Candida sp. SY16의 휴식세포를 이용한 생물계면활성제 Mannosylerythritol Lipid의 생산 (Production of a Biosurfactant Mannosylerythritol Lipid by Resting Cell of Candida sp. SY16.)

  • Candida sp. SY16의 휴식세포를 이용하여 탄소원만 포함되 있는 증류수로부터 많은 양의 생물계면활성제(mannosylerythritol lipid: MEL)를 생산할 수 있었다. 플라스크를 이용한 생산반응의 경우, 20 g/l의 휴식세포를 75g/1의 soybean oil이 포함되있고, pH는 4~5범위에서 반응할 때, 가장 높은 생산수율을 얻을 수 있었으며, 높은 농도($PO_4$-P 농도 198 $\mu\textrm{g}$/ml 이상)의 인은 MEL 생산을 저해하는 것으로 나타났다. 최적화된 조건으로 jar fermentor를 이용하여 휴식세포를 반응한 경우 생물계면활성제를 120 h 반응 후, 58 g/l로 얻을 수 있었으며, 그 생산성은 성장세포를 이용한 회분식 발효의 경우보다 높았고, 배양시간도 단축시킬 수 있었다.

  • C$_2$화합물 가운데 Ethanol, acetic acid 및 acetaldehyde을 공통적으로 동화할 수 있는 효모를 토양 및 하수 진흙으로부터 분리한 후 형태학적, 배양적 및 생리학적 특성을 고려하여 그 분류학적 위치를 검토하였다. 그 결과 분리효모는 다극 출아법에 의한 영양 증식을 하였으며 자낭포자를 형성하지 않고 또한 색소가 형성되지 않는 점으로 보아 Candida속에 속하였으며 Candida sp. JY-5로 명명하였다.

항진균성 지의류 내생 곰팡이 및 항진균성 물질의 탐색 (Detection of Antifungal Endolichenic Fungi and Antifungal Compound)

  • 순천대학교 지의류 은행(Lichen Bioresources Center)에서 571종의 지의류 내생 곰팡이를 분양받아 Candida albicans에 대한 항진균 활성을 조사하여 4종을 선정하였다. 선정된 4종의 지의류 내생 곰팡이의 배양액 내에 존재하는 항진균 활성을 알아보기 위해 disc diffusion method 방법을 이용한 결과 Stereocaulon sp. (1429,1430)가 C. albicans에 대한 항진균 활성이 가장 좋았으며, 다음으로는 Graphis sp. (1245), Cryptosporiopsis diversispora (0156) 순으로 활성이 좋게 나타났으며, 이들 지의류 내생곰팡이가 고체배지와 액체배지에서도 항진균 활성 물질을 생성하였다. 고체배지에서와 액체배지에서 공통적으로 활성이 있는 지의류 내생 곰팡이 배양액의 chloroform, ethyl acetate 용매 추출물의 C. albicans의 항진균 활성도를 측정한 결과 4종 모두 chloroform 추출물에서 높은 활성도를 나타냈다. Candida albicans, Candida glabrata, Candida tropicalis, Candida lusitaniae에 대한 항진균 활성은 Stereocaulon sp. (1429, 1430)의 chloroform 추출물에서 모두 98% 이상의 높은 저해율을 보였고 Stereocaulon sp. (1429) 배양액의 chloroform 추출액과 ethyl acetate 추출액을 GC/MS를 이용해 성분을 분석한 결과 hexadecanonic acid-methyl ester, 14-octadecenoic acid-methyl ester, octadecenoic acid-methyl ester가 새로운 항진균 물질일 가능성을 보여주었다.

