A Study on the Visible Injury of some Herbaceous Plants by $SO_2$ gas (수종(數種) 초본류(草本類)의 $SO_2$ 가스에 의한 가시피해특징(可視被害特徵)에 관(關)한 연구(硏究))

  • $SO_2$ gas injuries for 19 varieties of 10 species of plants were investigated within a environmentally controlled growth chamber. Visible injuries were observed when exposing the plants either 0, 0.2, 0.4, 0.7 or 1.5ppm of $SO_2$ gas for 5 days from 9 : 00 to 17 : 00 everyday. The results obtained were as follows: 1. Light brown spots were shown on Raphanus sativus(Chunchu, Taeback, Jinjudaepyung), Brassica campestris(Manchun, Josaengmiho, Seoul, Jeonseung), and Capsicum annum(Searona, Hongsanho, Hongilpum); brown spots on Sesamum indicum(Pungnyun, Kwangsan); reddish brown shades on Cleosia cristata; and black brown spots on Perilla frutescens, Calendula officinalis, Chrysanthemum morifolium, and Salvia splendens, respectively. 2. The spotty injuries induced by $SO_2$ gas injuries for 19 varieties of 10 species of plants were investigated within a environmentally controlled growth chamber. Brassica campestris(Jeonseung), Capsicum annum(Searona, Hongilpum), and Cleosia cristata, however, showed the injury at 1.5ppm of $SO_2$ gas concentration. 4.   • Gray leaf spot of tomato was observed on tomato plants growing in a middle of a pepper field where pepper plants were badly infected with gray leaf spot. Both Stemphylium solani and S. lycopersici were isolated from the lesion. Both species were highly pathogenic on tomato and pepper. This is the first report in Korea of occurrence of gray leaf spot of tomato caused by Stemphylium solani Weber.

Characterization of A cDNA encoding A Novel Phenazine Compound in Hot Pepper

  • From the PMMV (pepper mild mottle virus)-inducible ESTs differentially expressed in Capsicum chinense PI257284, we isolated a full-length cDNA (CcPHZF: Capsicum chinense phenazine), encoding a phenazine biosynthesis protein which catalyzes the hydroxylation of phenozine-1-carboxylic acid to 2-hydroxyphenazine-1-carboxylic acid. Phenazine compound has been known to exhibit broad-spectrum of antibiotic activity against various species of bacteria and fungus. The entire region of CcPHZF is 879 bp in length and the open reading frame predicted a polypeptide of 292 amino acids. The homolog of CcPHZF is not Present in database except clones of AC004044 and NM100203 from Arabidopsis with 58 and 59%, respectively. Genomic Southern analysis indicated that the pepper genome contains a single copy of CcPHZF. The CcPHZF was strongly induced in the pepper leaves 3 days after PMMV treatment, when HR occurs on the leaf surface. Characterization of CcPHZF is underway to investigate if the CcPHZF is related to disease resistance against pathogens.

  • We surveyed on the host plants of Fungus gnat, Bradysia app. and found 21 species in the greenhouse and field. These are as follows: Cucumis sativus L., Cucumis melo L., Citrullus lanatus T., Cucurbita moschata F., Lycopersicon esculentum M., Capsicum annuum L (Pepper), Capsicum annuum L (Paprika), Lillium longiflorum T., Dianthus caryophyllus L., Rosa hybrida H., Gerbera jamesonii B., Chrysanthemum morifolium R, Phalaenopsis schilleriane R., Gladiolus grandiflours H., Zingiber officinale R., Cnidium officinale M., Canavalia gladiata DC., Angelica utilis M., Polygonatum odoratum D., Pinus densiflora S., and Pinus thunbergii P. Fungus gnat larvae cause damages to the root and promote decay and wilt by feeding on the roots and burrowing in plant tissue.

Marker Development for Erect versus Pendant-Orientated Fruit in Capsicum annuum L.

  • The erect habit of fruit setting is a unique characteristic of ornamental peppers and wild pepper species. The erect habit is known to be controlled by the up locus on pepper (Capsicum annuum L.) chromosome 12. The result of a genetic analysis using Saengryeog 211 (pendant), Saengryeog 213 (erect), and their $F_1$ and $BC_1$ progeny demonstrated that up is a recessive gene. To develop an up-linked marker, bulked segregant analysis (BSA) and amplified fragment length polymorphism (AFLP) were employed using 108 $F_{2:3}$ individuals. The closest AFLP marker, $A2C7_{469}$, was located at a genetic distance of 1.7 cM from the up locus and was converted into a cleaved amplified polymorphic sequence (CAPS) marker. This marker was mapped at a genetic distance of 4.3 cM from the up locus. When the CAPS was applied to seven ornamental lines and 27 breeding lines with erect fruit, these genotypes of 28 lines were correctly predicted. Thus, the CAPS marker will be useful for marker-assisted selection (MAS) of pepper breeding lines with the up allele at the early seedling stage.

