Reduction of Bacillus cereus in Cooked Rice Treated with Sanitizers and Disinfectants

  • This study aimed to identify effective washing and sanitation programs to minimize the contamination of cooked rice by B. cereus. As such, the effectiveness of five sanitizers, including QAC, alcohol, chlorine, CaO, and $H_2O_2$, was evaluated in relation to the survivability of B. cereus spores in cooked rice and resulting sensory properties of the rice. The water-treated cooked rice showed remaining B. cereus spores at 1.09 $log_{10}CFU/g$. In contrast, treatment with the minimum inhibitory concentrations of the sanitizers, such as 200 ppm of QAC, 50% of alcohol, 100 ppm of chlorine, 650 ppm of CaO, and 500 ppm of $H_2O_2$, destroyed all the spores in the cooked rice below a non-detection limit (ND< 0.15 CFU/g). The sensory properties of the sanitizer-treated (1,000 ppm of $H_2O_2$, 100 ppm of chlorine, and 800 ppm of CaO) cooked rice did not differ significantly from those of the water-treated cooked rice. As a result, 500 ppm of $H_2O_2$, 650 ppm of CaO, and 100 ppm of chlorine were found to effectively eliminate B. cereus spores in rice while cooking.

Protective Effects of Ursolic Acid on Osteoblastic Differentiation via Activation of IER3/Nrf2

  • Background: Oxidative stress is a known to be associated with in the pathogenesis of many inflammatory diseases, including periodontitis. Ursolic acid is a pentacyclic triterpenoid with has antimicrobial, antioxidative, and anticancer properties. However, the role of ursolic acid in the regulating of osteogenesis remains undetermined. This study was aimed to elucidate the crucial osteogenic effects of ursolic acid and its ability to inhibit oxidative stress by targeting the immediate early response 3 (IER3)/nuclear factor erythroid 2-related factor 2 (Nrf2) pathway. Methods: Cell proliferation was determined using water-soluble tetrazolium salt assay, cell differentiation was evaluated by alkaline phosphatase (ALP) activity, and formation of calcium nodules was detected using alizarin red S stain. Generation of reactive oxygen species (ROS) was determined using by DCFH-DA fluorescence dye in hydrogen peroxide ($H_2O_2$)-treated MG-63 cells. Expression levels of IER3, Nrf2, and heme oxygenase-1 (HO-1) were analyzed using western blot analysis. Results: Our results showed that ursolic acid up-regulated the proliferation of osteoblasts without any cytotoxic effects, and promoted ALP activity and mineralization. $H_2O_2$-induced ROS generation was found to be significantly inhibited on treatment with ursolic acid. Furthermore, in $H_2O_2$-treated cells, the expression of the early response genes: IER3, Nrf2, and Nrf2-related phase II enzyme (HO-1) was enhanced in the presence of ursolic acid. Conclusion: The key findings of the present study elucidate the protective effects of ursolic acid against oxidative stress conditions in osteoblasts via the IER3/Nrf2 pathway. Thus, ursolic acid may be developed as a preventative and therapeutic agent for mineral homeostasis and inflammatory diseases caused due to oxidative injury.

CaO2 적용에 따른 퇴적물의 P fraction 변화와 수질에 미치는 영향 (The Effect of CaO2 Application on the Change of Sedimentary Phosphorus Fraction and Water Quality)

  • This study was investigated to improve the phosphorus release and water quality by transformation of sedimentary P fraction for application of $CaO_2$. For the experiment, 0.5% (w/w) of $CaO_2$ was homogenized in the sediment and incubated with the control for 20 days. The analytical results showed that pH increased with $CaO_2$ and redox potential (ORP) was improved in the sediment of the reactor. The growth rate of chlorophyll-a was lower in the $CaO_2$ reactor and Dissolved Oxygen (DO) of overlying water maintained higher than that of the control. Total phosphorus (T-P) concentration in the overlying water increased from the initial concentration to 0.304mg/L in the control at 20 days. The reactor of $CaO_2$ was lowered by 29.3%. Ex-P, Fe-P and Ca-P in sediment P fraction were increased with the $CaO_2$. The formation of bound Fe-P and Ca-P in the sediments seemed to control the release of P by removing the Soluble Reactive Phosphorus (SRP) presented in the pore water. From the result, this indicated that the reduction of P release from the sediments seems to be effective in suppressing the eutrophication of P and improving the oxygen condition in the water quality with the application of $CaO_2$.

