Effects of Plant Growth Regulators for In Vitro Propagation of Cannabis sativa varieties (대마 품종별 기내 증식을 위한 식물생장조절제 처리 효과)

  • 대마(Cannabis sativa)는 카나비노이드(cannabinoids) 계열의 성분으로 인한 약리효능 뿐만 아니라 섬유, 식품 등의 분야에서도 식물소재로 활용도가 높은 것으로 알려져 있다. 최근 의료용 대마의 경우 규제완화가 예정됨에 따라 관련 산업분야에서도 안정적 원료 소재 생산에 대한 연구가 요구되고 있다. 대마는 실생번식이 가능한 종이나, 의료용 등 소재의 안정적 공급을 위해서는 균일한 cannabidiol(CBD) 성분의 확보가 가능한 영양증식 방법이 대안으로 사용될 수 있다. 따라서 본 연구에서는 영양증식 방법 중 조직배양 기술을 이용한 대마 품종별(Cherry Blossom, Queen Dream과 Hot Blonde) 증식법 구명을 위하여 식물생장조절제의 종류 및 농도에 따른 기내생육특성을 조사하였다. 세 품종 모두 1 mg l-1 thidiazuron (TDZ) 단독처리 시 신초(> 2cm)가 가장 많이 유도되었다. 단 Queen Dream 품종은 0.5 mg l-1 와 TDZ 0.2 mg l-1 NAA 혼용처리구에서 많은 수의 신초가 유도되었으나 상대적으로 신초의 길이(< 2cm)는 짧은 것으로 확인할 수 있었다. 발생된 신초는 MS 기본배지에서 발근을 유도한 후 토양에 순화하여 정상적인 식물체로 발달되었다. 이와 같은 식물생장조절제 처리를 포함한 식물조직배양 방법은 유용 개체·품종의 증식 및 의료·산업용 원료 소재의 안정적 생산에 활용될 수 있다.

Monosaccharides from industrial hemp (Cannabis sativa L.) woody core pretreatment with ammonium hydroxide soaking treatment followed by enzymatic saccharification

  • Ammonia soaking treatment was introduced for hemp woody core pretreatment to increase enzymatic saccharification of polysaccharides. Portions of the xylan, cellulose, and lignin were removed by aqueous ammonia soaking, which improved the enzymatic saccharification of cellulose and xylan. Following ammonia soaking, 37% ($50^{\circ}C$-6 day treatment) to 61% ($90^{\circ}C$-16 h treatment) of the cellulose was converted to glucose and 33% ($50^{\circ}C$-6 day treatment) to 48% ($90^{\circ}C$-16 h treatment) of the xylan to xylose. Cellulose responded better to enzymatic saccharification than did xylan after the ammonia soaking treatment. Aqueous ammonia soaking pretreatment was more effective than electron beam irradiation for increasing enzymatic saccharification of xylan and cellulose in hemp woody core.

LED광원별 새싹대마(Cannabis sativa L.) 생육 및 염증세포에 대한 세포독성 확인

  • 현재, ‘마약류 관리에 관한 법률’에 의해 규제되고 있는 대마는 “대마초와 그 수지 및 이를 원료로 제조된 제품 일체”에 대한 판매 및 유통이 엄격히 금지되어있다. 예외로, “대마초의 종자·뿌리 및 성숙한 대마초의 줄기와 그 제품”은 규정예외 되어있지만, 이를 식품 및 화장품 등의 산업재료로 활용하기 위해서는 대마의 새싹 및 근경부의 생산량과 안전성이 확보되어야만 한다. 그러므로, 본 연구에서는 동일 생육조건하에 LED광원에 따른 대마(Cannabis sativa L.)의 생육 및 생산량 차이를 확인하였다. 또한, LED광원 별 새싹대마의 독성을 확인하기 위해 대식세포인 RAW 264.7에 추출물을 처리하여 세포독성을 평가하였다. 실험결과, 청색광 처리 새싹대마가 생체 중 12.8 g로 가장 높았다 (적색광 10.8 g, 차광 8.8 g). 광원 별 세포독성은 모든 광원별 새싹대마 추출물에서 0.1 ~ 1 mg/ml까지 농도에서 독성이 없음을 확인하였다. 결과적으로, 빛을 처리한 조건의 새싹대마가 차광된 조건의 새싹대마보다 생산량이 증대되었으며, 광원별 새싹대마에서 세포독성이 없음을 확인하였다. 그러므로, 본 연구를 통해 대마 새싹이 식품 및 산업적 소재로서의 경쟁성과 안전성을 가지고 인체에 유익한 소재로 활용될 수 있는 가능성을 보였다.