  • "Determinate" and "indeterminate" inflorescences in plants are controlled by a single recessive gene, for example, SELF-PRUNING (SP) in Solanum lycopersicum, TERMINAL FLOWER1 in Arabidopsis, CENTRORADIALIS in Antirrhinum, and CENTRORADIALIS-like gene in tobacco. Pepper (Capsicum annuum L.) is an indeterminate species in which shoots grow indefinitely. In this study, we cloned and characterized the pepper SP-like gene (CaSP). RT-PCR revealed that the CaSP transcript accumulates to higher levels in floral buds than in other organs. Comparison of genomic DNA and cDNA sequences from indeterminate and determinate pepper plants revealed the insertion of a single base in the first exon of CaSP in the determinate pepper plants. CaSP is annotated in linkage group 8 (chromosome 6) of the SNU2 pepper genetic map and showed similar synteny to SP in tomato. Transgenic tobacco plants overexpressing CaSP displayed late-flowering phenotypes similar to the phenotypes caused by overexpression of CaSP orthologs in other plants. Collectively, these results suggest that pepper CaSP is an ortholog of SP in tomato.

Effects of Organic Farming on Communities of Arbuscular Mycorrhizal Fungi

  • Red pepper (Capsicum annum L.) roots and soils representing different agricultural management practices such as conventional (CON), no-chemical (NOC), and organic farming systems (ORG) were collected from 32 farm field sites in Kyunggi, Korea to investigate the effects of these agricultural practices on arbuscular mycorrhizal (AM) symbiosis. ORG inoculum significantly increased plant growth compared to inoculum from CON and NOC. A community analysis of AM fungi (AMF) using morphological features of spores revealed that AMF spore abundance and species diversity were significantly higher in ORG than in CON. Additionally, a community analysis of AMF colonizing roots using a molecular technique revealed higher AMF diversity in ORG than in CON. These results suggest that agricultural practices significantly influence AM fungal community structure and mycorrhizal inoculum potential.

An Etiological Study on the Anthracnose Fungus of Pepper Caused by Colletotrichum dematium in Korea (고추에서 분리(分離)한 탄저병균(炭疽病菌) Colletotrichum dematium에 대한 병원학적(病原學的) 연구(硏究))

  • In order to investigate an etiological characteristics of the pepper anthracnose fungus infected with Collectotrichum dematium, this experiment was undertaken. Isolation percent of the fungus was 55.3% from the seed sample of Jewon and 0.3% from that of Eumsung Kun, according to seed health blotter method. Although the acervulus was similar to the known two Colletotricum species, C. acutatum and C. gloeosporioides, setae was not only straightly extended above the acervulus, but also shaped as falcate conidia ranging $12{\sim}31.2{\times}2.4{\sim}4.8{\mu}m$. Acervului of the fungus were evenly scattered on the surface of pepper seed coat, and caused seedling blight after seed germination. Pre-and post-emergence seedling blight as well as foliar lesion and fruit rot was confirmed by inoculating conidial suspension. No typical anthracnose sysmptom was obtained from soybean seedlings, welsch onion and spinach seedlings upon inoculation. Therefore, this fungus is said to the undescribed form species of the fungus of pepper in Korea (Colletotricum dematium f. sp. capsicum).

Agroinfiltration-based Potato Virus X Replicons to Dissect the Requirements of Viral Infection

  • Extensive research of the Potato virus X(PVX) has been performed in in vitro transcription system using the bacteriophage T7 promoter. We constructed an efficient T-DNA based binary vector, pSNU1, and modified vectors carrying PVX replicons. The suitability of the construct to transiently express PVX RNA using Agrobacterium tumefaciens was tested by analysis of infectivity in plants. The expressed PVX RNA was infectous and systemically spread in three plant species including Nicotiana benthamiana, N. tabacum cv. Xanthi-nc, and Capsicum annuum cv. Chilsungcho. The PVX full length construct, pSPVXp31, was caused severe mosaic symptoms on N. benthamiana, severe necrotic lesions on C. annuum while milder symptoms and delayed mosaic symptoms were appeared on the systemic leaves on N. tabaccum. RT-PCR analysis confirmed the presence of PVX RNAs on both inoculated and systemic leaves in all three plant species tested. Our results indicated that PVX replicons were efficiently expressed PVX RNA in at least three tested species. Further investigation win be needed to elucidate the mechanism of PVX replication, translation, movement and assembly/disassembly processes.

Capsaicin-Induced Apoptosis and Reduced Release of Reactive Oxygen Species in MBT-2 Murine Bladder Tumor Cells

  • Bladder cancer is a common cancer with high risk of recurrence and mortality. Intravesicle chemotherapy after trans-urethral resection is required to prevent tumor recurrence and progression. It has been known that antioxidants enhance the antitumor effect of bacillus Calmette-Guerin (BCG), the most effective intravesical bladder cancer treatment. Capsaicin, the major pungent ingredient in genus Capsicum, has recently been tried as an intravesical drug for overactive bladder and it has also been shown to induce apoptotic cell death in many cancer cells. In this study, we investigated the apoptosis-inducing effect and alterations in the cellular redox state of capsaicin in MBT-2 murine bladder tumor cells. Capsaicin induced apoptotic MBT-2 cell death in a time- and dose-dependent manner. The capsaicin-induced apoptosis was blocked by the pretreatment with Z-VAD-fmk, a broad-range caspase inhibitor, or Ac-DEVD-CHO, a caspase-3 inhibitor. In addition to the caspase-3 activation, capsaicin also induced cytochrome c release and decrease in Bcl-2 protein expression with no changes in the level of Bax. Furthermore, capsaicin at the concentration of inducing apoptosis also markedly reduced the level of reactive oxygen species and lipid peroxidation, implying that capsaicin may enhance the antitumor effect of BCG in bladder cancer treatment. These results further suggest that capsaicin may be a valuable intravesical chemotherapeutic agent for bladder cancers.