모단피의 PC12 cell 산화억제 효과 및 neuronal 유전자 발현 profile 분석에 대한 연구 (Effect of Moutan Cortex Radicis on gene expression profile of differentiated PC12 rat cells oxidative-stressed with hydrogen peroxide)

  • Yukmijihwang-tang has been widely used as an and-aging herbal medicine for hundred years in Asian countries. Numerous studies show that Yukmijihwangtang has anti-oxidative effect both in vivo and in vitro. It has been reported that Moutan Cortex Radicis extract (MCR) was the most effective herb in Yukmijihwang-tang on undifferentiated PC12 cells upon oxidative-stressed with hydrogen peroxide. The purpose of this study is to; 1) evaluate the recovery of neuronal damage by assessing the anti-oxidant effect of MCR on PC12 cells differentiated with nerve growth factor (NGF), 2) identify candidate genes responsible for anti-oxidative effect on differentiated PC12 cells by oligonucleotide chip microarray. PC12 cells, which were differentiated by treating with NGF, were treated without or with hydrogen peroxide in the presence or absence of various concentration of MCR. Cell survival was determined by using MTS assay. Measurement of intracellular reactive oxygen species (ROS) generation was determined using the H2DCFDA assay The viability of cells treated with MCR was significantly recovered from stressed PC12 cell. In addition, wide rage of concentrations of MCR shows dose-dependent inhibitory effect on ROS production in oxidative-stressed cells. Total RNAs of cells without treatment(Control group), only treated with H₂O₂ (stressed group) and treated with both H₂O₂ and of MCR (MCR group) were isolated, and cDNAs was synthesized using oligoT7(dT) primer. The fragmented cRNAs, synthesized from cDNAs, were applied to Affymetrix GeneChip Rat Neurobiology U34 Array. mRNA of Calcium/calmodulin-dependent protein kinase II delta subunit(CaMKII), neuron glucose transporter (GLUT3) and myelin/oligodendrocyte glycoprotein(MOG) were downregulated in Stressed group comparing to Control group. P2X2-5 receptor (P2X2R-5), P2X2-4 receptor (P2X2R-4), c-fos, 25 kDa synaptosomal attachment protein(SNAP-25a) and GLUT3 were downregulated, whereas A2 adenosine receptor (A2AR), cathechol-O-methyltransferase(COMT), glucose transporter 1 (GLUT1), EST223333, heme oxygenase (HO), VGF, UI-R-CO-ja-a-07-0-Ul.s1 and macrophage migration inhibitory factor (MIF) were upregulated in MCA group comparing to Control group. Expression of Putative potassium channel subunit protein (ACK4), P2X2A-5, P2X2A-4, Interferon-gamma inducing factor isoform alpha precursor (IL-18α), EST199031, P2XR, P2X2 purinoceptor isoform e (P2X2R-e), Precursor interleukin 18 (IL-18) were downregulated, whereas MOO, EST223333, GLUT-1, MIF, Neuronatin alpha, UI-R-C0-ja-a-07-0-Ul.s1, A2. adenosine receptor, COMT, neuron-specific enolase (NSE), HO, VGF, A rat novel protein which is expressed with nerve injury (E12625) were upregulated in MCR group comparing to Stressed group. The results suggest that decreased viability and AOS production of PC12 cell by H₂O₂ may be, at lease, mediated by impaired glucose transporter expression. It is implicated that the MCR treatment protect PC12 cell from oxidative stress via following mechanisms; improving glucose transport into the cell, enhancing expression of anti-oxidative genes and protecting from dopamine cytotoxicity by increment of COMT and MIF expression. The list of differentially expressed genes may implicate further insight on the action and mechanism behind the anti-oxidative effects of herbal extract Moutan Cortex Radicis.

치약용 약효제의 적용시간에 따른 실험실적 효능 연구 (Evaluation of In-Vitro Efficacy of Active Ingredients in Dentifrice Used for Different Treatment Times)

  • 페이스트 치약의 구강 내 사용시간(1~3분)과 사용시간을 길게 조절할 수 있는 부착형 치약의 사용시간을 고려하여 치약제의 유효성분을 처리시간 조건별로(1분, 3분, 10분, 30분 및 60분) 미백 효과, 항치석 효과, 재광화 효과 및 상아세관의 폐쇄 효과를 실험실적으로 평가한 결과 다음과 같은 결과를 도출하였다. 처리 시간대별 명도 변화값(${\Delta}L$)은 1분 $5.50{\pm}1.51$에서 60분 $27.35{\pm}0.54$였고 치석으로부터 제거된 칼슘 양(ppm)은 1분 $4.23{\pm}0.14$에서 60분 $25.15{\pm}0.32$였으며 경도 증가값은 1분 $1.96{\pm}1.44$에서 60분 $12.55{\pm}2.09$였다. 상아세관의 폐쇄 정도는 10분 처리에서 일부 상아세관의 폐쇄가 관찰되기 시작하여 60분 처리에서는 대부분의 상아세관의 폐쇄가 관찰되었다. 본 실험조건에서는 처리시간을 10분에서 60분 처리시 1분에서 3분 처리할 때보다 최소 1.57배에서 최대 8.26배까지 효능을 증가시켰으며 재광화 효과, 항치석 효과 및 상아세관 폐쇄 효과에서는 사용시간 10분 이상에서 적절하였고 미백효과에서는 30분 이상이 보다 효과적일 것으로 생각되었다.

