) 7. Total cholesterol in serum significantly increased in the Inzinsarnmultang-injected groups after $CCl_4$-intoxication.(p<0.05) Those results indicate that the Inzinsammultang Herbal-acupuncture have significant effects on the liver injury induced by $CCl_4$. So it is expected that the Inzinsammultang Herbal-acupuncture can be used to cure inflammations and recover the functions of damaged liver cells.

Effects of Rhynchosia molubilis Saponin on Hepatotoxicity and Pathology (간독성과 병리학적인 면에서 노두 사포닌의 효과)

  • This study was carried out to investigate the effects of Rhynchosia molubilis saponin on carbon tetrachloride ($CCl_4$)-induced hepatotoxicity. Sprague-Dawley rats were intraperitoneally administered the Rhynchosia molubilis saponin at 100 mg/kg every day for two weeks, then $CCl_4$ (3.3 ml/kg) was injected into rats. 12 hours later, they were anesthesized with ether and dissected. Rhynchosia molubilis saponin-administered group showed 59.92% and 62.28% of inhibitory effects on aspartate aminotransferase (AST) and alanine aminotransferase (ALT) activities compared to $CCl_4$-treated group (p<0.05). Malonedialdehyde (MDA) levels of Rhynchosia molubilis Saponin-administered and $CCl_4$-treated (RSC) group in liver homogenate and mitochondria were significantly inhibited to 61.83%, 81.11 %, respectively, compared to $CCl_4$-treated group (p<0.05). Superoxide dismutase (SOD) activities of RSC group in liver homogenate and mitochondria were significantly inhibited to 66.53%, 31.04%, respectively, compared to $CCl_4$-treated group (p<0.05). GPx activities of RSC group in liver homogenate and mitochondria were significantly inhibited to 72.74%, 72.68%, respectively, compared to $CCl_4$-treated group (p<0.05). The histological examinations showed that the liver cell necrosis and centrilobular congestion aggregation induced by $CCl_4$ were dearly eliminated by the administration of Rhynchosia molubilis saponin. These results suggest that Rhynchosia molubilis saponin could have the protective effects against hepatotoxicity.

Protective Effects of Angelica keiskei Extracts Against D-Galactosamine (GalN)-induced Hepatotoxicity in Rats

  • Although the vegetable Angelica keiskei (AK) has widely been utilized for the purpose of general health improvement among Korean population, its functionalities are not very well defined. In this study, we examined the effects of methanol extract of AK in rats on the biochemical changes induced by two hepatotoxins, D-galactosamine (GalN) and carbon tetrachloride ($CCl_4$). AK was orally administered once daily for 7 days to male rats at 200 and 500 mg/kg, before hepatotoxins. Effects of AK were assessed 24 hr later. AK pretreatments at 200 and 500 mg/kg significantly blunted GalN-induced elevation in liver lipid peroxidation, plasma aspartate-transaminase (AST) and alanine-transaminase (ALT) activities. AK also prevented, after 500 mg/kg but not after 200 mg/kg, the GalN-induced elevation in triglyceride, total cholesterol and LDL-cholesterol levels. Differently from against GalN-induced toxicity, AK did further elevate the $CCl_4$-induced rise in AST, ALT and lipid peroxidation. These results suggest that AK, when pre-administered prior to GalN, exerted protective effects against GalN-induced hepatotoxicity, in contrast however, AK exacerbated that induced by $CCl_4$. To explore possible mechanism for the toxicity-potentiating effects of AK on $CCl_4$, the activity of hepatic drug metabolism after AK treatment was assessed. It was observed that AK increased the activity of aniline hydroxaylase, a cytochrome P450 isoenzyme responsible for metabolic activation of $CCl_4$. This finding suggests that hepatoprotective effects of AK are not equally expected depending on hepatotoxins employed.

  • The level of plasticizer such as diethyl phthalate (DEP), di-n-propyl phthalate (DprP), di-n-butyl phthalate (DBP), di-n-pentyl phthalate (DPP), butylbenzyl phthalate (BBP), di-(2-ethylhexyl) phthalate (DEHP), dicyclohexyl phthalate (DCHP) and di-(2-ethylhexyl) adipate (DEHA) was determined in disposable sanitary gloves. Samples of disposable sanitary gloves were taken at retail shops, and their overall extractions in carbon tetrachloride were measured, after establishment of soxhlet appratus in 6 hours. All of 8 samples of disposable sanitary gloves at retail shops were polyethylene (PE). All of the manufactures was voluntarily labelled their boxes of gloves in line Korea Food Code. The level of phthalate esters and DEHA in all of 8 samples were not exceed the detection limit. The detection limits of DEP, DprP, DBP, DPP, BBP, DEHP, DCHP and DEHA were 1.8, 1.9, 1.3, 1.1, 0.9, 0.7, 2.1 and 0.8 mg/L, respectively.

