Three strains capable of degrading a chlorophenol were isolated by selective enrichment from soils contaminated with industrial wastewater. A Pseudomonas solanacearum TCP114 could use 2,4,6-trichlorophenol (TCP) as sole carbon and energy source, while two strains of Pseudomonas testosteroni CPW301 and Arthrobacter ureafaciens CPR706 could use 4-CP. All isolates also grew well on phenol. The degradation of one component by a pure strain was strongly affected by the presence of other compounds in the medium, CPW301 and CPR706 entirely lost the ability to degrade 4-CP and phenol in the presence of TCP. TCP114 also lost the ability to degrade phenol when 4-CP was added to the culture medium. These restrictions on the degradability could be overcome by employing defined mixed cultures (TCP114 and one strain of 4-CP degrading strains). All three components were successfully degraded by defined mixed cultures through their cooperative activities. It was also demonstrated that defined mixed cultures could be immobilized by using calcium alginate for the semi-continuous degradation of the three component mixture. Immobilization could not only accelerate the degradation rate, but also allowed the reuse of the cell mass several times without loss of the cells' degrading capabilities.
In this study, in order to develop a continuous production process of lactosucrose in a packed-bed reactor, Sterigmatomyces elviae ATCC 18894 was selected and mutated. The mutant strain of S. elviae showed 54.3% higher lactosucrose production than the wild type. Reaction conditions such as temperature, pH, substrate concentration and flow rate were also optimized. Under optimized reaction conditions ($50^{\circ}C$, pH 6.0, 25% sucrose and 25% lactose as substrate, flow rate 1.2 ml/min), the maximum concentration of lactosucrose (192 g/l) was obtained. In a packed-bed reactor, continuous production of lactosucrose was performed using S. elviae mutant immobilized in calcium alginate, and about 180 g/l of lactosucrose production was achieved for 48 days.

This work described some essential factors necessary for micro-propagation of banana for mass production of synthetic seeds for germ plasm conservation, and how peroxides activity of conserved tissue was influenced. Shoot tips of field grown plants were used to obtain shoot clusters on shoot proliferation medium (MS medium supplemented with 5 mg/l BAP). Using longitudinally-split shoot tip technique, 18720, 8640, 7488, 2016 plantlets were obtained from one shoot tip of Maghraby, Grand Naine, Balady, and Williams, respectively, in six subculture, one month each, on solid medium. Shoot tips excised from in vitro grown plantlets were encapsulated in calcium-alginate beads and stored at $4^{\circ}C$ for one month on half-strength MS basal medium without growth regulators or sugars. After one month all the viable-conserved synseeds formed shoots when they were transferred to MS basal medium, some of them showed synchronous formation of shoot and root systems in one week. Plants retrieved from encapsulated shoot tips were hardened off and transferred to soil.
이 연구에서는 탄산칼슘 형성 박테리아의 내생포자를 접종한 알지네이트 겔과 포자현탁액 형태의 균열치유제를 모르타르에 첨가하여 균열치유성능을 비교, 분석함으로써 균열치유제 제조방법별 균열치유성능을 분석하였다. 또한 박스형 암거 형태의 실 구조물에 적용하여 실험실 환경뿐만 아니라 개발된 균열치유제가 실제 현장에 적용될 수 있는 환경을 조성하여 실구조물 스케일에서 균열치유성능을 검증하고자 하였다. 건조 방식을 달리한 두 가지 형태의 알지네이트 겔로 구성된 균열치유제를 분석한 결과, 건열건조 방식의 균열치유제는 균열치유 성능을 나타내었으나, 동결건조 방식의 경우 얼음 결정에 의해 다수의 포자가 사멸되어 균열치유성능을 잃는 것으로 나타났다. 실스케일 구조물의 균열부에서 추출된 균열치유 추정물질의 SEM 사진과 XRD 패턴 분석 결과 균열치유제에 적용된 균열치유 미생물이 생성한 탄산칼슘 결정 중 하나인 calcite인 것으로 나타났으며, 미생물에 의한 균열치유메커니즘이 실구조물에서 구현될 수 있음을 확인하였다.
