Preparation of Biodegradable Porous Calcium Metaphosphate Matrix (생분해성 다공질 Calcium Metaphosphate Matrix의 제조)

  • It is well known that new tissue or blood vessel is grown into a porous calcium phosphate ceramics used as a bone graft substitute due to their excellent biocompatibility. In this study, the most chemically stable porous $\beta$-crystalline form in various forms of calcium metaphosphate, Ca(PO$_3$)$_2$is prepared by the controlled thermolysis of monocalcium phosphate, Ca(H$_2$PO$_4$)$_2$.The diameter of cylindrical pores formed during cooling was controlled by a holding time at the melting point of a monocalcium phosphate and by the change of a crystallization temperature, to obtain the most appropriate size (about 200$\mu$m) of pores for the application of bone substitutes and matricuts. It was observed that the increasing holding time at the melting point of monocalcium phosphate results in the decreases of cylindrical pore sizes.

  • In the development of biomaterials as a substitute of hard tissues, the biocompatibility and osteoconductivity of the biomaterial are considered to be one of the most significant considerations. These biological properties of a material can be greatly improved by the modification of the surface properties by the depositing calcium phosphate thin films on the material since calcium phosphate films possess similar chemical compositions to hard tissues. The success of a material as a biomaterial will be determined by the interaction of the surface of the material with the adhesion molecules which induce cellular adhesion and biological responses of the adherent cells. Depending on the adsorption mechanisms and adsorbed conformation of the adhesion molecules on the surface of the biomaterial, cellular responses, such as adhesion, proliferation and differentiation of osteoblast cells, can be promoted or restricted. It has been reported that materials of which surfaces were modified with thin films of calcium phosphate appeared to be more osteoconductive. Rapid formations of bone nodule in addition to higher differentiations of osteoblast have been observed on the calcium phosphate thin films.

  • In this study, we found a simple surface biofunctionalization of biphasic calcium phosphate (BCP) based on the high affinity between alendronate and the calcium ions of BCP, and the strong interaction between heparin and bone morphogenic protein-2 (BMP-2). The biofunctionalized BCP did not be precipitated well and display a remarkable enhancement of osteogenic activity of human adipose-derived stem cells by showing increased alkaline phosphatase (ALP), calcium deposition and osteogenic-related genes (i.e., Runx-2, ALP, osteocalcin, and osteopontin), and bone regeneration in the calvarial defect model. Therefore, this simple surface technique can be used to easily functionalize various calcium phosphates.

Isolation of Insoluble Phosphate-Solubilizing Bacteria and Optimum Condition for Solubilization (인광석 가용화 세균의 분리 및 가용화 최적조건)

  • 850 strains of phosphate-solubilizing bacteria were isolated from soil of Chung-nam and Daejeon region using 0.5% calcium phosphate containing medium. The HS-2 strain with the highest rock phosphate-solubilizing activity was selected and identified as Azotobacter sp. HS-2 based on the microbiological characteristics. The optimum culture temperature and initial pH of medium for solubilization of rock phosphate were $30^{\circ}C$ and pH 6.0-7.0, respectively. Addition of oxalic acid(0.5M) into the PDB-rock phosphate medium increased 50% solubilization of rock phosphate.

  • We have developed a TOF-MEIS system using 70~100 keV He+. A TOF-MEIS system was designed and constructed to minimize the ion beam damage effect by utilizing a pulsed ion beam with a pulse width < 1 ns and a TOF delay-line-detector with an 120 mm diameter and a time resolution of 180 ps. The TOF-MEIS is an useful tool for interfacial analysis of the composition and structure of nano and bio systems. Our recent applications are reported. We investigated the effect with Polyaspartic Acid (pAsp) and Osteocalcin on the initial bone growth of calcium hydroxyl appatite on a carboxyl terminated surface. When pAsp is not added to the self-assembled monolayers of Ca 2mM with Phosphate 1.2 mM, the growth procedure of calcium hydroxyl appatite cannot be monitored due to its rapid growth. When pAsp is added to the SAMs, the initial grow stage of the Ca-P can be monitored so that the chemical composition and their nucleus size can be analyzed. Firstly discovered the existence of 1-nm-sized abnormal calcium-rich clusters (Ca/P ~ 3) comprised of three calcium ions and one phosphate ion. First-principles studies demonstrated that the clusters can be stabilized through the passivation of the non-collagenous-protein mimicking carboxyl-ligands, and it progressively changes their compositional ratio toward that of a bulk phase (Ca/P~1.67) with a concurrent increase in their size to ~2 nm. Moreover, we found that the stoichiometry of the clusters and their growth behavior can be directed by the surrounding proteins, such as osteocalcin.