더미 미생물용출에 의한 폐-광석으로부터 유용금속 용해 특성과 Fe 제거와 전기분해를 이용한 금속구리분말 회수 (The Characteristic Dissolution of Valuable Metals from Mine-Waste Rock by Heap Bioleaching, and the Recovery of Metallic Copper Powder with Fe Removal and Electrowinning)

  • 폐-광석으로부터 금속구리분말을 회수하기 위하여 더미 미생물용출, Fe 제거와 전기분해실험을 수행하였다. Cu가 0.034% 함유된 폐-광석시료에 대하여 더미 용출실험을 수행한 결과, Cu 용출률은 박테리아 용출-용액에서 61%, 황산 용출-용액에서 62%로 나타났다. Fe를 효과적으로 제거하기 위하여 더미 용출-용액에 NaOH, $H_2O_2$$Ca(OH)_2$를 각각 적용한 결과 $H_2O_2$가 가장 효과적인 Fe 제거제로 선정되었다. 전해질 용액을 준비하기 위하여 $H_2O_2$를 더미 용출-용액에 처리한 결과 박테리아 용출-용액에서 Fe가 99%, 황산 용출-용액에서 60%로 제거된 반면에 Cu 제거율은 각각 5%와 7%로 나타났다. 이 용액에 대하여 전기분해 실험을 수행한 결과 Cu 회수율이 박테리아 용출-용액에서 98%, 황산 용출-용액에서 76%로 나타났다. 모수석 형태의 금속구리분말이 양쪽 용출-용액에서 회수되었다.

시판 소건 샛줄멸의 식품성분 및 관능 특성 (Food Components and Sensual Characterization of Commercial Plain-dried Silver-stripe Round Herring)

  • 제주특별자치도의 특산품으로 개발한 소건 샛줄멸의 효율적 이용, 마케팅 및 규격화를 위한 기초자료로 활용할 목적으로 소건 샛줄멸의 식품성분 특성에 대하여 살펴보았고, 아울러 이를 자건 샛줄멸 및 자건 멸치의 식품성분과도 비교, 검토하였다. 수분함량 및 염도는 소건 샛줄멸이 각각 31.9% 및 7.6%로, 자건 샛줄멸(각각 27.2% 및 12.3%) 및 자건 멸치(각각 22.8% 및 15.4%)에 비하여 수분함량의 경우 높았고, 염도의 경우 낮았다. 과산화물값, 지방산 조성, 황색도 및 갈변도의 결과로 미루어 보아 지질의 산화정도는 소건 샛줄멸이 자건 샛줄멸 및 자건 멸치에 비하여 높았다. 소건 샛줄멸이 자건 샛줄멸 및 자건 멸치에 비하여 총아미노산 함량의 경우 낮았으나, 칼슘과 인 함량의 경우 높았다. 열수 가용성 질소, 엑스분 질소, 유리아미노산 및 taste value의 결과로 미루어 보아 소건 샛줄멸의 맛은 강도 및 경향에서 모두 자건 샛줄멸뿐만이 아니라 자건 멸치에 비하여도 우수 하였다. 소건 샛줄멸은 자건 샛줄멸에 비하여 맛 및 형상에서 우수하였으나 반드시 비린내에 대한 개선이 이루어져야 할 것으로 판단되었다.

옥수수전분 제조공정의 부산물인 maize germ의 지질 및 아미노산 조성 (Chemical Composition of Maize Germs Separated in the Manufacture of Cornstarch)