락툴로오스는 기존에 화학적인 이성화법을 통해 생산해왔던 기능성 당으로서 프로바이오틱스나 장내균총 개선을 위한 의약품으로 활용되어 왔다. 최근 락툴로오스 화학전환법의 단점인 촉매제거와 부산물제거 에너지손실등의 문제를 해결할 수 있는 생물촉매를 이용한 락툴로오스 전환법이 대두되었다. 본연구에서는 유당의 낮은 용해도와 락툴로오스의 효율적전환을 위해 최적의 효소를 선별하여 무작위 돌연변이법으로 유전자를 개량하여 열내성이 $75^{\circ}C$까지 증진되고 활성이 1.3배 향상된 효소를 선별하였다. 이 효소를 정제하여 사용하는 대신 본 연구에서는 과량 발현시킨 대장균을 Ca-alginate로 고정화하여 $70^{\circ}C$에서 200 g/l의 유당과 회분식으로 반응시켜 43%의 전환 수율을 확인하였다. 반복회분식 실험에서 고정화된 담체는 비교적 안정적이었으며 4회 반복반응 후에도 80% 이상의 활성을 유지하고 있었다. 산업적인 방법을 개발하기 위해 고정화 담체를 이용한 반응기의 운전 최적화와 담체의 안정화를 증진시키는 추가적인 연구가 필요하지만, 본 연구에서는 열내성 특성을 이용하여 정제된 효소가 아닌 효소를 발현하는 세포자체를 고정화 시킴으로써 경제성있는 생산에 대한 방법론을 제시하였다.
현재 남해안 일대에서 과량 생산되고 있는 양식산 굴의 소비 촉진을 위하여 영양과 건강 기능성을 함유하면서 신세대의 기호에 맞는 굴 조미 건제품의 제조를 시도하였다. 최적조건에서 저장성 있는 조미 반건조 굴 가공품의 제조를 위하여 동결 굴을 해동한 다음 굴 특유의 비린내를 차폐하기 위하여 무즙($5{\pm}3^{\circ}C$, 2시간)에 침지, 수세 및 탈수하고 5분동안 증자하였다. 이어서 탈수(10분) 및 가열$(105^{\circ}C)$한 다음 조미액(간장 33.6, 밀가루 7.2, 물 40.5, 솔비톨 8.8, 미림 9.9)에서 5분 동안 침지한 후 열풍건조기($45^{\circ}C$, 6시간)에서 건조처리 하였다. 그리고 1% sodium alginate 용액에 침지, 건조($45^{\circ}C$, 1시간)한 다음 레토르트 파우치 필름에 충전, 밀봉한 후 저장성 부여를 위하여 $F_0$ value가 5.4분이 되도록 살균하여 조미 반건조 굴 가공품을 제조하였다. 최적 조건에서 제조된 코팅 및 조미처리 굴 가공품은 대조 제품(무코팅 및 무조미 건조 굴 가공품)에 비하여 수분 및 조회분이 약간 높았고, 조지방 및 조단백질은 낮았으며, 색조는 어두우면서 경도는 높았다. 코팅 처리 조미 반건조 굴제품은 또한 필수 아미노산의 함량이 높으면서 곡류 제한 아미노산인 lysine과 n-3계 지방산의 조성비가 높았다.