  • 충남 금산군과 전남 완도군, 제주도 송악산 등지에서 서식하고 있는 야생화들로부터 Rhodosporidium fluviale 201-C-1과 Kluyveromyces thermotolerans 150-JE-2(1), Rodosporidium paludigenum과 Candida sp. 80-J-3등의 국내 미기록 효모들을 분리, 선발하여 이들의 미생물학적 특성들을 조사하였다. Rhodosporidium fluviale 201-C-1은 내염성의 무포자 효모로 의균사를 형성하였고 urease 활성을 갖고 있었다. Kluyveromyces thermotolerans 150-JE-2(1)는 내당성과 내염성을 가진 무포자효모로서 $30^{\circ}C$, pH 4.0~6.0에서 잘 생육하였다. Candida sp. 80-J-3과 Rhodosporidium paludigenum 86-J-1 균주들은 모두 무성생식을 하는 무포자 효모들이었고 YPD 배지, Vitamin-free 배지와 5% NaCl을 함유한 YPD 배지 등에서 잘 생육하였다. 또한 Candida sp. 80-J-3 균주의 urease 활성은 음성이었으나 Rhodosporidium paludigenum 86-J-1 균주는 urease 활성이 양성이면서 pH 7.0~8.0에서 잘 생육하였다.

  • 인공종묘 생산 철갑상어 사육과정에 복부팽만을 특징으로 하는 질병이 발생하였다. 병어는 위와 소화관의 가스의 이상 축적에 의한 것으로 특히, 위의 분문부에서의 가스 축적이 현저하기 때문에 이상 유영을 하다가 폐사하게 된다. 병어의 위, 양어지의 배출수와 잔사에서는 원인체로 추정되는 Candida sp.가 검출되었다. 먹이와 함께 섭취된 Candida가 대사 과정 중에 발생하는 가스가 위에 축적됨으로서 위가팽창하게 되고, 일부는 식도와 부레로 이동한 것으로 생각된다.

임상가검물에서 분리한 Candida sp.의 항진균제 Ketoconazole, 5-Fluorocytosine 및 Amphotericin B의 단독 혹은 복합처리에 의한 항진균력에 대한 연구 (In Vitro Studies of Ketoconazole in Combination with the 5-Fluorocytosine and Amphotericin B against Candida sp. Isolated from Clinical Specimens)

  • The antifungal activities of amphotericin B, 5-fluorocytosine, and ketoconazole in combination of amphotericin B/ketoconazole and 5-fluorocytosine/ketoconazole were determined against 42 strains of Candida spp. isolated from oral cavity. Among 42 strains of Candida species, 36 strains of Candida albicans, 2 strains of Candida parapsilosis and Candida tropicalis 1 strain of Candida krusei and Candida stellatoidea were identified. The minimum inhibitory concentrations(MICs) of amphotericin B, 5-fluorocytosine and ketoconazole for these strains were ranged from 0.05-1.56 mcg/ml, 12.5->100.0 mcg/ml and 0.2-50.0 mcg/ml. In all of the experimental strains, amphotericin B had the greatest antifungal activity on a dilution basis. When a microtiter checkerboard technique was used 5-fluorocytosine acted synergistically with ketoconazole against all strains, whereas amphotericin B has a reduced effect. The killing curve experiments with on strain of Candida albicans WMC-85024 demonstrated that the combination of amphotericin B/ketoconazole and 5-fluorocytosine/ketoconazole produced a decrease in number of colony forming unit of >3 logs in 72 hours.

토양으로부터 Candida albicans 의 균사형태에 선택적인 활성을 나타내는 미생물로부터 항진균 물질의 탐색 (Screening of Antifungal Compounds from Microorganisms with Preferential Activity against the Mycelial Phase of Candida albicans)

  • To search and develop the compounds exhibiting antifungal activities against the mycelial phase of Candida albicans, approximately 2,900 microorganisms isolated from soil were examined for antifungal activity. Among them, a strain with preferential activity against the mycelial phase of Candida albicans was isolated and identified as Streptomyces sp. A393. Isolation and purification of compounds A393 showing antifungal activity against the mycelial phase of C. albicans were performed using XAD-7 column chromatography, silica gel chromatography, preparative thin- layer silica gel chromatography, and HPLC. The molecular weights of compounds isolated from Streptomyces sp. A393 were determined as 774, 790, 804 and 820. These compounds appeared to have a structure of macrolide antibiotics, oligomycin A, B, C and E. Especially, oligomycin E, which is formerly reported to have no antifungal activity, showed antifungal activity against the mycelial phase of Candida albicans.

