The purpose of this study is to set up the analytical method of new persistent organic pollutants (POPs) such as chlordecone, endosulfan, ${\alpha}$-HCH, ${\beta}$-HCH, ${\gamma}$-HCH. The analytical methods for these compounds listed as new POPs by the Stockholm Convention need to be newly established. Therefore, we proposed the analytical method for 5 organic chlorinated pesticides (OCPs) and then applied the analytical method to environmental samples. To do this, the pre-treatment such as florisil and activated carbon cleanup process in the Korean official method for classic POPs had been reviewed. All of compounds except chlordecone were pre-treated simultaneously with reviewed cleanup process and detected by GC/MS and HRGC/HRMS respectively. There is a problem that chlordecone could not get a high sensitivity by GC analysis, but in this study GC/MS method was proposed.
Pt/C powder which was used as electrocatalyst in a Phosphoric Acid Fuel Cell(PAFC) was fabricated by colloid method. It was reported that the sulfur from reductant, $Na_2S_2O_4$, worked as a poison against catalyst during long term operation. To remove these sulfurs, we try to treat Pt/C powder by three different methods. First, we tried to remove the sulfur according to temperature and time in $H_2$ atmosphere. As the heat treatment temperature is raised up, the effect of the removal is increased but the electrode performance is decreased because of the growth of Pt particle size. The optimal heat treatment temperature is $400^{\circ}C$, the size of Pt particle is approximately $35{\sim}40{\AA}$ and the electrode performance is $360mA/cm^2$ at 0.7 V. At $400^{\circ}C$, even though the time of heat treatment is extended, size of Pt, amounts of remaining sulfur and electrode performance is almost constant. Secondly, when we removed in a crucible at $900^{\circ}C$ the removal of the sulfur was not better, but the size of Pt particle, approximately $80{\AA}$, was smaller than that of heat treatment in $H_2$ atmosphere at $900^{\circ}C$. Lastly we treated with solvents such as acetone, benzene, and carbon disulfide. It was observed that sulfur components were removed partly by extraction with solvents, the electrode performances were similar each other.

Korean regional network of tower flux sites, KoFlux, has been initiated to better understand $CO_2$, water and energy exchange between ecosystems and the atmosphere, and to contribute to regional, continental, and global observation networks such as FLUXNET and CEOP. Due to heterogeneous surface characteristics, most of KoFlux towers are located in non-ideal sites. In order to quantify carbon and energy exchange and to scale them up from plot scales to a region scale, applications of various methods combining measurement and modeling are needed. In an attempt to infer regional-scale flux, four methods (i.e., tower flux, convective boundary layer (CBL) budget method, MM5 mesoscale model, and NCAR/NCEP reanalysis data) were employed to estimate sensible heat flux representing different surface areas. Our preliminary results showed that (1) sensible heat flux from the tower in Haenam farmland revealed heterogeneous surface characteristics of the site; (2) sensible heat flux from CBL method was sensitive to the estimation of advection; and (3) MM5 mesoscale model produced regional fluxes that were comparable to tower fluxes. In view of the spatial heterogeneity of the site and inherent differences in spatial scale between the methods, however, the spatial representativeness of tower flux need to be quantified based on footprint climatology, geographic information system, and the patch scale analysis of satellite images of the study site.
In this study, we have performed several tests for composite plastic materials to be applied on interior materials of a vehicle to identify their combustion characteristics using cone calorimeter, smoke density chamber and toxicity index chamber. We have prepared a total of 12 samples for 4 major parts of a vehicle wherein each major part has 3 different materials. The results of cone calorimeter test showed ignition time of PVC sheet and PVC leather were 2s. The 8 samples showed under less than 10s of ignition time. The sample comprising Nylon and PE had the biggest maximum heat release rate of 635 $kW/m^2$. The sample comprising Rubber showed the smallest maximum heat release rate but with the biggest total heat release. The results of smoke density chamber test showed the sample that is made up with Rubber had the biggest specific optical smoke density. The sample comprising PVC leather and PUR showed the biggest VOF4 which enables the initial smoke production. The results of toxicity index test showed that all samples contained carbon dioxide content exceeding its lethal concentration. The sample comprising PVC showed high content of hydrogen chloride and hydrogen bromide. The PVC sheet showed the biggest toxicity index calculated by using lethal concentration and test results. Toxicity index of all sample wes over 1.
Bacterial colonies which were able to oxidize the manganese were isolated from six soil samples in Byungchon area. Among them, one bacterial strain was selected for this study based on its high manganese oxidation activity. This selected bacterial strain was identified as Pseudomonas sp. MN5 through physiological-biochemical test and analysis of its 16s rRNA sequence. This selected bacterial strain was able to utilize fructose and maltose, but they doesn't utilizing various carbohydrates as a sole carbon source. Pseudomonas sp. MN5 showed a very sensitive to antibiotics such as kanamycin, chloramphenicol, streptomycin and tetracycline, but a high resistance up to mg/ml unit to heavy metals such as lithium, manganese and barium. Optimal manganese oxidation condition of Pseudomonas sp. MN5 was pH 7.5 and manganese oxidation activity was inhibited by proteinase K and boiling treatment. The manganese oxidizing protein produced by Pseudomonas sp. MN5 was purified by ammonium sulfate precipitation, HiTrap Q FF anion exchange chromatography and G3000sw $_{XL}$ gel filtration chromatography. By sodium dodecyl sulfate polyacrylamide gel electrophoresis, three manganese oxidizing protein with estimated molecular weights of 15 kDa, 46.7 kDa and 63.5 kDa were detected. Also, it was estimated that manganese oxidizing protein produced by Pseudomonas sp. MN5 were a kind of porin proteins through internal sequence and N-terminal sequence analysis.