  • 옥수수전분을 제조하는 과정에서 얻어지는 부산물의 산업적 이용과 공장 폐기물로 인한 환경오염을 최소화시키기 위한 기초자료를 얻기 위하여 옥수수를 파쇄하고 비중의 차로 전분부와 분리하여 모아지는 배아함유 폐기물의 조성분을 분석 정량하여 얻은 결과는 다음과 같다. 1. 일반성분으로 조단백질이 25.3%, 조지방이 19.2%로 나타났으며 시료 100g당 열량은 512kca1로 우수하였다. 2. 무기성품의 함량은 Ca, P이 비교적 낮았으며, K, Mg, Zn, Mn은 높은 값을 나타내었다. 3. 유지의 화학적 특성으로는 요오드가는 44.0 검화가 292.3, 산가 1.5, 과산화가 3.3이었으며 종자나 배아의 지방질 특성과 비슷하였다. 4. Silicic acid column chromatography에 의하여 분획된 지질은 대부분 중성지질 이었으며 다음으로 당지질, 인지질 순이었다. 5. 전지질, 유리지질 및 결합지질의 주요 지방산은 linoleic$(60{\sim}62.5%)$, oleic$(20{\sim}22.5%)$ 및 palmitic acid$(10{\sim}13.5%)$이었으며 불포화지방산이 포화지방산 보다 훨씬 많았다. 6. 질소화합물은 불용성 단백태질소함량이 32.5%로 가장 많고, 수용성 단백태질소 12.7% 및 peptide태 질소화합물은 3.3%로 적은 함량을 보였다. 7. 단백질의 주요 amino산은 glutamic, glycine, alanine 및 aspartic acid등 이었으며 곡류에 결핍되기 쉬운 lysine을 포함한 필수아미노산도 고루 함유되어 있었다. 8. SDS-disc electrophoresis 결과 $5{\sim}6$개의 단백질 band를 확인하였으며, $14,500{\sim}24,500$ 정도의 분자량을 가진 단백질이 주종을 이루고 있음을 알 수 있었다.

Comparative Efficacy of Synthetic and Natural Tenderizers on Quality Characteristics of Restructured Spent Hen Meat Slices (RSHS)

  • In the present study, comparative efficacy of natural as well as synthetic tenderizers on the quality characteristics of restructured spent hen meat slices (RSHS) was studied. Four different batches of RSHS viz. Control (without any tenderizer), T1 (1.25% calcium chloride replacing salt in formulation), T2 and T3 (1.5% each of pineapple rind and fig powder, replacing binder in the formulation) were developed in pre-standardized formulation. Vacuum tumbling was performed for 2.5 h and cooked product (RSHS) was assayed for quality attributes. Samples were packaged in aerobic conditions, stored for 21 days under refrigeration ($4{\pm}1^{\circ}C$) and were evaluated for pH, oxidative and microbial quality parameters at regular interval of 7 days. Water holding capacity of T2 was recorded the highest and significantly higher (p<0.05) than all other samples. The textural attributes of T2 were comparable to T1 but significantly higher (p<0.05) than C and T3. The colour attributes ($L^*$, $a^*$, and $b^*$ value) of T2 and T3 were improved due to use of natural tenderizers. During sensory evaluation, tenderness scores for T2 samples were recorded the highest. Throughout storage period, thiobarbituric acid reactive substances (TBARS), free fatty acids (FFA) and peroxide value (PV) followed an increasing trend for control as well as treated products; however, T2 showed a significantly (p<0.05) lower value than control and other treated samples. It can be concluded that good quality RSHS with better storage stability could be prepared by utilizing 1.5% pineapple rind powder as natural tenderizer.

벼 침수토중 직파 재배 연구 I. 온도 및 파종 심도에 따른 출아 및 초기 생육 (Direct Seeding Cultivation on Submerged Paddy in Rice I. Seedling Emergence and Early Growth under Different Temperature and Seeding Depth)

  • 벼 침수토중 직파재배의 기술확립을 위하여 1985년 작물시험장에서 벼 종자에 과산화칼슘(CaO$_2$) 분의처리 효과와 온도 및 파종 심도에 따른 품종별 출아 및 초기생육을 검토한 결과를 요약하면 다음과 같다. 1. 과산화칼슘 종자 분의는 무처리에 비하여 출아 및 입모가 현저히 좋았으며 초기 생육도 좋았다. 2. 벼 종자의 과산화칼슘 분의 효과는 고온(29/21$^{\circ}C$)보다는 저온(12/12$^{\circ}C$, 17/17$^{\circ}C$) 조건에서 컸다. 3. 파종심도에 따른 출아율은 토중 1 cm 깊이에서 가장 좋았다. 4. 침수토중 직파조건하에서 출아율이 높은 품종은 일반계는 천마벼, 남양벼, 기호벼, 추정벼, 낙동벼, 동진벼 이고 다수계 품종은 태백벼, 삼강벼, 가야벼 등이었다. 5. 침수토중 직파시에는 저온하에서 출률이 높은 품종의 선정 및 출아구을 높히기 위한 검토가 앞으로 중요한 과제가 될 것으로 생각된다.