Clinical Features of Otomycosis Co-occurring with Chronic Otitis Media and the Causative Fungi

  • Background: Otomycosis is a fungal infection that comprises 7~10% of outer ear infections. Although the occurrence is higher in humid climates, relatively few studies have investigated otomycosis occurrences in humid environments. While recurrent chronic otitis media discharge in the ear creates a milieu in which otomycosis is likely to occur, investigations of otomycosis co-occurring with chronic otitis media have been rare. Objective: To examine the characteristics of patients with otomycosis co-occurring with chronic otitis media and identify causative fungi. Methods: The study included 60 patients with chronic otitis media who presented typical otomycosis findings in the outer ear canal and the presence of fungi. Patients were treated in the department of otolaryngology, Daegu Catholic University Medical Center, between July 2011 and June 2018. Results: The mean patient age was 57.77 years, and our study included 20 men and 40 women (p=0.010). The lesion was on the right in 39 patients and on the left in 21 (p=0.020). Ear discharge was the most common chief complaint at diagnosis. Of the 54 patients over age 19, 10 had diabetes (18.5%). Aspergillus was causative in 29 patients and Candida in 31. Aspergillus niger was identified in 15 patients, Aspergillus sp. in 14, Candida parapsilosis in 12, Candida sp. in six, and Candida albicans in five. Conclusion: Otomycosis and chronic otitis media co-occurrences increase with age. The Aspergillus and Candida genera were similar in proportion. A. niger was the most common Aspergillus species, while C. parapsilosis was the most common Candida.

Direct Fermentation of D-Xylose to Ethanol by Candida sp. BT001

  • A yeast strain, BT001, which can directly ferment D-xylose to ethanol was isolated from forest soils, and then identified as Candida sp. Cultural conditions for the optimum ethanol production, along with the effects of aeration on cell growth and ethanol production were investigated. Aeration stimulated the cell growth and the volumetric rate of ethanol production, but decreased the ethanol yield. Optimum temperature and initial pH for the ethanol production were $33{\circ}^C$ and 6.0, respectively. In a shake flask culture, this strain produced 52.3 g ethanol per liter from 12%(w/v) D-xylose after incubation for 96 hours. Ethanol yield was 0.436 g per g D-xylose consumed. This corresponds to 85.8% of theoretical yield. Also, this yeast strain produced ethanol from D-galactose, D-glucose and D-mannose, but not from L-arabinose and L-rhamnose. Among these sugars, D-glucose was the fastest in being converted to ethanol sugars.

Molecular Cloning and Characterization of Mn-Superoxide Dismutase Gene from Candida sp.

  • The manganese-containing superoxide dismutase (MnSOD) is a major component of the cellular defence mechanisms against the toxic effects of the superoxide radical. Within the framework of studies on oxidative stress=responsible enzymes in the Candida sp., the gene encoding the MnSOD was isolated and examined in this study. A specific primer was designed based on conserved regions of MnSOD sequences from other organisms, and was used to isolate the gene by PCR on reverse-transcribed Candida poly($A^{+}$) RNA. The PCR product was used to screen a Candida genomic lambda library and the nucleotide wequence of positive clone was determined. The deduced primary sequence encodes a 25kDa protein which has the conserved residues for enzyme activity and metal binding. The 28 N-terminal amino acids encoded by the Candida cDNA comprise a putatice mitochondrial transit peptide. Potential regulatory elements were identified in the 5' flanking sequences. Northern blot analysis showed that the transcription of the MnSOD gene is induced 5-to 10-fold in response to mercury, cadmium ions and hydrogen peroxide.