Effect of Timing of Ethephon Treatment on the Formation of Female Flowers and Seeds from Male Plant of Hemp (Cannabis sativa L.)

  • Hemp (Cannabis sativa L.) is a dioecious plant, although monoecious plants are bred in some cultivars for fiber or seed production. Recently, hemp has received attention for medicinal use with some cannabinoids, including cannabidiol. Self-fertilization for breeding inbred lines is difficult because of dioeciousness and anemophily in hemp. This experiment was conducted to develop a self-fertilization method by forming female flowers and seeds from male plants of dioecious hemp. To induce the formation of female flowers on male plants, 500 mg L-1 of ethephon was sprayed on plants at soon, seven and fourteen days after primordia formation. The plant ratio of female flowers formation and the number of harvested seeds were increased by ethephon treatment. Female flowers of male plants have 5 stigmas in contrast to the dual stigma of female 1plants. Male plant seeds were lighter and smaller than those from female plants. Although the germination rate was lower than that of normal seeds from female plants, the seeds from male plants germinated to grow seedlings. Thus, we suggest that hemp plants should be treated with ethephon at soon after primordia formation to induce the formation of more female flowers on the male plants.

A Study on the Potency and Stability of the Cream Containing Cannabidiol Extracted Cannabis sativa L. from Korea (국내산 대마로부터 추출한 칸나비디올 함유 크림의 역가 및 안정성 평가)

  • In this study, the stability of cream containing 1% of cannabidiol (CBD) with more than 99% extracted from domestic Cannabis sativa was evaluated. The pH, hardness, content, and chromaticity were measured at intervals of 2 weeks under storage conditions for each temperature for 12 weeks (4 ℃, 25 ℃, 37 ℃, and 45 ℃). The pH of the experimental group containing 1% of CBD tended to decrease more than that of the control group. As a result of measuring the change in the content of CBD in the cream under each temperature for 12 weeks using HPLC, the difference in the amount of decrement according to temperature was confirmed. It was confirmed that as a result of measuring the chromaticity using color difference meter, the yellowness of the cream containing CBD increased according to the increase by the storage period and temperature compared to the control group. From the above results, it was confirmed that the content decrease and color change according to the temperature of the pH 5 ~ 6 cream formulation containing CBD. When CBD is applied to medicine and cosmetic formulations in the future, it is considered that research and development should be conducted in consideration of the change in the quantity and physicochemical quality of CBD under the temperature.

Densified Pellet Fuel Using Woody Core of Industrial Hemp (Cannabis sativa L.) as an Agricultural waste (농업부산물인 산업용 대마(Cannabis sativa L.) 목부를 이용한 고밀화 펠릿 연료)

  • We prepared densified wood pellet by agricultural waste. The hemp woody core was used as replacing wood resource. Hemp was separated into the bast fiber and the woody core by hot steaming treatment. The hemp woody core had a similar lignin content(19.4%) and carbohydrate composition with hardwood(20-25% lignin in hardwood), respectively. Also, the hemp had a low ash content(0.5%), which resulted in a low ash formation in pellet burning. Heating value of the hemp pellet(18.40 MJ/kg) had a very similar to the pellet made by hardwoods. The hemp woody core could be replaced the hardwood for densified wood pellet.