The ability of probiotics to adhere to the intestinal epithelium likely plays an important role in their colonization of the gastrointestinal tract. Here, we performed high-throughput screening (HTS) for suitable characteristics of potential probiotic bacteria using attachment and colonization ability through a C. elegans surrogate in vivo model. A total of 100 strains of lactic acid bacteria (LAB) isolated from infant feces were subjected to the colonization assay using C. elegans intestine. Based on colonization ability, we showed that nine isolates have a high attachment ability during whole experimental periods (up to 168 h), compared to Lactobacillus rhamnosus strain GG as a control. Also, through the use of an in vitro cell attachment model, nine isolates revealed highly binding activity to the mucus layer. Next, the selected 9 isolates were assayed for their survival ability when exposed to acidic and bile conditions as well as cholesterol reduction and the utilization of prebiotic substrates. As a result, the isolated nine strains were determined to be highly resistant to acid and bile conditions. In addition, they have significant activity for the reduction of cholesterol and utilization of several prebiotic substrates as a carbon source. Finally, the selected nine strains were identified by either L. rhamnosus or L. plantarum (4 strains for L. rhamnosus and 5 strains for L. plantarum, respectively). Taken together, we propose that the direct colonization of probiotics using C. elegans may be applicable to the rapid screening of valuable probiotic strains in vivo.
In order to utilize the agricultural waste products, two strains of mold producing powerful cellulolytic enzyme were sereened from various soils, composts, rotten wood and others. The optimum condition of cellulase production was studied. The results obtained were summarized as follows. 1. Two strains of mold which showed remarkably high cellulolytic activity were identified as Aspergillus niger-SM 6 and Trichoderma viride-SM 10. 2. The highest cellulase production was obtained at pH 5.0-6.0 in 5 days. 3. Cellulase production in strain Aspergillus niger-SM 6 increased with the addition of C.M.C., $(NH_4)_2SO_4$, C.S.L., orange peel powder and rice hull. The rice hull, treated with 3N NaOH at $120^{\circ}C$ for 15 min. and neutralized with various acids, was used. Up to 50% of wheat bran could be substituted by the treated rice hull without any decrease of cellulase activity. 4. In the strain of Trichoderma viride-SM 10, cellulase production increased with the addition of C.M.C., $NH_4NO_3$, Vitamin-free casamino acid and orange peel powder, while the other carbon, nitrogen, phosphate sources, natural nutrients and organic substances gave no remarkable effect.

Biodegradation of the C. I. Reactive Blue 114 was investigated in an upflow anaerobic sludge blanket (UASB) reactor. Important parameters studied include dye concentration, the kind and concentration of carbon source, hydraulic retention time (HRT), and influent pH. Glucose was found to be a better co-substrate than the mixture of volatile fatty acids (VFAs), although its concentration did not affect dye removal efficiency in the range of $1000{\sim}3000mg/{\ell}$. When HRT increased from 6 hr to 24 hr, dye removal efficiency increased up to 12 hr and remained almost constant thereafter at about 40%. When influent pH was varied in the range of 6.0~8.0, the effluent pH was varied in the range of 6.8~7.5 with maximum efficiency at pH 7.0. The highest dye removal rate obtained was $52mg/{\ell}{\cdot}day$, while the maximum dye load to meet the discharge limit of color intensity was estimated to be $46mg/{\ell}{\cdot}day$ at HRT of 12 hr and an influent glucose concentration of $2200mg/{\ell}$.

An integrated database(DB) system was developed to manage the marine geological data and geophysical data acquired from around the Korean peninsula from 2009 to 2013. Geological data such as size analysis data, columnar section images, X-ray images, heavy metal data, and organic carbon data of sediment samples, were collected in the form of text files, excel files, PDF files and image files. Geophysical data such as seismic data, magnetic data, and gravity data were gathered in the form of SEG-Y binary files, image files and text files. We collected scientific data from research projects funded by the Ministry of Oceans and Fisheries, data produced by domestic marine organizations, and public data provided by foreign organizations. All the collected data were validated manually and stored in the archive DB according to data processing procedures. A geographic information system was developed to manage the spatial information and provide data effectively using the map interface. Geographic information system(GIS) software was used to import the position data from text files, manipulate spatial data, and produce shape files. A GIS DB was set up using the Oracle database system and ArcGIS spatial data engine. A client/server GIS application was developed to support data search, data provision, and visualization of scientific data. It provided complex search functions and on-the-fly visualization using ChartFX and specially developed programs. The system is currently being maintained and newly collected data is added to the DB system every year.

Recently, the market of skin-care device has been steadily growing up. In this paper, we tried to develop a personal compound stimulus device more competitive than existing products. As the compound stimulus, biochemical stimulus of herbal extraction fluid, thermal stimulus of plate-shaped carbon fiber heater, and optical stimulus of near infrared LED were selected. By some evaluation tests, the thermal stimulation part and the optical stimulation part were found to be developed properly. Additionally, the efficacy of the mixed stimulus of thermal and optical stimulation was tested in C2C12 mouse myoblast. Through RT-PCR analysis, it was found that, by the developed compound stimulus, the expression of collagen I mRNA and collagen III mRNA increased by 4.9 and 1.3 times respectively.