Preparation of Natural Polymer-CaP Composite Films (천연 고분자-칼슘 포스페이트 복합 박막 제조)

  • We investigated the surface modification method for the preparation of organic-inorganic hybrid composite thin film. Gelatin obtained from the decomposition of collagen was allowed to adsorb in a polystyrene tissue culture dish for 2 h to from layers of gelatin. Supersaturated ionic solution of calcium and phosphorus was injected on the gelatin adsorbed layer to form calcium phosphate thin film. During the initial period of incubation, nucleates were formed. With increase of the incubation time, CaP (calcium phosphate) thin film grew on the surface of the culture dish. The gelatin/CaP thin film displayed the highly porous three-dimensional surface structure. Attenuated, total reflectance Fourier transform, infra-red spectroscopy (ATR-FTIR) was used to analyze the chemical properties of CaP film. The analysis demonstrated that the CaP film formed at initial period of treatment appeared to be amorphous. With increase of incubation time, the crystallinity of the film was slightly increased, but the presence of the peaks for the low crystalline CaP confirmed that the CaP thin film prepared in this study was poorly crystallized.

Solubilization of Insoluble Phosphates by Aspergillus sp. PS-104 Isolated from Soil (토양에서 분리한 Aspergillus sp. PS-104 균주에 의한 난용성 인산염 분해)

  • Phosphate-solubilizing microorganisms were isolated from soil around Kyungnam and Kyungbook regions using potato dextrose agar-calcium phosphate medium. A fungus with the greatest phosphate-solubilizing activity was selected and identified to Aspergillus sp. PS-104, based on the morphological characteristics of conidiophore and conidia; unbranching type of conidiophore, terminally swelling of conidiophore and septate of mycelium, in malt extract agar and potato dextrose agar media. The optimum temperature and initial pH to solubilize rock phosphate in potato dextrose broth-rock phosphate medium were $30^{\circ}C$ and pH 7.0, respectively. In these optimum conditions, phosphate-solubilizing activities of Aspergillus sp. PS-104 against four twos of insoluble phosphate, tricalcium phosphate, aluminium phosphate, hydroxyapatite and rock phosphate, were quantitatively determined. As result, the maximum phosphate-solubilizing activity was obtained with tricalcium-phosphate (1,900 ppm) while minimum activity was obtained with hydroxyapatite (320 ppm). Futhermore, phosphate-solubilizing activity of Aspergillus sp. PS-104 was found higher when treated with nitrates as compared to the ammonium salts as a nitrogen sources.

Effect of Amino Acid Solution for Cell Growth and MPS Activity of Mineral Phosphate Microorganisms. (아미노산액 처리에 의한 인산가용화균주의 생육 및 가용화 효과)

  • Phosphate-solubilizing activities of the two strains (Burkholderia sp. DA23 and Klebsiella sp. DA7l-1) against tri-calcium phosphate and hydroxyapatite were quantitatively determined. Two strains were found to solubilize two types of insoluble phosphate different amounts of amino acid solutions in liquid culture. MPS ability of the strains was increased with concentration of amino acid addition. The optimal solubilization condition of insoluble phosphate in sucrose minimal medium were 0.1% amino acid solution, respectively. The efficiency of amino acid addition was no difference between the two types of insoluble phosphate, tri-calcium phosphate and hydroxyapatite.

Dissolution on the Surface of Bioceramics Prepared by Commercial Calcium Phosphate Powders (상용 인산칼슘계 분말로 제조된 생체세라믹스의 표면용해 특성)

  • In this study, dissolution characteristics of four types of commercial calcium phosphate ceramics were investigated in distilled water with respect to chemical composition and microstructure. For all samples, no significant damage was observed after 3 days of immersion. Following the 7 days of immersion, surface dissolution of the ceramics containing a crystalline phase susceptible to water such as TCP, even pure hydroxyapatite, was initiated at grain boundaries and the dissolution was extended interior to the material along the grain boundaries. In the considerably dissolved area, there was grain separation followed by the formation of 20 $\mu\textrm{m}$ of cavities. In at least one case, the residual pores on the surface appeared to initiate dissolution. In a dissolved area, a crack during the fracture propagates along the grain boundaries resulting in intergranular fracture, while transgranular fracture occurs in a dense area without significant dissolution.

Studies on the Dissolution Rate of Phenylbutazone Deposited on Excipients by Solvent Deposition Method (용매침착법을 이용한 페닐부타존의 용출속도에 관한 연구)

  • A dissolution characteristics of phenylbutazone deposited on Avicel and dibasic calcium phosphate by solvent deposition method were studied. The solvent deposition was confirmed by scanning electron microscopy. Avicel was superior to dibasic calcium phosphate as excipient in dissolution rate. Total amount of phenylbutazone dissolved from Avicel deposition system at 30minutes were enhanced 1.2-1.6 times compared with physical mixtures of them. The dissolution rate of 10% solvent deposition system was highest and that of 75% solvent deposition system was lowest in Avicel system and dibasic calcium phosphate system. Dissolution profile of commercial products was dependent on manufacturing conditions and dissolution rate of 10% Avicel system was greater than that of commercial products.