Edible films containing antimicrobial agents can be used as safe alternatives to preserve food products. Essential oils are well-recognized antimicrobials. However, their low water solubility, volatility and high sensitivity to oxygen and light limit their application in food preservation. These limitations could be overcome by embedding these essential oils in complexed product matrices exploiting the encapsulation efficiency of β-cyclodextrin. This study focused on the maximization of β-cyclodextrin production using cyclodextrin glucanotransferase (CGTase) and the evaluation of its encapsulation efficacy to fabricate edible antimicrobial films. Response surface methodology (RSM) was used to optimize CGTase production by Brevibacillus brevis AMI-2 isolated from mangrove sediments. This enzyme was partially purified using a starch adsorption method and entrapped in calcium alginate. Cyclodextrin produced by the immobilized enzyme was then confirmed using high performance thin layer chromatography, and its encapsulation efficiency was investigated. The clove oil/β-cyclodextrin inclusion complexes were prepared using the coprecipitation method, and incorporated into chitosan films, and subjected to antimicrobial testing. Results revealed that β-cyclodextrin was produced as a major product of the enzymatic reaction. In addition, the incorporation of clove oil/β-cyclodextrin inclusion complexes significantly increased the antimicrobial activity of chitosan films against Staphylococcus aureus, Staphylococcus epidermidis, Salmonella Typhimurium, Escherichia coli, and Candida albicans. In conclusion, B. brevis AMI-2 is a promising source for CGTase to synthesize β-cyclodextrin with considerable encapsulation efficiency. Further, the obtained results suggest that chitosan films containing clove oils encapsulated in β-cyclodextrin could serve as edible antimicrobial food-packaging materials to combat microbial contamination.
Xylitol을 당수용체로하여 당알콜 올리고당 gluosyl-xylitol을 생산하기 위하여 갭술고정화 전세포 CGTase를 제조하고자 하였다. CGTase를 생산하기 위하여 Bacillus macerans를 배양하는 경우 organic form의 질소원을 사용하는 경우 inorganic form의 질소원을 사용하는 경우보다 더 많은 CGTase를 생산하였고 배양도중 탄소원인 starch가 분해되는 동안 CGTase가 생성되었다. B. macerans에 의하여 생산된 CGTase는 80% 이상이 extracellular cnzyme 이며, intracellular enzyme은 20% 이내이었다. E. coh, C. glutamicum, S. cerevisiae 등과 달리 캡슐내부에 B. macerans를 접종하고 캡슐내부에서 고농도로 배양할 수 없었다. 배양액속에 존재하는 CGTase는 다른 이온성 물질들로 인하여 활성탄, Ambolite, Sephadex 등의 흡착제에 흡착시킬 수 없었다. 미생물을 배양한 배양책 전체를 10배로 농축하여 캡슐내에 고정화함으로써 캡슐고정화 전세포 CGTase를 제조할 수 있었다. 농축배양액을 이요하여 제조된 캡슐고정화 전세포 CGTase는 hydrolysis, intermolecular transglycosylation을 수행하였으며, xylitol을 당수용체로 하고 dextrin을 당공여체ㅗ 하여 glucosyl-xylitol을 생산하였다.

Since the skin covers most surfaces of the body, it is susceptible to damage, which can be fatal depending on the degree of injury to the skin because it defends against external attack and protects internal structures. Various types of artificial skin are being studied for transplantation to repair damaged skin, and recently, the production of replaceable skin using three-dimensional (3D) bioprinting technology has also been investigated. In this study, skin tissue was produced using a 3D bioprinter with human skin cell lines and cells extracted from mouse skin, and the printing conditions were optimized. Gelatin was used as a bioink, and fibrinogen and alginate were used for tissue hardening after printing. Printed skin tissue maintained a survival rate of 90% or more when cultured for 14 days. Culture conditions were established using 8 mM calcium chloride treatment and the skin tissue was exposed to air to optimize epidermal cell differentiation. The skin tissue was cultured for 14 days after differentiation induction by this optimized culture method, and immunofluorescent staining was performed using epidermal cell differentiation markers to investigate whether the epidermal cells had differentiated. After differentiation, loricrin, which is normally found in terminally differentiated epidermal cells, was observed in the cells at the tip of the epidermal layer, and cytokeratin 14 was expressed in the lower cells of the epidermis layer. Collectively, this study may provide optimized conditions for bioprinting and keratinization for three-dimensional skin production.