Protective Effects of Plant Extracts on the Hepatocytes of Rat Treated with Carbon Tetrachloride (사염화탄소를 투여한 흰쥐의 간세포에 대한 식물 추출물들의 보호효과)

  • To investigate the effects of plant extracts on the protection against liver damage by $CCl_4$ in rat, two kinds of experiment were performed, firstly by the primary hepatocyte culture and secondly by the animal feeding. The primary hepatocyte culture with the extracts of pine leaf, soybean sprout and mugwort showed significantly low activities (p<0.01∼0.05) of aspartate aminotransferase (AST) and alanine aminotransferase (ALT), indicating an excellent protective effect against liver damage by $CCl_4$. In the second experiment, the microsomal malondialdehyde (MDA) contents of the above same groups were also significantly lower (p<0.01) than the $CCl_4$-treated group without plant extracts, but shiitake showed less effect. Among four kinds of plant extracts, extracts of pine leaf and mugwort showed also much higher activities of the microsomal cytochrome P-450 in comparison to soybean sprout and shiitake. In the test of xanthine oxidase (XOD) activity, all of three groups except shiitake showed significantly low activities (p<0.01). These consistent results in vitro and in vivo suggest that the extracts of pine leaf, soybean sprout and mugwort may have strong protective effects against liver damage induced by the potential toxicants such as $CCl_4$.

Effect of Green Tea Catechin on Acute Hepatotoxicity in Rats (랫트의 간 손상에 대한 녹차카테킨의 보호 및 치료효과)

  • Green tea catechin (GTC) is known to have a wide variety of pharmacological activites. In the present study, the effects of GTC on acute hepatotoxicity induced by carbon tetrachloride ($CCl_4$) and galactosamine were examined in rats. Two doses (50 or 100 mg/kg) of GTC were administered to rats orally for 3 days befor or after the induction of hepatotoxicity. A hepatotoxicity was induced by the inpraperitoneal injection of the $CCl_4$ (0.5 ml/kg) or galactosamine (400 mg/kg). GTC(50 mg/kg) reduced the aspartate aminotransferase (AST) and alanine aminotransferase (ALT) level of the $CCl_4$-intoxicated rats in the pre-treatment group (from 262${\pm}$11, 80${\pm}$19 to 153${\pm}$22, 55${\pm}$25), and also in the post-treatment group (from 156${\pm}$40, 105${\pm}$3 to 106${\pm}$22, 55${\pm}$9), respectively. And GTC (50 mg/kg) also reduced the levels of AST and ALT in both pre-treatment (from 576${\pm}$24, 276${\pm}$68 to 236${\pm}$13, 115${\pm}$13) and post-treatment (from 233${\pm}$54, 137${\pm}$11 to 119${\pm}$23, 44${\pm}$17) when induced by galactosamine. GTC also showed the inhibition of pathogenesis of hepatocyte of $CCl_{4^-}$ and galatosamine-intoxicated rat. These results suggest that green tea catechin (GTC) may be useful fur the prevention and therapy of hepatotoxic pathogenesis.

Therapeutic Effects of Hovenia Dulcis Thunb Extract on $CCl_4$ Induced Liver and Kidney Damage in Rats (Carbon Tetrachloride로 유발된 Rat의 간장과 신장 손상시 헛개나무 추출액의 치료효과)

  • Hovenia dulcis Thunb (HDT) has been known folk medicine and has been used as therapeutic drug in the treatment of liver disease. Also it has been used as a detoxifying agents for alcoholic poisoning and promoting diuresis. However, there has not been any study on therapeutic effect of Hovenia dulcis extract on $CCl_4$ induced liver and kidney damage in rats. In this study, we report on therapeutic effects of Hovenia dulcis extract on $CCl_4$ induced liver and kidney damage in rats. Rats were divided into four groups of eighteen animals. Control group (DW) was administrated with distilled water 2.5 mL/kg per peritonial administration and then $CCl_4$ group (CCl) was administrated $CCl_4$ 2.5 mL/kg per peritonial administration, $CCl_4$+HDT extract group ($CCl_4$+HDT) was administrated HDT extrat (100 mg/kg) after $CCl_4$ 2.5 mL/kg administration, $CCl_4$+Silymarin group ($CCl_4$+Sily) was administrated Silymarin (50 mg/kg) after $CCl_4$ 2.5 mL/kg administration. The complete blood cell (CBC) count of RBC, WBC, PCV, Hb, MCH, MCV, MCHC and blood chemistry profile of AST, ALT, GGT, ALP, Total choloesterol, Tryglyceride, Total bilirubin, Amylase, Glucose, BUN, Creatinine, Lipase and pathologic changes were observed for 7 days after administration of D.W., $CCl_4$, $CCl_4$+HDT extract, $CCl_4$+Silymarin. The results are as follows : 1. RBC and PCV were significantly (p < 0.01) increased in all groups compared to D.W. but hemoglobin, MCH, MCV and MCHC were not showed significant difference during experimental periods. 2. AST, ALT, T-cholesterol, T-bilirubin, TG were significantly (p < 0.05) increased in all groups on day 3 compared to D.W. and were normal on day 7. 3. ALP was significantly (p < 0.05) decreased in $CCl_4$+HDT group on day 3 but Amylase was not showed significant difference during experimental periods. 4. BUN was significantly (p < 0.05) increased in $CCl_4$ group on day 7, but $CCl_4$+HDT group and $CCl_4$+Sily group were normal. Creatninie was significantly (p < 0.05) increased in $CCl_4$ group on day 3 and normal on day 7 but $CCl_4$+HDT group and $CCl_4$+Sily group were not showed significant difference during experimental periods.
